2.Study on Electrochemical Behaviors and Determination of Xanthine and Uric Acid on Modified Glassy Carbon Electrode

The silver doped poly ( L-lysine ) modified glassy carbon electrode was fabricated by cyclic voltammetry, the surface of the electrode was characterized by scanning electron microscopy and electrochemical impedance spectroscopy. The electrochemical behaviors and the simultaneous detection of xanthine and uric acid were studied by cyclic voltammetry and differential pulse voltammetry. The results indicated that this modified electrode exhibited excellent electrocatalytic activity towards the oxidation of xanthine and uric acid. The stable oxidation peaks of xanthine and uric acid appeared with the peak potential of 0. 980V and 0. 600V respectively at the modified electrode in pH 3. 0 phosphate buffer solution. The oxidation peaks of xanthine and uric acid were separated at 380 mV. Under the optimum conditions, the linear ranges for the determination of xanthine and uric acid were 1 . 00 × 10-6-2 . 50 × 10-4 mol/L respectively by differential pulse voltammetry. The detection limits were 5. 0×10-7mol/L. The method has been applied to the simultaneous detection of xanthine and uric acid in healthy human urine with satisfactory results.
3.Clinical characters of different degrees nonalcoholic fatty liver and the relationships with blood uric acid

Objective To analyze the clinical characters of the different degrees nonalcoholic fatty liver (NAFLD) and the relationships with blood uric acid.Methods Two hundred and eighty-two cases body examinational and hospitalized patients were selected.According to whether complicated with NAFLD and liver steatosis degree,patients were divided into 4 Groups.Group A was control group that without NAFLD,Group B was mild NAFLD group,Group C was moderate NAFLD group and Group D was severe NAFLD group.Clinical data,blood lipids,blood uric acid and other biochemical indicators of four groups were compared.Results In 282 cases of patients,there were 78 cases (27.66%) without NAFLD patients,94 cases (33.33%) mild NAFLD,92 cases (32.62%) moderate NAFLD and 18 cases (6.38%) severe NAFLD.As the severity of fatty liver increased,body mass index (BMI) higher and higher ((24.51 ± 2.55),(27.06± 2.81),(28.37± 3.23),(30.71 ± 2.96) kg/m2),and the differences among four groups were statistically significant (F =35.976,P <0.05).Gglycerin trilaurate (TG),low density lipoprotein cholesterol (LDL-C) of D group were hihger than other groups ((3.04± 1.22) mmol/L vs.(1.73± 1.46),(2.59 ± 1.78),(2.81 ± 1.94) mmol/L,(2.99± 0.52) mmol/L vs.(2.28±0.78),(2.47±0.75),(2.41±0.74) mmol/L),high density lipoprotein cholesterol (HDL-C) was lower compared with group A,B,C (1.00± 0.23) mmol/L vs.(1.15± 0.24),(1.05± 0.21),(1.00 ±0.21) mmol/L),the differences were statistically significant(F=6.818,4.597,6.414;P<0.05).In 4 groups the blood uric acid(UA) of groups B,C,D was higher than group A,group D was higher than group B,C,and the differences were statistically significant((316.13±71.86),(356.98±83.00),(355.37±81.01),(409.67 ±50.90) μmol/L;F=8.797;P<0.05).Conclnsion Obese patients are more susceptible to NAFLD,and NAFLD with a certain degree of lipid and uric acid metabolism,and the severity of NAFLD has a positive correlations with lipids and serum uric acid levels.It is particularly obvious in patients with severe NAFLD,which prompting that blood uric acid level can be used as an independent risk factor in the pathogenesis of NAFLD,and as an auxiliary judge for NAFLD severity index.
4.Research progress in visualizing single molecule in live cells

Our knowledge of molecular events and interactions in cell biology and molecular biology are mostly based on findings in nonliving body condition while ignoring the whole process of the events,which inevitably involves false assumption that all molecules behave in the same environment and in the same way. With the emergence of new fluorescent molecular probes prepared through linking new fluorescent material such as quantum dots (QDs) to micromolecule antibody such as antibody simulacrum, the development of methods for transducing extrinsic fluorescent probes into live cells,the application of laser technology, the finer fluorescent micro-imaging system, and the fast and sensitive fluorescence collection system in the application of life sciences, it is now possible to image the trajectory, kinetic behavior, and interaction of biomolecules in live cells. Visualizing single molecule in live cells has opened a new chapter for cell biology and molecular biology. Recent research developments and foreground of single molecule visualizing are discussed in this article.
6.Research progress of Ghrelin and obesity,insulin resistance and type 2 diabetes mellitus

Ghrelin is a small active peptide secreted by gastric fundus cell. As an endogenous ligand of growth hormone receptor,it helps to promote growth hormone secretion,increase food intake and participate in energy metabolism. Recently,studies have indicated that Ghrelin,together with other peripheral signals,participate in the regulation of glucose and lipid metabolism. Insulin resistance (IR) is an important pathophysiological basis of the development of type 2 diabetes mellitus (T2DM),while obesity is the common risk factor of both IR and T2DM. It has been confirmed that Ghrelin has a close and complex connection with obesity and T2DM. Further study about Ghrelin may open up a new way to the diagnosis and treatment of obesity and T2DM.
7.Correlation between sensitiviy of neoadjuvant chemotherapy and multidrug-resistance in breast carcinoma

8.The titer of ANCA in relation to disease activity in patients with microscopic potyarteritis

Objective To study whether ANCA liter is sensitive serological marker reflecting vasculitis disease activity. Methods MPO-ANCA and IF-ANCA tilers variation in five sera specimen with active MPA diseases were studied after methylprednisolone and cyclophosphamide pulse therapy. The clinical and pathological features of 5 patients with MPA were investigeted comparing pre-treatment with post-treatment. Results (1) Serum ANCA were found positive and increased liter in 5 patients wilh active MPA disease. (2) Titer decreased after Ireatment, while renal function improved. (3) After half year, MPO-ANCA was negative in 5 patients, IF-ANCA was negative in 2 patients. Conclusion There is a close correlation between serum ANCA titer and MPA renal disease activity.
10.The Effect of Exercise Training on Plasma TNF-? and IL-1? in Rats after Mental Stress

Objective:To explore the effect of exercise training on plasma TNF-? and IL-?concentration in rats after mental stress.Methods:SD rats were conducted on exercise training with different load for 8 weeks and exerted on mental stress for 2 weeks in the later stage of(raining,TNT-? and IL-? concentration were measured. Results:①TNF-? and IL-1 ? concentration in mental stress was significantly lower than that in control group.②Af- ter 8 weeks training,there was a tendency towards rising in IL-1 ? concentration in 30min training group,but not re- markable.IL-1 ? concentration in 60rain training group were significantly higher than that in control group,but TNF-? concentration were not changed obviously,TNF-?and IL-1? concentration in stress and 30min training group and in stress and 60min training group were significantly higher than that in mental stress group.Concluslon:It indicated that exercise training might increase the release of endogenous TNF-? and IL-1? and maintain the stabi- lization of immune function under mental stress state.
