1.Effects of 70 pearl flavor pill associating with acupuncture on nerve function in acute cerebral infarction patients

Objective To analyse the effect of 70 pearl flavor pill associating with acupuncture on nerve function in acute cerebral infarction patients.Methods A total of 128 patients with acute cerebral infarction were enrolled in this study in our hospital from January 2015 to June 2016 and randomly divided into two group.Two groups were treated with routine treatment,the observation group was treated another with 70 pearl flavor pill combined with acupuncture program.Results The total effective of observation group was 92.19%,higher than that of control group 75.00%(P<0.05).After treatment,NIHSS score,FMA motor function score,Serum S-100βprotein,hs-CRP level of observation group were better than that of control group (P<0.05).Conclusion 70 pearl flavor pill associating with acupuncture on the basis of routine treatment can improve hemorheology,reduce inflammation and improve the therapeutic effect and it has no obvious side effects.
2.Clinical analysis of central venous catheter related infection (CRI)

Objective To investigate the characteristics of pathogens and risk factors of the catheterassociated infection (CAI) in emergency ICU (EICU) in order to design an appropriately therapeutic strategy for the future.Methods From January 2008 to December 2010,a total of 1363 patients were enrolled for this retrospective study.Blood sample taken from the vein with indwelling catheterization and the tips of catheters cut in 5 cm after withdrawn from the veins in 1363 patients were collected for bacterial culture.Results Of 1363 catheters,pathogens were found in 147 (10.79%) after venous catheterization.The daily occurrences of CAI were 3.05 ones per 1000 catheters.Of 147 cases of infection,46.94% pathogens were gram-negative bacilli,40.14% gram-positive cocci,and 12.92% fungi.Unconditional Logistic regression analysis suggested that repeated catheterization,femoral vein catheterization,the application of multi-lumen catheter and long-term indwelling catheterization were the independent risk factors responsible for CAI.Conclusions The risk factors responsible for catheter related infections should be controlled to prevent the occurrence of nosocomial infection.
3.Effect of taurine on the differentiation of 3T3-L1 preadipocytes

AIM: Some studies found that the glucose uptake ability of diabetes mellitus mice is stronger after feeding with taurine. Whether the taurine can affect adipocytes or not deserves further studies. This article investigates the effects of taurine on the differentiation of 3T3-L1 preadipocytes and detects the mechanics. METHODS: Experiments were performed in the Laboratory Center of Xiangya Third Hospital of Central South University from July to September 2006. ①3T3L1 preadipocytes were provided by Shanghai Cell Bank of China Academy of Sciences. ②3T3-L1 preadipocytes were cultured in high glucose DMEM medium containing fetal bovine serum of 0.10 volume fraction, 108 u/L benzylpenicillin and 80?1010 U/L streptaquaine. After confluence, cells at 5?107 L-1 were incubated in culture flask, and induced with 0.5 mmol/L IBMX, 0.5 mg/L insulin and 1 ?mol/L dexamethasone. The cells in an experimental group were intervened with taurine, whereas these in a control group were not treated with taurine. Oil O staining was used to observe the development of adipose cells. C3T3-L1 preadipocytes were treated with 10,20 mmol/L taurine respectively for 24 and 48 hours. RNA and protein were extracted in the control group and the experimental group. ③Genes related to adipose cells development was examined by RT-PCR and Western-blot. RESULTS: ①The number of cells stained by oil O was less in the experimental group than the control group in 3T3-L1 preadipocytes treated with 10 mmol/L taurine for 14 days. ②After C3T3-L1 preadipocytes were treated with 10,20 mmol/L taurine respectively for 24 and 48 hours, there were no changes in expression of Insig-2 protein, PPAR, Insing-2, adiponectin, adiponectin receptor, GLUT-4, AP-2 mRNA. CONCLUSION: Taurine inhibits the differentiation of 3T3-L1 preadipocytes, but the mechanics still need to be investigated.
4.Problem and correction of the application of foreign instrument with higher false-positive prenatal screening for neural tube defects

Objective To establish appropriate correction models of serum marker α -fetoprotein (AFP) multiple of the median (MOM) for prenatal screening on patients with neural tube defect (NTD) at the second trimester to replace the foreign data currendy used in Beijing with a higher sensitivity.Methods A sample of 5815 mid-gestation pregnant women with normal pregnancy outcome were selected including 5557 cases of age < 35 years and 258 cases of age ≥35 years.The regression relationship between AFP median and gestational age was builded by exponential model and the regression relationship between MOM and weight was builded by power model in age < 35 years.The regression relationship between AFP median and gestational age was builded by S model and the regression relation ship between MOM and weight was builded by quadratic model in age ≥35 years.The normal reference values in two age sections were estimated approximately by percentiles method.Results Corrected AFP MOM values with mean value (age < 35 years:1.001; age ≥35 years:1.113) and median value (age < 35 years:0.934;age ≥35 years:0.990) obtained by regression analysis were more close to 1.00 than the data from the software[mean value(age < 35 years:1.164; age ≥35 years:1.254) and median value (age <35 years:1.093; age ≥35 years:1.149)].There were significant differences (age <35 years:t =65.120,P =0.000;age ≥35 years:t =9.812,P =0.000).Conclusion Correction model for maternal serum marker is more applicable for Beijing population at the second-trimester prenatal screening.
5.Acupuncture at neck Jiaji acupoints combined with moxibustion for clinical study of cervical vertigo

Objective To observe the effect of acupuncture at neck Jiaji acupoints combined with moxibustion to cervical vertigo.MethodsIn Wuhan Chinese and Western Union Hospital from August 2015 to November 2016, 80 patients with cervical vertigo were randomly divided into two groups, the treatment group was treated by needling cervical Jiaji points, at the same time with moxibustion on neck Jiaji acupoints, the control group was given drug treatment.ResultsThe total effective rate in the treatment group was 95%, but the control group total effectiveness was 72.5%, the effect of the two groups had significant difference(P<0.05).ConclusionAcupuncture at Neck Jiaji points combined with moxibustion treatment had the functions of relieving rigidity of muscles and activating collaterals,dispelling cold and dredging channel blockage, relieveing local muscle spasm,improveing blood circulationand improve the function of the blood supply to the brain to vertigo.
6.Effects of Selenium and Zinc on the Absorption, Excretion and Accumulation of Fluoride in Rats

ve To investigate the effects of selenium and zinc on the absorption, excretion and accumu-lation of fluoride in rats. Methods The contents of fluoride in serum, excrement, urine and bone were determined in Wistar rats drinking distilled water containing 100 mg/L NaF and orally perfused jointly with 0.1 mg/(kg? d) Na2SeO3 and/or 14.8 mg/(kg?d) ZnSO4 one time per two days continuously for 90 days. Results Na2SeO3 and/or ZnSO4 could increase the concentration of fluoride in urine, decrease the concentration of fluoride in serum and the content of fluoride in bone of rats. Exposure to ZnSO4 and joint exposure to Na2SeO3 and ZnSO4 could increase the content of fluoride in excrement. Conclusion ZnSO4 could inhibit the absorption of fluoride in intestine, Na2SeO3 and /or ZnSO4 could promote the excretion of urine fluoride and restrain the accumulation of fluoride in bone of rats.
7.Inhibition of the vasculogenic mimicry formation by targeting fibroblast growth factor receptor in glioma

Objective To explore the effect of fibroblast growth factor receptor(FGFR)inhibitor BGJ398 on the vasculogenic mimicry(VM)formation of glioma cells. Methods The phosphor-FGFR(pFGFR)was de-tected by Western blot,the expressions of MMP2 and MMP14 were detected by Western blot and immunocytochem-istry;the VM formation of U87MG and U251MG was tested by tube formation assay;subcutaneously implanted tu-mor model in nude mouse was established and tumor sections were CD34/PAS double-stained to detect the forma-tion of VM in vivo. Results Western blot showed that pFGFR in the experimental groups decreased significantly (P < 0.05);western blot and immunohistochemical staining showed that the expression of MMP2 and MMP14 in the experimental groups decreased significantly compared to the control group. In the tube formation assay ,the tube formation of U87MG and U251MG cells were restrained. In the subcutaneously implanted tumor model ,the VM number of the experimental group(13.85 ± 3.96)was significantly lower than that in the control group(26.40 ± 5.06,P < 0.05). Conclusions In vivo and in vitro experiments confirmed that BGJ398 can inhibit the activa-tion of FGFR,and inhibit the VM formation of glioma cells. These indicate FGFR signaling pathway is involved in the formation of VM.
8.The protective effect of exogenous bone marrow mesenchymal stem cells on pulmonary fibrosis induced by paraquat

Objective To explore the protective effect and possible mechanism of bone marrow mesenchymal stem cells (BMSCs) on pulmonary fibrosis induced by paraquat (PQ) in rats.Methods A total of 54 male SD rats were randomly (random number) divided into 3 groups (n =18 in each).Group A (normal control group),group B [PQ + phosphate buffer solution (PBS)],group C (PQ + BMSCs group).Rats in group B and C were induced to get pulmonary fibrosis by intragastric administration of PQ in a dosage of 120 mg/kg.In addition,BMSCs was given to rats in group C by injection in a dose of 1 × 106/mL via the vena caudalis,whereas an equal volume of PBS (1 mL) was given to rats in group B by injection instead.The survival rats in each groups were sacrificed separately at 7 days,14 days and 28 days after administration of PQ.The lower lobe of left lung were taken to observe histopathological changes with HE staining and Masson staining,and the pulmonary fibrosis was scored by using the Szapiel classification of alveolitis.At the same time,the lower lobe of right lung was harvested to detect the hydroxyproline (HYP),TGF-β1 and HGF in lung tissue by using immunohistochemistry.Results The pathological changes in lung tissue of rats were inflammatory change in alveoli space filled with massive amount of exudate obviously in group B at 7 days after exposure to PQ,but in group C,the inflammatory changes were much milder than those in group B.The exudation and edema in lunge alveoli were mitigated in group B at 14 days after exposure to PQ,but pulmonary interstitial fibers were increased.The degree of pulmonary interstitial fibrosis in group C was milder than that in group B.In group B,there were obvious pathological changes including destroyed alveoli septum,enlarged alveoli,thickened alveoli septum and the deposition of collagen fibers,disarranged alveolar structure at 28 days after exposure to PQ.The deposition off collagen fiber was slighter with well observed basic alveolar structure in group C than that in group B.The levels of HYP in lung tissue at different intervals in B group were escalating with time after exposure to PQ,and reached a peak at 28 day,which were significantly higher than those in group A (P < 0.01).The levels of HYP in lung tissue at different intervals in C group were increasing with time after exposure to PQ,and reached a peak at 28 day,which were significantly lower than those in group B (P < 0.01).The levels of TGF-β1 from different intervals in B group were increased greatly with wider distribution over large portion of lung structure than those in group A,and reached a peak at 7 days,then declined with time,which were significantly higher than those in group A (P < 0.05).The levels of TGF-β1 at different intervals in group C were significantly lower than those in group B (P < 0.05).The levels of HGF at different intervals in B group increased with time,but there was no significant difference among those at different intervals (P >0.05).The levels of HGF at different intervals in group B were little bit higher than those in group A,but there was no significant difference between them (P > 0.05).The levels of HGF at different intervals in group C increased with time with significant differences among different intervals (P < 0.05).The levels of HGF at 3 intervals in group C were significantly different from those of group A and group B group (P <0.05).Conclusions BMSCs can alleviate pulmonary fibrosis induced by PQ,and increase the survival rate,which may be attributed to decreasing the level of TGF-β1 and increasing the level of HGF.
9.The effects of calmodulin kinase Ⅱ inhibitor on hypertrophic cardiac myocytes

Objective To investigate the effect of the calmodulin kinase Ⅱ Inhibitor KN-93 on L-typecalcium current(ICa,L)and intracellular calcium concentration([Ca2+]i)in hypertrophic cardiac myocytes.Methods Forty-eight female New Zealand white rabbits were randomized(random number)into four groups(12 animals in each group):the sham operation group(sham group),the left ventricular hypertrophy group(LVH group),the myocardial hypertrophy + KN-93 group(KN-93 group),and the myocardial hypertrophy + KN-92 group(KN-92 group).Myocardial hypertrophy in the rabbits was established by coarctation of the abdominal aorta.In the sham group,the abdominal aorta was dissociated without coarctation.Eight weeks after coarctation,single ventricular myocytes were isolated by enzymaticdissociation,and ICa,L was recorded using perforated-patch recording(PPR)techniques.[Ca2+]i was measured using single-cell calcium imaging with the fluorescence calcium indicator dye fura-2/AM.Results Cardiac hypertrophy was successfully established after 8 weeks of coarctation of the abdominal aorta.The peak ICa,L in the LVH group and the sham group was(1.38 ± 0.3)nA and(0.87 ± 0.1)nA at 0 mV,respectively(P <0.01,n =12).There was no significant difference in Ica,L density between the LVH group and the sham group[(6.7 ±1.0)pA/pF vs.(6.3±0.7)pA/pF; P≥0.05,n=12].The addition of either KN-92(0.5 μmol/L)or KN-93(0.5 μmol/L)to the perfusing solution caused a modest steady-state inhibition of peak ICaL(9.4% ±2.8%,KN-92; 10.5% ±3%,KN-93)(P≥0.05,n =12)at 0 mV.However,at a higher concentration(1 μmol/L),KN-93 more potently inhibited peak ICa,L(40%±4.9%)compared to KN-92(13.4% ± 3.7% ; P < 0.01,n =12).Resting[Ca2+]i levels in fura-2-loaded myocytes isolated from the sham,LVH,KN-92,and KN-93 groups were(98 ± 12.3)nmol/L,(154 ± 26.2)nmol/L,(147 ± 29.6)nmol/L,and(108 ± 21.2)nmol/L,respectively.Conclusions The CaMK Ⅱ specific inhibitor,KN-93,can effectively block ICa,L and reduce intracellular calcium overload in hypertrophic cardiac myocytes.This action may account for the antiarrhythmic effect of KN-93 in hypertrophic ventricular myocardium.
10.Changes and clinical significance of ischemia modified albumin in patients with cardiopulmonary resuscitation

Objective To study the changes in the level of serum IMA in patients with cardiac arrest followed by CPR,and explore the value of serum IMA in predicting prognosis of these patients after CPR.Methods (1) A total of 36 patients (25 men and 11 women) after CPR following cardiac arrest were recruited into experimental group (group C).Of them,15 cases (11 men and 4 women) and 21 cases (14men and 7 women) were assigned into resuscitation group (group R) and non resuscitation group (group NR),respectively,according to the presence or absence of ROSC.Meanwhile,another 60 healthy subjects (39 men and 21 women) matched for age and gender were recruited into healthy control group (group H).(2) The determination of the serum IMA level and comparisons of changes in serum IMA levels among groups were carried out.Results (1) The level of serum IMA in group C was higher than that in group H (P < 0.05) ; The serum IMA level in group R was lower than that in group NR and higher than that in group H (P <0.05 for both).(2) The area under the ROC curve for serum IMA in predicting the prognosis of patients treated with CPR after cardiac arrest was 0.73.When the cut off value was 128.25 U/mL,the sensitivity and specificity were 57.1% and 93.3% respectively; the positive predictive value and negative predictive value were 92.3% and 60.9% ; positive likelihood ratio and negative likelihood ratio were 8.5 and 0.5 ; the correct diagnosis index was 72.2%,and Youden index was 50.5%.Conclusions Serum IMA has a certain value in prediction of prognosis in patients treated with CPR following cardiac arrest.
