1.The effect of ultrashortwave therapy on the expression of BMP-2 in the early stages of steroid-induced avascular necrosis of the femoral head

Objective To observe the effect of ultrashortwave therapy on the expression of BMP-2 in steroid-induced avascular necrosis of the femoral head(SANFH). Methods Thirty New-Zealand white rabbits were randomly divided into two groups:a normal group(4 rabbits)and an intervention group(26 rabbits).An animal model of steroid-induced avascular necrosis of the femoral head was induced through the intravenous injection of horse blood serum and the intraperitoneal injection of methylprednisolone in the intervention group.X-ray examination confirmed the successful induction of SANFH in 20 of the rabbits,who were then subdivided at random into an uhrashortwave therapy group(1 0 rabbits)and a model group(1 0 rabbits).The rabbits in the ultrashortwave group were treated with ultrashortwave therapy in two 1 5-day courses,with a 5-day interval between them. Non-thermal ultrashortwave therapy was given in the first course,then mild thermal uhrashortwave therapy was given in the second.Those in the model group were reared without ultrashortwave therapy.All the animals were sacrificed immediately after the end of the last treatment,and their femoral heads were sampled to observe the expression of BMP-2 using real time fluorescent quantitative PCR. Results The expression of BMP-2 mRNA in the intervention group was only one fifth of that observed in the normal group.It was 2.7 to 10 times that observed in the model group.Both differences were significant at the 1% confidence level. Conclusion Ultrashortwave therapy can promote the expression of BMP-2 and improve reactivation in avascular necrosis of the femoral head.
2.Value of the procedure of percutaneous catheter drainage guided by ultrasound in treatment on deep perianal abscess

Objective To discuss the best method to treat deep perianal abscess and to investigate the clinical efficacy and value of the procedure of percutaneous catheter drainage guided by ultrasound in treatment.Ninteen patients with deep abscess underwent the procedure from Jan.2012 to May.2013.Methods Clinical data of 19 patients were analyzed retrospectively,including 16 male cases and 3 female cases.Results The average age is 39.8.All patients were cured without complications.The average length of stay is 7.8 days,a mean follow-up of 6.6 months after discharge,no patient with recurrence or fistula.Conclusion No specific performance of colorectal carcinoid,surgical resection is the most effective method of the treatment of colorectal carcinoid,the procedure should be individual treatment.However,the long term prognosis need to be explored in the future.

Chemical genetics is the science which takes the small molecular compounds as tools to solve the genetics problems or to disturb/adjust normal biological process so as to find out protein functions. Because the small molecules have the diverse chemical characters and the ability to identify the target proteins, they also can be filtrated on the basis of phenotype. So the methods of chemical genetics have been applied in almost all of the researches on biology and medicine. In this paper, the methods to acquire small molecular compounds are introduced. The enormous progress achieved in the field of combinatorial chemistry, which has allowed the rapid production of a large number of chemically diverse molecules, is an important prerequisite to make chemical libraries available to academic researchers. And the applications of the compounds in early embryo development, cell differentiation, on-set and course of disease are discussed, too. The application of small molecules has an enormous impact on our understanding of cell biology. There are many examples where small molecules, in combination with genetic screens, have facilitated the dissection of complex cellular processes.

5.Clinical observation of rimazolom for painless gastroscopy sedation in outpatients

OBJECTIVE To obser ve the efficacy and safety of rimazo lom for painless gastroscopy sedation in outpatients. METHODS Totally 84 patients who underwent painless gastroscopy were collected from the outpatient department of the Central Hospital of Enshi Tujia and Miao Autonomous Prefecture from March to June in 2021. By random number table method combined with envelope allocation concealment method ,they were randomly divided into observation group and control group ,with 42 cases in each group. The patients in the observation group were slowly injected with Sufentanil citrate injection 0.1 μg/kg+Rimazole toluenesulfonate for injection 0.2 mg/kg. Patients in the control group were slowly injected with Sufentanil citrate injection 0.1 μg/kg+ Propofol emulsion injection 2 mg/kg. Gastroscopy was performed after the patient ’s consciousness disappeared. The sedative efficiency,sedative onset time ,recovery time and the occurrence of adverse drug reaction were observed in 2 groups. The heart rate(HR),mean arterial pressure (MAP),pulse oxygen saturation (SpO2),modified observer ’s assessment of alertness/sedation (MOAA/S)score and Narcotrend score were recorded in 2 groups after entering the room (T0),after anesthesia induction (T1), when gastroscope entered the throat (T2),at the end of gastroscope withdrawal (T3),5 min after gastroscopy (T4). RESULTS There was no significant difference in the effective rate of sedation (100%),the incidence of respiratory depression , nausea and vomiting between the two groups (P>0.05). The qq.com onset time of sedation in the observation group was longer than control group ,and the recovery time and the incidence ofhypotension,hypotension to be tre ated,injection pain and bradycardia in observation group were significantly shorter or lower than control group (P<0.05). At T 0,there was no significant difference in HR ,MAP,SpO2,MOAA/S score or Narcotrend score between two groups (P>0.05). From T 1 to T 4,the HR of control group was significantly lower than that of the same group at T 0,and significantly lower than observation group at the same time(P<0.05). From T 1 to T 3,the MAP of two groups were significantly lower than the same group at T 0(P<0.05),but there were no significant differences between two groups and between T 4 and T 0(P>0.05). There was no significant difference in SpO 2 at different time points between two groups and HR at different time points in observation group (P>0.05). From T 1 to T 3,MOAA/S score and Narcotrend score of two groups were significantly lower than the same group at T 0,while the MOAA/S score and Narcotrend score at T 1 and T 3 and Narcotrend score at T 3 of observation group were significantly higher than control group at the same time (P<0.05),and the Narcotrend score of observation group at T 2 was significantly lower than control group at the same time(P<0.05);at T 4,there were no significant differences in MOAA/S score and Narcotrend score between two groups (P> 0.05). CONCLUSIONS Remazolam shows good sedative effect and safety for painless gastroscopy.
6.Propeptide-mediated protein folding: mechanism and its impact on lipase.

The formation of most proteins consists of two steps: the synthesis of precursor proteins and the synthesis of functional proteins. In these processes, propeptides play important roles in assisting protein folding or inhibiting its activity. As an important polypeptide chain coded by a gene sequence in lipase gene, propeptide usually functions as an intramolecular chaperone, assisting enzyme molecule folding. Meanwhile, some specific sites on propeptide such as glycosylated sites, have important effect on the activity, stability in extreme environment, methanol resistance and the substrate specificity of the lipase. Studying the mechanism of propeptide-mediated protein folding, as well as the influence of propeptide on lipases, will allow to regulate lipase by alternating the propeptide folding behavior and in turn pave new ways for protein engineering research.
