2.Inhibitory effect of LIGHT-Fc gene transfection on human esophageal squamous carcinoma cell line Eca109

Objective To investigate the in vitro inhibitory effect of LIGHT on human esophageal carcinoma cells. Methods LIGHT Fc expression vector was transfected into human esophageal squamous carcinoma cell line, Eca109. The inhibitory effect of LIGHT gene on cell growth was detected by MTT and cell growth curve. The expressions of LT?R and HVEM were detected by RT PCR. Results Expression of LIGHT Fc gene could inhibit Eca109 cell proliferation. The growth curve of Eca109/LIGHT was significantly lower than that of the control group in the culture medium containing 1% FCS. MTT test showed that there was significant difference in cell viability between Eca109/LIGHT and the control group ( P
3.Finite element analysis of maxillary central incisor all-ceramic crowns in different occlusal states

BACKGROUND:The strength of al-ceramic crowns is affected by many factors. At present there are many studies concerning the neck edge shape, cutting thickness, binder and convergence angle, but little has been reported on the effects of different occlusions on al-ceramic crowns. OBJECTIVE:To explore the maxilary central incisor al-ceramic crowns in different occlusal states by finite element analysis. METHODS: Three-dimensional finite element models of the maxilary central incisor al-ceramic crowns were established. Empress II and In-Ceram Zirconia were selected. The normal overbite position and deep overbite position were applied to force a load of 230 N, with the long axis of the tooth at a 45° angle. Distribution of inner stress and edge stress was analyzed at different occlusal states. RESULTS AND CONCLUSION:(1) The stress distribution of al-ceramic crowns under different occlusion relationship was different. The stress peaks of cementum, alveolar bone and periodontium in the deep overbite were lower than those in the normal overbite. The stress peaks of al-ceramic crowns and binder were higher than in the deep overbite than the normal overbite. The stress peak of the dentin in the Empress II group was higher in the deep overbite than in the normal overbite. The stress peak of the dentin in the In-Ceram Zirconia group was lower in the deep overbite than in the normal overbite. Different al-ceramic crowns had no influence on the stress distribution, but in the deep overbite, the stress was mainly concentrated in the occlusal contact area, and in the normal overbite, the stress was mainly concentrated in the occlusal contact area and at the labial cervical margin. (2) The stress distribution of different al-ceramic crowns under the same occlusal state was different. In the normal overbite, different al-ceramic crowns had no evident influence on the stress peak, and in the deep overbite, there was a certain effect of different al-ceramic crowns on the stress peak, but there was no significant difference. The stress peaks of blinder and dentin were obviously affected by al-ceramic crowns. The stress peaks of blinder and dentin in Empress II group were higher than those in the In-Ceram Zirconia group.
4.Inhibitory effect of recombinant LIGHT-Fc gene on the proliferation of human esophageal squamous carcinoma cell line Eca109

Objective To investigate the inhibitory effect of recombinant LIGHT-Fe gene on the proliferation of human esophageal carcinoma cell line Eea109. Methods LIGHT-Fc expression vector was transfected into human esophageal squamous carcinoma cell line Eca109 by using DOTAP liposomal transfection reagents. The effects of LIGHT-Fc gene on the proliferation of esophageal carcinoma cell line Eca109 in vitro were detected by cell growth curve and MTr assay. Forty-five nude mice were equally divided into Eea109/Wt group, Eca109/neo group and Eca109/LIGHT group. Carcinogenesis and pathological expression of the esophageal carcinoma tissues were observed. Results The expressions of LIGHT receptors were detected in Eca109 cells. The proliferation of Eca109 cells was inhibited after trasfecting LIGHT-Fc gene into Eca109 cells. The numbers of tumors generated in Eea109/Wt group, Eca109/neo group and Eca109/LIGHT group were 12, 11 and 5, with statistical significance between Eca109/LIGHT group and the other two groups (X2 =6.652, 4.821, P <0.05). The result of histopatholagical examination indicated that the tissue necrosis appeared significantly in tumors derived from Eea109/LIGHT cells. Conclusions The growth of esophageal squamous carcinoma cell line Eca109 can be suppressed by LIGHT-Fc gene whether in vitro or in vivo.
5.Effects of ethyl gallate on sepsis-induced acute lung injury in rats

Objective To evaluate the effects of ethyl gallate (EG ) on sepsis-induced acute lung injury (ALI) in rats .Methods Forty-eight healthy male Wistar rats ,aged 10-12 weeks ,weighing 150-250 g ,were randomly divided into 3 groups (n=16 each) using a random number table :control group (group C) ,group ALI , and group EG .The animals were anesthetized with intraperitoneal pentobarbital sodium 50 mg/kg .Sepsis was induced by cecal ligation and puncture (CLP) .EG 1 ml/h (50 mg·kg-1 ·h-1 ) was infused intravenously via the femoral vein for 4 h starting from 6 h after CLP in group EG .The equal volume of normal saline was given instead in group ALI . The rats were sacrificed at 10 and 24 h after CLP (T1 ,2 ) , and the lungs were removed for determination of wet/dry lung weight ratio (W/D ratio ) , superoxide dismutase (SOD ) activity , contents of malondialdehyde (MDA ) and glutathione (GSH ) , and 3-NT expression (by immuno-histochemistry ) in lung tissues .The pathological changes of the lung were examined with light microscope .Results Compared with group C ,the W/D ratio ,MDA content and 3-NT expression were significantly increased and GSH content was decreased at T1 ,2 ,the SOD activity was significantly decreased at T2 in group ALI ,and the MDA content and 3-NT expression were increased at T1 ,and the W/D ratio ,MDA content and 3-NT expression were increased and GSH content was decreased at T2 in group EG ( P<0.05) .Compared with group ALI ,the MDA content was significantly decreased at T1 ,and the W/D ratio ,MDA content and 3-NT expression were decreased ,and GSH content and SOD activity were increased at T2 in group EG ( P<0.05) .The pathological changes of the lung were obvious in group ALI , and significantly attenuated in group EG .Conclusion EG can attenuate sepsis-induced ALI by antioxidative effects in rats .
6.Drug-resistant Gene of Meticillin-resistant Coagulase Negative Staphylococcus in Children Septicemia

OBJECTIVE To study the distribution of drug-resistant gene of meticillin-resistant coagulase negative Staphylococcus(MRCNS) in children septicemia.METHODS The total MRCNS isolates were 40,and whether in there harbored genes mec,erm and qac was studied.RESULTS Among 40 CNS strains,in there harbored genes mec,erm,and qac were 38(95.0%),30(75.0%) and 18(45.0%),respectively.CONCLUSIONS MRCNS in children septicemia where harbor drug resistance genes is very serious,so we should pay great heed to its effective control.
7.Biotransformation of polyphyllins by Curvularia lunata fermentation

Objective To find the microorganisms which hydrolyze sugar residues of steroidal saponins at C-3 position and to obtain their glucosyl-derivatives.Methods The enzymes secreted by fermentation of Curvularia lunata((3.438 1)) in the culture medium were employed to transform polyphyllin Ⅴ(compound Ⅰ) and polyphyllin Ⅵ(compound Ⅱ).The products were separated by means of chromatography on C_(18) column and their structures were elucidated on the basis of spectral analyses.Results Compounds Ⅰ and Ⅱ could be transformed by C.lunata((3.438 1)) and the main products were identified as diosgenin-3-O-?-D-glucopyranoside,named trillin(compound Ⅲ) and pennogenin-3-O-?-D-glucopyranoside(compound Ⅳ),respectively.Conclusion The terminal rhamnosyls of polyphyllin Ⅴ and polyphyllin Ⅵ at C-3 position could be hydrolyzed selectively by C.lunata((3.438 1)) for the first time.

Objective:To evaluate the clinical effects of Pumpkin Polysaccharide Granules for diabetes Ⅱ.Methods:The clinical effects were evaluated on the governing principle of clinical investigation of diabetes treatment with new Chinese medicinal. In experiment the clinical effects of Pumpkin Polysaccharide Granules (as a treatment group) were compared with that of Xiaoke pill (as a control group).Results:The total effective rate of treatment group was 87.13%, 10 cases of then marked effect, 16 cases effect, 4 cases no improvement. 9.Uniform-design in optimizing conditions of supercritical fluid extraction to Daphne odora Thurrb fresh flowers

Objective: To study the best condition of extracting effective component in Daphne odora Thurrb fresh flower by CO 2 supercritical fluid extraction(SFE). Methods: Uniform design was taken in these 4 factors: extracting pressure, extracting temperature, releasing pressure and releasing temperature, together with the experiments of 7 levels in each factor. Results: The best conditon in SFE: extracting pressure 35MPa, extracting temperature 40 ?C , releasing pressure over 8MPa and releasing temperature 36 ?C . Conclusion: This method is simple, highly selective and efficient in extracting effective component of Daphne odora thurrb fresh flowers.
10.Antibiotic Resistance Changing of Esherichia coli in Third Grade Hospitals of Hubei Province:A Dynamic Investigation

OBJECTIVE To investigate dynamically antibiotic resistance changing of Escherichia coli in Hubei province and provide the reference for clinical rational application of antibiotics.METHODS The WHONET4-5 software was used to analyze the antibiotic resistant rates of 2901 strains of E.coli isolated from all kinds of clinical specimen of children from 2003 to 2006 in the third grade hospitals of Hubei province.RESULTS Drug-resistance rate of E.coli in children group was lower than that in adult group.Except for 100.0% susceptibility to carbopenems such as imipenem and Meropenem,in the four years,drug-resistance of the other antibiotics showed ascending tendency.Ampicillin showed the highest drug-resistance rate(84%-90%).The lower(3%-25%) were amikacin,cefoxitin,ceftazidime,cefepime.CONCLUSIONS Antibiotic resistance of E.coli in children in Hubei province showed the ascending tendency.We should take effective measurement to control the spread and prevalence of the drug-resistant bacteria.
