1.Combination therapy of periodontal intrabony defects with demineralized freeze-dried bone powder and platelet-rich plasma

Objective:To evaluate the effect of decalcified freeze-dried bone allograft(DFDBA) and DFDBA with platelet-rich plasma(PRP) in the treatment of periodontal infrabony defects.Methods: Fifteen periodontal infrabony defects(30 sites) in 12 patients with periodontitis(9 patients with chronic periodontitis and 3 patients with aggressive periodontitis) were selected.Three months after initial therapy,they were assigned to either the DFDBA group(10 defects with 20 sites) or the DFDBA with PRP group(5 defects with 10 sites).The patients were evaluated for plaque index(PI),bleeding index(BI),probing depths(PD) and clinical attachment loss(CAL) before the treatment and at 6 months after periodontal bone graft surgery.Results: The PI,BI,PD and CAL in the DFDBA group were 1.7,2.7,6.0 mm and 7.0 mm at baseline;1.5,1.8,3.9 mm and 4.4 mm at the 6 months after periodontal surgery.The PD,CAL and BI in DFDBA with PRP group at baseline were 6.2 mm,7.1 mm and 2.9;3.2 mm,3.6 mm and 1.7 at the 6 months after periodontal surgery.The PI did not change significantly after surgery in both groups.While both groups showed significant improvement(P

AIM: To explore the kinetics and thermodynamics parameters in the extraction of flavonoids from Fagopyrum tartarjcum . METHODS: Rutin was adopted as marker substance to create a regression equation between Rutin concentration and absorbance by meaus of UV. RESULTS: According to Fick diffusion principle Ⅱ, extraction process was analysed to gain parameters, such as K, Ea, Y, t 1/2 , K ?. CONCLUSION: The method would be useful for the process design and the choice of operation condition to extract flavonoids from Fagopyrum tartarjcum.
4.Study on application of massive blood transfusion program in massive hemorrhage and early stage of coagulopathy

Objective To study the effect of massive blood transfusion program(MTP) in early massive blood transfusion and preventing coagulopathy.Methods Fifty cases of massive blood transfusion were clinically collected and performed the statistical analysis and processing before and after intervention according to the blood transfusion scheme of MTP.The statistical comparison was performed by using the data before and after intervention.Results PLT was decreased with the increase of transfusing of RBC.After transfusing 15 U of RBC,the PT test,INR and APTT were increased significant(P<0.05).After transfusing 20 U of RBC,the Fib was decreased significant(P<0.05).The MTP had statistical difference between before and after intervention(P<0.01).Conclusion The MTP application has an important significance to prevent the occurrence of coagulopathy and evaluate the coagulation status.
5.Influence of intense pulsed light on the secretion of TGF-β1 in cultured human fibroblasts and intervention of JNK inhibitor

Objective To determine the influence of intense pulsed light (IPL) on the secretion of TGF-β1 in cultured human fibroblasts and the intervention of JNK inhibitor.Methods The callan foreskin fibroblasts were cultured and divided into 2 groups. In the IPL treatment group, cells were irradiated with IPL with fluences of0 (negative control), 10, 18, 27, 36, and 36 J/cm2 × 2 (irradiated with IPL with fluences of 36 J/cm2 twice). In the IPL + inhibitor group, cells were irradiated with IPL with fluences of 36 J/cm2 after incubation with the inhibitor SP600125 for 2 h. TGF-β1 in the culture supernatant was evaluated 48 h after the irradiation using enzyme-linked immunosorbent assay. Results Compared with the negative control, TGF-β1 in the culture supernatant decreased at the IPL irradiation of 10, 18, 27, and 36 J / cm2, whereas TGF-β1 increased at the IPL irradiation of 36 J/cm2× 2. In the IPL + inhibitor group, the concentration of TGF-β1 in the culture supernatant decreased compared with the controls (P<0.05). Conclusion IPL can suppress the secretion of TGF-β1 at the lower fluence and promote the secretion at a higher fluence. JNK inhibitor may play an inhibitive role when IPL regulates the TGF-β1 secretion in cultured human fibroblasts. IPL may stimulate TGF-β1 secretion of the fibroblast cells in human skin via JNK signal pathway.
6.Clinical efficacy of Paroxetine combined with mid-frequency electrical pulse acupoint stimulation for premature ejaculation.

OBJECTIVETo investigate the clinical value of Paroxetine combined with mid-frequency electrical pulse acupoint stimulation (EPAS) in the treatment of premature ejaculation (PE).
METHODSTotally 69 PE patients were equally assigned to receive oral Paroxetine 20 mg/d, mid-frequency EPAS, or oral Paroxetine 10 mg/d combined with mid-frequency EPAS (P + EPAS) , all for 8 weeks. We obtained the intravaginal ejaculation latency time (IELT) and Chinese Index of Premature Ejaculation (CIPE-5) scores of the patients before and after treatment, and compared adverse reactions among the three groups of patients.
RESULTSOne patient of the Paroxetine group gave up treatment because of abdominal pain and nausea. Compared with the baseline, the patients in the Paroxetine, EPAS, and P + EPAS groups all showed markedly increased IELT ([0.92 ± 0.11] vs [4.07 ± 0.11] min, P < 0.01; [0.92 ± 0.12] VS [2.78 ± 0.17] min P < 0.05; [0.91 ± 0.09] vs [5.31 ± 0.13], P < 0.01) and decreased CIPE-5 scores (12.5 ± 3.0 vs 22.0 ± 2.1, P < 0.01; 12.8 ± 2.9 vs 19.5 ± 1.9, P > 0.05; 13.1 ± 2.8 vs 25.2 ± 2.1, P 0.01), with statistically significant differences between the P + EPAS group and the other two (P < 0.05). The total effectiveness rate was 95.7% in the P + EPAS group, remarkably higher than in the Paroxetine (72.7%, P < 0.05) and the EPAS group (47.8, P < 0.01).
CONCLUSIONOral Paroxetine combined with mid-frequency EPAS has a higher safety and efficacy than either Paroxetine or EPAS alone in the treatment of PE.

8.Study on the SFE condition for curcumin in Curcuma longa.

OBJECTIVETo optimize the conditions of supercritical fluid extraction (SFE) for curcumin in Curcuma longa.
METHODOptimum extraction conditions were studied by orthogonal tests. The extracts were analyzed by HPLC.
RESULTThe optimal extraction conditions were pressure 25 MPa, temperature 55 degrees C, static time 4 h, dynamic time 5 h, flow rate of CO2 3.5 L x min(-1), co-solvent ethanol 30% (mL x g(-1)).
CONCLUSIONIt is feasible to extract curcumin by SFE.

9.Serotonin excites arcuate neurons directly but inhibits them through intercalated GABAergic neurons.

Effects of serotonin (5-HT) on spontaneous discharges of single hypothalamic arcuate neurons were observed in rat brain slices by extracellular recordings. The results showed that (1) of 149 neurons selected randomly and tested for 5-HT application, 33 (22.2%) were excited, 82 (55.0%) were inhibited, and 34 (22.8%) showed biphasic responses or failed to respond; (2) substitution of low Ca(2+)-high Mg(2+) artificial cerebrospinal fluid (aCSF) for normal aCSF abolished the 5-HT-induced inhibitory effect but failed to affect the 5-HT-induced excitatory effect; (3) cyproheptadine, a non-selective 5-HT receptor antagonist, could block either the 5-HT-induced excitatory or inhibitory effects in all neurons tested; and (4) bicuculline, a GABA(A)-receptor antagonist, blocked the 5-HT-induced inhibitory effect. These results imply (1) 5-HT excites arcuate neurons through a mechanism that is insensitive to the decreased extracellular Ca(2+), suggesting a direct postsynaptic action of 5-HT on the 5-HT-receptors located in the membrane of the neurons recorded; and (2) 5-HT might elicit the inhibitory effect through a Ca(2+)-sensitive release of GABA from intercalated GABAergic local neurons that are excited first by 5-HT.
