1.Effects of different general anesthesia protocols on immune function in patients with oral malignant tumor

Objective To investigate the effects of different general anesthesia protocols on immune function in patients with oral malignant tumor. Methods Sixty ASA Ⅰ or Ⅱ patients undergoing elective radical operation for oral malignant tumor were randomly divided into 3 groups ( n = 20 each): group Ⅰ total intravenous anesthesia (TIVA); group Ⅱ combined intravenous-inhalational anesthesia (IV-INH) and group Ⅱ inhalational anesthesia (INN). Anesthesia was induced and maintained with propofol and remifentanll in group Ⅰ; with sevoflurane,propofol and remifentanil in group Ⅱ and with sevoflurane and remifentanil in group Ⅲ. Peripheral venous blood samples were taken at 30 min before (To) and 1 h (T1), 3 h (T2 ) and 5 h (T3) after induction of anesthesia, the end of operation (T4 ) and at 24 h (T5 ), 48 h (T6 ) and 72 h (T7) after operation for determination of the percentages of T lymphocyte subsets (CD3+ , CD4+ , CD8+ , CD4+/CD8+ ratio). Natural killer (NK) cells (CD16+ ,CD56+ ) and B lymphocyte (CD19+ ) with flow cytometer. Results The percentages of CD3+ , CD4+ , NK cells, B lymphocyte and CD4+/CD8+ ratio were significantly decreased during and after operation at T1-5 in all groups and the percentges of CD3+ ,CD4+ ,NK cells and CD4+/CD3+ ratio were decreased at T6 in groups Ⅱ and Ⅲ as compared with the baseline values before anesthesia at To. The percentage of CD4+ cells and CD4+/CD8+ratio were significantly lower during anesthesia at T2,3,6 in group Ⅱ than in group Ⅰ . The percentages of CD4 +and NK cells and CD4+/CD8+ ratio were significantly higher after operation at T4,5 in group Ⅱ than in group Ⅲ.The percentages of CD3 + , CD4 + , NK cells and CD4 +/CD8 + ratio were significantly lower at T2-6 in group Ⅲthan in group Ⅰ . Conclusion TIVA with midazolam, propofol and remifentanil has less impact on immune function than inhalational and combined intravenous-inhalational anesthesia in patients with oral malignant tumor under-going elective radical operation.
2.Clinical analysis of percutaneous transforaminal endoscopic surgery for elderly patients with lumbar spinal stenosis

Objective To determine the effect of percutaneous transforaminal endoscopic surgery for elderly patients with lumbar spinal stenosis .Methods 80 senile patients with lumbar spinal stenosis were collected .According to the treatment methods ,they were divided into observation group and control group .40 patients in the observation group underwent percutaneous transforaminal endoscopic treatment ,40 patients in the control group underwent conventional surgical treatment .The operation time , blood loss , hospitalization time , X -ray exposure time , postoperative visual analogue scale( VAS) score and disability index score were compared between the two groups .Results Compared with the control group,the operative time[(61.7 ±12.4)min vs.(89.3 ±31.2)min],bleeding volume[(22.3 ± 7.2) mL vs.(192.7 ±49.1) mL] and hospitalization time [(5.3 ±1.7) d vs.(11.2 ±3.7) d] of the observation group were significantly less,while the X -ray irradiation time [(17.1 ±3.9) min vs.(10.1 ±3.1) min] was significantly longer.As to VAS incision scores,,postoperative 12h[(4.1 ±1.5) points vs.(8.3 ±2.0) points], postoperative 24h[(2.3 ±1.2)points vs.(7.7 ±2.1)points],postoperative 48h[(1.3 ±0.6)points vs.(5.2 ± 1.6)points]and postoperative 72h[(0.5 ±1.9)points vs.(3.1 ±1.1)points]in the observation group were significantly lower than those in the control group .Compared with the control group ,the ODI scores at 6 months after operation[(11.2 ±3.6)points vs.(17.2 ±4.4)points],12 months after operation[(6.1 ±1.3)points vs.(10.3 ± 2.5)points]and 24 months after operation[(2.9 ±0.8)points vs.(6.4 ±2.1)points]in the observation group were significantly lower,the differences were statistically significant (all P <0.05).Conclusion Minimally invasive treatment of lumbar spinal stenosis with minimally invasive surgery has minimal trauma , less bleeding and shorter hospitalization time.It can significantly improve the symptoms of the incision pain and the functional activities of the limbs,and improve the quality of life of the patients ,and it is worthy of further popularization and application .
3.Determination of binding ability of PaP3’s terminase large subunit to cos site

Objective To investigate the binding ability of the terminase large subunit of Pseudomonas aeruginosa bacteriophage PaP3 to the cos site. Methods The gene tls was amplified from the genome of bacteriophage PaP3 by PCR and subcloned into pMD18-T vector. Then the gene tls cut down from the vector was inserted into the plasmid pQE31 which could give a 6-His tag at the N-terminal of the expressed protein. The recombinant vector pQE-tls was transformed to E.coli. JM109, after induction with IPTG, the expressed bacteria were resuspended and sonicated, then after centrifugation, the inclusion body was obtained. The inclusion body was dissolved with lysis buffer, then the tagged protein was purified by Ni-NTA affinity chromatography and renatured by dialysis. Finally the DNA-binding ability of the fusion protein rTLS was determined by EMSA. Results The expression plasmid pQE31-tls was successfully constructed, and the target protein yield was up to 30% of the total bacterial proteins. After purification and renaturation, the fusion protein rTLS can partially bind the cos fragment. Conclusion The fusion protein rTLS was successfully expressed, purified and renatured. The rTLS has the specific DNA-binding activity. The present work lays the foundation for the further research of the gene tls.
4.Clinical Study on Stage Treatment withTiao He Ying Wei Needling for Insomnia

Objective To observe the clinical efficacy of stage treatment withTiao He Ying Wei(regulating Ying-nutritional and Wei-defensive qi) needling in treating insomnia.Method A hundred insomnia patients presenting difficulty falling asleep were randomized into group A1 and B1, 50 cases each; 100 insomnia patients presenting difficulty maintaining sleep were randomized into group A2 and B2, 50 cases in each group; 100 insomnia patients presenting early-morning awakening were randomized into group A3 and B3, 50 cases each. Group A1, A2 and A3 were treated withTiao He Ying Wei needling, while group B1, B2 and B3 were treated with conventional medication. The Symptoms score and cerebral blood flow indicators were observed before and after the intervention.Result After the treatment, the symptoms scores were significantly changed in each group (P<0.05). The improvement of symptoms score in group A1 was superior to that in group B1 (P<0.05); the improvement of symptoms score in group A2 was superior to that in group B2 (P<0.05); the improvement of symptoms score in group A3 was superior to that in group B3 (P<0.05). The cerebral blood flow indicators (middle cerebral artery, posterior cerebral artery, anterior cerebral artery, and basilar artery) were significantly changed after the treatment in group A1, A2 and A3 (P<0.05). After the treatment, there were significant differences in comparing the cerebral blood flow indicators between group A1 and B1, A2 and B2, and A3 and B3 (P<0.05).Conclusion Stage treatment withTiao He Ying Wei needling can improve the sleep quality of insomnia patients.
5.A survey on the learning interest of college students majoring in Bioinformatics

Objective:To understand the learning interest of college students majoring in Bioinformatics in Shenyang Medical College and its influencing factors,so as to provide a strong basis for future teaching reform and curriculum setting.Method:After the actual teaching,a self-made questionnaire was used to conduct a quantitative survey on the first-year students majoring in Bioinformatics of Shenyang Medical College to understand their learning interest and analyze its influencing factors.Results:The questionnaire survey found that students majoring in Bioinformatics had different interest points for all 9 courses,different teaching links and different teaching methods in the second semester of freshman.Students had a general high interest in Introduction to Bioinformatics and Introduction to Biometrics.In the Introduction to Bioinformatics course,students were more interested in the history of bioinformatics,but less interested in algorithm-related knowledge points.In the Introduction to Biometrics course,students were more interested in the application cases of biometric recognition,but less interested in laws and regulations of biometrics.The interest of students in learning mainly lied in whether it is conducive to the postgraduate entrance examination.Conclusion:Through strengthen the education of bioinformatics related courses,enhance students'understanding of bioinformatics knowledge and improve their interest in Bioinformatics by analyzing the results of students'questionnaire.

Industry-university-research cooperation is not only the core of technological innovation,but also an important way to enhance industrial competitiveness and achieve high-quality development.Industry-university-research cooperation in Liaoning Province has achieved significant results in promoting technological innovation and economic development,but there are still some problems and challenges.The main problems include insufficient depth of industry-university-research cooperation,scattered innovation resources,lack of long-term stable cooperation mechanisms,as well as talent loss and lack of high-quality innovative talents.Through systematically sorting out the existing models of industry-university-research cooperation,it proposes a series of targeted and operable countermeasures and suggestions.These measures and suggestions provide solid theoretical support for the healthy development of industry-university-research cooperation in Liaoning Province.
7.Expression and clinical significance of serum glycogen synthase kinase-3β and mothers against decapentaplegic homolog 4 in patients with acute coronary syndrome

Objective:To investigate the relationship between the expression levels of serum glycogen synthase kinase-3β (GSK-3β) and mothers against decapentaplegic homolog 4 (SMAD4) in patients with acute coronary syndrome (ACS) and the severity and prognosis of coronary artery disease.Methods:A total of 192 ACS patients admitted to Shandong First Medical University Affiliated Qingdao Hospital from June 2020 to May 2022 were selected as the ACS group, while 192 non ACS patients admitted to the same hospital were selected as the non ACS group. Enzyme-linked immunosorbent assay method was applied to determine the expression levels of serum GSK-3β and SMAD4 in two groups. The Gensini score was applied to evaluate the degree of coronary artery disease in patients, Spearman method was applied to analyze the correlation between serum GSK-3β, SMAD4 expression levels and Gensini score in ACS patients. Receiver operating characteristic (ROC) curve was applied to analyze the predictive efficacy of serum GSK-3β and SMAD4 levels on the prognosis of ACS patients.Results:The serum levels of GSK-3β and SMAD4 in the ACS group were significantly higher than those in the non ACS group: (2.70 ± 0.40) μg/L vs. (2.24 ± 0.41) μg/L, (12.19 ± 2.10) μg/L vs. (9.79 ± 2.82) μg/L, and there were statistical differences ( P<0.05). The serum levels of GSK-3β and SMAD4 in ACS patients with mild, moderate and severe coronary artery disease increased sequentially ( P<0.05). Spearman analysis showed that serum GSK-3β and SMAD4 levels in ACS patients were positively correlated with Gensini total score ( r = 0.569 and 0.587, P<0.01). In ACS patients, 48 cases had poor prognosis, and the incidence of poor prognosis was 25.00% (48/192); 144 cases had a good prognosis. The serum levels of GSK-3β and SMAD4 in ACS patients with poor prognosis were significantly higher than those in ACS patients with good prognosis: (3.15 ± 0.53) μg/L vs. (2.55 ± 0.36) μg/L, (14.03 ± 2.08) μg/L vs. (11.58 ± 2.11) μg/L, and there were statistical differences ( P<0.05). The ROC curve result indicated that the area under the curve (AUC) for predicting poor prognosis in ACS patients with serum GSK-3β and SMAD4 levels alone and in combination was 0.799, 0.784 and 0.858, respectively, the AUC predicted by the combination of the two was obviously higher than the AUC predicted separately ( Z = 2.04 and 2.75, P = 0.041 and 0.006). Conclusions:The expression levels of GSK-3β and SMAD4 in the serum of ACS patients are abnormally elevated. They are positively correlated with the degree of coronary artery disease in ACS patients, and both have good predictive power for adverse prognosis in ACS patients, while the combined use of the two has better predictive performance.
