1.Behavioral Problems and Quality of Life in Children with Asthma

Objective:To explore differences in beha viors and quality of life between asthmatic and non-asthmatic children . Methods:Be haviors and quality of life were assessed in the 84 children with asthma and healthy children,4-11 years of age .Results :Behavioral problems were more prevalent in asthmatics than non-asthmatics.The major beha vior al problems in children with asthma were recession, physical complaints,anxiety/ de pression,and social intercourse.The children with asthma were less satisfact ory than healthy children with respect to quality of life especially on anxiety,depressi on and somatic feelings.Conclusion:Asthma contribut es to the developmen t of behavioral problem and adversely affects the children's quality of life.
2.Antagonistic Effect of Ziyin Xiehuo Recipe on Estrogen-like Activity of Environmental Endocrine Disruptors Exposed Rats

Objective To explore the antagonistic effect of Ziyin Xiehuo Recipe(ZYXHR,Recipe for nourishing kidney yin and purging ministerial fire) on estrogen-like activity of environmental endocrine disruptors,nonylphenol(NP) and mixture of NP and bisphenol A(BPA). Methods Totally 30 three-week-old female SD rats were randomly divided into control group(fed with corn oil),exposed group A (feeding NP100 mg / kg ),exposed group B (feeding NP 50 mg/kg+ BPA 200 mg/kg ),treatment group A (NP 100 mg/kg + ZYXHR),treatment group B (NP 50 mg/kg+ BPA 200 mg/kg + ZYXHR). After 15 days of treatment,the uterine weight gain and PCNA protein expression were detected. Results As compared with those of the control group,Uterine wet weight,uterus organ coefficient,thickness of endometrium and smooth muscle,and the height of endometrial glands and glandular epithelium significantly increased in exposed group A and B (P
3.Behavioral problems and interventions in child with asthma

objective To investigate the differences in behaviors between asthmatic and nonasthmatic children , and to evaluate the psychological interventions. Method Behaviors were investigated in the 84 children with asthma and 84 healthy children, After psychological interventions of 3~4 months, the effects were evaluated in 18 asthmatic children. Results Behavioral problems were more prevalent in asthmatics than nonasthmatics,behavioral problems were different in asthmatic children of different severity and gender,psychological interventions as well as drug therapy have beneficial effects on controling of asthmatic symptoms. Conclusions Asthma contributes to the development of behavioral problems,Psychological intervention in child with asthma has many positive effects on controling the symptom and miniming the bad behaviors.
4.Reliability and validity of the center for epidemiologic studies depression scale in rural populations

Objective To assess the reliability and validity of the center for epidemiological studies depression scale (CES-D) in rural populations. Methods CES-D was administered to 330 rural residents in Jinzhou and Zhuanghe district of Dalian city. Hamilton Rating Scale for Depression (HAM-D) was used as standards to assess the discriminant validity and criterion validity. Statistical analysis included Pearson's correlations and the exploratory factor analysis. Results The Cronbach α was 0.74,and the average item-total correlation was 0.59. Two factors were extracted by using exploratory factor analysis:affective and somatic symptoms and interpersonal problems. The range of factor loading was between 0.42 and 0.83. CES-D mean scores were significantly different between the depressive group (22.80±10.28) and non-depressive group (15.36±5.76. t =2.60,P =0.01). The correlation coefficient between CES-D and HAM-D was 0.50( P <0.01). Conclusion The reliability and validity of CES-D for rural populations is acceptable.
5.Progress of cellular and vaccine immunotherapy in multiple myeloma

Allogeneic hematopoietic stem cell transplantation and donor lymphocyte infusion for multiple myeloma (MM) can induce graft-versus-myeloma immunity and long-term survival,but limited efficacy and associated toxicities have prevented its widespread application.Cellular immunotherapies and vaccines are explored to induce more specific,reliable,and potent antimyeloma immune responses with less treatmentrelated risk.Advances in molecular biology,basic and applied immunology,have led to several promising approaches such as genetically engineered T cells with chimeric antigen receptors and T-cell receptors targeting myeloma-specific epitopes,vaccine primed ex vivo expanded autologous T cells,expanded marrowinfiltrating lymphocytes,and plasma cell/dendritic cell fusion vaccines.The combination of these emerging therapies to immunomodulatory drugs and inhibitors of programmed death-1 T-cell regulatory pathways could improve the outcome for MM patients.This article reviews the latest progress of cellular and vaccine immunotherapy for MM at the 58th American Society of Hematology (ASH) Annual Meeting,and discusses how these therapies might integrate and synergize with existing treatment paradigms.
8.Clinical study on intestinal fatty acid binding protein and the endotoxin in early diagnosis of intestinal barrier dysfunction

Objective To screen the high specific and sensitive monitoring indications in the diagnosis of intestinal barrier dysfunction.Methods A total of 70 critical patients with intestinal barrier dysfunction and acute physiology and chronic health evaluation (APACHE) Ⅱ score≥8 and over the same period 41 patients without intestinal barrier dysfunction and APACHE Ⅱ score≤6 were recruited.The general information,histories,symptoms,physical signs,24 hours urine output and the condition of mechanical ventilation treatment were recorded.The venous blood was taken for bacteria culture,white blood cell counting,creatinine level,diamine oxidase (DAO) activity,D-lactic acid,intestinal fatty acid binding protein (IFABP) and endotoxin level testing.The urine was taken for urinary IFABP level testing.Twenty-four hours urine was reserved for 24 hours total urinary IFABP testing.The factors which might influence intestinal barrier dysfunction were analyzed by univariate analysis and multivariate analysis.The measurement data were analyzed by t test and the count data were analyzed by x2 test.The factors were screened according to receiver operating characteristic (ROC) curve.Results The factors related with intestinal barrier dysfunction were white blood cell counting (OR=3.971,P=0.046),plasma endotoxin level (OR=7.857,P=0.005)and 24 hours total urinary IFABP (OR=11.154,P=0.001).The areas under the ROC curve (AUC)of plasma endotoxin level and 24 hours total urinary IFABP were 0.852 and 0.820 respectively (both P<0.01).The critical value was 8.0 pg/ml and 17.12 ng respectively.The sensitivity was 97.8% and 84.4%.The specificity was 66.7% and 72.7%.Conclusion Once critical patients presented certain gastrointestinal symptoms and physical signs with plasma endotoxin level >8.0 pg/ml and or 24 hours total urinary IFABP >17.12 ng,which might indicate intestinal barrier dysfunction.
9.Drug Quality Representation Analysis of Preparations of Aurea Helianthus based on Anti-myocardial Ischemia Effect

This study was aimed to select the best preparation form of Aurea Helianthus in order to provide a scientific basis for research and development of anti-myocardial ischemia drugs.The content of effective index components of preparation of Aurea Helianthus was detected using HPLC-PDA.Acute myocardial ischemia rat model introduced by injecting pituitrin was used to investigate the effect of anti-myocardial ischemia.The quality of preparation from Aurea Helianthus was characterized based on pharmacy and efficacy.And the characterized results were given correlation analysis.The results showed that the HPLC method for quantitative determination was verified by methodology.The average recoveries were in the range of 97.04%-102.44%.All three preparations of Aurea Helianthus had the efficacy of anti-myocardial ischemia in varying degrees.The enrichment had the highest effect with lowest extraction rate and the lowest dosage,which was the best form in medicinal-preparation application of Aurea Helianthus.It was concluded that the content of related drug components was higher with unique proportion in enrichment of Aurea Helianthus which was prepared based on drug system proved a high quality medicine.Therefore,the key to the improvement of efficacy was to explore the fittest proportion and improve the content of related pharmaceutical ingredients pertinently according to drug system.
10.Metabolism of 3-cyanomethyl-4-methyl-DCK, a new anti-HIV candidate, in human intestinal microsomes.

The biotransformation, CYP reaction phenotyping, the impact of CYP inhibitors and enzyme kinetics of 3-cyanomethyl-4-methyl-DCK (CMDCK), a new anti-HIV preclinical candidate belonging to DCK analogs, were investigated in human intestinal microsomes and recombinant cytochrome P450 (CYP) enzymes. CMDCK (4 micromol L(-1)) was incubated with a panel of rCYP enzymes (CYP1A2, 2C9, 2C19, 2D6 and 3A4) in vitro. The remaining parent drug in incubates was quantitatively analyzed by a LC-MS method. CYP3A4 was identified as the principal CYP isoenzyme responsible for its metabolism in intestinal microsomes. The major metabolic pathway of CMDCK was oxidation and a number of oxidative metabolites were screened with LC-MS. The Km, Vmax, CLint and T1/2 of CMDCK obtained from human intestinal microsome were 45.6 micromol L(-1), 0.33 micromol L(-1) min(-1), 12.1 mL min(-1) kg(-1) and 25.7 min, respectively. Intestinal clearance of CMDCK was estimated from in vitro data to be 3.3 mL min(-1) kg(-1), and was almost equal to the intestinal blood flow rate (4.6 mL min(-1) kg(-1)). The selective CYP3A4 inhibitors, ketoconazole, troleandomycin and ritonavir demonstrated significant inhibitory effects on CMDCK intestinal metabolism, which suggested that co-administration of CMDCK with potent CYP3A inhibitors, such as ritonavir, might decrease its intestinal metabolic clearance and subsequently improve its bioavailability in body.
