2.Impact of different beam set-up methods on quality of intensity modulated radiation therapy in nonsmall cell lung cancer

Objective To investigate whether the change of beam set-up methods will influence the dosimetric quality of intensity modulated radiation therapy (IMRT) for non-small cell lung cancer (NSCLC).Methods Twenty-one stage Ⅰ-Ⅲ NSCLC patients were selected for this study.The technique of step and shoot was used and three different beam set-up methods were chosen for IMRT planning,including IMRT-7 with nine equal-spaced beams angled 0°,51°,102°,153°,204°,255°and 306°; IMRT-5 with five equal-spaced beams angled 0°,72°,144°,216°and 288°; and IMRT-5m which was created from IMRT-7 but excluded 2 fields (51°and 102° were omitted if there was lesion in the right lung,while 255°and 306° were excluded if there was lesion in the left lung).The dose constrains ofnormal lungs for IMRT were set according to V5-V60 of normal lungs obtained from the same patient's actually treated 3D-CRT dose volume histogram.The prescription dose for IMRT started from 65 Gy,and then escalated or decreased step by step by 2 Gy once a time until the best plan was obtained.Results For normal lung dose,IMRT-5m had lower V5-V25 than the other two groups; but there was no significant difference in V30-V40.IMRT-5 was the worst for V45-V60; and mean lung dose was lowest in IMRT-5m.Dose parameters of esophagus and spinal cord,target conformity index,and total monitor units were all similar among difference plans.IMRT-5m had lowest heart V40 compared to the other two groups.For target heterogeneity index,IMRT-5 was higher than IMRT-7,but there were no significant differences among IMRT-5m,IMRT-5 and IMRT-7.Compared to 3D-CRT,the prescription dose could be increased by (5.1 ±4.6) Gy for IMRT-7,(3.1 ±5.3) Gy for IMRT-5,and (5.5 ±4.8)Gy for IMRT-5m.Conclusion Fewer beams and modified beam angles could result in similar,even better plan quality.
3.Inhibition of autophgay enhances resveratrol-induced apoptosis of human chondrosarcoma cells

AIM:Toinvestigatewhetherautophagyisup-regulatedwhenresveratrol(Res)inducesapoptosis in chondrosarcoma , and to study the effects of autophagy inhibitor combined with Res on chondrosarcoma .METHODS:SW1353 cells were divided into 4 groups: control group, Res group, 3-methyladenine (3MA) group, and Res +3MA group.Electron microscopy was used to observe the autophagyosomes in control group and Res group .At the same time, the viability of the cells in the 4 groups was detected by CCK-8 assay.TUNEL staining and Western blotting (for determi-ning the levels of cleaved caspase-3, Bax and Bcl-2) were used to reflect levels of apoptosis in all groups .The expression of autophagy-related proteins Beclin 1, LC3-Ⅱ and p62 was detected by Western blotting .RESULTS: Exposure of the cells to Res resulted in a decrease in cell viability and an increase in the level of apoptosis ( P<0.05 ) .Compared with control group, the level of apoptosis was increased but the autophagy was decreased (P <0.05).Compared with Res group, the cell viability and the level of autophagy were decreased and the level of apoptosis was increased ( P<0.05 ) . CONCLUSION:Resveratrol induces apoptosis and autophagy , and inhibition of autophgay enhances resveratrol-induced apoptosis in chondrosarcoma .
4.Effect of Electrical Stimulation of Vagus Nerve on the Expression of Calcium-Binding Protein S100A8 in Septic Shock Rats

Objective:To establish septic shock rat model and to investigate the effect of electrical stimulation of vagus nerve on the expression level of plasma calcium-binding protein S100A8 in septic shock rats.Methods:Thirty male adult rats were per-formed cecal ligation and puncture (CLP)to establish septic shock model and then randomly divided into six group:Group I, sham electric acupuncture and normal;Group II,electric acupuncture and normal;Group III,sham electric acupuncture and CLP;Group IV,electric acupuncture and CLP;Group V,vagotomy,sham electric acupuncture and CLP;Group VI,vagoto-my,electric acupuncture and CLP.In Group III,IV,V and VI,indwelling catheter was placed in the common carotid artery for monitoring mean arterial blood pressure (MAP)continuously.Plasma calcium-binding protein S100A8 was determined by ELISA.Results:In groups with CLP,MAP gradually decreased,and serum S100A8 level at postoperative 12 h significantly increased (P <0.01).Compared to that in groups with CLP,the decreased extent of MAP reduced in rats treated with electri-cal stimulation (P <0.01).Moreover,the decreased extent of MAP reduced and the level of plasma calcium-binding protein S100A8 significantly decreased in Group VI,compared to that in Group V (P <0.01).Conclusions:The electrical stimulation of vagus nerve can alleviate gradual decrease of MAP which was caused by CLP induced septic shock in rats and reduce the level of plasma calcium-binding protein S100A8,so that it contributes to anti-shock treatment as supplementary role.
5.A successfully treated case of colchicine poisoning with lethal dose

At present, there is no specific antidote for colchicine intoxication, and 0.8 mg/kg is its lethal dose.The prognosis of colchicine intoxication patients is closely related to the dosage, but the individual difference is very great. A 38-year-old man with colchicine poisoning was admitted to the Seventh People's Hospital of Shanghai University of Traditional Chinese Medicine, who had ingested 80 mg colchicine tablets (1.19 mg/kg) orally for 4 hours. He was immediately put on gastric lavage, enema, and catharsis. Continuous blood purification was performed for 34 hours and 22 minutes, with a combination of hemoperfusion (HP) and continuous veno-venous hemofiltration dialysis (CVVHDF). He also received a large dose of the glucocorticoid with 80 mg of methylprednisolone injected intravenously every 8 hours and organ function support. The patient was hospitalized for 2 weeks and discharged with improvement. The successful treatment of this case was reported for reference.
6.Effect of orthodontic traction on the microstructure of dental enamel.

OBJECTIVE:
To investigate the effect of orthodontic traction on the microstructure of dental enamel.
METHODS:
Forty-eight isolated premolars were randomly divided into 6 groups (=8), including Group A (blank control group), in which the teeth were bonded with the orthodontic brackets without any loading force; Groups B1, B2, and B3 where the teeth were bonded with the orthodontic brackets using clinical adhesives and loaded with 50 g force for 6 months, 200 g force for 6 months, and 200 g force for 1 month, respectively; and Groups C1 and C2, where the teeth were bonded with straight wire brackets using light curing bonding and chemical curing bonding techniques, respectively. All the teeth were embedded with non-decalcified epoxy resin. Scanning electron microscope (SEM), atomic force microscope (AFM), and energy spectrometer (EDS) were used to analyze interface morphology and elemental composition of the teeth sliced with a hard tissue microtome.
RESULTS:
Compared with those in Group A, the teeth in the other 5 groups showed increased adhesive residue index with microcracks and void structures on the enamel surface under SEM; AFM revealed microcracks on the enamel surface with angles to the grinding direction. A larger loading force on the bracket resulted in more microcracks on the enamel interface. The interface roughness differed significantly between Groups A and C2, and the peak-to-valley distance differed significantly between Groups A, C, and C2.
CONCLUSIONS
Orthodontic traction can cause changes in the microstructure of normal dental enamel.
