Objective To predict and screen the B-cell epitopes on nucleoprotein(NP)of human avian H5N1 virus strain. Methods As NP nucleotide sequences of strain A/GD/01/06(H5N1)were sequenced,B-cell epitopes were predicted by the analysis of the flexible regions of secondary structure for NPprotein and by screening on B-cell epitopes for NP protein with methods of Hydrophilicity Plot by KyteDoolittle,Surface Probability Plot by Emini, and Antigenic Index by Jameson-Wolf.Then a screening method was established by Hierarchical cluster,Bivariate correlate and Quartiles in SPSS13.0.and the variation of amino acids in NP protein was appraised in epitope prediction.Resuits The computer-predicted most possible epitopes for NP protein were located within or nearby its N-terminal 317-326,452-457,467-473,367-370,491-498,375-378,171-177,48-53,245-250.76-104 and so on.NP protein in A/GD/01/06(H5N1)increased a glycoprotein domain(NES368-370)by the substitution of N370S and changed the bio-features.Conclusion Stepwise prediction of the B-cell epitopes for the NP protein based on multiple parameters is helpful for the molecular-immunology and drug-screening,and the substitution of N370 S in NP of A/GD/01/06(H5N1)varied its antigenicity but didn't change the epitope size(SNEN367-370).

Acta Nutrimenta Sinica 1956;0(01):-
Objective: To study the influence of high cell incubating temperature on AMP-activated protein kinase(AMPK) activity and lipid metabolites of piglets hepatocytes in vitro.Method: Primary hepatocytes of piglets about age 55d were separated and cultured under 37 ℃(control) or 42 ℃(heat stress). The anabolic and catabolic products of [14C]-oleic acid were detected for hepatocytes and culture media at 60min,120min and 180min.There were 9 replicates per time point.Result: Heat stress activated AMPK activity and enhanced fatty acid oxidation. The production of [14C]-CO2 and [14C]-acid soluble metabolites(ASM) was higher in heat stress group than in the control. At the same time, heat stress depressed the incorporation of [14C]-oleate into phospholipids, monoglycerides, triglycerides, cholesterol and cholesteryl ester. Conclusion: Heat stress activated AMPK activity and enhanced the formation of anabolic products and depressed catabolic products in piglets hepatocytes in vitro.

BACKGROUND: Serum creatinine (SCr) is the most commonly used parameter to estimate renal function impairement, but there are some shortcomings. Many factors including age, gender, drug, diet, muscle mass and metabolic rate can influence SCr, leading to an inaccurate estimation of kidney impairment. Studies have shown that cystatin C (CysC) is not affected by factors such as muscle mass, age, gender, diet, inflammation or tumor. The present study was undertaken to compare the sensitivity of CysC and SCr in evaluating renal function impairment at early stage of shock. METHODS: Seventy-one patients aged 38.3±21.4 years, who had been treated at the Emergency Medicine Department of the First Affiliated Hospital, Sun Yat-sen University between February 2006 and June 2007, were studied. They were divided into groups A, B, C, and D according to the shock time. Serum sample was drawn from each patient at 1, 2, 3, 4 hours after shock to determine SCr and CysC. CysC and SCr were determined again at 72 hours and 7 days after shock. RESULTS: CysC increased earlier than SCr in the 71 patients, and CysC decreased slower than SCr when shock was corrected. CysC increased at 1 hour after shock. There was a negative correlationship between CysC, SCr and glomerular filtration rate (GFR), especially at early stage of shock. CONCLUSIONS: There is renal injury at early stage of shock. CysC is more sensitive than SCr in assessing renal function at the early stage of shock.

OBJECTIVETo investigate the prognostic factors for non-small cell lung cancer(NSCLC)in patients under 40 years of age.
METHODSThe clinicopathological data of 148 young patients with NSCLC were retrospectively analyzed. Kaplan-Meier and Cox regression analyses were used to analyze the relationship between prognostic factors and survival time.
RESULTSThe patients were followed-up for 6 - 148 months, and the follow-up rate was 100%. In the whole group, 122 patients died and 26 cases were surviving. The 1-, 3- and 5-year survival rates were 54.7%, 10.4% and 5.6%, respectively. The median survival time (MST) was 14.7 months. Kaplan-Meier analysis showed that Karnofsky performance status (KPS), clinical stage, treatment modality and serum CEA were related with prognosis (P < 0.05). Multivariate analysis indicated that KPS, clinical stage, treatment modality and serum CEA were independent prognostic factors (P < 0.05).
CONCLUSIONSKPS, CEA, clinical stage and treatment modalities are independent prognostic factors in young NSCLC patients.

Objective To explore the role of Beclin 1 and PTEN in gastric caicinoma genesis and the effects on prognosis. Methods The expression of Beclin 1 and PTEN in 199 gastric caicinoma specimens and corresponding adjacent noncancerous tissues were examined by tissue microarrays and immunohistochemistary, and the relation with gastric cancer was analyzed. The rate of Beclin 1 and PTEN expression in 15 fresh gastric carcinoma samples and corresponding adjacent noncancerous tissues were detected by Western blot. All the samples were from Changzheng Hospital. Results The Results of immunohistochemistary showed that the rates of Beclin 1 and PTEN positive expression in cancinoma tissues were 47.2% (94/199) and 55.8% (111/199),both were lower than that of adjacent noncancinoma tissues (94.5%,188/199 and 92.5%,184/199; P < 0.01).The lower expression of Beclin 1 and PTEN in gastric carcinoma were associated with gender, differentiation degree, depth of tumor invasion, lymph node metastases and disease stage(P<0.05).There was a positive correlation between Beclin 1 and PTEN expression in gastric carcinoma tissues (r =0.680,P<0.01). The survival analysis indicated that Beclin 1 and PTEN were independent factors in determining the prognosis of gastric cancinoma patients. The 5-year survival rate of Beclin 1 positive patients was 67.0% (63/94),and of negative patients was 33.3% (35/105).The 5-year survival rate of PTEN positive patients was 71.2% (79/111),and of negative patients was 21.6% (19/88) ( all P<0.001).The Results of Western blot indicated that the expression of Beclin 1 and PTEN in gastric carcinoma tissues were significantly lower than that in the adjacent noncarcinoma tissues ( all P<0.001).Conclusion The abnormal expression of Beclin 1 and PTEN may be related to carcinogenesis and the development of gastric carcinoma.
