1.Purification and identification of recombinant human interferon-lamda 1

BACKGROUND:In 2003,a new family of interferon(IFN) III is discovered,that is,IFN-?s including three subtypes:IFN-?1,2 and 3.OBJECTIVE:To analyze the recombinant human interferon-?1(rhIFN-?1) expressed in Pichia pastoris,further more,to identify its physical and chemical characteristics,expression of receptors,and anti-tumor activities.DESIGN,TIME AND SETTING:The single sample observation was performed at the Department of Biochemistry and Molecular Biology,Institute of Basic Medical Sciences from September 2006 to July 2008.MATERIALS:The barms pAO815-4?F-IFN?1/GS115 contained rhIFN?1 expressive plasmids,human cervical carcinoma cell Hela and HCT116 cells were preserved in the state key lab for Molecular Biology from Institute of Basic Medical Sciences Chinese Academy of Medical Sciences METHODS:The Pichia pastoris contained rhIFN-?1 expressive plasmids was cultured and induced to express IFN-?1.The positive ion exchange chromatography and gel chromatography were used to purify the expression product.MAIN OUTCOME MEASURES:The physico-chemical characters of the purified product were tested.The product's IL-10R2 and IL-28R were investigated by RT-PCR and real-time quantitative PCR and its antitumor activity was tested by MTT analysis.RESULTS:The expressed product rhIFN-?1 yield reached to 9.23 mg/L,and its purity was near 90%.The isoelectric point of rhIFN-1 was 8.01,with molecular weight of 20 960.The expressions of IL-10R2 and IL-28R were detected in U937 cell.rhIFN-?1 could decrease the cell proliferation just like IFN-?2a,but this function was unclearly in the IL-28R cell.CONCLUSION:rhIFN-?1 can be highly expressed in Pichia pastoris,and its product has the physical and chemical characteristics consistent with natural IFN-?1,and the anti-tumor activity is similar to IFN-?2a,however,it is weak in decreasing the proliferation of relatively lowerintensity of receptor distribution in marrow-hematopoiesis system.
2.Predictive value of serum HDAC3,CysC and albumin levels on a large area of myocardial infarction

Objective To investigate the predictive value of serum histone deacetylase 3(HDAC3) ,cystatin C(CysC) and albu‐min levels on a large area of myocardial infarction .Methods According to whether heart failure and (or) cardiogenic shock occur‐ring during hospitalization ,102 patients with acute myocardial infarction(AMI) were divided into the two groups :the non - compli‐cating heart failure and (or) cardiogenic shock group(n= 63) and the complicating heart failure and (or) cardiogenic shock group(n= 39) .Then according to whether the creatine kinase(CK‐MB) peak value was greater than 200 IU /L ,102 AMI patients were di‐vided into two groups :CK‐MB peak values ≥ 200 IU /L group(n= 58) and the CK‐MB peak values < 200 IU / L group(n= 44) .The serum HDAC3 ,CysC and albumin levels were detected at early morning on 2 d of admission in all patients .Results Compared with the non - complicating heart failure and (or) cardiogenic shock group ,serum HDAC3 and CysC levels in the the complicating heart failure and (or) cardiogenic shock group were significantly increased(P< 0 .01) ,while serum albumin protein level was significantly in the occurrence of heart failure and(or)cardiogenic shock group .Compared with the CK‐MB peak value < 200 IU /L group ,serum HDAC3 and CysC levels in the CK‐MB peak value > 200 IU /L group were significantly increased (P< 0 .01) ,while serum albumin level was significantly decreased(P< 0 .01) .Conclusion Serum HDAC3 ,CysC and albumin levels have certain predictive value on a large area of myocardial infarction and conduce to judge the prognosis of patients .
3.Clinical efficacy of low-level laser therapy for androgenetic alopecia

Objective To evaluate the efficacy and safety of low?level laser therapy(LLLT)for androgenetic alopecia, and to compare the therapeutic effect of LLLT alone versus in combination with finasteride tablets. Methods Thirty?nine male patients were randomly divided into the LLLT group(n=21)and combination group(n=18)to be treated with LLLT alone or in combination with oral finasteride tablets(1 mg/d)for 6 months. LLLT was given twice a week, and each session lasted 30 minutes. All the patients were photographed and asked to fill a questionnaire about subjective symptoms, and hair density (the number of hairs per unit area on the scalp) was determined by using a dermatoscope to evaluate the grade of alopecia, before the treatment, and 3 and 6 months after the treatment. Results The hair density in the LLLT group was significantly higher after 6 months than after 3 months of treatment and before the treatment(184.59 ± 21.17 vs. 169.24 ± 29.21 and 166.67 ± 32.94 hairs/cm2, both P<0.05), but was insignificantly different between before and after 3 months of treatment (P > 0.05). The hair density in the combination group significantly increased after 6 and 3 months of treatment compared with that before the treatment(201.80 ± 16.55 and 186.39 ± 17.97 vs. 157.85 ± 27.97 hairs/cm2, both P < 0.05), and was significantly higher after 6 months than after 3 months of treatment(P<0.05). No significant difference was observed in hair density between the two groups before the treatment(P>0.05), but the combination group showed increased hair density compared with the LLLT group after 3 and 6 months of treatment(both P<0.05). After 6 months of treatment, LLLT alone was effective in 17 patients, but ineffective in 4 patients, and the combination therapy was effective in 16 patients, but ineffective in 2 patients, with no significant difference in the response rate between the LLLT group and combination group (χ2 = 0.057, P > 0.05). During the treatment, 24(62%)patients felt less greasy, and 22(56%)reported less hair loss, with no discomforts reported by the patients except local warm feeling. Conclusion LLLT is indeed effective for the treatment of androgenetic alopecia with no adverse reactions.
4.Clinical application of venous indwelling needle

Objective:Venous indwelling needle has been widely used in clinical treatment as a replacement of scalp acupuncture, and keeps constantly upgrading. Emergency use and nutrient supply by intravenous infusion through venous indwelling needle. The article discusses venous indwelling needle in clinical treatment for dermatological patients. Methods:It compares different complications through two different puncture and sealing methods of venous indwelling needle. Results:It summarizes highly possible complications during and after operating venous indwelling needle, and it proves that the latest puncture and sealing methods of venous indwelling needle could be better for those patients with skin damage, according to the different states of their skin and vein. Conclusion:For dermatological patients, their skin and vein could be protected effectively through the latest clinical methods of venous indwelling needle. The occurrence of many complications and patients’ pain could also be reduced.
5.Follicular Occlusion Triad:a Case Report and Pedigree Analysis

Objective To study the clinical features of follicular occlusion triad, and whether it is a hereditary disease. Methods Based on the clinical examination of a case who developed squamous cell carcinoma secondary to follicular occlusion triad, the pedigree of the patient was surveyed and analyzed. Results There were a total of thirteen patients in this pedigree, the age of onset was about 20 years old. The clinical features and laboratory examination of the proband was consistent with follicular occlusion triad. Conclusions Hereditary factor is important in the pathogenesis in follicular occlusion triad,and the disease maybe an autosomal dominant inherited disease.
6.Clinical efficiency and safety of thiazolidinediones on treatment of patients with nonalcoholic fatty fiver disease: A Meta analysis

Objective To evaluate the clinical efficiency and safety of thiazolidinediones (TZDs) on treat-ment of patients with nonalcoholic fatty liver disease (NAFLD). Methods The published controlled clinical trials about TZDs on treatment of patients with NAFLD from 1998 to 2008 were identified by searching in the full-text data base,such as Medline, PubMed, Ovid, EMCC, CBM Disc, CNKI, Wanfang, Weipu data and so on, as well as by searching of cross references from original articles and reviews. The enrollment standard was formulated strictly ac-cording to the designs of trials, and Meta analysis was performed by software package of Review Manage 4.2. Further-more, the statistic test, sensitivity and publication bias about the combined statistic data were revealed by referring to forest-graph and funnel-plot. Results All of the 473 patients in 18 controlled clinical trials were enrolled in this study. The weighted mean diffrerence of serum ALT, AST,TC,TG and BMI, HOMA-IR before and after treatment with TZDs 32. 24 (18.98-45.49) (Z=4.77,P<0.05),20.32 (2.30-38.35) (Z=2.21,P<0.05),0.26 (-0.01-0.54)(Z=1.88,P=0.06),0.63 (0.33-0.93) (Z=4.14,P<0.01),and -0.06 (-0.66-0.55) (Z=0.19, P=0.85),2.63 (1.33-3.93) (Z=3.96, P<0.01). In all papers, the improvement of hepatic tissue after treatment with TZDs in patients with NAFLD was observed in only 6 articles, which demonstrated the hepatic steatosis, inflammation and fibrosis [WMD (95% CI) 1.59 (1.08-2.10), 1.45 (0.90-2.00), 0.72 (0.12-1.32), Z= 6.08,5.16,2.33,P<0.05],especially the degree of NASH activity index,whose WMD (95% CI) was 4.21 (3.13-5.28) and Z=7.66,P<0.01, were improved significantly in the 104 NASH patients taken this kinds of drugs. There were 7 patients exiting from the all chnical trial, and the main adverse effects in patients accepting treat-ment with TZDs were debility, dizziness, edema of lower extremity, weight gain, indisposition of gastrointestinal tract, elevated liver enzyme and so on, which were similar to the control group. Conclusions The therapeutic effects of TZDs on NAFLD is confirmed. Not only are the serum liver enzyme,TC,TG and HOMA-IR in patients with NAFLD improved, but this new euglycemic agent has no acutely adverse effect in clinical application,which confirms the clin-ical efficiency and safety of TZDs on treatment of patients with NAFLD.

8.Comparison of multiple-injection and single-injection techniques ultrasound-guided anterior sciatic nerve block

Objective To compare the difference of ultrasound-guided anterior sciatic nerve block by multiple injection and single injection.Methods Among 90 patients undergoing lower limb operation,aged 18-68 years old,47 patients were males and 43 patients were females,all falling into ASA Ⅰ-Ⅲ.The patients were randomized to ultrasound-guided anterior sciatic nerve block by multiple injection (group M,n=45) single injection (group S,n=45).Local anesthetic of 0.5% ropivacaine hydrochloride 20 ml was used.The width,thickness and depth of sciatic nerve and blocking performance time were recorded.Sensory and motor blockade on the operated limb were evaluated every 10 minutes after injection of local anesthetic for 30 minutes and complications were observed in two groups.Results The width,thinkness and depth of sciatic nerve on the ultrasound screen were not statistically significant in the two groups.The performance time of group M was longer than that of group S (P<0.05).The proportion of patients with sensory blockade of each nerve significantly differed between the two groups: for superficial peroneal nerve at 10,20,and 30 minutes,for the sural nerve at 20,30 minutes,for the tibial nerve at 30 minutes,that of group M was higher than that of group S (P<0.05).A significantly greater rate of motor blockade was observed for the dorsal flexion of foot and toes extension in group M than that of group S at 30 minutes (P<0.05).The complications of nausea,vomitting and tinnitus were not different in two groups.Conclusion Multiple injection needs more operation time compared with single injection,but its onset time of sensory and motor blockade is quicker,the complications were not different.
9.Effects of ischemic preconditioning on oxidative stress and mitochondrial function in young and old rat myocardium with ischemia/reperfusion

[ ABSTRACT] AIM: To study the protective effect of ischemia preconditioning ( IPC ) on ischemia/reperfusion ( IR)-damaged myocardium in young and old rats .METHODS: Male Wistar rats aged at 3 months ( young ) and 20 months ( old) were used to establish myocardial IPC model and IR model with the method of Langendorff heart perfusion . The rats were divided into young ischemia/reperfusion ( YIR) group, young ischemic preconditioning ( YPC) group, old ischemia/reperfusion ( OIR) group and old ischemic preconditioning ( OPC) group.Transmission electron microscopy was used to observe the ultrastructural changes of myocardial tissue and myocardial mitochondria .The myocardial infarction area was determined by TTC staining .The lactate dehydrogenase ( LDH) content in coronary effluent fluid and the levels of su-peroxide dismutase ( SOD) and malondialdehyde ( MDA) in myocardial tissues were detected by the method of colorimetry . The levels of nitrated and carbonylated proteins in myocardial tissue were measured by ELISA .The myocardial cell apopto-sis was analyzed by TUNEL assay .The mitochondrial respiratory function and mitochondrial permeability transition pore o-pening induced by calcium load were evaluated by oxygen electrode method .RESULTS: Compared with YIR group , the myocardial infarction area in YPC group was obviously smaller , SOD activity in myocardial tissues increased , LDH activity in coronary effluent fluid and the content of MDA decreased , and the levels of nitrated and carbonylated proteins in the car-diac tissues reduced .In YPC group, the mitochondrial membrane structure appeared intact , cristae of the mitochondria showed close arrangement , and the matrix was compressed under the electron microscope .Myocardial mitochondrial respir-atory control rate , state Ⅲoxygen consumption and the P/O ratio in YIR group all significantly increased , proton leak de-creased, mitochondrial swelling induced by calcium distinctly reduced , and myocardial apoptosis rate declined .No signifi-cant difference of the above indexes between OIR group and OPC group was observed .Compared with YPC group , myocar-dial ultrastructural damage increased clearly , cardiac oxidative stress increased , mitochondrial respiratory function de-clined, and cell apoptosis and necrosis increased in OPC group .CONCLUSION:Ischemic preconditioning has protective effect against myocardial IR injury in young rat hearts , while old rat hearts were less sensitive to ischemic preconditioning , leading to bluntness of cardioprotection with IPC in aging hearts .This may be related to mitochondrial injury and severe cel-lular apoptosis caused by increase of cardiac oxidative stress levels in the aging ischemic preconditioning heart .
10.Expression increase of cyclooxygenase 2 in alcoholic liver disease

Objective To explore the effect of cyclooxygenase 2(COX-2) in the pathogenesis of alcoholic liver disease(ALD).Methods A model of ALD was developed with Wistar rats by intragastric alcohol.Liver samples were collected by the end of 4th,8th,12th and 16th week respectively.The concentrations of triglyceride(TG),oxygen free radical(ORF),superoxide dismutase(SOD),anti-superoxide anion(ASOA),6-ketone-prostaglandin F1?(6-K-PGF1?),thromboxane B2(TXB2) and malondiadehyde(MDA) in the liver homogenate were measured by biochemistry chromatometry.Pathology of hepatic tissue was examined by hematoxylin eosin,sudan Ⅳ and sirius red staining.The protein and mRNA expression of COX-2 in liver tissues were examined by immunohistochemical method and reverse transcription-polymerase chain raction(RT-PCR) respectively.Results By the end of 4th,8th,12th and 16th week,the content of COX-2 mRNA in liver tissue of model rats were 1.29,1.70,1.92 and 3.38 times the same as normal rats(P
