1.Analysis on the effect of herbal reyanbao combined with rehabilitation training on the rehabilitation of patients with lumbar disc herniation

Objective To study the effect of herbal reyanbao combined with rehabilitation training on the rehabilitation of patients with lumbar disc herniation.Methods 120 cases with lumbar disc prolapse as the research object from January 2015 to December 2016 in Wuhan integrated traditional Chinese and western medicine hospital were divided into two groups(the control group and the observation group).The control group were treated with routine conservative+conventional nursing plan, and the observe group were given herbal reyanbao combined with rehabilitation training.Clinical data were compared in the two groups.Results The clinical effect and rehabilitation degree in the observation group were better than those in the control group, the difference was statistically significant(P< 0.05).Conclusion It can help patients with lumbar disc herniation recover as soon as possible which herbal reyanbao combined with rehabilitation training were used on the rehabilitation of patients, and it is worthy of application.
2.Transport of limonin in rat intestine in situ and Caco-2 cells in vitro.

Limonin existed in citrus fruits has been shown to have anti-bacterial, anti-viral, anti-feedant, anti-nociceptive, anti-inflammatory activities and anti-carcinogenic activities. But the clinical use is limited by its low bioavailability. The aim of this study is to observe the absorption and secretion transport mechanisms of limonin in intestine which can pave the way for the further study and clinical use. The transport characteristics and mechanisms of limonin in rat were studied by in situ intestine perfusion and in vitro Caco-2 cells method. The intestinal absorption of limonin was probably via a facilitated diffusion pathway which was poor and without segment-selection. Verapamil and ketoconazole improved the absorption remarkably according to the result of in vitro Caco-2 cells study; however, probenecid had no significant effect on the absorption. The P-gp efflux and CYP3A4 metabolism were involved in the poor intestinal absorption and low bioavailability of limonin. The exploration of the intestinal absorption mechanism is crucial to the design of dosage form and clinical use of limonin.
3.Impact of hypothyroidism on sperm motility of male rats

Objective To study the impact of hypothyroidism on sperm motility in male rat.Methods According to body weight,20 male Wistar rats were randomly divided into control group and hypothyroidism group (1 ml/100 g/day,0.1% propylthiouracil by intragastric administration for 60 days) 10 rats in eachgroup.Body weight of these rats was observed every 3 days.After the last intragastric administration,all rats were killed.The levels of thyroid hormones [total triiodothyronine (T3),total thyroxine (T4),and thyroid stimulating hormone (TSH)] were measured by radioimmunoassay.Sperm motility parameters[average path velocity(VAP),straight fine velocity (VSL),straightness (STR),amplitude of lateral head displacement (ALH),sperm density(p),curvilinear velocity (VCL),linearity(LIN),wobble (WOB),mean angular deviation (MAD) and beat cross frequency (BCF)] were measured by a WLJY-9000 color-detection system.Results Compared with the control groups[(298.20 ± 12.15) g,(344.00 ± 13.73)g],the weights of hypothyroidism group of the 30 days[(239.00 ± 15.02) g] and the 60 days [(232.67 ± 17.86)g] were significantly lower(t =7.704,11.380,all P < 0.05).The levels of T3[(373.3 ± 101.3) ng/L] and T4 [(4.00 ± 0.89) × 103 ng/ml] of hypothyroidism group were significantly decreased compared with that of the control groups [(1000.0 ± 273.5)ng/L,(44.33 ± 7.84)× 103 ng/L,t =5.262,12.520,all P < 0.05].Level of TSH[(5.77 ± 0.89) × 103 U/L] of hypothyroidism group was significantly increased compared with that of the control group[(1.87 ± 0.70) × 103 U/L,t =8.413,P < 0.05].Values of VAP[(27.45 ± 1.59)μm/s],VSL [(21.08 ± 1.10)μm/s],STR[(70.53 ± 3.48)%] and ALH[(1.96 ± 0.26)μm] of hypothyroidism group were significantly increased compared with that of the control groups[(24.38 ± 2.59)μm/s,(17.99±2.06)μm/s,(65.93 ± 2.71)%,(1.53 ± 0.27)μm,t =2.687,2.404,2.420,3.175,all P < 0.05].p[(5.07 ± 0.74)109/L] of hypothyroidism group was significantly decreased compared with that of the control group [(8.76 ± 1.01)109/L,t =6.463,P < 0.05].VCL[(52.83 ± 5.56)μm/s],LIN[(38.58 ± 3.41)%],WOB[(52.64 ± 3.24)%],MAD [(64.21 ± 6.71) radian/s] and BCF [(8.93 ± 0.62) Hz] of hypothyroidism group were not significantly different compared with that of the control groups[(49.92 ± 6.43) μm/s,(36.52 ± 2.73)%,(52.49 ± 3.49)%,(62.77 ± 7.34)radia/s,(9.32 ± 0.61)Hz,t =0.805,1.089,0.037,0.341,1.033,all P > 0.05].Conclusion Hypothyroidism can affect sperm activity in male rats,decrease sperm density and cause damage to the reproductive system.
4.Effects of recombinant human parathyroid hormone (1-34) and elcatonin on bone metabolism and serum SPARC in postmenopausal women with osteoporosis

ObjectiveTo investigate the effect of rhPTH (1-34) and elcatonin on bone metabolism and serum secreted protein acidic and rich in cysteine ( SPARC ) in postmenopausal women with osteoporosis.Methods One hundred and twenty-four postmenopausal women with osteoporosis were randomly divided into 2 groups:One group was treated with recombinant human parathyroid hormone ( 1-34 ) [ rhPTH ( 1-34 ) ] 200 U/d by subcutaneous injection (PTH group,n =89 )and another group was treated with elcatonin 20 U/week by intramuscular injection (CT group,n =35 ) for 12 months.All patients received a basic therapy with oral calcium ( Ca 600 mg+ Vit D3125 U,q..d.).The bone mineral density ( BMD ) of lumbar spine( L2-4 ),the left femoral neck,greater trochanter,and Ward's triangle,serum calcium and phosphate were measured by baseline,6 months' and 12 months.Levels of serum bone-specific alkaline phosphatase( BSAP),serum secreted protein acidic and rich in cysteine (SPARC)were determined by an ELISA assay.ResultsBy 12 months,rhPTH ( 1-34 ) treatment significantly increased the lumbar spine L2-4 BMD 7.9% (P<0.05),serum calcium 8.3 % ( P< 0.05 ),serum BSAP 93.4% ( P< 0.05 ),serum SPARC by 12.6%[ ( 195.68±59.57 vs 173.81 ±81.33 ) pμg/L,P<0.05 ].Elcatonin therapy increased the lumbar spine L2-4 BMD by 3.2% (P<0.05) at the end of 12 months,but elcatonin did not influence serum calcium,BSAP and SPARC.The rhPTH( 1-34 ) increased lumbar spine L2-4 BMD more than elcatonin did at 12 months( P<0.05 ).ConclusionrhPTH (1-34) could promote the bone anabolism more effectively than elcatonin did.Serum SPARC may play an important role in promoting osteogenesis by rhPTH.
5.Comparison of experimental autoimmune encephalomyelitis models induced by two kinds of proteolipid proteins

BACKGROUND: Experimental autoimmune encephalomyelitis has become the most classical animal model for multiple sclerosis. However, the experimental autoimmune encephalomyelitis model of China presented one-way course of disease. By using proteolipid protein 139-151 and proteolipid protein 178-191, relapse remitting experimental autoimmune encephalomyelitis models may be induced in SJL/J mice which were susceptible to immune, which have similar clinical situation, course and histologicallterations to multiple sclerosis.OBJECTIVE: To establish the relapse remitting experimental autoimmune encephalomyelitis mouse model induced by proteolipid protein, which has similar clinical situation, course and histological alterations to multiple sclerosis.DESIGN: Completely randomized controlled study.SETTING: The centre of Neuro-information, and Neurological Institute,General Hospital of Chinese PLA.MATERIALS: The study was carried out at the Laboratory of Neuro-pathology, General Hospital of Chinese PLA, from February to June 2004.Sixty female SJL/J mice with 8-12 weeks old were selected and randomly divided into proteolipid protein 139-151 group and proteolipid protein-178-191 group with 30 in each.INTERVENTIONS: After injected with proteolipid protein-139-151 or proteolipid protein-178-191, the models of relapse remitting experimental autoimmune encephalomyelitis were induced, and the body weight and neurological signs of each female SJL/J mouse were viewed. The tissue morphological changes of models were observed with hematoxylin and eosin and uxol fast blue stain.MAIN OUTCOME MEASURES: The neurological symptoms and signs,features of relapse and remitting and the perivascular inflammatory cell infiltration, demyelinated lesion of the model of experimental autoimmune encephalomyelitis mouse induced by two proteolipid protein peptides.RESULTS: All 60 mice entered the final analysis. ① Significant neurological symptoms, signs and features of relapse and remitting was manifested in the model of experimental autoimmune encephalomyelitis mouse induced by two proteolipid protein peptides. Obvious phenomena of perivascular inflammatory cuffing, satellitism, predominant perivascular inflammatory cell infiltration and demyelinated lesion were found in spinal and cerebral tissue. ②Changes of body mass: Before immunity, the body mass of mice in two groups was( 17. 84 ± 2.59) g and (17. 88 ± 0.52) g respectively. Onset of relapse of the mice in proteolipid protein-178-191 group was earlier and faster, their body mass had no distinctive change after immunization and the mean body mass was(23.52 ± 2.37) g till the 60th day. Meanwhile, Onset of relapse of the mice in proteolipid protein-139-151 group was later and slower. After the immunity, the body mass of mice was little decrease, and the body mass was (16. 70 ±0.46) g on the 60th day. ③ Neural functional scores: The highest functional scores in the two groups were not different(3.86 ± 1.10vs 3.71 ±1.05, t=0.49, P=0.628).CONCLUSION: The two different antigenic peptides of proteolipid protein can all cause the autoimmune response of central nervous system. Both models have the same characters of relapse and remitting and the severity has no significant difference. But compared with proteolipid protein 139- 151 group,onset and recover of the experimental autoimmune encephalomyelitis of the mice in proteolipid protein 178-191 group were earlier, as well as weight variance was larger, which maybe due to the different structure of two peptides.
6.Antibacterial activity of Cu ions released from 316L-Cu antibacterial stainless steel

BACKGROUND:316L-Cu antibacterial stainless steel is made by adding a certain amount of copper into the stainless steel fol owed by a special heat treatment to uniformly disperse copper-rich precipitates in stainless steel substrate, thereby harvesting the antibacterial properties. OBJECTIVE:To evaluate the antibacterial activity of the Cu ions released from 316L type Cu-bearing antibacterial stainless steels against Porphyromonas gingivalis, thereby providing biomedical evidence for its clinical application. METHODS:The medical 316L stainless steel samples at a surface area to volume ratio of 0.1 cm2/L were soaked in simulated body fluids at 37 ℃ for 1-10 days. A graphite furnace atomic absorption spectrophotometer was employed to detect the amount of Cu release in the simulated body fluids each day and then the rate of Cu release per day could be determined. The antibacterial activities of the steel samples were evaluated by a standard film-covered method under a scanning electron microscope. RESULTS AND CONCLUSION:The daily Cu releasing amount from the 316L-Cu stainless steel within 10 days was significantly higher than that of 316L stainless steel, and al the values remained nearly constant. With time, the sterilizing rate of 316L-Cu stainless steel was gradual y increased, and reached 100%until the 10th hour. Porphyromonas gingivalis showed some morphological changes at 3 hours after treated with 316L-Cu stainless steel, appeared with cleavage at 6 hours, and mostly disintegrated into pieces at 9 hours. The results indicated that the 316L-Cu antibacterial stainless steel showed excel ent antibacterial property against Porphyromonas gingivalis, slowly release Cu irons, and alter the surrounding microenvironment, which is a highly promising biomaterial and has good clinical value.
7.Data Mining and Analysis of Acupuncture Treatment for Diabetes

ObjectiveTo explore the characteristics of and regularities in the application of points for acupuncture treatment of diabetes in clinical literature.MethodOne hundred and two pieces of clinical literature on acupuncture treatment for diabetes published from 1998 to 2013 were sorted out, sifted, entered and statistically analyzed using modern computer and data mining technology to summarize the characteristics of and regularities in the application of acupoints in the literature.ResultSanyinjiao was the most frequently used point for acupuncture treatment of diabetes (68). Two points[Pishu(BL20)and Shenshu(BL23)], three points[Pishu, Shenshu andSanyinjiao(SP6); Weiwanxiashu(Ex-B3), Pishu and Shenshu], four points[Zusanli(ST36),Pishu, Shenshu and Sanyinjiao]and five points[Zusanli, Pishu, Shenshu, Weiwanxiashu and Sanyinjiao]were the most frequently used point combinations. The most frequently used meridian was the Bladder Meridian of Foot-Taiyang (260). The selected points were distributed on the back and the lower limb. The proportion of used specific points reached 74.5%. Five Shu points and Back-Shu points were used most frequently. The proportion of using manual acupuncture was 57.6%. Treatments with the cooperation of Dietary control and exercise conditioning accounted for 64.4%.ConclusionAcupuncture treatment of diabetes is mainly based on regulating the middle Jiao, with the upper Jiao and the lower Jiao taken into account. Specific points are the main part of point combinations. Manual acupuncture is the main treatment and meanwhile, attention is paid to adjusting life style.
9.Effect of Gli2 on apoptosis and ROS level in renal tubular epithelial cells under high glucose

Objective:To investigate the effect of Gli2 on apoptosis and ROS level in renal tubular epithelial cells under high glucose. Methods:The renal tubular epithelial cell NRK-52E was used as the object of study,Gli2 overexpression vector and control vector were transfected,the non transfected cells were also used as controls,RT-PCR and Western blot detected Gli2 levels,culture of non transfected cells by high glucose and low glucose cell culture medium, cells transfected with Gli2 overexpression vector were cultured in high glucose medium,apoptosis was detected by flow cytometry,the ROS content was detected by DCFH-DA probe method, the content of SOD was detected by xanthine oxidase method; Bax, Cleaved Caspase-3 and Smo levels were detected by Western blot. Results:The Gli2 mRNA and protein levels of NRK-52E cells transfected with Gli2 overexpression vector were significantly higher than that of control cells(P<0. 01),the Gli2 mRNA and protein levels in the NRK-52E cells transfected with the control vector were not different from those of the control cells(P>0. 05). The apoptosis rate of NRK-52E cells without transfection was elevated after high glucose culture,elevated ROS content,SOD content decreased,the levels of Bax,Cleaved Caspase-3 were elevated in the cells,Smo levels declined,compared with the control cells,the difference was statistically significant(P<0. 01). After overexpression of Gli2 over-expression vector,NRK-52E cells were cultured with high glucose,the rate of apoptosis decreased,ROS content decreased,elevated SOD content,the levels of Bax,Cleaved Caspase-3 decreased,elevated levels of Smo,compared with NRK-52E cells cultured with simple high glucose,the difference was statistically significant(P<0. 01). Conclusion:Apoptosis and oxidative damage in renal tubular epithelial cells induced by high glucose, Gli2 can decrease the apoptosis of renal tubular epithelial cells induced by high glucose, mitigate oxidative damage.
10.Effects of Formaldehyde Escaped From Plywood Exposure on Neurotransmitter in the Brain of Mice

Objective To investigate the effects of repeated exposure by inhalation to several concentrations of formaldehyde 5 hours/day 7 consecutive days on the content of neurotransmitters in the brain of adult female BALB/c mouse. Methods The 24 mice were randomly divided into 3 groups(groups A to C) 8 in each and exposed respectively to formaldehyde of 0.02-0.07 mg/m3 controls 0.99-1.2 mg/m3(3 m2 plywood/m3 room)and 3.44-3.63 mg/m3(10 m2 plywood/m3 room). After sacrifice the brain were excised and the neurotransmitters were examined. Results Asp、NE、Glu、E and Ach in animals of the exposed groups were significantly decreased but GABA was increased. Conclusion Exposure of formaldehyde escaped from plywood may result in neurochemical change such as Asp NE GluE Ach and GABA in the brain of mouse.
