2.Research progress of correlation between blood-stasis syndrome and inflammation.

Through summarizing the literatures concerning basic and clinical study on the correlation between blood-stasis syndrome (BSS) and inflammation, reviewing close correlation of BSS with C-reaction protein, serum interleukin-6, tumor necrosis factor and adhesion molecules, it was found that promoting blood circulation and removing stasis approaches could effect vitally in clinical treatment of inflammation, and the inflammation reaction shows certain effect of mediation in animal model of BSS. Accordingly, the important role played by inflammatory reaction in the occurrence and development of BSS is summarized in the paper.

3.Inhibition of osthole for resorption of rats femur tissue in vitro.

OBJECTIVETo investigate osthole effect on femoral tissue resorption activity of rat in vitro.
METHODSSix SD rats weighted (80 ± 5) g were used to isolate and culture femoral tissue (diaphyses and metaphysis) in vitro. The cultured tissue were devided into control group, estradiol group and osthole group. The femoral tissue was treated with final concentration of 1 x 10(-5) mol/L osthole and 1 x 10(-8) mol/L estradiol culture in vitro at 48 hours after cultured. Tartrate-resistant acid phosphatase (StrACP) activity, glucose and Lactic acid content, StrACP, MCSF (Macrophage colony stimulating factor) and CTSK (Cathepsin K) mRNA was detected by Real-Time RT-PCR were detected.
RESULTSConcetration of Alkaline phosphatase activity were 2226 and 2498 in 1 x 10(-5) mol/L osthole and 1 x 10(-8) mol/L estradiol respectively. As compared with control group, the activity of StrACP of 1 x 10(-5) mol/L osthole and 1 x 10(-8) mol/L estradiol were inhibited at 6, 9, 12 days (P < 0.05); under treatment of in l x 10(-5) mol/L osthole, the content of Lactic acid were increased and the content of glucose were decreased at 3, 6, 9 days (P < 0.05); StrACP, MCSF and CTSK mRNA expression level were inhibited at 6, 9 days (P < 0.05).
CONCLUSIONOsthole can inhibit bone resorption and raise the level of nutrition metabolism of femurs tissue.

4.Appraisal of the prognosis in patients with acute myocardial infarction treated with primary percutaneous coronary intervention.

Acute myocardial infarction (AMI) is still the leading factor causing crippling and death in cardiovascular disease. Percutaneous coronary intervention (PCI) can significantly reduce inpatient mortality and incidence of complication. But owing to the existence of restenosis, in-stent thrombosis, etc., recurrent post-PCI cardiovascular events and high repeatability of hospitalization, as well as its crippling rate and mortality, remain a serious threat to the society and the patients' family. Therefore, the appraisal and intervention in post-PCI associated risk factors has presently become one of the foci in clinical research. To improve the near- and long-term prognosis and quality of life in post-PCI AMI patients, further improvement of the evaluation system in risk factors and prognosis is necessary in order to provide a theoretical basis for early application of intervention in high-risk patients in clinical practice. This thesis mainly dissertates some explicit and valuable factors for clinical prognosis evaluation in recent studies, involving C-reactive protein (CRP), tumor necrosis factor-alpha (TNF-alpha), brain natriuretic peptide (BNP), atrial natriuretic peptide (ANP), Chinese medicine (TCM) syndrome, their correlation with clinical state and course of AMI, and their importance in clinical prognosis.

5.Differential gene expression profiles in coronary heart disease patients of blood stasis syndrome in traditional Chinese medicine and clinical role of target gene.

OBJECTIVETo investigate the differential gene expression profiles in coronary heart disease (CHD) patients of blood-stasis syndrome (BSS) by oligonucleotide microarray technique, and the clinical significance of target gene.
METHODSSubjects were assigned to CHD patients with BSS (n=8), CHD patients without BSS (n=8), and BSS patients without CHD (n=8) based on coronary angiography and the diagnostic criteria of BSS. The sex- and age-matched healthy volunteers (n=8) were enrolled as the control group. Venous blood samples were collected for RNA extraction; Test-3 chip was employed to examine the quality of samples. Then, the samples were hybridized with Affymetrix U133 Plus 2.0 array to compare the gene expression profiles among the four groups. Gene-array scanner and gene chip operating software were applied to screen out hybridization signals and analyze gene expression, respectively. Based on the comparison of the samples of the four groups, the differential genes related with CHD and BSS were analyzed with Gene Ontology (GO) and pathway, and target genes selected were confirmed by real-time reverse transcription polymerase chain reaction (RT-PCR). Thirty CHD patients with BSS were selected according to the former criteria and 40 healthy as controls. The serum concentration of interleukin-8 (IL-8) was determined by double-antibody sandwich avidin-biotin peroxidase complex enzyme-linked (ABC-ELISA).
RESULTSA total of 107 differential genes were found being associated with CHD, including 48 up-regulated genes and 59 down-regulated genes. Among these 107 differential genes, 14 genes (13.1%) were found related to inflammatory reaction and immune response through GO analysis. In the pathway analysis, 4 of 15 conspicuous pathways were referred to the inflammation and immune response. Among 48 differential genes related to BSS, 26 genes were up-regulated, and 22 were down-regulated. Five of the 48 genes (10.4%) and 5 of 10 significant pathways were involved in inflammation and immunity. The results of real-time RT-PCR proved the accuracy of the gene chip. The patients have markedly higher level of serum IL-8 compared to the controls (P<0.05).
CONCLUSIONThe correlation of inflammatory- and immune-related genes with CHD patients of BSS was revealed at the level of nucleic acid, and the target gene IL-8 may play a role in the pathobiology of CHD with BSS.

6.Comparison of effects of kaempferide and anhydroicaritin on biomineralization of cultured osteoblasts.

This study is to compare the effects of kaempferide and anhydroicaritin on biomineralization of rat osteoblasts (ROB) in vitro. Calvarias were dissected aseptically from newborn SD rats, the osteoblasts were obtained by enzyme digestion and were cultured in MEM containing 10% FBS. The medium was changed every three days, and serial subculture was performed when cells covered with 90% of the dish. Kaempferide and anhydroicaritin were separately added with final concentrations of 1 x 10(-4), 1 x 10(-5), 1 x 10(-6) and 1 x 10(-7) mol x L(-1) under the conditions of osteogenic differentiation. The proliferation was measured by MTT, and the optimal concentration was detected by the ALP activity at the 9th day after osteogenic induction culture. The osteogenic indexes of kaempferide, anhydroicaritin and control group with the optimal concentration were compared. The result showed that the anhydroicaritin at concentration of 1 x 10(-5) mol x L(-1) had significantly promoted the activity of ALP, calcium content and osteocalcin content, increased the number of CFU-F(ALP) and mineralized nodules, enhanced the mRNA level of BMP-2, OSX and Runx-2, which are key genes of osteogenic differentiation, and raised the protein content of collagen-I. However, the kaempferide group had not significantly represented the ability that promoted osteogenic differentiation of ROB. The difference of osteogenic differentiation on ROB between kaempferide and anhydroicaritin was caused by the prenyl group on C-8 of icariin.

7.Effects of rat serum containing total flavonoid extract of Epimedium sagittatum on development of osteoblasts.

OBJECTIVETo investigate the effects of total flavonoid extract of Epimedium sagittatum (TFE) on the proliferation and differentiation of newborn rat calvarial osteoblasts (ROB).
METHODTFE was supplemented into the culture medium of ROB at 0. 1, 1, 10 and 100 microg x mL(-1) respectively. The serum of rats administered TFES (SRAT) was also added into the medium in a parallel treatment at 2.5%, 5% and 10% respectively. Their effects on cell proliferation and differentiation was studied by MTT and the analysis of osteogenic differentiation marks.
RESULTTFE had no appreciable and on cell proliferation and differentiation at any concentration. However, 2.5% and 5% SRAT stimulated cell proliferation strongly and, 5% SRAT significantly promoted the maturation and function of osteoblast by improving the alkaline phosphatase activity, osteocalcin secretion, calcium deposition and the number of mineralized nodular structures.
CONCLUSIONThe metabolites of TFE should be the anti-osteoporosis constitutes of Epimedium sagittatum.

8.Short-term clinical outcomes of neonates of secundiparous mothers: a single-center cohort study.

OBJECTIVETo investigate the short-term clinical outcomes of neonates of secundiparous mothers, and to provide a reference for the clinical practice after the change in birth policy.
METHODSA cohort study was performed for the parturients and their neonates born in Yongkang Maternal and Child Health Care Hospital in Zhejiang, China between June 2015 and April 2016. According to the parity of the mother, the neonates were divided into primiparous group and secundiparous group. The short-term clinical outcomes of neonates were compared between the two groups.
RESULTSA total of 4 091 neonates who met the inclusion criteria were enrolled, and there were 2 023 neonates in the primiparous group and 2 068 in the secundiparous group. In the secundiparous group, most mothers (57.16%) were aged 26-34 years, and 16.49% were aged above 35 years. Compared with the primiparous group, the secundiparous group had a significantly higher rate of births by cesarean section (42.55% vs 25.06%; P<0.05). The percentages of neonatal respiratory distress syndrome (NRDS; 8.6% vs 3.4%) and transient tachypnea of the newborn (TTN; 7.2% vs 2.6%; P<0.05) in hospitalized neonates from the secundiparous group were significantly higher than in those from the primiparous group.
CONCLUSIONSElderly parturient women are not the major population of secundiparous parturients. The neonates of secundiparous mothers have a higher rate of births by cesarean section, which might be associated with increased percentages of NRDS and TTN in hospitalized neonates from the secundiparous mothers.

9.Effect of time and temperature on count of peripheral blood cells in newborns

Objective To study the effect of time and temperature on the count of peripheral blood cells in newborns.Methods The peripheral blood of 100 cases of newborns who were admitted in the First Affiliated Hospital of Hebei North University from January 2016 to June 2016 were collected,and measured with Sysmex XS-500i automatic blood cell analyzer immediately,then were split into two parts and stored at 4 ℃ and room temperature respectively.The measurement was repeated in 24 and 48 h,and the groups were compared on WBC,RBC,PLT,HGB,IG% and IG#.Results There were statistically significant differences between the instantly measuring result and those in 24 and 48 h under room temperature (P<0.05).In case of 4 ℃,the instantly measuring result had no obvious difference with that in 24 h (P>0.05),while statistical difference with that in 48 h (P<0.05).Conclusion Newborns' peripheral blood can be stored at 4 ℃C,and the counting results will not be affected for WBC,RBC,PLT,HGB,IG% and IG# within 24 h.
10.Effects of Chinese herbs on multiple ion channels in isolated ventricular myocytes.

BACKGROUNDShensong Yangxin (SSYX) is one of the compound recipe of Chinese materia medica. This study was conducted to investigate the effects of SSYX on sodium current (I(Na)), L-type calcium current (I(Ca, L)), transient outward potassium current (I(to)), delayed rectifier current (I(K)), and inward rectifier potassium currents (I(K1)) in isolated ventricular myocytes.
METHODSWhole cell patch-clamp technique was used to study ion channel currents in enzymatically isolated guinea pig or rat ventricular myocytes.
RESULTSSSYX decreased peak I(Na) by (44.84 +/- 7.65)% from 27.21 +/- 5.35 to 14.88 +/- 2.75 pA/pF (n = 5, P < 0.05). The medicine significantly inhibited the I(Ca, L). At concentrations of 0.25, 0.50, and 1.00 g/100 ml, the peak I(Ca, L) was reduced by (19.22 +/- 1.10)%, (44.82 +/- 6.50)% and (50.69 +/- 5.64)%, respectively (n = 5, all P < 0.05). SSYX lifted the I - V curve of both I(Na) and I(Ca, L) without changing the threshold, peak and reversal potentials. At the concentration of 0.5%, the drug blocked the transient component of I(to) by 50.60% at membrane voltage of 60 mV and negatively shifted the inactive curve and delayed the recovery from channel inactivation. The tail current density of I(K) was decreased by (30.77 +/- 1.11)% (n = 5, P < 0.05) at membrane voltage of 50 mV after exposure to the medicine and the time-dependent activity of I(K) was also inhibited. Similar to the effect on I(K), the SSYX inhibited I(K1) by 33.10% at the test potential of -100 mV with little effect on reversal potential and the rectification property.
CONCLUSIONSThe experiments revealed that SSYX could block multiple ion channels such as I(Na) I(Ca, L), I(k), I(to) and I(K1), which may change the action potential duration and contribute to some of its antiarrhythmic effects.
