1.TVT-O in the treatment of stress urinary incontinence: long-term evaluation and urodynamic study

Objective To evaluate the long-term outcomes,complications and urodynamic parameters of using tension-free vaginal tape-obturator(TVT-O)in the treatment of patients with stress urinary incontinence.Methods Preoperative and postoperative evaluations including urodynamic data and quality of life were performed for 24 patients with stress urinary incontinence who were enrolled and treated with TVT-O between May 2007 and June 2011.Results Patients completed long-term postoperative follow-up from 12 to 60 months after surgery.Twenty-one(87.5%)patients achieved long-term subjective success and 22(91.7%)achieved objective success.Surgical satisfaction and quality of life was high.Long-term postoperative abdominal leak point pressure of 23 patient was greater than 100 cm H2O,and the remaining parameters preoperative and postoperative showed no significant differences.Voiding difficulty and external reflection voding desire were the main long-term complications.Conclusions TVT-O is with few complications and high patient satisfaction.It is a simple,safe and effective procedure for treatment of patients with stress urinary incontinence.Long follow-up period is important for the comprehensive evaluation of TVT-O.Postoperative urodynamic study has significance in complications,diagnosis and evaluation.
2.CCL21/CCR7 axis and mechanisms of tumor invasion and metastasis

Secondary lymphoid tissue chemokine (CCL21) is a double-edged sword, which exerts antitumor, anti-infection immune response by binding to the receptor CCR7 on the surface of the multiple immune cells. However, a variety of tumor cells also express the receptor CCR7, the combination of CCL21 with CCR7promotes the invasion and metastasis of tumor cells, leading to the facilitation of tumor development. Therefore,exploring the mechanism(s) of tumor invasion and metastasis might be helpful for use of CCL21 as tumor gene therapy through blocking of CCL21's promotion of tumor invasion and metastasis.
3.Expression and Correlated Analysis of Autophagy-related Proteins in Human Squamous Cell Carcinoma and Adenocarcinoma of Lung

Objective To explore expression of Beclin1, autophagy-related protein 1 (Atg1), mammalian target of ra-pamycin (mTOR) in squamous cell carcinoma (SCC) and adenocarcinoma of lung and to analyse relationship among these three factors. Methods Immunohistochemical stainning was applied to detect expression of Beclin1, Atg1 and mTOR in 82 samples of SCC and adenocarcinoma of lung (lung cancer group) and 17 samples of paraneoplastic normal lung tissues (con-trol group). Correlations of expression of Beclin1, Atg1 and mTOR were analyzed in SCC and adenocarcinoma of lung. Re-sults The expression of Beclin1 was significantly lower in lung cancer group than that in control group(52.44%vs 94.12%, P<0.01). There were significant differences of the differentiation, TNM stages and lymph node metastasis in lung cancer group compared with those in control group. And Beclin1 expression showed a decreasing trend with malignant and invasive of tumors. mTOR expression in lung cancer tissue with lymph nod metastasis is higher than that in the tissue without lymph node metastasis(P<0.01). Atg1 expression in lung cancer tissue with larger mass is less than that in lung cancer tissue with small lumps(P<0.01). Beclin1 Expression was positively correlated with the Atg1 expression in SCC(r=0.609,P<0.01). Beclin1 and Atg1 expressions were both negatively correlated with mTOR expression in SCC(r=-0.617,-0.444,P<0.01). Beclin1 expression was positively correlated with Atg1 expression in adenocarcinoma(r=0.458,P<0.01). Beclin1 and Atg1 expression were both negatively correlated with mTOR expression in adenocarcinoma(r=-0.497,-0.541,P<0.01). Conclu-sion Beclin1, Atg1 and mTOR expression showed a strong correlation in lung cancer, and clinicopathological change of lung SCC and adenocarcinoma may be related to autophagy.
4.Effect of basic fibroblast growth factor on human periodontal ligament cells cultured in vitro

BACKGROUND:Basic fibroblast growth factor is a pluripotent cytokine that can promote the proliferation of mesodermal and neuroectodermal cells.
OBJECTIVE:To observe the effect of basic fibroblast growth factor in human periodontal ligament cells cultured in vitro.
METHODS:Human periodontal ligament cells at passage 5 were inoculated into the 96-wel plates at the density of 1×108/L, and were randomly divided into four groups. The cells were cultured inα-MEM containing 15%fetal bovine serum and 0, 1, 10, 100μg/L basic fibroblast growth factor, respectively. At 1, 3, 5, 7 days of the culture, the cellproliferation was determined, and the activity of alkaline phosphates was detected at 1 and 7 days.
RESULTS AND CONCLUSION:There were significant differences in the proliferation of human periodontal ligament cells among the four groups (F=6.586, P=0.024). As the increase of the basic fibroblast growth factor concentrations, the absorbance value was gradual y increased and reached the peak in 100μg/L basic fibroblast growth factor group (P<0.05). The alkaline phosphatase activity in basic fibroblast growth factor groups was lower than that of the control group (P=0.000), the higher the concentration was, the lower activity was (P<0.05). Results show that basic fibroblast growth factor can promote the proliferation of human periodontal ligament cells and inhibit the activity of alkaline phosphatase, and the effect is concentration-dependent.
5. Demand and structure analysis of operation box for commanding military medical service

9.Transplanted hepatocytes and collagen hydrogel units subcutaneously establish engineered hepatic tissue

BACKGROUND:Colagen hydrogel provides good matrix support for hepatocyte growth and tissue reconstruction, and the colagen-based engineered tissue is easy to merge the growth and form integrated tissue. OBJECTIVE:To improve the thickness of engineered hepatic tissue by dissociating hepatocytes/colagen hydrogel composite into smal hepatic units that accumulate in the subcutaneous cavity. METHODS:Freshly isolated hepatocytes from rats were mixed with colagen hydrogel to establish hepatocytes/colagen hydrogel composite. The hepatocytes/colagen hydrogel composite was dissociated into smal hepatic units after being cured. The undissociated hepatocytes/colagen hydrogel composite was taken as a control. Six Spraque-Dawley rats were enroled. Three of them were subjected to a two-thirds partial hepatectomy to induce liver regeneration. Dissociated and undissociated hepatocytes/colagen hydrogel composites were implanted into the bilateral inguinal subcutaneous cavity. Dissociated and undissociated hepatocytes/colagen hydrogel composites were implanted into the bilateral inguinal subcutaneous cavity of the other three rats. At the 7th day after transplantation, engineered hepatic tissue formation was evaluated using hematoxylin-eosin staining, immunohistochemical staining and India ink perfusion methods. RESULTS AND CONCLUSION:The grafts in these two groups al formed vascular engineered hepatic tissue in the subcutaneous cavity, but after the smal hepatic units merged, a large piece of vascular engineered hepatic tissue formed. The hepatic tissue thickness was up to 4 mm. The whole piece of implanted liver grafts only formed smal pieces of hepatic tissues, with only several layers of cels. Immunohistochemistry staining confirmed that the hepatocytes in vascular engineered hepatic tissue had the characteristics and functions of natural hepatocytes. Partial hepatectomy experiment showed that engineered hepatic tissue had the ability to respond to regenerative stimulus of partial hepatectomy. These results show that dissociating the hepatocytes/colagen hydrogel grafts into smal units that accumlate in the subcutaneous cavity can increase the thickness of the engineered hepatic tissue.
10.fMRI study on the impact of passive hyperthermia on the executive control function

Objective To explore the causal influences between activated regions in executive control network when participants were performing attention network test (ANT)under passive hyperthermia using Granger causality analysis.Methods Thirty male healthy participants which were randomly divided into control group (n=1 5)and experimental group (n=1 5)performed ANT task during two thermal conditions:hyperthermic condition (50℃)and normothermic condition (20℃)during functional magnetic reso-nance imaging.The data processing was performed using statistical parametric mapping 8 (SPM8).Granger causality analysis was performed on the resulted activated regions,as well as the correlations of the GC values with the reaction time in both groups.Results Between-group comparisons on Granger causality analysis revealed significantly enhanced influences of anterior cingulate gyrus and right inferior parietal lobule on dorsolateral prefrontal cortex,and bidirectional influences between dorsolateral prefrontal cortex and right superior parietal lobule,but decreased influence between right superior parietal lobule and right inferior parietal lobule.The correlation analysis showed significant correlation between GC value of right superior parietal lobule to dorsolateral prefrontal cortex and the reaction time in control group (r=-0.558,P =0.031),but yet there was no significant correlation in experimental group (r=0.355,P =0.257).Conclusion Heat stress affected the causal influences within the executive control network,which subse-quently disrupted the executive control performance during passive hyperthermia.
