As a transmembrane protein, β-amyloid precursor protein(β-APP) distributes extensively in the central nervous system, has the effect of neurotrophic, and neuroprotective, promote neurite growth and synaptogenesis, β-amyloid(Aβ) is the digestion products of its precursor-APP in the pathological conditions, and it is the main component of senile plaques-the main pathological changes of Alzheimer' s disease (AD), its toxic effects can also induce neuronal apoptosis, The expression of the two proteins after brain injuried has a close relationship with the injury, cognitive dysfunction, Alzheimer' s disease and the pathophysiological changes of central nervous system. To explore its expression in the brain after traumatic brain injury can determine the degree of injury, assess the prognosis and open up new avenues for the treatment of traumatic brain injury.
2.Optimal flow velocity of transcranial doppler in diagnosing and grading middle cerebral artery stenosis in comparison with magnetic resonance angiography

Objective To evaluate the best cutoff points of flow velocity for diagnosing and grading middle cerebral artery (MCA) stenosis as compared with magnetic resonance angiography (MRA) in a large group of asymptomatic patients. Methods We diagnosed 148 MCA stenostic patients with conventional method using EME-TC2000 transcranial Doppler (TCD) by a single reseach technician. All patients were given a MRA within 1 week after TCD examination. The severity of MCA stenosis was categorized as normal-mild (75% and void of flow signal) on the basis of the amount of signal loss and the lumen reduction of MCA in MRA . According to the different systolic velocities as following ≥130 cm/s, ≥140 cm/s , ≥150 cm/s, ≥160 cm/s, ≥ 170 cm/s, we drown different ROC(receiver operator characteristic) curves and determined which cutoff point was the best for diagnosing and grading the MCA stenosis.Results The numbers of vessel with normal, moderate and severe stenosis were 75, 112,109 respectively in MRA, their mean MCA velocities are (121?22) cm/s, (155?21) cm/s, (199?43) cm/s respectively in TCD, there was a obvious difference of systolic velocity among these three groups. Through calculating the bigest areas under ROC, we concluded that the optimal cutoff points for diagnosing MCA stenosis was 140 cm/s (areas under ROC was 0.87, P
3.Study of using urokinase for injection to lysis endometriosis ovarian cyst under sonography guided puncture

Objective To establish a method for lysis coagula rapidly within endometriosis cyst to ease aspiration difficulty while ultrasound-guided puncture and aspiration therapy. Methods Urokinase for injection with different concentration was used to lysis the coagula within endometriosis ovarian cyst with thick content. The effect of urokinase on the coagula, the wall of cyst and the aspiration rate of content were investigated. Results The area of coagula in unit area was significantly small in urokinase group compared with that in control group[( 95.6? 17.5)?m2 vs ( 1692.3? 1028.3)?m2,F= 28.86,P= 0.000]. Urokinase can lysis the coagula appending to the wall but did not change the permeability of cyst wall. The aspiration rate was significantly faster in urokinase group than in control group[( 2.79? 2.61)ml/min vs ( 1.42? 0.58)ml/min; t= 2.4572,P= 0.0121]. Conclusions Urokinase can resolve the coagula within endometriosis ovarian cyst and on the cyst wall safely and effectively, making it easy for cyst content aspirating and improving the coagulation effect of ethanol.
4.The preliminary research on the characteristics of the cognitive function and the mechanism in frontal bottom laceration patients

Objective To investigate the possible pathogenesis of the cognitive function in unilateral frontal bottom laceration by follow-up study in patients after one month of the onset. Methods MMSE, Loewenstein Occupational Therapy Cognitive Assessment (LOTCA), Montreal Cognitive Assessment (MoCA), Wisconsin Card Sorting Test (WCST) scales were used to evaluate neurocognitie function in 42 patients after one month of onset of unilateral frontal bottom laceration and 45 normal controls. The wave amplitude and the latency of the endogenous composition N2, P3 of P300 were measured at the cognitive potential instrument. Level of AChE was determined by ELISA and active AChE was analyzed by the ration analyses. Stepwise multivariate regression analyzed the correlation of the overall cognitive function and the lever and active of AChE. Results The cognitive test scores in patients were significantly worse than those in normal controls. The ability of recite sentences, fluency of words, reading, understanding language,cognitive transfering decreases in the left frontal bottom laceration patients (Group A, 23 cases), while the ability of attention, action, organization, graphics depicting, abstract epitoming, logical thinking were all seriously impaired in the patients with right frontal bottom laceration (Group B, 19 cases). The latency of the endogenous composition N2, P3 in patients ( Group A: (322. 4 ± 17.0), (410. 1 ± 19.9) ms; Group B:( 308.4 ± 15.6), (385.5 ± 17.4) ms) is more lengthen ( F = 4. 084, P = 0. 018; F = 3.467, P = 0. 038 )than the normal controls ( (268.6 ± 14. 7 ), ( 369. 2 ± 15. 4 ) ms) and the wave amplitude is lower ( F =2. 986 ,P =0. 047 ;F =3. 313 ,P =0. 041 ). The latency of N2 ,P3 in Group A of is more lengthen than Group B, while the wave amplitude is higher. The difference of the active of AChE in patients and control groups had no statistical significance, however, the level of AChE in two groups had statistical significance. The comparison of the active and the total AChE in patients has also not statistical significance. The correlation of the overall cognitive function has the linear regression with the parts of the brain and the level of AChE ( rY1.2 = 0. 584, P = 0. 039; rY2.1 = 0. 726, P = 0. 017 ). The standardized regression coefficients showed the level of AChE has the biggest influence to the overall cognitive function ( |Beta| =0. 3601, rY2.1 =0. 726).Conclusions AChE may be one of the important factors in the cognitive function after frontal bottom laceration. The specific damages of cognitive function in unilateral frontal bottom laceration patients closely relate with the lesion locations in the injured frontal bottom laceration.
5.Study on ConA promoting phagocytosis, cytotoxicity and producing effectors of macrophages

Objective To study how ConA actives macrophages in vivo to produce cytotoxic effectors and its phagocytic functions,and cytotoxicity. Methods ConA was intraperitoneally injected(ip). Cock red blood cells(cRBC) were used to evaluate MΦ phagocytic activity,and S180 cells as target cells to analyze MΦ dependent cytotoxicity(MTC).Nitric oxide(NO),TNF-α and IL-1 levels of MΦ cultural supernatant were measured using griess reagent,L929 cells MTT method and thymocytes proliferation test respectively. Results ConA could promote MΦ to phagocytize cRBC and kill S180 cells,enhance the production of such factors as NO,TNF-α and IL-1 by MΦ. There was significant difference compared with PBS control group(P<0.01). Conclusions ConA could stimulate MΦ to produce effectors, which mediate immune regulation of ConA to MΦ.
7.DHA promotes NGF-induced neuronal differentiation in PC12 cells via activating BMP pathway

Aim To investigate the effect of DHA on NGF-induced neuronal differentiation of PC12 cells and explore the possible mechanism via regulating BMP pathway. Methods PC12 cells were treated with 100μg·L-1 NGF and 100 μg·L-1 NGF + 10 μmol· L-1 DHA for 3, 6 and 9 days respectively. The length and number of neurite were detected by immunofluores-cenc. DHA content was analyzed by gas chromatogra-phy in all groups. The protein expression of BMP4, BMP7 , BMPR-II and p-Smad 1/5/8 was determined by Western blot. Results The length of total primary neurite in NGF+DHA groups was obviously increased, longer than that in NGF group; DHA content in 10μmol · L-1 DHA group was higher than that in the control group;NGF+DHA groups also unregulated the protein expression of BMP4 , BMP7 , BMPR-II and p-Smad 1/5/8 . Conclusion DHA promotes NGF-in-duced neuronal differentiation in PC12 cells, which may be associated with the upregulation of BMP path-way protein.
8.Growth-associated protein-43 gene modified adipose-derived mesenchymal stem cells protect against retinitis pigmentosa

BACKGROUND: Gene therapy and cell transplantation therapy, especially their combined use, have certain therapeutic effects on retinitis pigmentosa. However, little attention has been paid to the combination of gene therapy and cell transplantation in the treatment of retinitis pigmentosa.OBJECTIVE: To investigate the protective effect of adipose-derived mesenchymal stem cells (ADMSCs) modified by growth-associated protein-43 (GAP-43) gene on the retina of rats with retinitis pigmentosa. METHODS: ADMSCs from rats with retinitis pigmentosa were isolated and cultured, and then transfected with GAP-43 lentiviral vector. Sixty retinitis pigmentosa rats were randomized into experimental group (subretinal injection of GAP-43-transfected ADMSCs), control group (subretinal injection of ADMSCs) and sham operation group (PBS injection). The expressions of GAP-43 protein, Rho protein and GS protein in the retina of rats were determined by western blot. The outer nuclear layer thickness of the retina was determined by hematoxylin-eosin staining. RESULTS AND CONCLUSION: After transfected cell transplantation, the expression of GAP-43 protein in the retina was gradually increased with time and showed significant differences at different time post-transplantation (P < 0.05).The expression levels of Rho protein and GS protein in the retina of experimental and control groups were higher than that in the sham operation group (P < 0.05). The expression of Rho protein in the retina of the experimental group was higher than that in the control group (P < 0.05). The expression of GS protein in the retina of the experimental group was lower than that in the control group (P < 0.05). The thickness of retinal outer nuclear layer was ranked as follows: the experimental group > the control group > the sham group, and there was significant difference between groups (P < 0.05). These results show that the ADMSCs modified by GAP-43 gene have protective effect on the retina of rats with retinitis pigmentosa.
9.Transvaginal Contrast-enhanced Sonography in the Differentiation of Benign from Malignant Adnexal Masses

Purpose:The study was designed to investigate the feasibility of contrast-enhanced ultrasound in the differentiation of benign and malignant adnexal masses.Materials and Methods:Sixty-nine consecutive patients with adnexal masses received trans vaginal contrast-enhanced ultrasound.The image and perfusion features were assessed.Results:All of 26 malignant tumors showed detectable contrast enhancement,including 24 cases with a quick,heterogeneous or branching pattern.Among 39 benign lesions,24 were cystic with circle or half-circle enhancement,including 5 cases with intra-cystic septum or papillae slightly enhanced.The other 15 cases were solid,8 of them had slightly dotted enhancement.There are significant difference in enhancement patterns between benign and malignant masses ( P < 0.0001).The 4 cases of borderline tumors showed progressive,heterogeneous enhancement.Conclusion:Contrast-enhanced ultrasound is of value in the differentiation of benign and malignant adnexal masses.
10.Total tumor perfusion value of CT in hepatocellular carcinoma with the tumor volume and Child-Pugh classification

Objective To investigate the correlation between tumor perfusion parameters and tumor volume and Child-Pugh classification in CT of hepatocellular carcinoma (HCC).Methods Fifty-six patients with hepatocellular carcinoma were selected to perform CT perfusion imaging.The parameters of the total tumor perfusion such as hepatic artery perfusion (HAP),portal vein perfusion (PVP)and hepatic perfusion index (HAPI)were calculated according to the degree of hepatic encephalopathy,albumin,bilirubin, clotting time,ascites for liver Child-Pugh classification.The relationship between the tumor or peritumoral perfusion parameters with tumor volume and Child-Pugh classification were analyzed.Results (1)There was no correlation between tumor or peritumoral perfusion with the tumor volume.(2)The difference of HAP,PVP and HAPI between the different Child-Pugh classification groups was statistically significant (P<0.000 1).(3)With the reduction of Child-Pugh classification,the tumor body HAP and HAPI values gradually decreased, while the PVP value increased gradually.Conclusion There is no correlation between the tumor volume of hepatocellular carcinoma with total tumor perfusion parameters.The differences in perfusion measurements between different Child-Pugh classification can intuitively and quantitatively reflect the reserve function of the liver.
