Journal of Geriatric Cardiology 2007;4(4):229-232
Objective The goal of this study was to examine the association between urotensin Ⅱ (U Ⅱ) concentration and the severity of coronary artery disease (CAD). Methods We studied U Ⅱ concentrations in 100 patients with known or suspected CAD referred for cardiac catheterization. Based on coronary angiograms, subjects were classified as having no or mild CAD (stenosis <50%) and significant CAD (stenosis=50%). Micheal score system was used to estimate the severity of CAD. Result U Ⅱ concentration in the significant CAD group had no difference compared with the no or mild CAD group (1.95±1.18 pmol/L vs 2.04±1.47 pmol/L, P>0.05),but higher in the severe group (score =9) than in the normal or nearly normal group (score<3)( 2.50±1.62 pmol/L vs 1.61±1.05 pmol/L,P=0.03). U Ⅱ concentration had no relationship with other known risk factors, but it correlated with CAD severity (r=0.213, P=0.034).In multiple regression analysis, U Ⅱ is one of the determinants of the severity of CAD, other than age, abnormal glucose, hypertension and gender. Conclusios U Ⅱ is elevated in severe CAD and there is a significant relationship between U Ⅱ concentration and CAD severity.

Objective To investigate the influence of infasound therapy on Raji cells. Methods The Raji cell line was cultivated routinely and grouped as an infrasound and a control groups.Infrasound 8TM was used as a therapeutic infrasound generator which worked in 3 modes(frequency range 4~20 Hz,infrasound energy less than 90 dB).The applicator of the infasound generator was put on 1.5~2.0 cm fom the surface of liquid in the dish containing Raji ceil.Raji cells would be treated for 15,30,60,and 90 minutes, then tested with trypan blue assay,MTT assay, flow cytometry anatysis, and scanning electron microscope(SEM)after 0,24,and 48 h cultivations, respectively. Results Trypan blue assay showed that there were no significant differences between the 2 groups(P>0.05).MTT assay showed that although optical density value of the infrasound groups seemed to be lower than that of the control group.the differences were not significant(P>0.05).Flow cytometry analysis showed that the rate of necrotic cells and apoptosis cells in all groups was less than 10%;and that the differences between all groups were not significant(P>0.05).The scanning electron microscopy showed that the cells treated by infasound exposure for 120 minutes and then cuhivated for 24 h showed that the prominent or micro-floss of the membrane become shorten and decreased. The surface of the membrane became smooth. Conclusion Infrasound(less than 90 dB)treatment in the experiment had no obvious influence on multiplication and apoptosis of Raji cells. But the membrane of Raji cell Would be affected directly by the infrasound, and the penetration of the membrane could be changed.

Objective: To study the expression of Fas, Fas ligand (FasL) and IFN ? in gastric cancer and its possible significance. Methods: Fifty eight gastric paraffin wax embedded cancer tissues and fifty three normal tissues adjacent gastric cancer were tested for the expression of Fas and FasL protein by immunohistochemistry and IFN ? mRNA by in situ hubridisation respectively. Results: The positive rate of Fas in cancer cells of gastric cancer tissues was significantly lower than that in gastric epithelial cells of the tissues adjacent cancer(19.0% and 64.2%, respectively; ? 2=23.46, P = 0.00). The positive rate of FasL in cancer cells of gastric cancer tissues was significantly higher than that in gastric epithelial cells of the tissues adjacent cancer(63.8% and 45.3%,respectively; ? 2=3.83, P =0.05). The positive rate of IFN ? in cancer cells of gastric cancer tissues was significantly lower than that in gastric epithelial cells of the tissues adjacent cancer(0.0% and 49.1%,respectively; ? 2=37.16, P =0.00). Conclusion: The Fas-FasL system was unbalanced, and the expression of IFN ? was low in gastric cancer cells in this study. These may be related to the carcinogenesis of gastric epithelial cells and might be responsible for the immune escape of these cells.

Objective To develop and validate a simple and reliable HPLC method for the analysis of L-carnitine in human seminal plasma and to investigate its clinical significance as a potentially useful index of infertile. Methods After proteins in seminal plasma are precipitated with acetonitrile,L-carnitine in seminal plasma was derivatized to form its ester.HPLC separation of the sample solution was performed on a Lichrospher SiO_2 column and detected by ultraviolet absorbance at 260 nm.A mobile phase composed of acetonitrile-citrie acid buffer(containing 12 mmol/L triethanolamine,pH 5.0)was found to be the most suitable for this separation at a flow rate of 1.2 ml/min and enabled the baseline separation of the L-carnitine from interferences with isocratic elution. The L-carnitine levels in seminal plasma were studied in both 87 patients with infertility and 30 control subjects. Results Under the chromatographic conditions described, the L-carnitine derivative had a retention time of approximately 13 min.Good separation and detectability of L-carnitine in human seminal plasma sample were obtained. The method proved to be linear in the range of L-carnitine from 0 ?mol/L to 1000 ?mol/L.The relative standard deviations of within-and between-assay for L-carnitine analysis were 1.23% and 1.36%,respectively.The recoveries were 91.6% -96.5% for the human seminal plasma samples.L-carnitine concentrations in the populations were(392.7?107.2)?mol/L in the fertile group(n=30),(270.0?83.9)?mol/L in asthenozoospermia group(n=29),(187.9? 43.9)?mol/L in oligozoospermia group(n=19)and(175.7?67.1)?mol/L in oligoasthenozoospermia group(n=39).The large difference(P0.05).Conclusion The determination of L-carnitine level in seminal plasma may prove useful as a potentially biochemical marker of fertility and this is a useful guidance for the clinic therapy and the mechanismic study on the male reproduction.
