OBJECTIVE:
The aim of this retrospective study was to analyze the recovery rate of sudden sensorineural hearing loss in children, and explore the prognostic factors in order to guide the clinical diagnosis and treatment.
METHOD:
A retrospective review was conducted for the prognosis of children with sudden sensorineural hearing loss during the past 5 years (from November 2010 to May 2015) in Chinese PLA General Hospital. This paper have a complete clinical data of 101 patients (113 ears)with sudden hearing loss, ranging from 0 to 18 years old Patients were divided into four groups according to hearing recovery and eight putative prognostic factors were analyzed.
RESULT:
Among 101 patients (113 ears), the ratio of male and female was 60:53. Treatment was initiated from 1 to 183 days after disease onset, with an average of (18.5 ± 22.1) d. Bilateral and unilateral hearing loss were 24 ears and 89 ears, respectively. The proportion of mild hearing loss, moderate hearing loss, severe hearing loss and profound hearing loss were 7.1%, 6.2%, 23.9% and 62.8%, respectively. Vertigo and tinnitus occurred in 54.9% and 77.9% of the patients, respectively. After the treatment, the complete recovery rate was 9.7% and the overall recovery rate was 36.3%. The degree of hearing loss, earlier treatment onset, sex and bilateral involvement were significantly associated with hearing recovery (P < 0.05).
CONCLUSION
Sudden sensorineural hearing loss in children was generally identified as severe and profound hearing loss, but after positive and timely treatment, it can be improved or even cured. The mild hearing loss, earlier treatment onset, unilateral hearing loss and female were positive prognostic factors. The concurrence of tinnitus or vertigo, the results of ABR and DPOAE had no significant influence on prognosis.

To analyze the biological characteristics of Pasteurella multocida in bovine respiratory tract and its prevalence in large-scale cattle farms, bacterial isolation, culture, and morphological observation were conducted on the lungs and liver samples of dead cows suffering from respiratory diseases in Hohhot,Inner Mongolia. The isolated strains were studied through biochemical testing,16S rRNA gene sequencing, specific primer PCR identification, capsule serotyping, pathogenicity testing, virulence gene testing, drug sensitivity testing, and drug resistance gene detection methods. The results showed that six strains of Pasteurella multocida serotype A were isolated and identi-fied from the lungs of diseased and dead cows. After sequencing the 16S rRNA sequence of the bac-teria,it was found that the six strains of Pasteurella multocida had the closest genetic relationship with the Chongqing isolate CQ2(CP033599.1).The results of mouse pathogenicity test and viru-lence gene detection showed that all isolates were pathogenic and carried at least 16 or more related virulence genes such as exbB,nanB,sodC,oma 87,etc.,but no hsf1 and toxA were detected. The results of drug sensitivity tests and resistance gene detection showed that the isolated strains were sensitive to different degrees of antibiotics such as ciprofloxacin, ofloxacin, and cefotaxime. They were resistant to streptomycin, clindamycin, and lincomycin, and resistance genes of str A,strB,and tet(H)were detected. The results indicate that there is a certain correlation between the pathoge-nicity and virulence genes, drug resistance phenotype, and drug resistance genes of Pasteurellamultocida type A in cattle. It is recommended to use quinolones(such as ciprofloxacin)and cepha-losporins(such as cefotaxime)antibacterial drugs in clinical practice, which can provide scientific basis and prevention and control plans for the prevention and treatment of respiratory diseases caused by Pasteurella multocida in cattle farms, and lay a foundation for the epidemiological mo-nitoring of bovine respiratory multocida pasteurellosis.
3.Application of metal-organic frameworks in implant surface modification
Chinese Journal of Tissue Engineering Research 2024;28(5):783-788
BACKGROUND:In recent years,metal-organic frameworks have been widely used in the biomedical field because of their unique properties, and metal-organic frameworks can be applied to implant surface modification for improving their biological properties and the surgery's success rate. OBJECTIVE:To introduce metal-organic frameworks commonly used in implant surface modification and the preparation and action mechanism of surgical implants modified by metal-organic frameworks. METHODS:The articles concerning metal-organic frameworks used in implant surface modification published from 2010 to 2023 were searched on PubMed,Web of Science and CNKI.Finally,64 articles were included for review. RESULTS AND CONCLUSION:(1)Metal-organic frameworks have the properties of high porosity, large specific surface area, and diverse spatial structure. As bioactive material,metal-organic frameworks have attracted much attention in tissue engineering and regenerative medicine.(2)The metal-organic frameworks commonly used in the field of implant surface modification include three types.Zeolitic imidazolate frameworks are commonly used as carriers, carrying various drugs and biomolecules. The MIL series are suitable for drug carriers and nanoenzymes.Bio-metal-organic frameworks have many applications because of their diverse structure and are characterized by excellent biosafety.(3)The preparation of implant materials modified by metal-organic frameworks can be mainly divided into two types:one is to incorporate metal-organic frameworks into pretreated implants;the other is to synthesize metal-organic frameworks in situ on pretreated implants.(4)Metal-organic frameworks on the implant surface could release their components, load drugs, catalyze active reactions,or combine with other materials, giving implants antibacterial and anti-inflammatory characteristics and promoting osseointegration. The preparation method of metal-organic framework coatings needs further improvement, and the metabolism and distribution of metal-organic framework coatings should be deeply explored, and its clinical application should be actively promoted.

Objective:To analyze the differential expression of silent information regulator transcript-1 (SIRT1) in tissues and cells of nasopharyngeal carcinoma (NPC), to explore the effects of SIRT1 on the proliferation and migration of NPC cells, as well as the effects on and mechanisms of lipid metabolism in NPC cells. Methods:Experimental subjects: In this study, tissue specimens were obtained from patients who visited the Department of Otolaryngology and performed nasopharyngeal tissue biopsy in the Affiliated Hospital of Nantong University from 2019 to 2020. Among them, 6 cases were male, 6 cases were female, age range: 27-72 years old, including 7 cases of NPC diagnosed by pathology and 5 cases of normal nasopharyngeal mucosa. Experimental methods and outcome measures: Western Blot and quantitative real time polymerase chain reaction (qRT-PCR) were used to detect the protein and mRNA levels of SIRT1. CNE2 cell line was selected for subsequent experiments. Cell viability and migratory ability were evaluated by CCK8, wound healing and Transwell assays respectively. Animal xenograft tumor model was used to explore the role of SIRT1 inhibitor Ex527 on tumor growth in nude mice. Oil red and Bodipy were used to stain intracellular lipids. For the mechanical investigation, the interactions between SIRT1 and hypoxia inducible factor-1α (HIF-1α) were analyzed by immunoprecipitation (IP) and chromatin immunoprecipitation (ChIP). Finally, statistical analysis was performed by SPSS 26.0 software, P<0.05 was considered statistically significant. Results:The levels of SIRT1 protein (1.005±0.168) and mRNA (5.829±2.395) in NPC tissues were higher than those in normal nasopharyngeal mucosa (0.181±0.042,1.995±1.605). Differences were statistically significant ( t values were 6.438 and 2.759, both P<0.05). The mRNA and protein levels of CNE1, CNE2, 5-8F and 6-10B cell lines were also higher than those in normal nasopharynx epithelial cell line NP69. Besides, overexpression of SIRT1 correlated with the proliferation and migration of NPC cells. The tumorigenesis ability of nude mice in the Ex527 group was lower than that in the control group. The low SIRT1 expression reduced the protein level of the key enzymes of liposynthesis in NPC cells, improved the expression of lipolysis enzymes, while HIF-1α overexpression promoted lipid synthesis enzymes in NPC cells. SIRT1 inhibited HIF-1α transcription by enhancing deacetylation levels. The binding ability of HIF-1α to SIRT1 promoter regions decreased when NPC cells were hypoxic. Conclusions:SIRT1 promotes the proliferation, migration and lipid metabolism of nasopharyngeal carcinoma cells, which might be expected to provide new theoretical basis for prognosis judgment and gene therapy.
