BACKGROUND: The invasion of highly metastatic carcinoma cells on reconstituted basement membrane component is considered as a common model for in vitro experimental studies of invasion and metastasis of malignant tumors. It has been reported that synthetic peptide influences some tumor cells. However, the effect of synthetic peptide on highly metastatic human lung giant-cell carcinoma cell(PG) has been rarely studied.OBJECTIVE: To study the effects of synthetic peptide RGD, YIGSR and their derivatives on invasive ability of PG in vitro.DESIGN: A completely randomized controlled trial was conducted.SETTING and MATERIALS: The human lung highly metastatic tumor and NIH3T3 cell line were both supplied by the Department of Pathology of Beijing Medical University. The synthetic peptides were provided by the Synthetic Laboratory of the Institute of Materia Medica of the Chinese Academy of Medical Sciences and Chengde Medical College. Transwell invasion chamber was purchased from Costor Company. The whole experiment was conducted at the Department of Biochemistry of Chengde Medical College.INTERVENTIONS: NIH3T3 cell line was used to prepare chemotactic factor. PG cells were seeded in Transwell invasion chamber. Under the effect of chemotactic factor, the effects of synthetic peptides on PG invasive ability were examined.MAIN OUTCOME MEASURES: PG cell number of passing through the reconstituted basement membrane was used as an index to evaluate the invasive ability of tumor cells.RESULTS: An appropriate number of PG cells were cultured for 10 hours with chemotactic factor, and the number ofinvasive cells reached a number range that can be applied to statistical analysis(50 to 100 invasive cells). After 100 mg/L of synthetic peptides RGD, YIGSR were incubated for 48 hours with PG cells, the rate of inhibition on invasive ability was 53.63% and 36.86% respectively, and 0% and 6.48% respectively after incubation for 10 hours.CONCLUSION: The PG cells were the proper cells for in vitro experimental studies of invasive ability of cancer cells for the purpose of screening of anti-tumor medicine. RGD and Juncha/Zacha-peptide YIGSR showed a strong activity in inhibiting tumor invasion and a potential value in prevention and treatment of malignant tumor development.

Restriction endonucleases are important molecular biology tools for DNA recombination.Because of the cleavage of DNA,their recombinant expression is difficult with low yields and complicated purification processes. In commercial productions, the technology that uses specific methylases to protect host DNA from digestion of the expressed restriction enzymes was cumbersome and practically limited. For solving this problem the expression of restriction enzyme Not Ⅰ was performed by using the DNA methylase M.Sss Ⅰ derived from Spiroplasma sp.MQ1 which specifically kept CpG sequence methylated. The methylated DNA was protected from the cutting of Not Ⅰ whose recognition sequence contained CpG.The gene of methylase M.Sss Ⅰ was introduced into Escherichia coli ER2566 and constitutively expressed, resulting in the CpG methylation pattern of the host DNA.Restriction enzyme Not Ⅰ was successfully expressed in this E.coli strain.Furthermore,by adding a purification tag to one terminus of the enzyme, recombinant Not Ⅰ was prepared as a highly purified and active product through two simple Ni-affinity and anion exchange chromatography steps. This expression system can be applied for the preparation of a series of restriction enzymes with CpG in their recognition sequences.

Objective To introduce the donor heart preservation techniques and study the clini- cal results in 26 patients undergoing orthotopic heart transplantations with long donor ischemia time (DIT) in our center. Methods From Sept.2004 to Aug.2006,26 patients underwent orthotopic heart transplantation at our center with DIT longer than 360 min.Crystalloid cardioplegia was infused through aortic root after aorta was cross clamped.UW solution was used when donor heart was removed. All patients underwent bicaval anastomotic cardiac transplantation.NO was inhaled through ventilator during and after operation in patients with pulmonary hypertension. Results DIT was 360- 560 (mean 401?43) min. There was no operative death in this group.Echocardiography and X-ray examinations in one month were normal. One case of abnormal coronary orifice was subjected to re- anastomosis of aorta. One case of renal failure post-operation was subjected to blood dialysis.Conclu- sion With proper donor heart procurement and preservation, recipient selection, anastomotic technique, efficient postoperative surveillance and pulmonary hypertension treatment, transplanted patients with long DIT can obtain the satisfactory short-term outcome.
8. Microenvironment and repair methods of peripheral nervous system injury
Chinese Journal of Tissue Engineering Research 2021;25(4):651-656
BACKGROUND: Peripheral nerve injury has been very common in clinical work. Although microsurgical technique can restore the continuity of the injured nerve well, the nerve repair effect is still unsatisfactory due to the high degree of differentiation and low regeneration ability of the peripheral nerve tissue, which seriously affects the quality of life of patients. At present, there is no unified conclusion on the microenvironment of peripheral nerve injury, and there are many repair methods in common use. OBJECTIVE: To review the microenvironment of peripheral nerve injury and the repair methods of peripheral nerve injury. METHODS: A computer-based online search of PubMed and CNKI databases was performed for the articles published from January 1964 to September 2019. The key words were “peripheral nerve injury; microenvironment; microsurgical technique; small gap bridging” in English and Chinese, respectively. Finally, 57 eligible articles were included to review. RESULTS AND CONCLUSION: (1) After a series of animal experiments and clinical studies, changes in microenvironment such as the establishment of nerve regeneration channels, neurotrophic factors, immune response, inflammatory response, and hormone regulation have been confirmed to be important factors affecting peripheral nerve repair. (2) It is feasible to repair the peripheral nerve injury by using biological conduit small gap bridging instead of the traditional external and fascicular membrane.

Flammulin, an anti-tumor protein, was purified from the aqueous extract of basidiomes of Flammulina Velutipes. Purified flammulin emulsified with Freund's adjuvant was injected subcutaneously into New Zealand white rabbits. After several immune enhancements, these animals were bled and sera were separated. Antiserum against flammulin in Western blots were applied to determine if flammulin be present in the liquid state culture or fruiting body. The result showed that anti-flammulin serum could recognize the aqueous extract of fruiting body in SDS-PAGE gels under the reducing conditions, no flammulin was detected in mycelia of Flammulina Velutipes.

International Eye Science 2009;9(6):1030-1031
AIM: To compare the measurement of anterior chamber depth(ACD) inclusive of corneal thickness using intrao-cular lens(IOL) master and ultrasound biomicroscopy (UBM) and evaluate the repeatability of each method.METHODS: Two consecutive measurements of ACD were prospectively performed using IOL master and UBM in 60 eyes in 60 individuals. Mean values were compared using the paired t test. For each individual, ACD measure-ments was performed 5 times to estimate the repeatability of each method by a coefficient of variation(CV).RESULTS: The mean ACD was 2.95±0.25mm with the IOL master and 2.96±0.22mm with the UBM. This diffe-rence was not statistically significant (P=0.631).The coefficient of variation (CV) was 0.56%±0.26% and 0.65%± 0.36% in IOL master and UBM, respectively.CONCLUSION: The mean ACD of IOL master was the same as UBM. The repeatability of IOL master is better than UBM.
