2.Investigation and analysis of brucellosis in human in Shapotou District, Zhongwei City of Ningxia in 2008

Objective To understand brucellosis infective status of Shapotou District, Zhongwei City of Ningxia. Methods The methods of stratified random sampling of epidemiological survey were used among high risk groups in 2008, who underwent erythromycin red plate cohesion test(RBDT) and test-tube agglutination test (SAT), in accordance with the diagnostic criteria for confirming cases of brucellosis(GB 15988-1995). Results Totally an occupational group of 2480 brucellosis in 10 townships were investigated, and 604 sera were tested. Tweenty eight cases were infected, the infective rate was 4.64%(28/604), 15 cases were diagnosed as brucellosis patients, prevalence rate was 2.48% (15/604); cattle attendants had the highest infection rate of 6.45% (26/403). Conclusions Brucellosis epidemic in Shapotou District, Zhongwei City of Ningxia, and it is place related to vocation.
3.Effect of hydrogen-rich saline on neuropathic pain in rats

Objective To evaluate the effect of hydrogen-rich saline on neuropathic pain in rats.Methods One hundred and twenty male Sprague-Dawley rats,weighing 200-250 g,were randomly divided into 4 groups (n =30 each) using a random number table:sham operation group (S group); hydrogen-rich saline group (H group); chronic constrictive injury (CCI) group; CCI + hydrogen-rich saline group (CH group).Neuropathic pain was induced by CCI.The animals were anesthetized with intraperitoneal 10% chloral hydrate 300 mg/kg.The left sciatic nerve was exposed and 4 ligatures were placed on the sciatic nerve at 1 mm intervals.In H and CH groups,hydrogen-rich saline 10 ml/kg was injected intraperitoneally twice a day for 3 consecutive days starting from 1 day after CCI.The paw withdrawal threshold to mechanical stimulation with yon Frey filament (PWT) and paw withdrawal latency to thermal stimulation (PWL) were measured on the day before CCI (T0) and on 3,5 and 7 days after CCI (T1-3).The rats were sacrificed after the last measurement of pain threshold at T4.The left lumbar segments (L4-6) of the spinal cord were removed for determination of the contents of tumor necrosis factor-alpha (TNF-α),interleukin-1β (IL-1β),interleukin-6(IL-6) and high-mobility group box-1 (HMGB1) (by ELISA),and malondialdehyde (MDA) content and superoxide dismutase (SOD) and catalase (CAT) activities (using visible spectrophotometer).Results Compared with S group,PWT was significantly decreased,PWL was shortened,the contents of TNF-α,IL-1β,IL-6,HMGB1 and MDA were increased,and the activities of SOD and CAT were decreased at each time point after CCI in CCI and CH groups.Compared with CCI group,PWT was significantly increased,PWL was prolonged,the contents of TNF-α,IL-1β,IL-6,HMGB1 and MDA were decreased,and the activities of SOD and CAT were increased at all the time points after CCI in group CH.Conclusion Hydrogen-rich saline can alleviate neuropathic pain in rats via reducing the inflammatory response and lipid peroxidation response.
4.The role of membrane complement regulatory protein in transplantation of bone marrow mesenchymal stem cell

Objective To explore the effect of complement activation on bone marrow mesenchymal stem cells (BMSCs)and evaluate the effect after transfection of complement regulatory proteins.Methods Bone marrow aspirate was harvested from 10 cases of patients suffered from fractures.Mesenchymal stem ceils were isolated,indentified cultured and then experimented in vitro.The complement cytotoxicity on the mesenchymal stem cells in autologous serum was measured by Europium cytotoxicity assay.The samples were divided into BMSCs group,BMSCs+ autologous human serum (AHS) group and BMSCs+ inactivated autologous human serum (iAHS) group.The complement membrane attack complex (MAC) deposited on the membranes was detected by flow cytometry.Finally,the cytotoxicity on BMSCs was measured after transfected with membrane complement regulatory proteins (mCRPs).All samples were divided into BMSCs with mCRPs untransfected group and BMSCs with mCRPs transfected group.Results More than 95% of cells derived from bone marrow were identified to be mesenchymal stem cells through detection of cell surface markers by flow cytometry.The cytotoxicity of untreated cells was 0.41%± 1.48%.BMSCs harvested from the 10 patients all had cytotoxicity after incubated with autologous serum,and the cytotoxicity was 32.59%±2.73%,while cytotoxicity after incubated with complement inactivated autologous serum was 2.59%±3.08%,which was similar to control group.Complement attack complex (MAC) could be detected on the BMSCs incubated with autologous serum,which implied the complement activation.After transfection of mCRPs,the cytotoxicity of autologous serum on transfected cells was decreased.The cytotoxicity of untransfected cells (41.70%±4.47%) had significant difference compared to the cells transfected with CD55 (21.87%±2.19%),the cells transfected with CD59 (18.67%± 1.42%),and the cells transfected with CD46+CD55+CD59 (28.43%±2.14%).CD55,CD59 and CD46+CD55 +CD59 transfected groups could impair effectively the cytotoxicity from complement.However,the cytotoxicity impairment was less effective in CD46 transfected cells (39.30%±3.96%),which had no significant difference compared to untransfected cells.Conclusion Membrane complement regulatory proteins could effectively protect bone marrow mesenchymal stem cells from attacks by complement.

6.Biological characteristics of endothelial progenitor cells from the peripheral blood versus umbilical cord blood

BACKGROUND:Endothelial progenitor cells from the peripheral blood and umbilical cord blood are an essential source to repair vascular endothelial cells damaged by various diseases.
OBJECTIVE:To compare the biological characteristics of endothelial progenitor cells from peripheral blood and umbilical cord blood in vitro.
METHODS:Mononuclear cells were isolated from the umbilical cord blood and peripheral blood with density gradient centrifugation method and 6%hydroxyethyl starch precipitation combined with density gradient centrifugation method, respectively. Mononuclear cells were counted. Then the cells were implanted on the culture plate pre-paved with rat tail col agen at the concentration of 1×106/cm2, and cultured in an endothelial cellculture medium for 7 days.
RESULTS AND CONCLUSION:Isolated and cultured endothelial progenitor cells from the peripheral blood and umbilical cord blood had similar morphological characteristics in vitro. Under the optical microscope, with the increase of culturing days, most adherent cells were changed from round to spindle-shaped. Peripheral blood endothelial progenitor cells had cellcolony formation, and spindle cells from the umbilical cord blood arranged typical y in a line structure. After trypan blue staining and drawing of cellgrowth curves, the number of mononuclear cells and endothelial progenitor cells, survival rate and proliferative ability of endothelial progenitor cells from the peripheral blood were al lower than those from the umbilical cord blood (P<0.05). At day 3 after incubation, the proliferation of endothelial progenitor cells from the peripheral blood and umbilical cord blood reached the peak, and then showed a decreased tendency. Flow cytometry and Immunofluorescence staining showed that endothelial progenitor cells from the peripheral blood and umbilical cord blood could express CD34, CD133, and vascular endothelial cellfactor receptor 2. The endothelial progenitor cells from the peripheral blood and umbilical cord blood could swal ow acetylated low density lipoprotein and be combined with ulex europaeus agglutinin Ⅰ. The results confirmed that the endothelial progenitor cells from the peripheral blood and cord blood have similar biologicla characteristics, but the proliferation ability of endothelial progenitor cells from the cord blood is higher.
7.Analysis of distribution characteristics and drug resistance of pathogens isolated from ascites of patients in Children's Hospital Affiliated to Zhengzhou University, 2015-2021

Abstract: Objective To investigate the distribution and drug resistance of pathogenic bacteria separated from ascites of patients in Children’s Hospital Affiliated to Zhengzhou University from 2015 to 2021, and to provide a basis for rational clinical antimicrobial agents. Methods Bacterial culture, bacterial identification and drug sensitivity analysis were performed on 1 058 non-duplicate ascites culture specimens from January 2015 to December 2021. The clinica1 and microbiologica1 data were ana1yzed by WHONET 5.6 and SAS 9.4 Results Of the 1 058 specimens, 586 (55.39%) were positive for pathogenic bacteria, with a total of 781 strains isolated. There was no significant trend of increase or decrease in the positivity rate over different years. Male children (63.99%) were more prevalent than female children. Appendicitis (59.22%) was the most common disease and Escherichia coli was the most common causative bacteria. Among neonates (≤28 d), the bacteria with the highest detection rate were Klebsiella pneumoniae (23.50%) and Enterococcus faecium (23.50%), while among children (>28 d), the highest detection rate was Escherichia coli (35.98%). Gram-negative bacteria accounted for 64.79% of the 781 strains, mainly Escherichia coli (38.28%), Klebsiella pneumoniae (8.58%), and Pseudomonas aeruginosa (5.89%); Gram-positive bacteria accounted for 29.45%, mainly Enterococcus faecium (8.58%), Streptococcus constellatus (2.69%), and Enterococcus avium (2.43%); fungi accounted for 1.66% and anaerobic bacteria accounted for 4.10%. The resistance rates of Escherichia coli to cefoperazone/sulbactam, piperacillin/tazobactam, imipenem and meropenem were 6.02%, 4.35%, 4.35%, and 3.68%, respectively. The resistance rates of Klebsiella pneumoniae to these drugs were 59.70%, 59.70%, 50.75% and 53.73% respectively. Linezolid-resistant strains of Enterococcus faecium were found. Conclusion Appendicitis is the most common abdominal infection in children, and the distribution of ascites pathogens varies with ages and diseases. The pathogenic bacteria are mainly Gram-negative bacteria, and the drug resistance of Klebsiella pneumoniae was more serious. It is particularly important to use antibiotics correctly and rationally to reduce the emergence of drug resistant bacteria.
8.Comparative Study of the Effects of Application at Different Acupoints on the Quality of Life in Allergic Rhinitis Patients Based on Treating

Objective To observe the effects of application at different acupoints in improving the quality of life in allergic rhinitis patients. Method Seventy-seven patients with allergic rhinitis were randomized into a 12-point application group (39 cases) and a 6-point application group (38 cases), to receive Dong Bing Xia Zhi (treating cold-induced diseases in summer) application during each of the three periods of canicular days at corresponding points, 2 h each time. Rhinitis Quality of Life Questionnaire (RQLQ) was evaluated before and after the treatment, and half a year later. Result After the treatment, the total effective rate was 92.3% in the 12-point application group versus 71.1% in the 6-point application group, and the 12-point application group was significantly better (P<0.05); half a year later, the total effective rate was 87.2% in the 12-point application group versus 63.2% in the 6-point application group, and the 12-point group was significantly better (P<0.05); regarding the RQLQ, the 6-point application group didn't gain marked improvement in sleep quality after the treatment and half a year later (P>0.05), but the 12-point application group showed marked improvement (P<0.05); the rest component scores in RQLQ were significantly improved after the treatment and half a year later compared to those before the treatment in both groups (P<0.05); after the treatment, the 12-point application group was superior to the 6-point application group in comparing the reductionof all component scores and general score except the non-nose/eye symptoms score (P<0.05); half a year later, except the non-nose/eye symptoms and behavior scores, the 12-point application group was superior to the 6-point application group in comparing the reduction of all component scores and general score (P<0.05). Conclusion Dong Bing Xia Zhi acupoint application can effectively improve the quality of life in allergic rhinitis patients, especially with acupoints selected based on holism.

10.Comparison of treatment with micro endoscopic discectomy and posterior lumbar interbody fusion using single and double B-Twin expandable spinal spacer.

OBJECTIVETo compare the therapeutic effect of posterior lumbar interbody fusion by single and double B-Twin expandable spinal spacer with micro endoscopic discectomy (MED) for lumbar intervertebral disc protrusion accompanying degenerative instability.
METHODSFrom March 2006 to May 2008, 45 patients with lumbar intervertebral disc protrusion accompanying degenerative instability were admitted and managed with posterior lumbar interbody fusion by B-Twin expandable spinal spacer with MED. The patients were randomly assigned to treatment with single B-Twin (Single group, n = 24) or double B-Twin (Double group, n = 21). There were 16 males and 8 females, with an average age of 45.5 years (43 - 60 years) in Single group; 13 males and 8 females, with an average age of 43.7 years (44-61 years) in Double group. All the cases suffered from only one level disc protrusion, L(3-4) 2 cases, L(4-5) 29 cases and L₅-S₁ 14 cases. Clinical outcomes were evaluated with surgical time, blood loss, visual analogue scale (VAS) scores preoperatively, 1, 3, 6 month postoperatively. Oswestry disability questionnaire (ODI) of the preoperative, 1 month postoperative, and latest follow-up and the disk space heights.
RESULTSForty three patients were followed-up for 1 to 3 years after surgery. The mean surgical time of Double group was longer than Single group [(152 ± 32) min vs. (91 ± 15) min, P < 0.01]. The average blood loss in Double group was more than that in Single group [(146 ± 73) ml vs. (95 ± 58) ml, P < 0.01]. The mean time of hospital stay in Single group was similar to that in Double group [(11.0 ± 3.2) d vs. (10.9 ± 3.3) d, P > 0.05]. Both groups could keep the disk space heights till the last follow-up [(7.7 ± 1.8) mm vs. (8.5 ± 1.7) mm]. In the 6 months follow-up post operation, the VAS score decreased from (8.1 ± 1.8) to (2.0 ± 1.0) in Single group, and (8.1 ± 1.9) to (2.1 ± 1.0) in Double group. At the last follow-up, the ODI decreased from (36 ± 7)% to (10 ± 4)% in Single group and (37 ± 6)% to (9 ± 4)% in Double group, but there was no significant difference between the two groups (P > 0.05). All the cases achieved fusion at the last follow-up, 3 patients in Single group and 2 patients in Double group suffered from intractable low back pain. One of the fins broke in one patient without any uncomfortable feeling.
CONCLUSIONSCompared with the management of lumbar intervertebral disc protrusion accompanying degenerative instability by double B-Twin expandable spinal spacer with micro endoscopic discectomy, the single B-twin can get similar clinical outcomes, but shorter surgical time, less blood loss and less medical costs.
