1.Anti-proliferation effect of triptolide-eluting stent in rabbit models with iliac restenosis

Objective To investigate the anti-proliferation effect of triptolide-eluting stent, as well as its effectiveress, dose-effect relationship and safety. Methods Experimental stents were divided into 4 groups with 10 stents each, namely bare stent group, sirolimus-coated stent group, 5.6 ?g triptolide-coated stent group and 56 ?g triptolide-coated stent group. By morphometric and histopathologic analysis,we compared 5.6 ?g and 56 ?g triptolide-eluting stents with bare metal stents and sirolimus-eluting stents 28 days after stenting in rabbit iliac models of restenosis. Results The neointimal area of 5.6 ?g triptolide-eluting stents was similar to that of bare metal stents, but larger than that of sirolimus-eluting stents. The neointimal area of 56 ?g triptolide-eluting stents was smaller than that of bare metal stents and 5.6 ?g triptolide-eluting stents, but was similar to that of sirolimus-eluting stents. In all the groups, proliferation on both edges of the stents was insignificant. No toxic effect had been found in the experimental animals related to triptolide-eluting stent. Conclusion Triptolide-eluting stents may inhibit neointimal proliferation. The effect of inhibiting neointimal proliferation is dose dependent and with no adverse effect. These results suggest that triptolide-eluting stent can prevent restenosis within four weeks.
3.Relationship between tuberculosis and microbiota

Tuberculosis(TB) caused by the Mycobacterium tuberculosis(Mtb) is a worldwide public health threat.Microbiota in body affects human health and is involved in human diseases, and its clinical importance is begi-nning to be understood.In this review, studies on the relationship between the establishment of Mtb infection and microbiota as well as the development and antibiotic treatment of Mtb infection were discussed.Studies have shown that: (1) microbiota influences the establishment of Mtb infection; (2) co-infection of Helicobacter pylori alters susceptibility to Mtb infection and progression of active TB; (3) microbiota influences the progression of TB by regulating the nutritio-nal, metabolic and immune status of the host; (4) susceptibility to reinfection increases in TB patients treated with antibiotics, possibly due to T-cell epitope depletion of common intestinal non-Mtb Mycobacterium, the effects of antibio-tics are long-term in patients; (5) the occurrence of childhood TB is age-related and many factors such as co-infection and vaccine inoculation increase risk.An in-depth study of the relationship between the microbiota and TB will provide a new perspective on the prevention of TB.
4.Training can promote the recovery of gastrointestinal function after radical cystectomy for elderly bladder cancer patients

Objective:To explore the effect of comprehensive gastrointestinal training on the recovery of gastrointestinal function after radical cystectomy (Bricker′s operation).Methods:Seventy-one elderly bladder cancer patients who had undergone Bricker′s operation were randomly divided into a control group of 34 and an observation group of 37. Both groups were given routine treatment, while the observation group was additionally provided with comprehensive gastrointestinal training immediately after the operation. Plasma gastrin was measured before the operation and on the 1st and 3rd day afterward. The return of peristaltic sounds, the exhausting and defecation time, the incidence of intestinal obstruction and the average hospital stay were recorded. On the 7th, 14th, 21st and 28th day after the operation, both groups′ gastrointestinal functioning was scored by using Gastrointestinal Symptom Rating Scale (GSRS).Results:The levels of plasma gastrin in the observation group were significantly higher than before the surgery and significantly higher than those in the control group on the 1st and 3rd day after the surgery, though there had been no significant difference between the two groups before the operation. Compared with the control group, peristaltic sounds returned significantly earlier in the observation group (after 1.22±0.15d), and the group′s exhaust time (1.88±0.22d) and first defecation time (2.95±0.19d) were also better. The incidence of intestinal obstruction (23.53%) was significantly lower in the observation group as well. The observation group recorded lower average GSRS values than the control group throughout the observation and follow-up periods, and their average hospital stay was significantly shorter.Conclusion:Comprehensive gastrointestinal training can effectively promote recovery after radical bladder cystectomy.
5.Effects of Burdock Leaf Extract NBY-4 on Oxidative Damage of Vascular Endothelial Cells

Objective To investigate the function of burdock leaf extract NBY-4 in protecting vascular endothelial cells from oxidative damage.Methods The oxidative damage model of vascular endothelial cells was established with hydrogen peroxide(H2O2).ROS,LDH,SOD,apoptosis rate and the expression of apoptosis were measured;Further test the nuclear transcription factor-κB(NF-κB).Results In the model group,H2O2 increased the apoptosis rate of endothelial cells,increased the ratio of Bax/Bcl-2 and the expression of Caspase-3,promoted the production of ROS and LDH in endothelial cells,and inhibited the level of SOD,resulting in oxidative damage to endothelial cells(P<0.05).NBY-4 could slow down the process of these effects;At the same time,NBY-4 can reduce NF-κB in endothelial cells.Conclusion NBY-4 can inhibit endothelial cell apoptosis and antioxidant damage by NF-κB signaling pathway.
6.Reliability and validity of the Chinese version of Intermittent Catheterization Difficulty Questionnaire

Objective:To translate the Intermittent Catheterization Difficulty Questionnaire (ICDQ) into Chinese, and to test its reliability and validity in neurogenic bladder patients.Methods:After translation, back-translation, cross-cultural debugging, expert consultation and pre-investigation, a Chinese version of ICDQ was formed. A total of 248 patients with neurogenic bladder clean intermittent self-catheterization who were treated and followed up in the First Hospital of Shanxi Medical University in October 2022 were selected as research objects by the convenient sampling method, and the Chinese version of ICDQ was used for investigation. The critical ratio method was used for project analysis. Content validity and structure validity were used to test the validity of the questionnaire. Cronbach's α coefficient, split half reliability coefficient and retest reliability coefficient were used to test the reliability of the questionnaire. A total of 248 questionnaires were sent out in this study, and 238 were effectively collected, with a recovery rate of 95.97% (238/248) .Results:The Cronbach's α coefficient of the Chinese version of ICDQ was 0.857, the split half coefficient was 0.711, and the retest reliability coefficient was 0.954. The content validity index of the Chinese version of ICDQ item level was 0.860 to 1.000, and the content validity index of the scale level was 0.930. Seven common factors were extracted by exploratory factor analysis, and the cumulative variance contribution rate was 77.38%. The results of confirmatory factor analysis showed RMSEA < 0.080, GFI, AGFI, TLI, CFI > 0.800. Conclusions:Through confirmatory factor analysis, the Chinese version of ICDQ shows that the model fitting indicators of the scale meet the corresponding requirements, indicating that the scale has high structural validity and overall model fitting. It can be used as an evaluation tool for intermittent self catheterization difficulties in patients with neurogenic bladder.
7.Crystal structure and optical activity of 2-(3,4-dimethoxyphenyl)-4-oxo-4H-chromen-3-yl acetate

Objective To investigate the structure and optical activity of flavonoid derivative:2-(3,4-dimethoxyphenyl)-4-oxo-4 H-chromen-3-yl acetate(4).Methods The absolute configuration was obtained by X-ray single crystal diffraction.The optical rotation of the compound was determined by the SGW ?R-1 automatic polarimeter.Results X-ray single crystal diffrac-tion showed that compound 4 belongs to the orthorhombic space group P212121with a=7.763(2)?A,b=13.930(4)?A and c=14.906(4),α= β= γ=90.00°,V =1611.91(8)?A3,Z=8.There is no hydrogen bonding between the molecules in this crystal state.The molecules maintain their stable arrangement in space by van der Waals forces.It can be seen from the single-crystal data that there is a 33.9(2)° torsion of the dimethoxy-substituted benzene ring in the compound 4 with respect to the 4 H-chromene skeleton,suggesting that there may exist optical activity.Further experimental results of the optical rotation proved that the compound has a L-rotatory,specific rotation[α]19.1D = -5.077°.Conclusion The results of the single crystal indicated that the benzene ring substituted with dimethoxy group in compound 4 has a 33.9(2)° torsion relative to the 4 H-chromene skeleton,which results in chirality and optical activity.This is the first report that non-flavonoid-glycoside has optical activity.
8.Effect and mechanism of embryonic intervention with Zuogui pill on the glucose tolerance in offsprings of pregnant rats with gestational diabetes mellitus

OBJECTIVE To explore the effect and mechanism of embryonic intervention with Zuogui pill on the glucose tolerance in offsprings of pregnant rats with gestational diabetes mellitus. METHODS Pregnant rats were randomly divided into blank group, model group, positive control group (insulin glargine), Zuogui pill low-, medium- and high-dose groups (4.725, 9.45, 18.9 g/kg). In addition to the blank group, streptozotocin was injected intraperitoneally to establish a gestational diabetes mellitus rat model. From day 6 to day 18 of pregnancy, each group was given relevant medicine and distilled water intragastrically, once a day. After 21 days of birth, the body weight and body length of offsprings were recorded, and the area under the curve (AUC) was calculated through a glucose tolerance test. After 22 days of birth, fasting blood glucose (FBG), fasting insulin (FINS) levels in serum, insulin sensitivity index (ISI), and homeostasis model assessment of insulin resistance (HOMA-IR) were measured, and the morphological structure of pancreatic tissue was observed. The protein spectrum of pancreatic tissue was analyzed by tandem mass tag-based proteomics, and protein and mRNA expression levels of apolipoprotein A1(ApoA1), solute carrier family 27 member 1 (Slc27a1), kininogen 1(Kng1) and sodium/potassium-transporting ATPase subunit alpha 2 (Atp1a2), solute carrier family 7 member 5 (Slc7a5), solute carrier family 3 member 2 (Slc3a2), bile acid-coenzyme A: amino acid N- acyltransferase (Baat), eukaryotic translation initiation factor 2 subunit gamma (Eif2s3) were detected. RESULTS Compared with the model group, the body weight, body length and ISI of offsprings in the positive control group and Zuogui pill medium-dose group were significantly increased (P<0.05), while the glucose tolerance and islet cell proliferation were significantly improved, and the AUC, FBG, FINS and HOMA-IR were significantly decreased (P<0.05). There were 88 potential target proteins for the embryonic intervention of Zuogui pill in offsprings of pregnant rats with gestational diabetes mellitus,involving multiple pathways such as peroxisome proliferator-activated receptor (PPAR), cyclic guanosine monophosphate-protein kinase G (cGMP-PKG), fat digestion and absorption, and bile secretion. The proteins closely related to glucose metabolism and insulin resistance mainly included ApoA1, Slc27a1, Kng1, Atp1a2, Slc7a5, Slc3a2, Baat, and Eif2s3. Among them, compared with the model group, protein and mRNA expressions of Slc7a5, Slc3a2, and Baat in the pancreatic tissues of pregnant rat offsprings in the Zuogui pill medium-dose group were significantly up-regulated (P<0.05); protein and mRNA expressions of ApoA1, Slc27a1, Kng1, Atp1a2 and Eif2s3 were all significantly down-regulated (P<0.05). CONCLUSIONS The intervention of Zuogui pill in the embryonic period on offsprings of pregnant rats with gestational diabetes mellitus can improve their blood glucose levels and pancreatic pathological morphology. The mechanism may be related to the upregulation of the expressions of Slc7a5, Slc3a2, and Baat and the down-regulation of ApoA1, Slc27a1, Kng1, Atp1a2 and Eif2s3 expressions in the PPAR, cGMP-PKG, fat digestion and absorption, and bile secretion pathways.
