Objective To investigate the expression and prognostic significance of inhibitor of growth 4 (ING4) and tail-type homeobox transcription factor 2 (CDX2) in colorectal cancer. Methods The expressions of ING4 and CDX2 pro-teins were detected by immunohistochemistry in 99 tissue samples of colorectal cancer and 30 corresponding para-cancer-ous normal tissue samples. The data of clinic outcomes were collected. The correlations between the expressions of ING 4 and CDX2 and clinicopathological parameters were also analyzed. Results The positive expression rates of ING4 and CDX2 were 68.8%and 72.7%in colorectal cancer tissues, which were significantly lower than those of corresponding normal tissue samples (93.3% and 96.7%, P<0.05). There were significant differences in the differentiation, depth of invasion, lymph node metastasis and tumor stage between expressions of ING4 and CDX2 (P<0.05). The 5-year survival rate was significant-ly lower in ING4 negative group (35.5%) compared with that of ING4 positive group (77.9%). The 5-year survival rate was significantly lower in CDX2 negative group (48.1%) than that of CDX2 positive group (70.8%, P<0.05). The expression of ING4 was positively correlated with the expression of CDX2 in colorectal cancer. Conclusion The expressions of ING4 and CDX2 are strongly associated with the carcinogenesis, development and prognosis of the colorectal cancer,which suggests that ING4 and CDX2 might be used as prognostic markers for the colorectal cancer.

Background:Acute pancreatitis can induce intestinal barrier dysfunction in its early phase,which is closely related with the progression and prognosis of the disease. Intestinal mucus layer not only serves as a physical barrier between pathogens and epithelium,but also plays a critical role in the maintenance of intestinal barrier function. Aims:To investigate the expression and role of mucin 2 (MUC2)in injured intestinal barrier in rats with acute necrotizing pancreatitis (ANP). Methods:Forty-two male Sprague-Dawley rats were randomly divided into two groups:the sham operation (SO)group and ANP group,which were induced via a retrograde injection of 3. 5% sodium taurocholate into biliopancreatic duct. Blood,pancreas and colon samples were obtained 6,12 and 24 hours after establishing the ANP model for determination of serum amylase and D-lactate (an indicator of intestinal permeability)and histopathological examination. PAS/ AB staining was used to observe the colon mucus layer and goblet cells,and the expressions of MUC2 and inflammatory cytokines in colonic tissue were detected by real-time PCR. Results:ANP models were successfully established. In ANP group,obvious colonic injury,increased intestinal permeability,thinner colon mucus layer,reduced mucin-containing goblet cells,down-regulated MUC2 mRNA and up-regulated tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β)and interferon-γ (IFN-γ)mRNAs were observed at each time point as compared with those in SO group (P < 0. 05). Spearman correlation coefficient revealed that MUC2 expression was negatively correlated with the intestinal permeability and expression of inflammatory cytokines in ANP group (P < 0. 05). Conclusions:Transcription of MUC2 is significantly down-regulated in colonic tissue of ANP rats,and might be associated with increased intestinal permeability and excessive expression of inflammatory cytokines in early phase of ANP.

Postmenopausal osteoporosis(PMOP)is a primary metabolic disorder of bone in postmenopausal women which is characterized by osteopenia, destruction of bone microstructure, increased bone fragility and fracture.PMOP is the major cause of joint pain and disability in middle-aged and elderly women.The research shows that early biomarkers can provide clinical evidence for early diagnosis of PMOP, assess fracture risk and evaluate treatment effect.In this paper, the latest advances in the application of PMOP biomarkers are described from the aspects of PMOP hormone correlation index and bone metabolism.

Objective: To observe the material basis and mechanism of action of Kai-Xin-San (KXS) in regulating antidepression of neurotrophic factors. Methods: KXS eluted by ethanol on macroporous resin was prepared. The antidepressive effect of different components was compared by tailing suspension test and forced swimming test of mice. The levels of nerve growth factor (NGF) and brain-derived neurotrophic factor (BDNF) in hippocampus were determined by ELISA. The rat astrocyte glioma C6 cell line and the rat adrenal pheochromocytoma PC12 cell line were used to evaluate the effects of different ethanol elution sites on the expression of NGF and BDNF and the differentiation of PC12 cells.Results: All of the ethanol elution components from KXS exerted anti-depressive effects by shorting the immobile time of tailing suspension and forced swimming of mice and 70％ ethanol elution components exerted best efficacy. This site also could increase expressions of NGF and BDNF on C6 glioma cell line. The 10％ ethanol elution site had the strongest ability to promote PC12 cell differentiation. Ginsenosides were the main effectuve ingredients for promoting the expression of neurotrophic factors. Conclusion: Regulation of neurotrophic factors might be the prominent action mechanism of KXS exerting anti-depressive effects.

OBJECTIVETo investigate the outcomes of autologous stem cell transplantation (ASCT) for patients with aggressive peripheral T-cell lymphoma (PTCLs) in advanced stage.

METHODSThe clinical data of 25 patients in complete remission (CR) with aggressive PTCLs received ASCT from May 1997 to June 2013 were retrospectively analyzed.

RESULTS① Of the 25 cases, 16 were unspecified PTCL (PTCL-U), 4 with angioimmunoblastic T cell lymphoma (AITL), 3 with anaplastic large cell lymphoma (ALCL) and 2 with hepatosplenic T cell lymphoma (HSTL), with a median age of 30(12-54) years old. Ratio of male to female is 16∶9. The distribution of stages was 8 cases with stage Ⅲ and 17 patients with stage Ⅳ. Nine patients presented with bone marrow involvement. Before ASCT, 18 patients were in CR1 and 7 patients were in CR2. ②Two patients with HSTL in stage ⅣB and IPI score 4/5 in CR1 relapsed and died within 12 months after ASCT. At a median follow-up of 38 (range 14-110) months, the estimated 3-year probability of PFS and OS for the other 23 patients was (63.1 ± 10.5)% and (71.8 ± 9.9)%, respectively. The patients in first CR had a better survival than the patients in second CR. The 3-year probability of PFS were (74.9 ± 11.0)% vs (33.3 ± 19.2)% (P=0.092) and OS were (80.2 ± 10.4)% vs (50.0 ± 20.4)% (P=0.043), respectively. The 3-year probability of PFS and OS were (40.0 ± 17.4)% and (53.3 ± 17.3)% in bone marrow involvement patients and the corresponding figure were (77.9 ± 11.3)% and (84.4 ± 10.2)% in non- bone marrow involvement patients.

CONCLUSIONASCT could improve the survival of aggressive PTCLs. Non CR1 status and bone marrow involvement had negative influence on OS in patients with aggressive PTCLs treated by ASCT. The prognosis was very poor in patients with HSTL and satisfactory regimens should be investigated.
