This study represents the first description of structural variations in the globin beta chains of 19 strains of inbred mice. The trypic pep tide mapping of beta chains of globin from BALB/c, BALB/cA, BALB/cJ, BALBcJ-nu, BALB/nu/ + ,BALB/cyJax,B10,B10A/sui,C57BL/6,C57BL/10snJ,SMMC/B,SMMC/C, C3H/He, CFW, DBA/2,SWI,Swars/nu/+,SSB and 129/sv strains of mice was established. The results showed that the position of peptide spots upon fingerprint mapping of beta chains was obviously different in comparison with each other. The different patterns of variability and polymorphism found in the various mouse species may then have come into being as a consequence of the action of selective forces on the various new beta chains which were created by some exchange processes.

Objective Approach to safe and effective lung protection strategies on ventricular septal defect infants during perioperation. Methods There were 4 groups as blank control group, ambroxol group, methylprednisolone group and ulinastatin group. We tried to find a safe and effective stratege on lung protection by comparing each other on the respiratory function indexes and inflammation biomarkers. Results Postoperation of VSD repair, respiratory function indexes such as Ppeak, oxygenation index inspiratory resistance and mechanical ventilation time were significantly different (P < 0.05) among the 4 groups. The inflammation biomarkers such as WBC,CRP and PCT had no differences the first day(P >0.05),and were statistically significant the third day (P < 0.05).Conclusions Ambroxol, methylprednisolone and ulinastatin all are effective drugs for lung protection.Considering about the clinical curative effect and side effects, ambroxol is more safe and effective relatively.

Objective:To investigate the peripheral mechanism by studying the histological changes of masseter muscles using HE stains and substance P(SP) and protein gene product 9.5(PGP9.5) immunohistochemical stains. Methods: Fifteen male Sprague-Dawley were randomly assigned into occlusal interference group(n=12) and control group(n=3).In occlusal interference group,0.4 mm thick crowns were bonded to the rats'first molar of the maxillary. In the control group, rats were anesthetized and mouths were forced open for about 5 min but restorations were not applied.1,5,10,and 21 d after 0.4 mm occlusal alteration treatment, mechanical pain thresholds of bilateral masseter muscles were quantitatively measured by modified electronic anesthesiometer in control group and occlusal interference group. The rats were euthanized by transcardiac perfusion after deep anesthetization at different time points. The paraffin sections of masseter muscles were made and processed for HE,SP,and PGP9.5 immunohistochemical staining. Results: Decreased head withdrawal threshold to mechanical pressure was detected in masseter muscles on both sides following occlusal interference.Histological stains of masseter muscles presented intact following occlusal interference, and no inflammatory cells were observed in both sides.Intensely stained PGP9.5 was observed at 1 d in occlusal interference groups and maintained until the end of the experiment.SP expression was the most obviously increased at 5 d in both sides and gradually decreased to the level of control. Conclusion: Experimental occlusal interference-induced masticatory muscle pain is associated with peripheral sensitization of nociceptive neurons rather than muscle damage and inflammation.

Objective To explore the inchoate changes in patients with Graves disease (GD) treated with 131I and appraise the therapeutic effect. Methods According to the thyroid size, patients’ age, thyroid 131I intake rate and course of the disease, 303 patients with GD were given individual 131I doses. Six months later, the symptom, signs, and thyroid hormone levels of every case were compared with those recorded before. Results The symptom and the level of thyroid hormones were significantly meliorated after 131I treatment. Six cases became goggle-eyed after treatment. Among the 303 patients, 249(82.18%) fully recovered, 21 (6.93%) partially remitted, 24(7.92%) developed hypothyroidism, and 9(2.97%) showed ineffective. Conclusion 131I treatment for GD with an individual dose can exert a satisfied therapeutic efficacy. It may as serve the first choice for GD treatment.

AIM: To establish human lymphocyte cell line secreting monoclonal antibody against Rhesus(D) antigen and analyse the nucleotide and deduced amino acid sequences of a human monoclonal anti-D Fab fragment. METHODS: By using PCR method, the cDNA of human anti-(Rhesus D) antibody(lgM ?)Fab fragment was amplified from an Epstein- Barr-virus-transformed cell line. Cloning and subsequent sequence analysis of the Fab fragment was performed. The deduced amino acid sequence was compared and analysed with previously published sequences. RESULTS : A band of approximate 700 and 650 base pairs was amplified using lgM heavy chain primers and ? light chain primers, respectively. Sequence analysis indicated that the deduced amino acid sequences was in agreement with the characterization of the amino acid present in the human lg Fab fragment. CONCLUSION: The cloning and sequencing of a human anti-Rhesus (D) antibody Fab fragment cDNA will make benefits for production of recombinant anti-Rhesus (D) antibody and prevention of Rh haemolytic disease in the newborn.

Objective To evaluate the effects of pulmonary recruitment (RM) in severe acute respiratory distress syndrome (ARDS) with different body position and positive end-expiratory pressure (PEEP) mode in stage of mechanical ventilation. Methods From June 2013 to June 2016, 80 cases of ICU hospitalized patients with clinical data were enrolled, and they lay in the prone or supine position (prone position group and supine position group, each of 40 cases). The hemodynamic parameters including heart rate (HR), central venous pressure (CVP) and peripheral arterial pressure, respiratory mechanics index including respiratory rate (RR), respiratory system compliance (Crs) and platform pressure (Pplate), and the changes of blood gas analysis index were monitored to observe the effects of RM in different positions with positive end expiratory pressure increasing method. Results The levels of oxygenation index and arterial partial pressure of oxygen (PaO2) were improved 3, 5 and 7 d after treatment compared with those 1 d after treatment (P<0.05), and they were improved significantly in prone position group than those in supine position group 3 and 5 d after treatment (P<0.05). The levels of RR, Crs and Pplate 15 min, 30 min, 1 h, 6 h, 12 h and 24 h after RM had significant differences compared with those before RM (P<0.01). The levels of HR, CVP, peripheral arterial pressure 15 min, 30 min, 1 h, 6 h, 12 h and 24 h after RM had no significant differences compared with those before RM (P>0.05). Conclusions For patients with severe ARDS, early implementation of prone position ventilation combined with PEEP increasing method re-expansion treatment is safe and reliable. It can effectively improve oxygenation level and help to improve the survival rate.

A system adapted for depot management in hospital based on RFID technology is introduced. With resolution of RFID from PHILIPS and RF transmission from Nordic VLSI Norway, the storage of medical materials and medical equipments can be identified, labeled and statistically controlled accurately and expediently. The system can not only save a mass of work force, but also improve work efficiency.

Objective Study on producing human monoclonal antibodies against Rhesus (D) antigen that was suitable for use as blood group typing reagent. Methods B lymphocytes from a Rh negative woman, which can produce anti D antibodies were transformed by Epstein Barr virus(EBV). Antibody secreting cells were enriched by RhD + group O erythrocytes and cloned by limited dilute method. By using one step emzymatic method on microplates, one thousand normal blood donors with a common Rh phenotype were tested with the supernatant of cell culture medium as well as a polyclonal human anti D and a commercial monoclonal anti D serum. Results Three human B lymphocyte lines secreting monoclonal antibodies to Rh (D) were established. One of them produced lgM antibody. The titer of the monoclonal antibodies was 64~128. Study on 1000 blood donors, the results did not show any discrepancy among the three different anti Rh(D) serum. Conclusion These monoclonal antibodies against D antigen could be used in Rh(D) typing.
