1.Analysis of risk factors of dementia after stroke in elderly patients.

Objective\ To explore the risk factors of dementia induced by stroke.Methods\ 258 elder patients with stroke had been followed up for three months,including clinical neuropathy and neuropsychological tests.Multivariate noncondition stepwise Logistic model analysis was made after single variable analysis.Results\ There were 83 patients with dementia induced by stroke,175 patients without dementia after stroke.The morbidity was 32.2%.The following eight factors were associated with dementia i.e.,age,multiple lesions,left lesion,large area of cerebral hemorrhage or cerebral infarction,cerebral atrophy,hypertension,the history of stroke,and bilateral lesions.Conclusions\ Dementia induced by stroke is caused by multiple factors.It is closely correlated to numbers of cerebral lesions,the location of cerebral lesions and the size of brain tissue necrosis.The primary conditions of patients'cerebral function are also closely associated with the occurence of dementia.
2.Correlation of survivin, p53 and Ki-67 in laryngeal cancer Hep-2 cell proliferation and invasion

Objective:To investigate the mechanism of survivin, p53 and Ki-67 on Hep-2 human laryngeal cancer endothelial cell proliferation and invasion.Methods:Laryngeal squamous cell carcinoma and paracancerous normal tissues were collected, total RNA was extracted from tissues,survivin,p53and Ki-67gene mRNA expression levels in laryngeal cancer and the adjacent tissues were detected by Real-time PCR. Human laryngeal cancer Hep-2 epithelial cells were selected,survivin gene was overexpressed, and cell proliferation was detected by MTT.p53 andKi-67gene expression changes in overexpressedsurvivin gene were detected by Western blot. Changes in Hep-2 cell invasive ability were studied whensurvivin was overexpressed as detected by Transwell invasion assay.Results: In the adjacent tissues, survivin,p53andKi-67 gene relative expression levels were 1.72 ± 0.9, 13.7 ± 5.7 and 5.7 ± 1.3, respectively; while in cancer tissues, gene relative expression levels were 53.7 ± 8.3, 66.7 ± 5.2 and 61.0 ± 3.1, respectively, which was significantly increased. As detected by MTT, relative cell survival rate within 12 h ofsurvivinoverexpression were: load control group, (88.5±1.6)%; overexpressed group, (90.3±1.9)%. Transwell invasion assay results indicated that overexpressedsurvivincould significantly increase the relative survival rate of cells. Conclusions:Expressions ofp53,Ki67 and survivin are increased in cancer; and there is a positive correlation betweensurvivin, p53andKi67 expressions in laryngeal carcinoma.
3.Budesonide/Formoterol in the Management of Patients with Mild or Moderate Asthma: Clinical Research

OBJECTIVE: To evaluate the efficacy and the safety of low dose budesonide/formoterol combination therapy vs.medium dose of budesonide(BUD) dry powder in the management of mild or moderate adult asthma.METHODS: A total of 62 patients with asthma were randomized to Group A(budesonide plus formcterol) or Group B(budesonide).Main outcome measures: daytime symptom and nocturnal symptom,peak expiratory flow(PEF),number of times of using rapid-acting ? 2 agonist and its ADR.RESULTS: After treatment,both groups had significant improvement in daytime symptom and nocturnal symptom(number of times of wakeup),PEF value and lung function,and the dosage of rapid-acting ? 2 agonist was decreased,showing significant differences between the two groups.The ADR was light in degree.CONCLUSION: Inhalation therapy of low dose of glucocorticoid plus long-acting ? 2 agonist is an optimal choice in the management of asthma.The efficacy of glucocorticoid could be enhanced more by combination with low dose and long-acting ? 2 agonist than by simply increasing its own dose.
4.Current situation of diabetes education and management in China at grassroots level

Diabetes mellitus is a worldwide public health problem,which is a serious threat to human health.WHO released the global diabetes report shows Chinese incidence of type 2 diabetes was explosive growth in the past few decades,but public awareness of diabetes is generally inadequate,especially at the grassroots level.Therefore,it is imperative to regulate the basic level of diabetes diagnosis and treatment.As one of the basic measures of diabetes treatment in our country,education and management of diabetes' foundation is weak,mainly for the lack of national basic medical and health institutions,dull form of education and lack of target,shortages of specialized diabetes staff.So in order to safeguards the success of primary diabetes education and management and improve the prevention of primary diabetes,it is necessary to establish a sound mechanism of reward system,make full use of various payment platform,strengthen the basic management of diabetes monitoring information construction and carry out the Internet education measures.This article reviews the current situation of education and management of primary diabetes.

AIM: To observe the change of nitric oxide (NO) generation system in the vascular adventitia, media and intima in septic shock rats. METHODS: The septic shock model was made in rats by caecal ligation and puncture. The intima, media and adventitia of the rat aorta were separated. OBJECTIVETo investigate the synergistic effect of transforming growth factor-beta1 (TGF-beta1) and platelet-derived growth factor-BB (PDGF-BB) on the expression of integrin beta3, in periodontal membrane of rat orthodontic tooth.
METHODSAn orthodontic tooth movement model was established. Up to 32 experimental rats were randomly divided into four groups according to a random number table. The four groups were injected with 1% PBS, TGF-beta1 (5 ng), PDGF-BB (10 ng), and combined TGF-beta1 (5 ng) and PDGF-BB (10 ng) in the buccal submucosal, respectively. The volume injected in each group was 0.1 mL. The animals were then sacrificed on the 10th day. The left maxillary first molar and periodontal tissue were taken. Different expressions of integrin beta3 were detected in periodontal tissues through immunohistochemistry. Mean optical density (OD) values of the positive fields were examined. The data obtained were analyzed through ANOVA. The data followed normal distribution, and were compared via t-test.
RESULTSCompared with the control groups, the expression of integrin beta3 was higher in the experimental groupin tension sides (P < 0.01). Significant differences in tension sides between the single-injection groups and the combined group were observed (P < 0.01). Compared with the control groups, the expression of integrin beta3 was higher in the experimental group in compression sides (P < 0.05). In addition, there was no significant differences in compression sides between the single-injection groups and the combined group (P > 0.05).
CONCLUSIONIn terms of local regulatory factors, TGF-beta1 combined with PDGF-BB enhance the expression of integrin beta3 in the periodontal membrane and accelerate periodontal remodeling. The synergistic effect of the two growth factors is better than the single growth factor.

7.Effects of xeroderma pigmentosum B gene on proliferation and apoptosis of vascular smooth muscle cells induced by interleukin-6

Objective To investigate effects of xeroderma pigmentosum B(XPB) gene on IL-6 induced proliferation and apoptosis in human vascular smooth muscle cells (VSMC).Methods Recombinant plasmid pcDNA3.1-XPB and vacant vector plasmid pcDNA3.1 were transfected stably into VSMC by liposome,and these cells were incubated with IL-6 at a 100 U/ml for 48 hours.The experiments were divided into six groups:blank control group; pcDNA3.1 group; pcDNA3.1-XPB group;IL-6 group; IL-6 + pcDNA3.1 group; IL-6 + pcDNA3.1-XPB group.The expression levels of XPB,Bcl-2,Bax and wild type p53 (wt-p53) were detected through reverse transcription polymerase chain reaction (RT-PCR) and Western blotting.The cell survival,cell cycle and apoptosis were examined with 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) and flow cytometry,respectively.Results The transfection of pcDNA3.1-XPB increased the expression of XPB,Bax and wt-p53 (P<0.05 or P<0.01),decreased the expression of Bcl-2 (P<0.05 or P<0.01),and reduced the IL-6 induced effects on decreasing the expression of Bax and wt-p53 and increasing the expression of Bcl-2(P<0.05 or P<0.01).The over expression of XPB inhibited the cell growth(q=2.95,P< 0.05),and reduced the positive effects of IL-6 on VSMC growth ( 102.6 +6.2) % vs.(124.5 + 7.9) %,q=3.49,P<0.05.The over expression of XPB increased the apoptosis rate of VSMC(P<0.01 ) and the cell amounts of G0/G1 phase (q=2.99,P< 0.05),decreased the cell amounts of S phase(q=3.05,P<0.05),and reduced the IL-6 induced effects on decreasing the apoptosis rate of VSMC(5.9±2.1)% vs.(0.3±0.1)%,q=7.53,P<0.01; the cell amounts of G0/G1 phase(70.9±6.7) % vs.(54.8±2.9) %,q=6.91,P<0.01 ;and on increasing the cell amounts of Sphase(20.2+3.6)% vs.(36.4+7.2)%,q=8.54,P<0.01.Conclusions XPB gene could inhibit VSMC proliferation,promote VSMC apoptosis,and reduce the effects that IL-6 promotes VSMC proliferation and inhibits VSMC apoptosis.Therefore,XPB gene is likely to be potential molecular target for treatment of atherosclerosis.

ABSTRACT:Objective To investigate the effects of total flavones of oldenlandia diffusa (FOD)on epithelial-mesenchymal transition in hepatocellular cancer cell line MHCC97-H.Methods TGF-β1 induced EMT in routinely cultured liver cancer cell line MHCC97-H;then MHCC97-H cell was divided into 5 groups:normal control group, TGF-β1 group,TGF-β1 + FOD group,TGF-β1 + 5-FU group,and TGF-β1 + FOD + 5-FU group.After 48 h of treatment,the invasion ability of MHCC97-H cell was detected by Transwell;the proteins of E-cadherin and vimentin were determined by Western blot.Results Compared with the normal form of MHCC97-H cell line,the cell had obvious long fusiform after TGF-β1 induction,and the invasion ability enhanced (P = 0.02 ).But after treatment,the invasion ability of MHCC97-H cell decreased in FOD group and 5-FU group compared with that in TGF-β1 group (P = 0.03,P = 0.02 ),and decreased more significantly in FOD + 5-FU group (P = 0.01 ).The expression of E-cadherin at the protein level decreased significantly (P = 0.01 )in TGF-β1 group,which was abolished in FOD group (P =0.03 )and 5-FU group (P = 0.02 ).The expression of vimentin at the protein level increased significantly (P =0.01)in TGF-β1 group,which was abolished in FOD group (P =0.04)and 5-FU group (P =0.03)and more obviously in FOD+5-FU group (P =0.01).Conclusion FOD can reverse the invasion of MHCC97-H cells in EMT induced by TGF-β1 through decreasing the expression of E-cadherin protein and inhibiting the epithelial-mesenchymal transition of MHCC97-H cell.
9.Protective effect of ligustrazine on lipopolysaccharide induced vascular hyperpermeability

Objective To investigate whether ligustrazine can reduce endotoxemia and inhibit the increase of lipopolysaccharide (LPS)-induced vascular permeability by suppressing Rho/ROCK signaling pathway in guinea pigs. Methods Eighteen of white male healthy guinea pigs were divided into three groups randomly and pretreated with intravenous injection of ligustrazine(3 or 6 mg/kg) or saline. 30 min later, LPS (100, 300, 1 000 μg/0.1 mL/site) and saline (0.1 mL/site) were administered intracutaneously at the dorsum of guinea pigs. Vascular permeability was determined by leakage area and absorbance of 610 nm (OD610) of Evans Blue dye after intracutaneous injection of LPS (100-1 000 μg/site). Results Dye leakage in the skin began to increase at 5 min, and revealed significant increase 2 h after LPS injection. Significant differences in the dye leakage area(F = 5.77, P < 0.05) and the OD610 (F = 7.736, P < 0.05) induced by 100, 300 and 1,000 ug/site of LPS administration were demonstrated between the control and ligustrazine groups (3 or 6 mg/kg). Conclusion Ligustrazine has protective effect on the hypermeability of endothelial cells induced by lipopolysaccharide, and inhibition of Rho/ROCK signaling pathway is a possible mechanism.
