1.Hematologic effects of linezolid in advanced aged patients

Objective To evaluate the hematologic effects of linezolid in advanced aged patients with hospital onset of infection.Methods The clinical characteristics and complete blood routine examination results of 45 elderly patients aged(92.9±5.2)years treated with linezolid over 600 mg of lowest dose everyday for more than 72 hours were retrospectively analyzed.Results Among the 45patients,20 patients(44.4%)had substantially lower platelet,6 patients(13.3%)lower haemoglobin,1 patient(2.2%)lower absolute neutrophil,and the linezolid treatment in 19 patients were discontinued due to hematological system adverse reactions.The significant drop in platelet occurred at(10.2±3.3)days after treatment.The significant drop in hemoglobin occurred at(9.8±3.0)d after treatment.The lowest count of platelet and hemoglobin took place at(13.1±3.9)d and (10.5±3.5)d after treatment,respectively.The drops in platelet and hemoglobin of some patients took place after cease of treatment.Stepwise logistic regression analysis revealed that treatment duration(OR =1.407,P<0.05)and creatinine clearance rate(OR=0.732,P<0.05)were the risk factors for thrombocytopenia.Conclusions The elderly patients using linezolid more likely suffer from adverse reactions of hematological system,particularly thrombocytopeniawhich is significantly more common in patients with renal insufficiency and prolongation of treatment time.The blood routine examination should be monitored closely when elderly patients are treated with linezolid.
2.Influence of therapy compliance on the prognosis of children with thalassaemia major

Objective The purpose of this study was to describe the influence of therapy compliance on prognosis of children with thalassaemia major (TM). Methods Children with TM was recruited by convenient sampling. The therapy compliance was measured by compliance index (CI) and serum ferritin (SF), The correlation between therapy compliance and incidence of complications were studied. Results Low therapy compliance existed in children with TM, 22.12% (CI) and 55.77% (SF). But no statistical difference existed in therapy compliance between boys and girls (P>0.05). Complication incidence in children with low therapy compliance was 85.19% (CI) and 100.00% (SF). The incidence of cardiac complication was 60.49% (CI) and 84.78% (SF), hepatic complication was 46.91% (CI) and 76.09% (SF), infectious complication was 8.64% (CI) and 10.86% (SF), growth retardation complication was 55.56% (CI) and 67.39% (SF), anemia complication was 43.21% (CI) and 21.74% (SF). The incidence rate of complications was lower in children with better therapy compliance than those with worse therapy compliance (P<0.01). Conclusions Children with TM had low therapy compliance and high incidence rate of complications. Low therapy increased the incidence rate of complications.
3.Acupuncture at neck Jiaji acupoints combined with moxibustion for clinical study of cervical vertigo

Objective To observe the effect of acupuncture at neck Jiaji acupoints combined with moxibustion to cervical vertigo.MethodsIn Wuhan Chinese and Western Union Hospital from August 2015 to November 2016, 80 patients with cervical vertigo were randomly divided into two groups, the treatment group was treated by needling cervical Jiaji points, at the same time with moxibustion on neck Jiaji acupoints, the control group was given drug treatment.ResultsThe total effective rate in the treatment group was 95%, but the control group total effectiveness was 72.5%, the effect of the two groups had significant difference(P<0.05).ConclusionAcupuncture at Neck Jiaji points combined with moxibustion treatment had the functions of relieving rigidity of muscles and activating collaterals,dispelling cold and dredging channel blockage, relieveing local muscle spasm,improveing blood circulationand improve the function of the blood supply to the brain to vertigo.
4.Dynamic changes of photorecrptor layer in eyes with acute central serous chorioretinopathy after laser treatment by fourier-domain optical coherence tomography

AIM: To dynamically observe the feeling change of the photorecrptor layer in the eyes with acute central serous chorioretinopathy ( CSCR ) krypton laser treatment by fourier - domain optical coherence tomography ( FD - OCT), and to study their correlation with the chang of vision.
METHODS: This is a retrospective case series study. The clinical diagnosis of 52 patients with monocular initial onset of central serous chorioretinopathy, krypton laser photocoagulation before treatment, after 1, 2, 4, 6, 8wk, 6mo, FD - OCT were performed to observe the morphological changes characteristic of photoreceptor layer and changes in vision.
RESULTS: After 1wk treatment, all cases were improved; 2wk, 6 cases were cured; 4wk, 38 cases were cured; 6wk, 41 cases were cured; 8wk, 45 cases were cured, the OCT showed macular retinal neuroepithelial layer ( RNL ) from fully absorbed; 6mo with the same 8wk. Before and after treatment in patients with best corrected visual acuity and from the height difference between the macular region of RNL was statistically significant (P<0. 05), there was a correlation between the changes of visual acuity after treatment and the macular detachment of RNL height (P<0. 05), Photoreceptor layer of complete and incomplete best corrected visual acuity difference was statistically significant (P<0. 01).
CONCLUSION: FD-OCT can dynamicaly observed acute central serous chorioretinopathy krypton laser treatment of photoreceptor ultrastruture changes. Photoreceptor layer of complete and incomplete best corrected visual acuity difference was statistically significant (P<0. 01).
5.Effect of PM2.5 on sympathetic innervation by stimulating the secretion of cardiac nerve growth factor in healthy rat hearts

ObjectiveTo assess the effect of PM2.5 on sympathetic innervation and the relationship with cardiac nerve growth factor in healthy rat hearts,identify the ability to the sympathetic nerve reconstruction,and explore the possible arrhythogenic mechanism of PM2.5.MethodsForty healthy SD rats were instilled into trachea with two different solutions twice per week for four weeks:control group with saline and experimental group with PM2.5 25 mg/kg.Then these rats were killed,and biatrial appendages,two ventricular anterior walls were left.Immune cytochemical staining of cardiac nerves was performed using anti-tyrosine hydroxylase antibodies and cardiac nerve growth factor was detected by western blotting.ResuitsCompared with the control group,both the density of sympathetic nerves and the expression of cardiac NGF protein in the experimental group were much higher in the left ventricular anterior wall ( P<0.01 and P<0.05) and in the right ventricular anterior wall ( P<0.01 and P<0.05).ConclusionsPM2.5 can induce regional sympathetic hyperinnervation in both ventricular anterior walls in healthy rats by stimulating the NGF protein secretion,and the sympathetic nerve reconstruction effect of PM2.5 was confirmed.
6.Gene expression profiling and functional analysis of cerebral artery after experimental subarachnoid hemorrhage

Objective To explore the difference of gene expression profiling between normal basilar arteries and basilar arteries of cerebral vasospasm (CVS) after subarachnoid hemorrhage (SAH) in rabbits. Methods cDNA chip of normal basilar arteries and basilar arteries of CVS after SAH in rabbits were downloaded from GEO database. The chip was analyzed and screened by Bioconductor software, and function enrichment and pathway analysis of the differentially expressed genes were analyzed by Cytoscape software. Then 6 adult male Japanese rabbits were used, and randomly divided into normal control group (n=3) and SAH model group (n=3). Rabbit SAH models were established by cisterna secondary-blood-injection method. RNA data of normal basilar artery specimens on the 0 day and basilar artery specimens after SAH on the 5-day were used to validate the parts of differentially expressed genes by qRT-PCR. Results A total of 4356 differentially expressed genes were found in normal basilar arteries and basilar arteries of CVS after SAH in rabbits. Among them, 920 genes were considered to be significant with P-value<0.05, such as GRIK1, MYH13, ZNF45, SAA3, RLN1, MSR1 and others. Function enrichment analysis indicated that the differentially expressed genes were involved in regulation of Ca2+transmembrane transporter activity, negative regulation of ion transmembrane transport, regulation of potassium ion transport, positive regulation of JAK-STAT signaling cascades and other biological processes. Pathway analysis showed that calcium signaling pathway, cGMP-PKG signaling pathway, HIF-1 signaling pathway, PI3K-Akt signaling pathway and other signaling pathways maybe related with the differentially expressed genes. qRT-PCR verification showed that the expression of MSR1 in SAH model group was consistent with that of the chip result. Conclusion The gene expressions of basilar arteries of CVS after SAH in rabbits are significantly different, and MSR1 gene can be used as a potential target for studying the pathological mechanism of CVS.
7.Effect of heche chongcao capsule on expression of TNF-α protein in rats with chronic obstructive pulmonary disease.

OBJECTIVETo observe the expression of tumor necrosis factor-alpha (TNF-α) protein in the lung tissue of rats with chronic obstructive pulmonary disease (COPD), and to evaluate the intervention and mechanism of Heche Chongcao Capsule (HCC).
METHODSThe COPD rat model was prepared by exposure to cigarettes smoke plus intratracheal injecting lipopolysaccharide (LPS). Forty successfully modeled SD rats were randomly divided into the COPD model group, the control group, the low dose HCC group, the medium dose HCC group, and the high dose HCC group, 8 in each group. Meanwhile, a normal control group consisting of 6 rats was also set up. HCC at 0.25, 0.5, and 1.0 g/kg was administered to rats in the 3 dose HCC groups respectively by gastrogavage combined with Theophylline Sustained Release Tablet (TSRT). Rats in the control group were administered with TSRT by gastrogavage at 4.5 mg/kg, 1 mL/100 g each time, once daily. All medication lasted for 4 successive weeks. Equal volume of distilled water was administered by gastrogavage to rats in the COPD model group and the normal control group. Morphological changes of the lung tissue were observed under microscope. The expression of TNF-α protein in the lung tissue were also detected using Real-time PCR.
RESULTSUnder electron microscope, the cilium in the tracheal epithelium was disorderly arranged, type I and II alveolar cells were degenerated, endoplasmic reticulum and mitochondria were swollen, the lamellar body was emptied, and free fragment could be seen inside alveolar space. Compared with the model group, all lesions were somewhat ameliorated in all medicated groups, especially in the medium dose HCC group. Compared with the model group, the expression of TNF-α protein decreased in all medicated groups, especially in the medium and low dose HCC groups (P < 0.05, P < 0.01). Compared with the control group, the expression of TNF-α protein decreased in the medium and low dose HCC groups (P < 0.05).
CONCLUSIONHCC could effectively regulate the expression of TNF-α protein and inhibit airway inflammation reaction in COPD rats.

8.Isolation and structural identification of flavonoids from Aurantii Fructus.

Aurantii Fructus is the dried and immature fruit of Citrus aurantium and its cultivars. To investigate the chemical constituents of Aurantii Fructus, the separation and purification of constituents were performed by column chromatography on silica gel LH-20, HW-40, ODS, PHPLC and PTLC. Fourteen flavonoids, including four flavone glycosides and ten polymethoxyflavones (PMFs) were isolated from the EtOAc fraction and Petroleum ether fraction of Aurantii Fructus and their structures were identified by physicochemical properties and spectral data (NMR and MS) as (2R) -and (2S)-6"-O-acetylprunin (1,2), naringenin-7-O-β-D-glucopyranside (3), 5,7,4'-trihydroxy-8,3'-dimethoxyflavone-3-O-6"-(3-hydroxyl-3-methylglutaroyl)-β-D-glucopyranoside(4), 4'-hydroxy-5,6, 7-trimethoxyflavone (5), natsudaidain (6), nobiletin (7), sinensetin (8), 5,6,7,4'-tetramethoxyflavone (9), 5,7,8,4'-tetramethoxyflavone (10), 3,5,6,7,8,3',4'-heptamethoxyflavone (11), tangeretin (12), 5-demethyl nobiletin (13), and 5-hydroxy-6,7,3', 4'-tetramethoxyflavone (14). Compound 3-5 s were isolated from this plant for the first time and compound 1 was a new one.

Objective To explore effect of endogeneous gangliosides(Gls) and integrin ?2?1 on protein phosphotyrosine expression of pp125 focal adhesion kinase (pp125FAK) after adhesion of SK-N-SH neuroblastoma cells to collagen(Col).Methods SK-N-SH cell line with high expression of integrin ?2?1 was cultured in presence of D-threo-1-phenyl-2-decanolamino-3-morphinoline-1-propanol(D-PDMP).Effect of endogeneous Gls,anti-?2 and anti-?1 monoclonal antibody on protein phosphotyrosine expression of pp125FAK during adhesion of SK-N-SH cells to Col were determined by immunoprecipitate and Western blotting.Results After 6 days,endogenous Gls in cells were almost depleted.Gls-depletion,anti?2 and anti-?1 monoclonal antibody were able to decrease pp125FAK expression of SK-N-SH cells adherent to Col respectively.GD2,the major component of neuroblastoma cell Gls could reco-ver pp125FAK expression to a certain degree.Conclusions Endogenous tumor Gls regulate protein phosphotyrosine expression of pp125FAK during adhesion of neuroblastoma cells to Col.It is suggested that tumor Gls may increase signal transduction of tumor cell integrin ?2?1 by increasing tyrosine phosphorylation of pp125FAK.
