1.Experimental study of apoptosis in chronic compressive injury of the spinal cord

Objective To explore the phenomenon of neural apoptosis after chronic compressive spinal cord injury. Methods A newly designed chronic progressive compressive spinal cord injury model of rat was adopted in this study. Forty-five Wister rats were divided into mild, moderate, and severe group according to the compressive degree of the spinal cord. The Feulgen stain and TUNEL were used to investigate the apoptosis and its characteristics in different kinds of neural cells. Results Apoptosis index in moderate injury group was the highest. Apoptotic cells largely located in ventral, lateral and dorsal column of the white matter. Most of them were oligodendrocytes. Positive neuron occasionally presented in laminae Ⅲ~Ⅸ part of the sections. Most of them located in dorsal gray horn. Conclusions Apoptosis is an important event in secondary pathophysiological process of chronic progressive compressive spinal cord injury. Apoptosis is one of the reasons for neural cell death. The apoptosis of oligodendrocytos may contribute to myelin sheath disruption of white matter.
2.Effects of Recombinant Human IGF-1 on Kidney in Diabetic Rats

PurposeTo observe the effects of rhIGF-1 on kidney in diabetic rats. MethodsUsing biochemistry, radioimmunoassay, molecular biology (RT-PCR). Results(1) 24 h UAER,24 h uriary volume in rhIGF-1 group is lower than that of diabetic control group; (2) The level of sertan IGF-1 ,kidney IGF-1 and IGF-1 mRNA in diabetic control group is lower than that of normal control group;The level of serum IGF-1 in rhIGF-1 group is higher than that of diabetic control group;no differences were found in the levels of kidney IGF-1 and kidney IGF-1 mRNA between diabetic control group and rhIGF-1 group; (3) The level of serum GH in diabetic control group is higher than that of normal control group; The level of serum GH in rhIGF-1 group is lower than that of diabetic control group; (4) rhIGF-1 might have some protective effects on diabetic nephropathy via electron microscope. ConclusionsrhIGF-1 doesn't increase the damage of diabetic kidney tissue.
3.Transfection of wild-type p14ARF gene leads to growth inhibition of human lung cancer cell lines

Objective To examine whether wild-type p14ARF gene is a candidate suppressor gene for lung cancer. Methods Human lung cancer cell lines having various endogenous backgrounds in INK4a, p53 and Rb genes were used as the recipients of the wild-type p14ARF gene. The expression of p14ARF mRNA and protein was detected with RT-PCR, immunohistochemistry and Western blot after G418 selection. Clones which expressed both p14ARF mRNA and protein were identified and selected for further experiements. By comparing with the parental and negative control cells treated with empty vectors, the effects of exogenously transfected p14ARF on cell division rate, cell cycle distribution and morphologic alteration were analyzed. In vivo evaluation of the growth rate was also made with the experiment of nude mice tumor formation. Results Upon transfection with p14ARF gene, cells were arrested at G1 or G1/G2 phase of cell cycle in three wtp53 lung cancer cell lines and their proliferation rates were also inhibited. Conclusion Human wild-type p14ARF gene has suppressive effect on abnormal proliferation of lung cancer cells, especially in some wtp53 lung cancer cells, and it might be an ideal candidate for gene therapy of human lung cancer.
