Objective To observe the effect of dexamethasone(Dex)on the proliferation and os- teogenic differentiation of human marrow stromal cells(MSCs)in vitro. Methods The primary human MSCs were isolated and cultured by Ficoll seperation culture in vitro. In subcultures, human MSCs were respectively treated with dexamethasone 10~(-9),10~(-8) and 10~(-7) mol/L.The proliferation of human MSCs was measured using MTF method;cytoplasmic alkaline phosphatase(ALP)activity was measured;the osteogenic marker osteopontin (OPN)mRNA were examined by reverse transcriptase polymerase chain reaction(RT-PCR).Results The op- tical density values in cultures treated with dexamethasone 10~(-8) and 10~(-7) mol/L for 8 days were significantly lower than those in the controls(P<0.05).Treatment of cells with Dex for 12 days led to a significant increase in cytoplasmic ALP activity(P<0.05)in a dose-dependent manner.Dex induced OPN mRNA.Conclusion Dex inhibits the proliferation of human MSCs and dexamethasone 10~(-7) mol/L leads to a strong decrease in cell number.Dex induces human MSCs differentiate to osteoblastic cells.

Objective To investigate the changes of cortical thickness and surface area in patients with bipolar depression(BD),and to explore the relationship between abnormal changes in gray matter and clinical symptoms. Methods 28 BD patients and 28 healthy controls underwent T1-weighted MRI.The Freesurfer software was used to process the T1 images, which used a set of automated sequences to analyze cortical thickness and surface area on 66 regions (33 regions of each hemisphere),and the correlation with clinical features was also calculated. Results Compared with controls,BD patients showed thinner cortical thickness in left medial orbitofrontal cortex((2.40±0.12) mm vs (2.55 ±0.18) mm,P=1.2× 10-3) and left rostral anterior cingulate((2.66±0.21) mm vs (2.88±0.27) mm,P=3.1 × 10-4),and smaller area of left cuneus((1 443.13± 131.00) mm2vs (6 634.70±600.16) mm2,P=2.7× 10-4) and right superior frontal gyrus ((6 634.70±600.16) mm2vs (7 300.50±653.39) mm2,P=1.3× 10-3).In addition, the negatively correlation was found between the cortical area of left cuneus and effective illness duration (r=-0.471,P=0.018),and the cortical thickness in left rostral anterior cingulate and total score of HAMD-17(17-item Hamilton Rating Scale for Depression) (r=-0.508,P=0.009).Conclusion There are abnormal altertion of cortical thickness and cortical areas of emotional circuit in bipolar depression, but the brain areas are not completely overlapping.Correlation analysis suggests that cortical thickness and area is related to different clinical features.

BACKGROUNDTo determine the inhibitory effect of antisense peptide nucleic acids (PNA) of telomerase on the growth of lung cancer cell lines.
METHODSThe synthesized modified antisense PNAs of telomerase were transfected into the lung cancer cell lines A549 and NCI-H446 respectively by lipofectamine transfection. Telomerase activity was detected by RT-PCR-ELLISA, and the cell counts were determined by MTT.
RESULTSSeventy two hours after transfection with antisense PNAs of telomerase, telomerase activity (A450 value) of A549 and NCI-H446 were down regulated from 0.582 ±0.039, 0.571±0.043 to 0.294±0.048 ( P < 0.01), 0.276±0.051 ( P < 0.01) respectively, and alive cell counts (A580 value) of them from 0.485± 0.009 , 0.513±0.015 to 0.191±0.027 ( P < 0.01), 0.138±0.046 ( P < 0.01) respectively. The growth of two lung cancer cell lines were significantly inhibited.
CONCLUSIONSAntisense PNA of telomerase might inhibit not only the telomerase activity, but also the growth of lung cancer cell lines in vitro.

BACKGROUNDTo evaluate the clinical significance of telomerase activity in lung cancer and to investigate the possibility of telomerase as cancer marker for lung cancer.
METHODSThe activity of telomerase was investigated by TRAP-PCR-ELISA in lung cancer tissues and corresponding adjacent noncancerous tissues obtained from resected specimens of 48 patients with lung cancer and 42 specimens of benign pulmonary lesions were examined simultaneously.
RESULTSTelomerase activity was detected in 42 (87.5%) of the 48 tumors and only 4 (8.3%) of the 48 adjacent noncancerous lung tissue samples (Chi-Square=13.029, P < 0.01), but in none of 42 specimens of benign pulmonary lesions (Chi-Square=14.016, P < 0.01). Correlation with pathological parameters showed that the expression of telomerase activity was associated with lymph node metastasis (93.5% vs 76.4% , Chi-Square=63.511, P < 0.01), but not with histological type, location and differenciated grade of tumor.
CONCLUSIONSTelomerase activation correlates with the carcinogenesis and aggressiveness of lung cancer. Telomerase might be one of the important diagnostic and prognostic factors in patients with lung cancer.
9.A study of risk factors and predictive factors in intraoperative floppy iris syndrome during phacoemulsification
International Eye Science 2017;17(11):2126-2129
AIM:To determine the incidence of intraoperative floppy iris syndrome (IFIS) in patients undergoing phacoemulsification in a Chinese hospital, and to assess new risk factors and predictive factors for IFIS.METHODS:A prospective, observational case series. In the consecutive cataract surgeries performed in one year, the medicine administration, pupil size (PS)before and after mydriasis, and signs of IFIS were recorded.RESULTS:Totally 807 patients (1068 eyes) underwent cataract surgeries.Among the 1068 eyes, signs of IFIS were noted in 34 eyes.Strong positive correlations were showed between finasteride (6.4%,OR =5.885),tamsulosin (25%,OR=21.578),reserpine (16.7%,OR=12.947),clozapine (66.7%,OR=139.467),post-panretinal photocoagulation (14.3%,OR =10.789) and IFIS.Pupil size was inversely related to IFIS incidence (P<0.001).A dilated pupil smaller than 7.25mm could predict iFiS (P<0.001).CONCLUSION:The incidence rate of IFIS was 3.18%.Reserpine,clozapine and post-panretinal photocoagulation emerged as new risk factors for IFIS.A small dilated pupil may imply IFIS occurrence.

Objective To investigate the variations of mtDNA from high and low metastatic mouse hepatocarcinoma cell sublines Hca-F and Hca-P, and the relationship between mutations of mtDNA and carcinogenesis. Methods The variations of D-loop, ND3 and tRNA Met+Glu+Ile gene fragments of mtDNA from Hca-F and Hca-P cells were analyzed by PCR-RFLP and sequencing techniques. Results No amplification fragment length polymorphism and restriction fragment length polymorphism were observed in tRNA Met+Glu+Ile , ND3 and D-loop of mtDNA from the 2 cell sublines. Sequence difference between these 2 cell sublines were found in mtDNA D-loop region by sequencing. Conclusions Genetic alteration of mtDNA non-coding region in tumors, which may reflect the environmental and genetic influences operative during tumor progression, can be linked to their tumorigenic phenotype.
