1.Significance of Specific Allergen Detection in Children with Asthma in Suzhou City

Objective To explore the main inhalational and alimentary allergens which triggers allergic asthma in children in Suzhou area.Methods The serum specific aspiration allergens were detected with enzyme-linked immunosorbent assay(ELISA) in 119 children with bronchial asthma,including 7 kinds of common inhalational allergens.The 6 kinds of alimentary allergens test was conducted in 65 children.Results The total inhalational allergens positive rate of the subject was 75.63%(90/119).The highest rate was dust mite 49.58%(59/119),then was house dust 21.02%(25/119),animal hair 19.33%(23/119).The positive rate of aspiration allergens increased with the patient′s age and showed a positive correlation.Positive rate of food allergens was 49.23%(32/65).Of them,the highest rate was fish 20.0%(13 cases),then was lobster and claw 16.92%(11 cases),egg 12.31%(8 cases).Conclusions Inhalational allergens are the main allergens in children with asthma in Suzhou district.The positive rate of aspiration allergens increases with the patient′s age.The most common allergen is dust mite.The allergen test plays a clinically significant and advisable role in treatment and prognosis in bronchial asthma in children.

4.Experimental occlusal interference induces the expression of protein gene products and substance P in masseter muscles of rats

Objective:To investigate the peripheral mechanism by studying the histological changes of masseter muscles using HE stains and substance P(SP) and protein gene product 9.5(PGP9.5) immunohistochemical stains.Methods: Fifteen male Sprague-Dawley were randomly assigned into occlusal interference group(n=12) and control group(n=3).In occlusal interference group,0.4 mm thick crowns were bonded to the rats'first molar of the maxillary.In the control group,rats were anesthetized and mouths were forced open for about 5 min but restorations were not applied.1,5,10,and 21 d after 0.4 mm occlusal alteration treatment,mechanical pain thresholds of bilateral masseter muscles were quantitatively measured by modified electronic anesthesiometer in control group and occlusal interference group.The rats were euthanized by transcardiac perfusion after deep anesthetization at different time points.The paraffin sections of masseter muscles were made and processed for HE,SP,and PGP9.5 immunohistochemical staining.Results: Decreased head withdrawal threshold to mechanical pressure was detected in masseter muscles on both sides following occlusal interference.Histological stains of masseter muscles presented intact following occlusal interference,and no inflammatory cells were observed in both sides.Intensely stained PGP9.5 was observed at 1 d in occlusal interference groups and maintained until the end of the experiment.SP expression was the most obviously increased at 5 d in both sides and gradually decreased to the level of control.Conclusion: Experimental occlusal interference-induced masticatory muscle pain is associated with peripheral sensitization of nociceptive neurons rather than muscle damage and inflammation.
5.Expression of imprinted gene PEG10 in human gastric adenocarcinoma tissues and significance

Objective To investigate the expression of a novel imprinted gene,PEG10,in human gastric adenocarcinoma,and the effect of PEG10 on cell growth and proliferation of gastric cancer cells.Methods The PEG10 mRNA expressions in 40 human gastric adenocarcinoma,the corresponding adjacent normal tissues and 6 nomal gastric tissues were detected by RT-PCR.The expression vectors of PEG10 were constructed and transfected into gastric cancer cell line MKN45 which had no endogenetic PEG10 expression.Cell growth ability was measured by MTT assay.Results High PEG10 mRNA expression level was detected in 9 of 20(45.0%) human gastric adenocarcinoma which was significantly higher than those of the matched normal tissues(10.0%)(P

Objective To analyze the clinical manifestations and the features of brain MRI and cerebrospinal fluid (CSF) findings in adult Chinese patients with anti-N-methyl-D-aspartate receptor (NMDAR) encephalitis.Methods We reviewed the clinical manifestations,brain MRI and CSF examinations of 29 patients who were diagnosed as anti-NMDAR encephalitis.Results The major clinical features of anti-NMDAR encephalitis patients included psychiatric symptoms (86%,25/29),seizures (83%,24/29),decreased consciousness (55%,16/29),involuntary movements (55%,16/29),central hypoventilation (34%,10/29),and hypersalivation (17%,5/29).Some patients also experienced autonomic instability,hemiplegia and aphasia.Underlying ovarian teratoma was identified in 14% of affected patients(4/29).Brain MRI was found abnormal in up to 62% patients (18/29),located in the temporal lobes,hippocampus,thalamus,brain stem,cingulate gyrus,frontal and parietal cortex,corpus callosum,internal capsule,basal ganglia and periventricular area.CSF findings were abnormal in 83% of patients with anti-NMDAR encephalitis.Oligoclonal banding in CSF was positive in 95% patients (19/20).The recurrence rate during 3 years was 31% (9/29).Conclusions Anti-NMDAR encephalitis is a treatable disease,yet with high recurrence rate.Its predominant clinical features are psychiatric symptoms and seizures,while involuntary movements,central hypoventilation and hypersalivation are its characteristic manifestations.Lesions in MRI are widespread,not only restricted to limbic lobe.
7.Effect of Acanthopanax senticosus injection on learning and memory of brain multi-infract rats

Objective To explore the effect of Acanthopanax senticosus injection (ASI) on brain multi-infract dementia rats. Methods Thirty-six male Wistar rats were randomly divided into 3 groups:control group, model group and ASI group. The multi-infract dementia rat models were set up by injecting mini-sludged blood in carotis internal arteries, learning and memory function of rats were determined by Morris water maze and Step-down test, the section of rat cerebral cortex and hippocampus were stained with haematoxylin eosin (HE). Results Compared with model group, latency in ASI group shortened significantly at 2nd,5th and 6th day, the distance shortened at 1st,2nd,5th and 6th day. The seeking tactics of ASI trgated rats improved in the Morris water maze test. The error times of ASI treated rats decreasd at 1st and 2nd day in Step-down test; ASI did not reduce significantly pathological changes of vascular dementia rats. Conclusion ASI has effect of treatment on multi-infract dementia rats.

9.Characteristics of ureteral stone position and dilatation of ureter in patients before treated with endourologic lithotripsy

Objective: To investigate the position and the size of ureteral stone in patients before treated with ureteroscopic lithotripsy or percutaneous nephrolithotomy (PCNL), as well as the dilatation of ureter caused by stone, which may be helpful for better understanding of pathogenesis of ureteral stone and improvement of the diagnosis and treatment in clinic.Methods: A total of 129 cases with ureteral stone for endourologic lithotripsy in Peking University People''s Hospital from Aug.2016 to Mar.2017 were included for the retrospective review.The CT data of the ureteral stones and the ureteral dilatation were collected, including the position of stone, the transverse and longitudinal diameter of stone, and the transverse diameter of dilated upper ureter and ureteropelvic junction (UPJ).The distribution of ureteral stones and dilatation in different parts of ureter were compared and analyzed.Results: All of the 129 cases of ureteral stone, stones were located at UPJ in 9 cases (7.0%), proximal ureter in 75 (58.0%), ureter crossing external iliac vessel (UEIV) in 6 (4.7%), distal ureter in 26 (20.2%), and ure-terovesical junction (UVJ) in 13 (10.1%).The mean transverse diameter of proximal ureteral stones was greater than that of distal ureteral stones [(8.47±2.36) mm vs.(6.74±1.99) mm, P<0.001], as the same as the mean longitudinal diameter [(11.00±4.41) mm vs.(7.50±4.28) mm, P<0.001].In 114 cases of dilated ureter for stone, the UPJ had a greater transverse diameter compared with the upper ureter [(14.39±6.09) mm vs.(11.45±3.85) mm, P<0.001].Conclusion: The most common location of stone is the proximal ureter in patients for endourologic lithotripsy, as the location in UEIV is rare.Both transverse and longitudinal diameters of stone in proximal ureter are greater than those in distal ureter.For dilated ureter, it is more severe in UPJ than in upper ureter.Traditionally, it is accepted that the stones lodge at 3 sites of natural narrowing in ureter, which may be questioned.
10.Expression of Survivin, RAGE and HMGB1 gene in human breast cancer and its clinical significance

Objective To investigate the expression of Survivin,receptor for advanced glycation endproduct(RAGE) and high mobility group protein B1 (HMGB1) gene in human breast cancer and their clinical significance.Methods The expression of Survivin,RAGE and HMGB1 gene was detected by Real-time PCR technology and fluorescence in situ hybridization (FISH) technology in the following tissue samples:50 early breast cancers,50 advanced breast cancers,and the corresponding adjacent normal mammary tissues.The relationship of their expression and several factors such as differentiation degree,invasion,lymph node metastasis and TNM stage of cancer was explored.Results The positive expression rate of Survivin,RAGE and HMGB1 gene was 62%,73% and 79% detected by Real-time PCR technology,78%,69% and 72% detected by FISH technology in breast cancer tissues.The overexpression of these genes was positively correlated with TNM stage and lymph node metastasis.The expression of Survivin gene was positively correlated with the expression of RAGE and HMGB1.Conclusion Overexpression of Survivin,RAGE and HMGB1 gene is of great significance in early diagnosis and prognosis of human breast cancer.
