1.Effects of adenosine A1 receptor-induced delayed preconditioning on heart preservation and its relation with NF-?B

Objective To study the potential improvement of donor heart storage by A1 adenosine receptor-induced delayed preconditioning and its relation with NF-?B. Methods A total of 64 male Wistar rats were randomly divided into 6 groups. Group A was pretreated with 2-chloro-N6-cyclopen-tyladenosine (CCPA), and Group B was only injected saline. Twenty-four hours later, the hearts in groups A and B were stored in St.Thomas solution for 4 h at 4 ℃ and reperfused with K-H buffer for 1 h. Group C was administered with CCPA after 15-minute saline injection, and Group D was injected with inhibitor of nuclear factor ?B, pyrrolidindiethyldithiocarbamate (PDTC) first and 15 min later injected with CCPA. Group E was only injected with saline and Group F with PDTC. Twenty-four hours later, the rat hearts were isolated, perfused with H-K buffer solution on the Langendorff apparatus, and subjected to hypothermic ischemia for 180 min with intermittent perfusion of modified St. Thomas solution, and reperfused with 37 ℃ H-K buffer for 60 min. Left ventricular function, myocardial CK-MB leakage, tissue levels of adenosine triphosphate were measured. Objective To study the inhibitory effect of cyclooxygenase 2 (COX 2) inhibitor celecoxib on the growth of hepatocellular carcinoma cell (HCC) line SMMC 7721 in vitro . Methods Growth suppression was evaluated by MTT method. Apoptosis related changes in morphology were observed by fluorescence microscopy (FM) and transmission electron microscopy (TEM). Apoptotic index (AI) was counted by the TDT mediated dUTP biotin nick end labeling (TUNEL) assay. The apoptotic rate was quantified by flow cytometry (FCM). Results Celecoxib inhibited the growth of HCC cells in a dose dependent manner. 4.Impact Factors on Nosocomial Respiratory Tract Infections after Operations under General Anesthesia with Tracheal Intubation

OBJECTIVE To probe into the impact factors of the nosocomial respiratory tract infections after the operations with tracheal intubation under general anesthesia in order to establish the prevention and treatment strategies and provide the scientific basis.METHODS By descriptive epidemiological method and case-control study method,80 cases with nosocomial respiratory tract infections in nearly 3 years who had tracheal intubation under general anesthesia were selected,and another 180 patients with non-nosocomial respiratory tract infections with tracheal intubation under general anesthesia were selected as the control.RESULTS The incidence of the nosocomial infections from 5923 patients of the operation with tracheal intubation under general anesthesia from 2005 to 2007 accounted for 10.1%.The incidence of the respiratory tract infections of operations under general anesthesia accounted for 5.25%.The nosocomial respiratory tract infection from the tracheal intubation under general anesthesia toped the list and accounted for 52.0%.It showed seven impact factors on the nosocomial respiratory tract infection after the operation with tracheal intubation under general anesthesia by the multi-factorial Logistic regression,respectively,with significant differences(P1).CONCLUSIONS There are many impact factors that the nosocomial respiratory tract infection(especially lower respiratory tract infection) resulting from the operation with tracheal intubation under general anesthesia.The study shows the main risk factors: age,the complications of the disease,surgical condition,the ventilation time of tracheal intubationunder the anesthesia,post-operative recovery time,smoking status,the time that the operated patients start coughing after the operation.Therefore,the prevention is especially important.
5.Drug-resistance analysis on 46 strains of Pseudomonas aeruginosa in children

Objective To investigate the distribution status of Pseudomonas aeruginosa (PA) in children in Suining area and the changes of drug-resistance spectrum to provide the basis for clinical antibacterial drugs selection .Methods The ATB microbiologi-cal system was adopted to identify the bacteria and the K-B method was used to conduct the drug sensitivity test .The detection situ-ation of PA in our hospital from January 2010 to December 2012 was performed the statistical analysis on the distribution in various specialties of pediatrics and drug resistance .Results Among 46 isolated strains of PA ,39 strains were mainly distributed in the specialties of respiration ,neonatology and pediatric intensive care unit (PICU) ,which accounted for 84 .8 % ,22 strains were isolated from sputum specimens ,which accounted for 47 .83% .The isolation rate in last three years showed the rising trend year by year , and the resistance to antibiotics was increased year after year .Conclusion PA is one of the main pathogens of clinical infection .We should strengthen its drug-resistance monitoring and use antibacterial drugs reasonably .
6.Expression and Significance of Telomerase Activity in Cervical Cancer

Objective: Our aim was to study the role of telomerase activation in the course of cervical carcinogenesis and progression.Methods:Telomeric repeat amplification protocol(TRAP) assay was used to measure telomerase activity in tissue samples with various cervical conditions:40 with cervical cancer, 50 with cervical intraepithelial neoplasia(CIN), 20 with normal cervice. Results:The positive rate of telomerase activity was 95.0%,44.0%, and 10.0% in cervical cancer, CIN, and normal cervices, respectively, which was significantly higher in cervical cancer than that in CIN and normal cervices, so was that in CIN than that in normal cervices (P<0.01) . The positive rate was 22.2%, 37.5%, and 75.0% in CINⅠ,Ⅱ, and Ⅲ, respectively, which was significantly higher in CINⅢ than that in CIN Ⅱand CINⅠ (P<0.01).Conclusion:Telomerase activation may relate to cervical carcinogenesis, which correlates well with the grade of cervical lesions.
7.The prevention and treatment of postoperative complications after portaazygous disconnection in patients with portal hypertension

Objective To review our experiences in portaazygous disconnection for the treatment of portal hypertension and to analyze the causes of postoperative complications. Methods We reviewed the results of 236 patients with portal hypertension who were treated with disconnection from April 1994 to July 2002. Results Postoperative complications occurred in 65 of all the patients(the incidence rate was 27.5%). Twenty-four patients experienced postoperative infection(10.2%),12 patients suffered from intraabdominal massive bleeding(5.1%),12 from massive ascites (5.1%),8 patients suffered from recurrent upper gastrointestinal bleeding (3.4%),7 patients experienced acute thrombosis of portal venous system(3.0%). Two patients suffered from multiple organ dysfunction syndrome (1.0%). The operative mortality was 3.4%(8/236). The main causes of death included intraabdominal massive bleeding and severe infection with MODS. Conclusions The occurrence of postoperative complications was related with the selection of patients,thorough portaazygous disconnection and perioperative management.
8.Effect of ginkgo biloba extract on the expression of postsynaptic density-95 and content of neurotransmitter amino acid in chronic cerebral ischemic rats

Objective To observe the impact of ginkgo biloba extract in rats with chronic cerebral ischemia on the expression of PSD-95 protein and the content of amino acid neurotransmitter.Methods A total of 42 SD rats were divided into the sham group (n=12), the model group (n=14) and the ginkgo biloba extract group (n=14) by random number table method. Cerebral ischemia rats were produced by permanent ligation of bilateral common carotid arteries . The rats in the ginkgo biloba group were intrgastric administrated with ginkgo biloba extract suspension 28 mg/kg daily for 40 days, since 2 hours later after the surgery. The rats in the sham and model groups were intragastric administrated with equal-Volume nomal saline daily for 40 days, since 2 hours later after the surgery. The expression of PSD-95 protein was detected by immunohistochemistry techniques with image analysis. The content of Glu and GABA in the thalamus was determinated by OPA-pre-column derivatization and HPLC fluorescence detection method.Results The expression of PSD-95 protein (cortex was 212.58 ± 45.02vs.244.20 ± 34.28, thalamus was 132.33 ± 28.32 vs.272.00 ± 62.14) were significantly lower in the cortex and thalamus of the model group than those of the sham group (P<0.01). The content of GABA (6 081.46 ± 2 388.91 mmol/Lvs.8 280.45 ± 3 388.49 mmol/L) in the thalamus of the model group rats was significantly lower than the sham group (P<0.05). Ginkgo biloba extract could significantly improve the expression of PSD-95 protein (cortex was 237.89 ± 34.41 vs.212.58 ± 45.02, thalamus was 226.18 ± 75.80 vs. 132.33 ± 28.32) in the cortex and thalamus of chronic cerebral ischemia rats (P<0.01), and significantly improve the content of Glu and GABA (Glu was 10 523.78 ± 3 639.72 mmol/L vs.6 081.46 ± 2 388.91 mmol/L, and GABA was 18 440.93 ± 7 476.88 mmol/Lvs.11 239.83 ± 4 411.79 mmol/L) in thalamus with chronic cerebral ischemic rats compared with the model group rats (P<0.01).Conclusion Ginkgo biloba extract could regulate the levels of Glu, GABA and selectly regulate the PSD-95 experssion in the cortex and thalamus of cerebral ischemia rats.
9.Progress and prospect of electrospun silk fibroin in construction of tissue-engineering scaffold.

Silk fibroin is a natural macromolecular fibroin. It has broad prospects in tissue engineering application due to its good physical and mechanical properties and good biocompatibility. This paper reviews its chemistry structure, property, the usage as matrix of tissue-engineering scaffold using electrospinning technology, and the influence on growth, proliferation and function of vascular endothelial cell, smooth muscle cell, keratinocyte and fibroblast. It also addresses the advantages and disadvantages of silk fibroin applied in tissue engineering study like artificial vascular, skin, bone stent etc. The potential applications on esophageal tissue engineering and regenerative medicine were discussed.

10.Feasibility of 3D-printing template-assisted and CT-guided 192Ir interstitial brachytherapy in the treatment of recurrent gynecologic tumors

Objective:To investigate the accuracy and feasibility of 3D-printing individualized template-guided and CT-guided 192Ir interstitial brachytherapy in the central recurrent gynecologic tumors by comparing pre-plan and intraoperative physical dosimetric parameters. Methods:This study involved 38 patients with central recurrent gynecologic tumors who underwent 3D printing individual template (3D-PIT)-assisted and CT-guided 192Ir interstitial brachytherapy in the Department of Radiation Oncology of the Peking University Third Hospital from Jan 2018 to Dec 2019.The prescription doses for the target tumor areas were 10-36 Gy to be delivered at 5-6 Gy/fraction for 2-6 fractions.The pre-plan and intraoperative dosimetric parameters were compared, including the minimum prescription doses delivered to 90% and 100% of target volume( D90, D100)and the mean percentage of volume receiving 100% of the prescription doses ( V100). Meanwhile, the doses delivered to 2 cm 3 ( D2 cm 3) of organs at risk (bladders, rectums, and colons) were analyzed.The quality parameters of the brachytherapy were studied, including conformity index (CI), homogeneity index (HI), and external index (EI) of the target volume.Perioperative complications were also observed. Results:A total of 194 treatments were included.During the treatment, 5-13 (median 6) needles were inserted, with a prescription dose of 5-6 Gy per fraction.There were no statistical differences between pre-plan and intraoperative D90, D100, V100, CI, HI, and EI as well as the D2 cm 3 of bladders and colons at risk ( P>0.05). In contrast, for the D2 cm 3 of rectums, the intraoperative dose was slightly higher than the pre-plan dose, showing a statistical difference ( t=-0.335, P=0.027). Conclusions:The 3D-PIT-assisted and CT-guided 192Ir interstitial brachytherapy at a high dose rate is accurate and feasible in the treatment of recurrent gynecologic tumors, meeting the pre-plan dose requirement.
