BACKGROUND: The selection of carrier plays an essential role in the research of applying tissue-engineering to fix the peripheral nerves. An ideal carrier would be one that is similar to extracellular matrix and that it has biocompatibility with in vivo cells.OBJECTIVE: To evaluate the biocompatibility of hippocampal neural stem cells with collagen and gelatin sponge in vitro and to probe into the feasibility of using the materials as biomaterial scaffold in peripheral nerve tissue engineering.DESIGN: A randomized and controlled trial.SETTING: Department of Otorhinolaryngology of the First Affiliated Hospital of Zhengzhou University; Department of Anatomy of the School of Basic Medical Sciences of Zhengzhou University.MATERIALS: The experiment was carried out from July to December 2005 at the Laboratory of Neurobiology of the Department of Anatomy of the School College of Basic Medical Sciences of Zhengzhou University.Twelve New born (＜ 24 hours) clean grade guinea pigs of either gender with a body mass of 50-70 g, were provided by the Experimental Animals Center of Zhengzhou University School of Medicine.METHODS: The new born (＜ 24 hours) guinea pigs were anesthetized intraperitoneally with 10 g/L chloral hydrate and sterilized in 0.75 volume fraction of alcohol. Hippocampal tissue was resected from the brain under a surgical microscope. Hippocampus neural stem cells were cultured in vitro. The cultured cells of two generations were suspended at a density of 1 ×1010L-1 and respectively combined with collagen and gelatin sponge.The number of cells was counted and histological changes were observed under an inverted phase microscope and scanning electron microscope after 7 days, and the adhesion rate of the two materials to the cells were measured.MAIN OUTCOME MEASURES: ①Growth of the neural stem cells and their adhesion to the collagen and gelatin sponge were observed and the total number of the cells and adhesion rate with carrier were measured. ②The morphological changes of differentiation cells by immunocytochemical staining.RESULTS: ①Hippocampus neural stem cells could grew on the collagen and gelatin sponge and attached to them gradually. The adsorption rate of collagen was higher than thai of gelatin sponge (37.17 % and 14.87 %,x2=4.819,P ＜ 0.05). ② There was no significant difference in the total number of the cells in the control group, collagen group and gelatin sponge group [(53.17±3.5)×104,(53.25±2.6)×104, (52.04±4.05)×104,F=0.233,P ＞ 0.05].CONCLUSION: Two biomaterials (collagen and glutin sponge), especially collagen, have good biocompatibility with hippocampus neural stem cells from the guinea pigs and can be used safely as scaffold materials in peripheral nerve tissue engineering.

Objective To approach the effect of prosthetic valve at preventing reflux esophagitis.Methods After the total gastrectomy and esophagojejunostomy,interrupted suture was apphed to form a prosthetic valve at output and input beyond Braun's stoma.Results Only one got light reflux esophagitis in 15 patients.Conclusion Prosthetic valve can prevent reflux esophagitis,and the operation is convenient and safe,which is deserved to be in generalization and approach the latest method.

Objective To determine the level of defensins(HNP_(1-3))and interleukin-8(IL-8)in colon tissue of patients with ulcerative colitis(UC)and to explore the relationship between HNP_(1-3)and IL-8 and the severity of disease.Methods The level of HNP_(1-3)and IL-8 in colon mucosa of UC patients was detected by immunohistochemistry method.Results The levels of HNP_(1-3)and IL-8 in colon mucosa of UC patients were much higher than that of normal controls.The expression of HNP_(1-3)and IL 8 of UC patients was higher in involved mueosa than in corresponding normal mucosa.The expression of HNP_(1-3) and IL-8 was related to the extent of lesion and the degree of disease.The expression of HNP_(1-3)was significantly correlated with that of IL-8(r_s＝0.957,P＜0.01).Conclusions The induction of HNP_(1-3) and IL-8 was involved in process of inflammation and development of UC.Regulating mechanisms of HNP_(1-3)expression might be related with the induction of IL-8.

As a member of growth arrest and DNA damage inducible gene family,GADD45αparticipats in the regulation of cell cycle,cell senescence,cell survival and apoptosis. GADD45αplays a critical role in the responses to cell injury induced by a variety of factors including cell stress and genotoxic chemicals. Different transcription factors and proteins are involved in transcriptional regulation of GADD45αgene. GADD45αprotein has been implicated in the regulation of genomic stability related cellular responses through interaction with other proteins. Genotoxicity test systems based on the char?acteristics of GADD45α in regulation of cell function,can be applied to the detection of potentially genotoxic compounds,which provides new ideas and methods about genotoxicity assessment. The molecular mechanism and research progress of GADD45α in genotoxicity test are summarized in this article.

Objective To characterize social-perceptual component and social-cognitive component of theory of mind(TOM) in patients with Parkinson's disease.Methods A total of 19 patients with idiopathic PD and 20 demographic-matched health subjects were included in this study.They were assessed by using Reading the Mind in the Eyes and Faux Pas task involving ToM socialperceptual component and social-cognitive component.The severity of PD was assessed by the Hoehn and Yahr scale.Results Significant difference was found between PD group and health controls in task involving ToM social-perceptual component (mind Reading:17.1 ± 3.1 vs.30.4 ± 1.7,F =278.756,P＜ 0.001),test involving social-cognitive component (Faux Pas Questions:29.0 ± 4.0 vs.41.2±4.4,F=81.647,P＜0.001).While the ability to identify sex and control questions of the Faux Pas showed no significant difference as compared to health controls group (F=3.508,P=0.069;F=1.063,P =0.309).Conclusions The ToM social-perceptual component and social-cognitive component are both impaired in patients with Parkinson's disease.

ObjectiveTo investigate the expression and the clinical significance of diacylglycerol kinase α (DGKα) and protein kinase C (PKC) in human hepatocellular carcinoma (HCC).MethodsDGKα and PKC expressions in the samples from 60 pathologically confirmed HCC patients were analyzed by immunohistochemistry. The relationship between DGKo expression and clinical pathology factors was analyzed.ResultsThe expression positive rates of DGKo and PKC were highest in normal liver tissues [90.0％ (9/10) and 100.0％ (10/10)].The positive rates were 81.7 ％ (49/60) and 71.7 ％ (43/60) in HCC tissues,respectively,and were 58.3 ％ (35/60) and 61.7 ％ (37/60) in carcinoma adjacent tissues,respectively.In three liver tissues,the positive rates of DGKα and PKC were significantly different (P ＜0.05).The location of both kinases in hepatocytes translocated from cytoplasm/nucleus to membrane.The expressions of DGKα and PKC were positively correlation(r =0.495, P ＜ 0.05), The DGKα expression was correlated to differentiation type,portal venous tumor thrombus and TNM staging(all P ＜ 0.05).ConclusionDGKa is expressed with high activity in advanced stage and poorly differentiated HCC. It may be promote the pathological process of HCC.

Objective To investigate the characteristics of the executive function and working memory in patients with mild cognitive impairment (MCI), and the relations between working memory and executive function and the effects of working memory on patient' s daily functioning.Methods Thirty patients with MCI and 30 healthy elderly subjects were tested with a wide neuropsychoingical battery of tests including those of executive function, working memory and other cognition domain tests.Executive function was evaluated by using the verbal fluency test (VFT), color trail test (CTT), digital span (DS).Results The results showed the presence of significant executive function and working memory impairment in MCI patients as compared with the healthy controls.The scores of executive function test in MCI patients were decreased significantly in CTT (130.8±58.2 vs 52.0±13.5), CTT-B (210.2±81.8 vs 121.0±33.4),DS (3.4±0.9 vs 4.2±1.1), VFT (8.9±5.4 vs 16.4±5.4) and visuo-objective working memory (0.73±0.12 vs 0.85±9.18) respectively (t = 7.108, 5.159,-2.544,-4.879, and-4.351, all P <0.01 or P < 0.05) .In the stepwise liner regression analysis objective working memory test made a significant independent contribution to ADL (β =-0.720, t =-3.571, P < 0.01).Conclusion The patients with MCI have the deficit in executive function and working memory.Visuo-objective working memory is closely correlated with general cognitive function such as daily function, resulting in daily function decline.
