1.Effect of dexmedetomidine on stress response and inflammatory reaction of patients with hysterectomy

Objective To study the effect of dexmedetomidine on stress response and inflammatory reaction of patients with hysterectomy.Methods 96 patients with hysterectomy who received therapy from October 2014 to October 2016 in Jinhua traditional Chinese medicine hospital were selected.According to random number table,those patients were divided into the observation group(n=48)and the control group(n=48).Two groups were treated with total intravenous anesthesia,the observation group was treated with intravenous infusion of dexmedetomidine1μg·kg-1 before anesthesia,pump is completed within 15 minutes,then continuous injection of 0.5μg·kg-1·h-1 speed to the 30min before the end of the operation; the control group was given the same way of infusion of normal saline.The operation time,blood loss,recovery time,extubation time and length of stay were recorded,and the hanges of mean arterial pressure(MAP),heart rate(HR),hydrogen peroxide(H2O2),malondialdehyde(MDA),total antioxidant status(TAS),tumor necrosis factor(TNF)-α,interferon(IFN)-γ,interleukin(IL)-10 at T0(before anesthesia),T1(at the end of pneumoperitoneum),T2(after the end of pneumoperitoneum 30min),T3(postoperative 24h)were recorded.Results There was no significant difference between the two groups in operation time,blood loss,recovery time,extubation time and length of stay; in the control group,the MAP was significantly reduce at T1,T2 was significantly higher,the HR was significantly reduce at T1(P<0.05); there was no significant change of MAP and HR levels in the observation group at each time point; the H2O2 and MDA in the observation group were lower than that of the control group at T1,T2 and T3,and the TAS was higher than that of the control group(P<0.05); the TNF-α,IFN-γ and IL-10 in the observation group were lower than that of the control group at T1,T2 and T3(P<0.05).Conclusion Intravenous infusion of dexmedetomidine before anesthesia is well for patients with hysterectomy,it'is helpful to maintain the hemodynamic stability,and effectively alleviate the stress response,inflammation reaction,it's worth promoting.
2.Correlation between antitumor activity of berberine and the inhibition of Wnt/?-catenin signal pathway

Aim To investigate the relationship between the antitumor activity of berberine and the inhibition of Wnt/?-catenin signaling pathway. Methods The antitumor activities of berberine and total alkaloids of Coptis were assessed for their ability to inhibit the proliferation and to induce apopotosis of human colon cancer lines. A well-established Tcf-4 responsive reporter was introduced into human colon cancer lines to test whether berberine and total alkaloids inhibited the ?-catenin/Tcf4 activity in these cancer cells.Results The proliferation of HCT116 and SW480 cells was inhibited substantially within the concentration ranges of 5 to 40 mg?L~(1) of berberine. 72 hours after treament both HCT116 and SW480 cells underwent apparent apopotosis as demonstrated by Hoechst 33258 staining. Total alkaloids of Coptis had similar effects to berberine when comparable concentrations of it with berberine were used.Berberine effectively and specifically inhibited the Tcf4 reporter activity, but not the positive control reporter at the same experimental conditions. Conclusion Our findings suggest that berberine may be mainly responsible for the antitumor activity of total alkaloids of Coptis and this effect may be mediated at least in part by its inhibitory effect on the Wnt/?-catenin signaling pathway, which is aberrantly activated in many types of human cancer.
3.Study on the Effects of Total Cost Prospective Payment System on the Consumption of Drugs

OBJECTIVE:To investigate the consumption of drugs after the inplementation of total cost prospective payment sys-tem(PPS),and to provide reference for hospital drug cost control. METHODS:The data of prescriptions collected from 9 hospital during 2007-2014 were divided into group A,B and C according to PPS,and then summarized statistically in respects of increase rate of total consumption sum,consumption sum ratio of major category;the drug cost per time of outpatient department,emergen-cy department and inpatient department were calculated as well as consumption sum ratio of self-paid drugs;the dosage per time of drugs in blood pressure and diabetes prescription were also calculated. RESULTS:The total consumption sum of drug increased slowly after the implementation of total cost PPS;the consumption sum ratio of major category kept stable,while that of anti-infec-tive agent decreased;the emergency drug cost per time achieved a negative growth,and outpatient and inpatient drug cost per time increased slightly. The proportion of self-paid drugs was relatively stable. The dosage per time of drugs in hypertension and diabetes prescriptions was stable,too. CONCLUSIONS:Total cost PPS is useful in controlling the rapid growth of drug costs,and promote the reasonable drug use. The consumption sum of self-paid drugs are well controlled. The increase of drug cost per time in outpa-tient and inpatient should arouse the attention of the relevant departments. In addition,it has no effect on drug dosage for the pa-tients with hypertension and diabetes.
4.Investigation of basic operation training in animal experiment in physiological scientific experiment teaching

5.Establishment and characterization of a healthy donor's ??T cell clone

Objective Establishment and characterization of healthy donor's ??T cell clones.Methods ??T cells were cloned by limiting dilution after positive sorting with 60Co irradiated allogeneic PBMC as feeder cells.Flow cytometry analysis and molecular biology technique were then used to identify ??T cell clones.MTT assay was used to verify their proliferation after incubated with epitope peptides recognized by ??T cells.Results A ??T cell clone had been established.The subtype of this clone was V ?9 V ?2 without expression of CD4 and CD8.Further studies indicated that epitope peptide EP6 could not only specifically bind to ??T cell clone but also trigger the proliferation of ??T cell clone.Conclusion A healthy donor's ??T cell clone was successfully established,which laid a solid foundation for further study on ??T cells.

7.Observations on the Efficacy of Electroacupuncture at Huatuo Jiaji Points plus Acupuncture Point Injection in Treating Postherpetic Neuralgia

Objective To investigate the clinical efficacy of electroacupuncture at Huatuo jiaji points plus acupuncture point injection in treating postherpetic neuralgia.Methods Sixty-two patients with postherpetic neuralgia were randomly allocated to a treatment group of 32 cases and a control group of 30 cases. The treatment group received electroacupuncture at Huatuo jiaji points plus acupuncture point injection and the control group, oral administration of carbamazepine tablets, vitamin B1 and vitamin B12. The pre-treatment and post-treatment clinical symptom score and VAS score were recorded in the two groups and the clinical therapeutic effects were compared between the two groups.Results The total efficacy rate was 93.8% in the treatment group and 66.7% in the control group; there was a statistically significant difference between the two groups (P<0.01). There were statistically significant pre-/post-treatment differences in clinical symptom item scores (pain, pruritus, burning sensation and sleep) in the two groups (P<0.05). There was a statistically significant pre-/post-treatment difference in the VAS score in the two groups (P<0.05). There was a statistically significant difference in pre-/post-treatment VAS score difference value between the treatment and control groups (P<0.05).Conclusion Electroacupuncture at Huatuo jiaji points plus acupuncture point injection is an effective way to treat postherpetic neuralgia.
8.The value of diffusion-weighted magnetic resonance imaging in gross tumor volume definition and tumor recurrence/metastasis prediction in nasopharyngeal carcinoma

Objective To investigate the value of diffusion-weighted magnetic resonance imaging (DWMRI) in gross tumor volume definition and localregional recurrence/distant metastasis prediction in nasopharyngeal carcinoma ( NPC). Methods From Jun. 2008 to Feb. 2011, each of 36 patients with or without Iocalregional recurrence and/or distant metastasis treated by intensity-modulated radiotherapy in our hospital was matched in this study. The difference of apparent diffusion coefficient ( ADC) values between these two groups was compared by t-test method. The conventional MRI and DWMRI before treatment were centrally reviewed to evaluated whether DWMRI would benefit to the definition of gross tumor volume.Results The mean ADC value were (0. 753 ±0. 091) x 10 -3 mm2/s and (0. 793 ±0. 094) x10-3 mm2/s in treatment failure group and control group, respectively ( t = - 1. 79 ,P = 0. 078) . The extension of gross tumor volume were much more outstanding in DWMRI when compared to conventional MRI. DWMRI predicted localregional recurrence more accuracy than conventional MRI in one patient . Conclusions DWMRI is help to definition of gross tumor volume. The ADC value before radiotherapy can not predict the localregional recurrence and/or distant metastasis for nasopharyngeal carcinoma.
9.Clinical value of radionuclinde bone imaging in spinal tuberculosis

[Objective]To evaluate the value of ECT in diagnosing and treatment the spinal tuberculosis.[Method]ECT by injecting 99Tcm-MDP to human body was used to know the developing of focus in spinal tuberculosis and whether other bone and joints tuberculosis were complicating simultaneously.[Result]The developing of 175 patients were positive,including multi-vertebral bodies tuberculosis 146 cases,which were composed of 2 vertebral bodies 78 case,3 vertebral bodies 26 cases,4 vertebral bodies and more 42 cases.Complicating tuberculosis of bones and joints 22 cases,articulatio sacroiliaca 29 cases.[Conclusion]ECT plays an important role in diaognosing the spinal tuberculosis and the tuberculose focus of body can be found and has the guidance for doctors to formulate the therapeutic regimem of spinal tuberculosis.
10.Anti-asthmatic effect inhibition of artesunate on the asthmatic model of guinea-pig

OBJECTIVE:To study the anti-asthmatic effects of artesunate on the durg-induced asthmat.METHOD:Drug-induced asthmatic model of guinea-pig. RESULTS:Using nubilous inhalation or intragastric administration,artesunate significantly inhibited the guinea-pig model asthma in a dose-dependent manner.It's ED50 were (99.95±7.69)mg.L-1(n=4) and (15.64±1.25)mg.kg-1.d-1(n=4) respectively. CONCLUSION:artesunate showed anti-athmatic effect in guinea-pig asthmatic model.
