1.Postconcussional syndrome and the development of its objective assessment techniques

The diagnosis of post-concussion symptoms (PCS) has been a controversial issue because of that its pathogenesis is often involved in both neural damage and psychosocial factors. Recent researches have examined the diagnostic and evaluating value of several available methods or tools, including clinical interview, conventional CT and magnetic resonance imaging (MRI), neuropsychologic testing and neurochemical test. In mild traumatic brain injury (MTBI), there exist diffusive damages to white matter distributed particularly near the gray matter-white matter boundary and in the long white-matter tracts in midline structures. The conventional imaging techniques are unable to detect any positive evidence due to the diffusive nature of these injuries. This paper is to review epidemiologic study, diagnostic criteria and relevant disputes, conventional diagnostic strategy and new MRI techniques, and then evaluate the application and prospective value of them in the diagnosis and rating of PCS and MTBI. Non-invasively quantitative assessments of modern brain imaging technique may be developed to sensitive and promising tools in the diagnosis and forensic evaluation of PCS.
2.New Strategy for anti-HBV therapy: blocking P-8 interaction.

Clinically being applied treatment against chronic hepatitis has three limitations: low response rates, severe adverse effects and a high rate of drug resistance. Hence, novel targets for antiviral therapy need to be developed so as to provide an armory of different strategies. During the replication of hepatitis B virus, the interaction of viral polymerase (P protein, also called P) and epsilonRNA is indispensable for the initiation of reverse transcription via protein priming and the pregenome RNA (pgRNA) packaging. Three strategies are currently developed for blocking P-epsilon interaction: heat shock protein inhibitors, epsilonaptamers and chemical compounds for blocking formation of P-epsilon complex. Previously, our group has for the first time worldwide in vitro screened several aptamers, which are able to interfere with the P-epsilon interaction. A strong inhibition against HBV was observed in vitro and in vivo experiments, respectively. In conclusion, the so far developed chemicals suppressing the P-epsilon interaction may bypass or overcome the viral resistance problems during clinic treatment and represent a highly attractive option for therapeutic intervention.

3.The changes of serum homocysteine, C-reactive protein levels in acute cerebral infarction patients and its clinical significance

Objective To investigate the changes of serum homocysteine ( Hcy),C-reactive protein ( hsCRP) levels in acute cerebral infarction patients and its significance.Methods 64 patients with acute cerebral infarction were selected as research group,and 60 healthy volunteers were selected as control group.Detected the acute cerebral infarction patients after 1d,the first 7d,14d Hcy,hs-CRP,total cholesterol,triglycerides,high density lipoprotein,low density lipoprotein levels and the control group the level of these indicators.Results Of total cholesterol,triglycerides,low density lipoprotein of the study group was higher than those of the control group of high-density lipoprotein(t =3.073,2.874,2.015,all P <0.05);after admission ld,treatment of the first 7d,14d showed no significant difference between the two groups ( t =2.223,2.268,all P < 0.05 ).Conclusion Hcy,hs-CRP and the occurrence of acute cerebralinfarction was closely related to the development,and testing to determine the severity of the disease for the guide had important clinical significance.
4.Preservation of isolated rabbit hearts after normothermia storage with stroma-free bovine hemoglobin solution

Objective To evaluate the protective effects of stroma-free bovine hemoglobin(SFBHB)on preservation of the donor heart.Methods 24 isolated perfused rabbit hearts were equally divided into two groups.In control group,the cardiac arrest were induced with St Thomas Ⅱ solution and repeated every 30 min.the myocardial temperature remained at 40℃.In experimental group,the isolated rabbit hearts were perfused continuously with St Thomas Ⅱ solution containing stroma-free bovine hemoglobin and the myocardial temperature remained at 37℃.The preservation lasted 6 h.Results There were significant differences between the two groups in the follows:(1)The mean values of the left ventricular developed pressure(LVDP)and coronary flow rate(CFR)were higher in experimental group(P<0.001);(2)The ATP level and myocardial oxygen uptake were significantly decreased in control group(P<0.001);(3)The mean values of released creatine phosphate kinase(CPK),lactec dehydrogenase(LDH)and myocardial contents of malondialdehyde(MDA)were higher in control group(P<0.01);(4)More water was present in the myocardial in control group(P<0.01);(5)The myocardial ultrastructural injuries were greater in control group.Conclusion The stroma-free bovine hemoglobin could offer great protective effects on preservation of the isolated rabbit hearts.
5.Effect of hyperbaric oxygen therapy on bone mineral density and biomechanics of new-formed bone in mandibular distraction gap in rabbits

Objective To observe the effect of hyperbaric oxygen therapy (HBOT) on mineral density and biomechanics of new-formed bone in distraction gap.Methods The mandibles were subjected to osteotomy and distractors were placed bilaterly in 30 rabbits,which were divided randomly into 2 groups.After 5 days for latency,the mandibles were distracted at a rate of 0.8 mm/day for 10 days,and then for consolidation.After surgery,group A received HBOT with daily distraction,whereas group B only distracted without HBOT served as control group.The animals in groups A and B were sacrificed 1 wk,2 wk,4 wk of consolidation,respectively.The mandibles were harvested and subjected to examination for new bone formation,and bone mineral density (BMD) and biomechanics of newly formed bone in the distraction gap.Results At one week of consolidation,no significant differences were observed between the BMD of newly formed bone in the two groups (P>0.05);however,at the other time points,BMD of newly formed bone in group A was significantly higher than that in group B (P<0.05).Moreover,at 4 weeks of consolidation,the strength,maximum fragile energy and modulus of elasticity of newly formed bone in group A were significantly higher than those in group B (P<0.05).Conclusions The results suggest that HBOT improves mineral density and strength of new formed bone in distraction gap.
6.Effects of resting pulmonary function and diffusing capacity on exercise capacity in COPD.

Objective To investigate the role of resting pulm on ary function (FEV 1,FVC,VC) and diffusing capacity (D LCO) on exercise limitation in patients with chronic obstructive pulmonary disease (COPD).Methods During the period of 2002-04~12 the results of resting pulmonary function test which were and diffusing capacity test performed on 26 subjects with stable COPD were reviewed.The symptom-limited maximum exercise (S LME) were performed.Dyspnea index (DI) and oxygen consumption (V? O 2/kg) were determined with a metabolic cart during exercise.Borg Scale s (BS) was used for counting degree of exertional dyspnea which appeared during exercise.At the end of SLME,arterial blood gas analysis was performed to determi ne PaO 2max,PaCO 2max and Phmax at that time.Results In stepwise multiple regression analysis,the variables significantly contributing to V?O 2max/kg were D LCO % and FEV 1/VC.Conclusion Lung function and diffusing capacity can be importan t determinants of exercise capacity in COPD.
7.Game Analysis on Muti-stakeholders in the Listing Decision of Drugs

OBJECTIVE: To provide proposal for government on dealing with the relationship between commercial and social property of drugs.METHODS: Multi-stakeholders subjects and their interrelation in the listing decision of drugs were investigated using game model.RESULTS & CONCLUSIONS: Innovation level of pharmaceutical industry (P),profitability of drugs operator (E),other factors affecting the proceeds of drug operators (K),additional gold cost of drugs (C1),additional cost of gold drugs sale (C2) were confirmed to be the key factors which affected the listing decision of drugs by pharmaceutical manufacturers.Governments are suggested to ensure the rational and fair use of drugs in public by policy supports,encouraging drug innovation and make-up on difference of drugs in medical institutions,optimizing benefit components of medical institutions,creating evaluation criteria for "listing value" and "necessity in clinic" of drugs.

9. Application of workflow technology to construct automatic rapid drug testing system

OBJECTIVE: To introduce the implementation method and design ideas to achieve automatic NIR (near-infrared spectroscopy) modeling and comparison function during the construction of "National Rapid Drug Testing Database Network Platform" and investigate and verify the feasibility of constructing the automatic rapid drug testing module using workflow technology. METHODS: Both the scientific data workflow and analysis tool Pipeline Pilot and the basic principle of two quick comparison algorithms of NIR were employed to construct a spectral processing and modeling workflow of drugs. Moreover, validation and comparison were carried out between our method and the instrument workstation software OPUS. RESULTS: The established workflow method was not only consistent with the results of OPUS, but also had the advantages of automation and easinessto use. CONCLUSION: The scientific workflow technology can be used to achieve automatic modeling and comparison function of NIR and easily introduce other chemometrics method in the future.
10.Construction of a general piggyBac transposon inducible cell immortalization vector and verification of its basic properties.

In order to construct generally efficient cell immortalization vector, pTP-hTERT, we modified the traditional piggyBac (PB) transposon using artificial synthesis, PCR and enzyme digestion. The modified vector contained the necessary transposon elements, a PB transposase expression cassette, a co-expression selectable element and a human telomerase reverse transcriptase (hTERT) expression cassette. The co-expression selectable element had two markers, enhanced green fluorescent protein (EGFP) gene and puromycin-resistance (Puro) gene, linked by porcine teschovirus-1 2A peptide (P2A). To validate the functionality of vector elements, we transfected pTP-hTERT into HEK293 cell, selected the positive cell clones and then conducted RT-PCR, Western blotting (WB) and Tail-PCR, methylene blue staining and statistic analysis on selected cells. The results of sequencing and cell culture show that the pTP-hTERT was constructed successfully and the positive cell could be selected by puromycin. The WB results, P2A cutting EGFP and Puro fusion protein with high efficiency, reflected the selectable element worked. The sequencing result of Tail-PCR confirmed the vector integrated into the genome through transposition. The results of methylene blue staining and statistic analysis indicated the clone of positive cells triggered by pTP-hTERT significantly increased (P < 0.01) compared with control group. The construction of pTP-hTERT provides an efficient tool for establishing immortalized cell lines and a demonstration for building other eukaryotic plasmids.
