1.Studies on the Genetic Variation of Two Mitochondrial DNA Molecules of Schistosoma japonicum

Objective To study the variation of Schistosoma japonicum through two mitochondrial DNA molecules. Methods Genomic DNA was isolated with kit, and the mitochondrial NADH dehydrogenase 1(ND1) and cytochrome c oxidase I (COI) gene fragments were amplified by polymerase chain reaction(PCR) and sequenced. The gene trees were constructed and the acquired data were analyzed with the help of bioinfotmatics. Results The gene trees showed that the Taiwan isolate and the mainland isolates can be divided in two groups: a group from the hilly region (Yunnan and Sichuan), another group from the lake region (Hunan, Jiangxi and Anhui); isolates from Hubei are at a different position on the gene trees. Conclusion There are variations among the geographic isolates of Schistosoma japonicum in China, nevertheless, they have close kinship.

The overall construction of experimental teaching model center is thus promoted.
3.The influence of IL-10-modified dendritic cells on proliferation and cytotoxicity of lymphocytes in experimental animals

Objective:To investigate the influence of dendritic cells modified by hIL-10 on proliferation and cytotoxicity of lymphocyte in experimental animals.Methods:The modified dendritic cells were injected intraperitoneally into C57BL/6 mice to sensitize it. Peripheral blood mononuclear cells (PBMC) of sensitized or unsensitized C57BL/6 mice act as reactive cells, dendritic cells modified or unmodified by hIL-10 act as stimulation cells. These two type cells co-cultured for 6 days. MTT assay was used to detect lymphocyte proliferation. Lactate dehydrogenase release method was used to examine the cytotoxic activity.Results:The results showed that IL-10 can inhibit lymphocyte proliferation reaction of unsensitized or sensitized C57BL/6 mice. Lymphocyte proliferation induced by modified dendritic cells is significant lower than that by unmodified dendritic cells. Modified dendritic cells can resist the cytotoxic activity of cytotoxic T lymphocyte.Conclusion:Dendritic cells stably expressing IL-10 can obviously decrease lymphocyte proliferation response of allogenic mice and reduce the cytotoxic activity.

Objective To compare the performance of four commercial anti-dsDNA antibody assays,i.e,BioPlex 2200 (BioPlex),Farr radioimmunoassay (Farr),MESACUP DNA-Ⅱ TEST ds [MBL-enzyme linked immunosorbent assay (ELISA)] and Anti-dsDNA-NcX ELISA (IgG) (EURO-ELISA),Antoantibodies Profile Assay Kit (HOB-Chemiluminescent Immunoassay) in disease activity assessment of systemic lupus erythematosus (SLE).Methods SLE patients (n=119) as well as healthy controls (n=200) and disease controls (n=100) were recruited and their serum anti-dsDNA antibodies were detected by BioPlex,Farr,MBL-ELISA,EURO-ELISA,and a standard Crithidia luciliae indirect immunofluorescence test (CLIFT).The consistency between above four methods to CLIFT was analyzed.The correlation of anti-dsDNA antibody level of these four methods to SLE disease activity was assessed.All data analyses were performed with Statistical product and service solutions (SPSS) 16.0 (SPSS.Inc) and GraphPad Prism 4.0.3 (GraphPad).Unless otherwise specified,all data in this study were expressed as mean±standard deviation.Cut-off values of the anti-dsDNA quantification methods were set by the manufacturers.Chi square and kappa coefficients were adopted to assess the agreement determination and correlation analysis between anti-dsDNA level and SLE disease activity (SLEDAI).Receiver-operator characteristic (ROC) curve analysis was used to compare the specificity and sensitivity of the anti-dsDNA assays.Student's t test was adopted for the comparison of anti-dsDNA levels by different methods between SLE and SLE+LN groups.A p value small than 0.05 was considered statistically significant.Results Using cut-off values set by the manufacturers,BioPlex demonstrated the highest overall agreement with CLIFT,while MBL-ELISA and EURO-ELISA showed the highest positive agreement with CLIFT.Disease activity correlation analysis showed that SLEDAI score correlated poorly with anti-dsDNA level in Farr assay,but strongly with the other three assays.Bioplex had a better performance in terms of SLE activity index corelation (r=0.297 6,P=0.001 2).Moreover,anti-dsDNA level differed in SLE patients with renal lupus nephritis in BioPlex assay (P=0.026 8),but not in the other assays.In ROC curve analysis,BioPlex showed the largest area under the curve (AUC) over other assays.Conclusion Bio Plex assay has better sensitivity and specificity than Farr,MBL-ELISA and EURO-ELISA and correlates well with SLE disease activity.

With the rapid development and expansion of the scale of the industry of Chinese materia medica,a large number of by-products in the process industrialization of Chinese materia medica have been produced,among which,the solid by-products of Chinese materia medica have been favoured by researchers due to the fact that they are rich in a large number of proteins,cellulose,hemicellulose,lignin,etc.,which can be used in the preparation of high value-added products.Therefore,the authors elaborated on the research on biochemical conversion,thermochemical conversion,resource oriented chemical components,preparation of biomass fuel,new composite materials and high-efficiency adsorbent of solid by-products in the process of industrialization of Chinese materia medica in recent years,aiming to provide theoretical basis for the comprehensive and high-value utilization of the solid by-products of Chinese materia medica and extension of the industrial chain.
7.Researches on multi-level rehabilitation service system in China: a bibliometrics analysis

ObjectiveTo analyze the trends, cooperation, topics and hotspots of researches about multi-level rehabilitation service system in China. MethodsThe literature on multi-level rehabilitation service system in China was searched and screened in databases of CNKI from 1983 to 2023. The number of the articles was described, and the cooperation, research hotspots and changing trend were analyzed using VOSviewer. ResultsA total of 4 643 articles were included. The number of the articles tended to increase and developed in stages. Nine groups with five or more researchers were found, and seven of them cooperated with each other. The most frequent keywords were community-based rehabilitation (occurrence 1 251 with connection strength 1 780), stroke (occurrence 674 with connection strength 1 126), family rehabilitation (occurrence 412 with connection strength 514), rehabilitation nursing (occurrence 178 with connection strength 240) and quality of life (occurrence 156 with connection strength 311). The researchers initially focused on disability rehabilitation, then focused on community-based rehabilitation and family rehabilitation, and gradually focused on the quality of life, activities of daily living, satisfaction, mental health, negative emotion and healthcare consortium in recent years. ConclusionThe researches about multi-level rehabilitation service system are developing in China, focusing on community-based rehabilitation, stroke, family rehabilitation, rehabilitation nursing and quality of life. The cooperation among scholar groups need to be strengthened. Quality of life, activities of daily living, satisfaction, mental health, negative emotion and healthcare consortium may be the hotspots in the future.

Objective:
To investigate the safety and efficacy of the Weitan Waifu patch on the postsurgical gastroparesis syndrome (PGS) of gastrointestinal cancer.
Methods:
The multi-center, double-blind, randomized controlled trial was conducted with superiority design. Patients with PGS of gastrointestinal cancer diagnosed in 4 AAA hospitals and the abdominal symptom manifested as cold syndrome by Chinese local syndrome differentiation were recruited. These patients were randomly divided into two groups according to 1∶1 proportion. Placebo or Weitan Waifu patch was applied in control group or intervention group, respectively, based on the basic treatments, including nutrition support, gastrointestinal decompression, promoting gastric dynamics medicine.Two acupuncture points (Zhongwan and Shenque) were stuck with placebo in control group or patch in treatment group. The intervention course was 14 days or reached the effective standard.
Results:
From July 15, 2013 to Jun 3, 2015, 128 participants were recruited and 120 eligible cases were included in the full analysis set (FAS), and 60 cases in each group. 88 cases were included in the per-protocol set (PPS), including 45 cases in the treatment group and 43 cases in the control group. In the FAS, the clinical effective rate in the treatment group was 68.3%, significantly superior than 41.7% of the control group (
