1.Mycoplasma Culture and Analysis of Susceptibility Test

OBJECTIVE To compare the detection rates of Ureaplasma urealyticum(Uu),by 4 kinds rits and find out the best one.METHODS Using four kinds of mycoplasma detection kits,namely are French mycoplasmaIST2,Zhuhai Livzon′s ICS,Dark horse′s IES and Shanghai Its Prachanda,under the same conditions,the mycoplasma of the Clinical specimens for 260 cases were tested.RESULTS The detection rate of mycoplasma(Uu) for 48 hours by four kinds of brands kits about mycoplasma liquid culture was as follows 45.4% for French MycoplasmaIST2,36.2% for Zhuhai Livzon′s ICS,37.7% for Dark horse′s IES and 36.8% for Shanghai Its prachanda.The results of susceptibility test: the reagent of IST2 had.the lowest resistance to josamycin,doxycycline,clarithromycin,ofloxacin and azithromycin,and had the better results than the other three kinds of reagents.Mycoplasma reagents form different companies had significant differences in resistance rates to five kinds of antibiotics.CONCLUSIONS The results suggest that when we cultivate clinical mycoplasma should be careful to select of the mycoplasma liquid culture identification kit.
2.Epidemiological features and influential factors of prehypertension in Yongchuan district of Chongqing

Objective To investigate the epidemiological features and influential factors of prehypertensionin Yongchuan district of Chongqing .Methods The cluster sampling method was adopted to conduct the survey on randomly selected 3 100 local people (≥18 years old) ,it concluded questionnaire ,physical examination ,blood specimen collection and so on .Results The prevalence of hypertension in Yongchuan district of Chongqing adults was 43 .0% .The prevalence of hypertension in male(49 .5% ) was higher significantly than that of female(35 .0% )(P<0 .05);The prevalence of prehypertension was increased with age up and then de‐clined in older age ,it was achieve highest at 40- <50 years old(50 .7% ) .The average age ,smoking rate ,drinking rate ,BMI ,waist circumference ,TC ,TG ,Purine trione of the prehypertension group was higher than that of the normal group(P<0 .05);the results of Logistic regression indicated that the risk factors of prehypertension weresmale ,higher BMI ,abdominal obesity ,hypercholesterol‐emia ,hypertriglyceridemia ,smoking regularly ,drinking regularly and so on(P<0 .05) ,physical activities was protective factor(P<0 .05) .Conclusion The prevalence of prehypertension in the adults of Yongchuan district of Chongqing is high ,relative intervention on these factors should be tookas soon as possible .
3.Expression of netrin-1 in placenta from patients with pre-eclampsia and the relation to placental angiogenesis

Objective To investigate the expression of netrin-1 in placenta from patients with pre- eclampsia and the relation to placental angiogenesis.Methods Twenty patients with pre-eclampsia(12 mild cases and 8 severe cases)and 20 normal late pregnant women were investigated.The expression of netrin-1 mRNA and protein was determined with RT-PCR and Western blotting respectively.The placenta vascular density was examined by immunohistochemical F8 staining.Results (1)The values of netrin-1 mRNA in normal group and pre-eclampsia groups were 0.51?0.08 and 0.41?0.06;The values of netrin- 1 protein in normal group and pre-eclampsia groups were 26.4?1.8 and 20.5?1.3(P

Aim To investigate the effects of β1-ad-renergic receptor (β1-AR ) on rapid component of the delayed rectifier potassium current ( IKr ) in ventricular
myocytes of guinea pigs with chronic heart failure ( CHF) . Methods The CHF model of guinea pigs was established by descending thoracic aortic banding .
Whole-cell patch-clamp technique was used to record IKr in ventricular myocytes. The effects ofβ1-AR on IKr in CHF ventricular myocytes were detected and its mechanisms were studied by pretreatment with protein kinase A ( PKA ) inhibitor and calmodulin kinase II( CaMK II) inhibitor. Results In CHF ventricular myocytes, xamoterol, the selectiveβ1-AR agonist, de-creased IKr by (52±8)% and prolonged action poten-tial duration. These effects were completely abolished by pretreatment of myocytes with CGP20712A, a selec-tive β1-AR antagonist. íamoterol only decreased IKr
by (28±3)% by pretreatment of CHF myocytes with specific PKA inhibitor KT5720 . KN93 , an inhibitor of CaMKII, did not attenuate the inhibitory effect on CHF ventricular myocytes. Conclusion IKr is inhibi-ted by β1-AR activation in CHF ventricular myocytes. PKA, but not CaMKII signaling pathway is involved in, at least in part, the inhibitory effect ofβ1-AR acti-vation on IKr.
5.Detection of Related Resistance Genes of Chloramphenicol and Tetracycline in Multidrug-resistant Pseudomonas aeruginosa

OBJECTIVE To explore the distribution of related resistance genes of chloramphenicol and tetracycline in Pseudomonas aeruqinosa isolates.METHODS The antibiotics susceptibility was tested by K-B method,catB cmlA,tetA tetB and smr-2 were detected by polymerase chain reaction(PCR),the gene cmlA was sequenced by PCR fluorescence spectrophotometry.RESULTS The drug-resistant rates of 20 strains of P.aeruginosa against 17 kinds of antibiotics ranged from 10.0% to 100.0%,and multidrug resistant strains were found.The gene of cmlA had been detected in 6 strains of 15 resistant P.aeruginosa isolates,but the genes of tetA,tetB and smr had not been detected.The cml was sequenced,and compared with GenBank,the result showed the gene fragment shared 99.0% homology in nucleotides with the GenBank sequences of cmlA7 and cmlA8.CONCLUSIONS Resistance to antibiotics in P.aeruginosa of our hospital is mainly related to cmlA.The resistance to chloramphenicol and tetracycline was not related to tetA,tetB and smr-2.
6.Image examination of renal injuries and analysis of renal explorative indications

Objective To study the image examination of renal injuries and discuss renal explorative indications so as to spare the kidney or nephron as much as possible and improve curative rate of diagnosis and treatment. Methods An analysis was done on 286 cases that included 231 cases with close injury, 54 with open injuries, one with iatrogenic injury and 91 with combined injuries. Of all, 212 cases were examined by B-ultrasonography, 163 by CT and 132 by intravenous urography(IVU) and 6 by digital subtraction angiography(DSA); 202 cases were treated with conservative treatment and 84 with operation. Results The diagnostic positive rates of IVU, B-ultrasonography and CT were 67.4%, 72.2% and 87.7%, respectively. Among the operation cases, 42 cases were treated by renal repair, 12 by partial nephrectomy and 30 by nephrectomy. The operation rate was 29.4% and the nephrectomy rate 35.5%. Interventional treatment of the kidney was carried out in three cases. Conclusions For renal injury cases, the first and most important step is to evaluate the injury condition so as to correctly determine whether an operation exploration is needed. The injury conditions and severity are mainly determined by the image examinations that change according to injury cause, injury type and clinical symptoms. Renal exploration or not, and the operation time exert great influence on renal reservation rate and complication rate.
7.Characteristic of salvianolic acid binding to bovine serum albumin and structure-performance relationship

Objective To study the characteristic of salvianolic acid(SA) binding to bovine serum albumin(BSA) and the structure-performance relationship.Methods The interactions between BSA and four natural SA(SAⅠ,rosmaric acid;SAⅡ,lithosperic acid;SAⅢ,salvianolic acid A;and SAⅣ,salvianolic acid B) were investigated by fluorescence and ultraviolet spectroscopy.Results The intrinsic fluorescence of BSA was quenched by SA via forming SA-BSA complexes and non-radiation energy transfer.The parameters of SA-BSA binding process,such as the static apparent association constant KA,the number of binding site n,the efficiency of energy transfer E,the spatial distance r were obtained,and the thermodynamic constants ?G,?H,and ?S were calculated.Conclusion The results indicate that SAⅢ bounds to BSA mainly through a hydrophobic force and the other three SA-BSA interactions are mainly driven by hydrogen bond and Van der Waals' force.Y,a comprehensive binding parameter constructing from the equation Y=lg (KA?E?n/r),could be used to reflect the interaction extent of SA-BSA system.The Y value changes with the number of free phenolic hydroxyl and molecule volume and decreases in the order of SAⅢ→SAⅣ→SAⅡ→SAⅠ.The results from correlation analysis indicate that it could be possible to estimate SA-BSA binding extent from hydrophobic parameter clogP and topo-polar surface area per unit of molecular weight tPSA/Mr of SA molecule.
8.High-intensity focused ultrasound with submicrobubbies to damage rabbit breast VX2 tumors

Objective To observed the effect of high-intensity focused ultrasound (HIFU) in combination of submicrobubble on ablation of breast tumor VX2 in rabbits.Methods Submicrobubble was prepared,and the appearance,distribution,diameter,and zeta potential were measured.The rabbits were injected with the tumor tissue suspension in bilateral breast tissues.HIFU(150 W,120 W,5 s,3 s) combined with submicrobubble to damage rabbit breast VX2 tumors.The coagulation necrosis was assessed by HE staining.The grayscale changes of the target area before and after HIFU irradiation were recorded automatically by computer.Results The diameter of the submicrobubble was 498.9 nm on average.HE staining showed the area of coagulation necrosis was significantly larger in the HIFU + submicrobubble group than the HIFU + PBS group.The injury range of 150 W irradiation was larger than that of 120 W irradiation,given the same irradiation time.The injury range of 5 s irradiation was larger than that of 3 s irradiation,given the same irradiation strength.The gray scale change in the target area was significantly higher in the HIFU + submicrobubble group than the HIFU + PBS group.Conclusions The submicrobubble exhibited sound imaging performance,and it can be use in the ultrasound therapeutics in the future.
9.Pharmacokinetics of thymopeptide in rabbit

Purpose The aim is to study the pharmacokinetic of iv thymopeptide in rabbit.Method Thymopeptide concentration of rabbit plasma was determined by HPLC and pharmacokinetic parameters were computerized with 3P87 program.Results and Conclusions The pharmacokinetic parameters of iv thymopeptide in rabbit were:Cmax=(30.06±9.27)μg/ml,t1/2(β)=(29.24±23.78)min, Ke=(0.032 3±0.013 8)min-1 and AUC 0→∞=(205.63±46.48)μg*min/ml.
10.Cloning,expression and purification of fragile X mental retardation protein

Objective Explore the conditions of the cloning,expression and purification of FMRP.Methods The plasmid pET22b(+)-FMR1,constructed by molecular cloning,was transformed into E.coli BL21(DE3) competent cells and induced to express FMRP by IPTG.Recombinant FMRP was purified by affinity chromatography,verified by Western-blot,and tested for its RNA binding ability.Results FMR1 cDNA was successfully cloned into pET22b(+) vector and expressed in E.coli BL21(DE3).A protein with Mr 79 000 was purified and confirmed to be FMRP.This protein retained the RNA binding ability of FMRP.Conclusion We successfully expressed recombinant hFMRP with high purity and activity in E.coli,which provided a reliable material to study the function of FMRP.
