OBJECTIVE:To observe the clinical efficacy of Xuebijing injection in the treatment of severe acute pancreatitis(SAP).METHODS:65 patients with severe acute pancreatitis were enrolled:30 patients in the control group were randomly as-signed to receive standard combined therapy of west medicines,and 35 in the treatment group to receive Xuebijing injection 100 mL qd by iv gtt plus the combined therapy as in the control group.The course of treatment in both groups was 7 days.RESUL-TS:The total efficiency in the treatment group was 94.3% and that in the control group was 83.4%,showing significant differences between the two groups(P
2.Effect of the expression level of long-chain non-coding RNA MEG3 and DNA demethylase TET2 on the aggressiveness of pituitary growth hormone adenoma

Objective:To investigate the role of long-chain non-coding RNA MEG3 and DNA demethylase TET2 in the invasive growth of pituitary growth hormone (GH) adenomas, and provide research directions for the subsequent exploration of molecular biological mechanisms.Methods:All of 60 patients with GH pituitary adenomas diagnosed in Dalian Central Hospital from January 2016 to November 2019 were collected, and the normal anterior pituitary tissue samples of 10 patients with traumatic brain injury without neurological or endocrine system diseases were collected as control group. Real-time reverse transcription-polymerase chain reaction (qPCR) was used to detect the expression of lnc RNA MEG3 and TET2 in normal pituitary tissue as well as invasive and non-invasive pituitary growth hormone adenomas, and the differences between each groups were analyzed. At the same time, age and gender were included in the research, and the effects of age and gender on the invasive growth of pituitary GH adenomas were analyzed.Results:The aggressive growth of MEG3 and TET2 was independent of patients′ age and gender. The expression of lncRNA MEG3 in invasive pituitary GH adenoma and non-invasive pituitary GH adenoma was significantly lower than that in normal control brain tissue. Analysis of the differences between the groups found that the expression level of lncRNA MEG3 in normal trauma control pituitary tissues, non-invasive GH adenomas, and invasive GH adenomas were sequentially decreased, and MEG3 expression level was related to the aggressive growth behavior of pituitary GH adenomas ( P<0.05). The expression levels of DNA demethylase TET2 in invasive pituitary GH adenomas and non-invasive pituitary GH adenomas were significantly lower than those in the normal control brain tissue, and the expression levels in the three groups of samples gradually decreased ( P<0.05). It suggested that the expression of lncRNA MEG3 was positively correlated with the expression of DNA demethylase TET2. Conclusions:The low expression of lncRNA MEG3 and DNA demethylase TET2 is closely related to the aggressiveness of pituitary growth hormone adenoma.

The paper uses computer multimedia technology to collect and switch audio and video signals, constructs the remote High Definition ( HD) surgery demonstration system which supports live broadcast, recorded broadcast, rebroadcast and video-on-demand depending on the network communication routing technology.It introduces functions, design scheme and composition of the system and points out that the system can realize HD video transmission and video talkback between the surgery scene and the classroom.Thus, tech-nical level of clinical surgery teaching can be enhanced.
4.Development and application of private cloud storage system

Objective To design a private cloud storage system to guarantee intranet and internet data security and realize data sharing between multi platforms.Methods The idea and solution for building the system were proposed by analyzing the difficulty during hospital informatization.The infrastructure based on the idle computing resources in the existing virtual pool and the application platform by private cloud storage software were involved in the development of the system,and related software and hardware were integrated after adjustment and customization,and then the system came into being with easy operation and high performance-cost ratio.Results The system developed realized freely data storage and sharing,and implemented file encryption to control the applied range.Conclusion The system has the functions of malicious code scanning,cooperative office system and data auto-backup,synchronization,transmission,sharing and etc,which executes intranet and intemet data sharing based on physical isolation.
5.Establishment of a nude mouse model of nasopharyngeal carcinoma lymph node metastasis and screening of the metastasis-related signature genes.

OBJECTIVETo establish a nude mouse model of nasopharyngeal carcinoma (NPC) lymph node metastasis and screen the signature genes associated with the metastasis.
METHODSThe NPC 5-8F-EGFP cells were inoculated into nude mice, from which a 5-8F-LN cell line with lymph node metastasis potential was obtained. The lymphatic metastasis-related signature genes of breast cancer and head and neck squamous cell carcinoma were screened by data mining method.
RESULTSThe NPC cell lines 5-8F and 6-10B showed 307 differentially expressed genes by microarray analysis, from which 20 overlapping genes were identified, and 3 overexpressed genes were found with probable metastasis potential, namely the ADM, IRF1, and CAV1 genes. Quantitative RT-PCR validated the data mining results in the 5-8F-EGFP, 6-10B-EGFP, NP69, and 5-8F-LN cell lines. The 3 NPC cell lines 5-8F-EGFP, 6-10B-EGFP and 5-8F-LN showed significantly higher expressions of IRF1 than NP69 cells (P=0.008, 0.022, and 0.006, respectively. The expression level of CAV1 in 5-8F-EGFP cells was significantly higher than that in 6-10B-EGFP cells (P=0.014), but ADM expression showed no significant difference between the 4 cell lines.
CONCLUSIONSIRF1 may play an important role in the progression of NPC. The overexpression of CAV1 in 5-8F-EGFP cells can be associated with the high metastatic potential of the cells.

6.Finite element analysis on the stress concentration effect of osteons in cortical bone

Objective To construct a three-dimensional (3D) solid model of the cortical bone including osteons,verify the stress concentration effect of osteons,simulate and predict the stress concentration location under fatigue using finite element analysis (FEA).Methods The 3D solid model of the cortical bone including osteons was constructed in Pro/E wildfire 5.0,and local stress and strain distributions in the cortical bone under different axial compression were calculated and analyzed in ANSYS 12.0.Fatigue simulation on the selected locations was conducted to evaluate fatigue status of the model subjected to different fatigue loading intensities.Results Obvious stress concentration at the junction of osteon and the interstitical bone appeared under axial compressive loads,and the percentage of pathological local strain in the cortical bone increased with the axial compression increasing.Fatigue simulation on the selected locations demonstrated that bone fatigue risk during physiological or daily activities was very low,while a high fatigue or fracture risk might occur during high-intensity exercises or training.Conclusions The 3 D solid model of the cortical bone including osteons is successfully established,the stress concentration effect of osteons is verified,and the location of bone fatigue damage under strenuous exercise and its risk are predicted.These experimental results can provide references for training management and athletic fatigue damage prevention in military recruits and long distance running athletes.
7.Protective effects of Huanglian Jiedu decoction on vascular endothelial function in type 2 diabetic rats.

OBJECTIVETo investigate the effects of Huanglian Jiedu (HLJD) decoction on vascular endothelial function in type 2 diabetic rats and explore the prophylactic and therapeutic significance and pharmacological mechanism of HLJD decoction in type 2 diabetic angiopathic complication.
METHODThe murine type 2 diabetes models were induced by the intravenous injection of a small dose of streptozotocin plus high fat and high caloric laboratory chow. Then modeled diabetic animals were divided into model group, HLJD group, and aspirin group. Normal ratsfed with routine chow were designated as normal group. The oral glucose tolerance test (OGTT) were performed in all animals, 9 weeks after treatment, the changes of murine body weights and levels of fasting blood glucose (FBG), serum total cholesterol (TC), triglycerides (TG), high density lipoprotein-cholesterol(HDL-C), fasting serum insulin (FINS), serum nitric oxide, plasma endothelin, angiotensin II and von Willebrand Factor (vWF) were determined 10 weeks after treatment.
RESULTCompared with model group, the result of OGTT of HLJD group was improved. The levels of the body weights, TC, TG and ET in HLJD group weredecreased (P < 0.05). The levels of FBG,INS, AngII and vWFwere significantly decreased (P < 0.01), and the levels of HDL-C and NO were obviously increased (P < 0.05), as compared with those in model group. Furthermore. The levels of FBG was lower in HLJD group than in aspirin group (P < 0.05), and the improvement of TG, HDL-C,NO, AngII, vWF levels in HLJD group was more greatly than that in aspirin group, but there was not significant difference between two groups (P > 0.05).
CONCLUSIONIt is suggested by the present results that HLJD decoction could protect vascular endothelium from early damage in type 2 diabetes. The protective effects of HLJD on endothelium might be related to its ability of reducing the blood glucose, adjusting plasma lipids profiles, improving insulin resistance, antagonizing inflammatory mediators and inducing endothelium-dependent vascular relaxation.

8.Inhibitory effect of retroviral vector containing anti-sense Smad4 gene on Ito cell line, LI90.

BACKGROUNDTransforming growth factor-beta1 (TGF-beta1) exerts strong fibrogenic potential in culture-activated HSCs. Smad4 is a key intracellular mediator for the transforming growth factor-beta (TGF-beta) superfamily of growth factors. The aim of this study was to assess the effects of the antisense Smad4 gene on Ito cell line, LI90.
METHODSThe recombinant retroviral vector pLXSN-Smad4 was constructed by cloning the rat antisense Smad4 cDNA into the retroviral vector pLXSN. Retroviruses with or without the antisense gene were obtained by transfecting pLXSN-Smad4 and pLXSN vectors into PA317 cells. Human hepatic stellate cells (HSCs) LI90 were infected with these retroviruses followed by selection with G418. The expression of Smad4 was detected by Northern and Western blots. Cell biological characteristics, including cell growth curve, 3H-TdR and 3H-proline uptake by HSCs and the production of extracellular matrix were assessed.
RESULTSmRNA and protein expressions of Smad4 in LI90 cells transfected with retrovirus containing the antisense Smad4 gene were much lower than those in LI90 cells transfected with empty vector or parental LI90 cells. Cells hypoexpressing the Smad4 gene exhibited a slower rate of growth, a lower uptake of 3H-TdR and 3H-proline (P < 0.01), and smaller production of th extracellular matrix, compared with parental LI90 cells and cells transfected with empty retrovirus.
CONCLUSIONSThe antisense Smad4 gene can suppress the expression of the Smad4 gene, reduce endogenous production of Smad4 mRNA and protein, block TGF-beta1 signaling pathway, inhibit activation of Ito cells, obstruct the growth of Ito cells, decrease the production of the extracellular matrix (ECM). Our results may provide a basis for the development of antifibrotic gene therapy.

9.Associations between genetic polymorphisms of glutathione S-transferase M1 and T1, smoking and susceptibility to colorectal cancer: a case-control study.

OBJECTIVETo evaluate the associations between genetic polymorphisms of glutathione S-transferase M1 and T1 (GSTM1 and GSTT1), smoking and susceptibility to colorectal cancer.
METHODSA case-control study of 126 patients and 343 healthy controls was conducted to investigate the role of GSTM1 and GSTT1 polymorphisms in colorectal cancer. Genotypes of GSTM1 and GSTT1 polymorphisms were analyzed by multiplex allele-specific polymerase chain reaction (PCR).
RESULTSThe frequencies of GSTM1 null and GSTT1 null genotypes were 55.5% and 20.4%, respectively. After adjustment for age and sex, among those with GSTT1 null genotype, the GSTM1 null genotype had a significant increased risk of rectal cancers compared to GSTM1 non-null genotype (OR=9.74, 95% CI, 1.13 - 83.85). A 2.22-fold risk of colon cancers was associated with GSTM1 null genotype compared to GSTM1 non-null genotype among current smokers (P >0.05). Individuals with GSTT1 null genotype and currently smoking had a significant risk of colon cancers (OR = 4.55, 95% CI, 1.14 - 18.17), and rectal cancers (OR = 4.60, 95% CI, 1.11 - 19.11).
CONCLUSIONThis study suggests that certain null GSTM1 and GSTT1 genotypes may be associated with an elevated risk of colorectal cancer which may be modified by interaction of the two genetic polymorphisms and cigarette smoking.

10.Threshold analysis of extent of resection for patients with gradeⅢgliomas using MRI

Objective To evaluate the prognostic significance of extent of resection (EOR) in patients with gradeⅢglioma, and the MRI sequence best related with tumor volume. Methods Ninety-six cases were studied retrospectively who were diagnosed as glioma of grade Ⅲ gliomas at Dalian Medical University Affiliated Hospital of Dalian Municipal Central Hospital during the period from 2010 to 2015. EOR was calculated using preoperative and postoperative contrast-enhanced T1-weighted and T2-weightedfluid attenuated inversion recovery (T2Flair) MR images. Univariate and multivariate analyses were performed to evaluate the correlation between EOR and overall survival (OS). Results The results of univariate analysis showed that patients with age<50 years(P=0.032), preoperative KPS>80 scores (P<0.01), no edema (P=0.005), postoperative chemotherapy (P<0.01) and T2Flair EOR≥68% (P<0.01) had better prognosis. Cox multivariate regression analysis showed that age(P=0.008), edema (P=0.003), postoperative chemotherapy(P=0.000) and T2Flair EOR (P=0.004) were independent factors of prognosis for grade Ⅲ glioma patients. Conclusions T2Flair-EOR is one of the most significant prognostic predictors in patients with grade Ⅲ gliomas. An important survival advantage is associated with T2Flair-EOR≥68% for patients with gradeⅢgliomas.
