1.Study of therapeutic efficacy of hyperbaric oxygen combined with chemotherapy on epithelial ovarian cancer
Chinese Journal of Postgraduates of Medicine 2010;33(33):21-24
Objective To discuss the therapentic efficacy of hyperbaric oxygen combined with PC program in newly diagnosed epithelial ovarian cancer patients. Methods Fifty-eight patients with epithelial ovarian cancer were divided into two groups by random digits table: HBO group(30 cases) and PC group(28cases). HBO group were exposed to hyperbaric oxygen of 2 standard atmospheric pressure 60 min, then given chemotherapy 25-30 min after extravehicular: cyclophosphamide 1000 mg/m2 + cisplatin 75 mg/m2.PC group with the same regimen without hyperbaric oxygen therapy were analyzed. The two groups were compared in the efficacy and 3-year survival rate, progression-free survival and adverse reactions. Results The total effective rate, not controlled rate, recurrence rate, recurrence time, 3-year survival rate in HBO group [83.3%(25/30),6.7%(2/30),33.3%(10/30), (21.0 ± 0.8) months,43.3%(13/30)] were better than those in PC group [67.9% (19/28), 17.9% (5/28), 46.4% (13/28), (18.0 ± 0.6) months, 17.9% (5/28)] (P <0.05), progression-free survival and overall survival time in HBO group were longer than those in PC group (P <0.05) and adverse reactions rate in HBO group was lower than that in PC group (P <0.05).Conclusions The hyperbaric oxygen combined with PC programs are better than PC programs in advanced epithelial ovarian cancer chemotherapy response rate, progression-free survival time and 3-year survival rates in ovarian cancer adjuvant chemotherapy. HBO can significantly reduce the PC's hematological toxicity and toxicity of the digestive system.

Objective: To investigate the effect of polypeptide extract from scorpion venom (PESV) on the matrix metalloproteinase2(MMP2)and matrix metalloproteinase9(MMP9)expression in leukemia-NOD/SCID mice, and the intervention mechanism of PESV in the multiplication and infiltration of leukemic cells thereof. Methods: In order to establish the animal model of outer marrow infiltration of human leukemia, bone marrow mononuclear cells of leukemia patients, irradiated 270 cGy on body by ~(137)Cs, were injected into NOD/SCID mice. The mice were randomly divided into five groups. The groups I, II and III were treated with different concentrations of PESV. Group Ⅳ was the model group injected by the normal saline solution. GroupⅤ was taken as control. The peripheral white blood cell count and blood smear were observed in groups. All of the mice were killed after four-week observation and MMP2 and MMP9 expressions were examined using Real timePCRmethod. Results: The expression levels of MMP2 and MMP9 were significantly lower in group I, group II and group III than that of the model group(P < 0.05). The expression levels of MMP2 and MMP9 were related to the concentration of PESV. Moreover, the peripheral white blood cell count and blood smear were more normal in mice treated with PESV than those of mice of model group. Conclusion: PESV inhibited the overexpression of MMP2 and MMP9 in leukemia-NOD/SCID mice, which significantly inhibited the multiplication and infiltration of leukemic cells.

This study is to observe the effects of sequoyitol on the expression of NADPH oxidase subunits p22 phox and p47 phox in rats with type 2 diabetic liver diseases. The model of high fat and high sugar diet as well as intraperitoneal injection of small dose of streptozotocin (STZ, 35 mg x kg(-1)) induced diabetic rat liver disease was used. After sequoyitol (50, 25 and 12.5 mg x kg(-1)) was administrated for 6 weeks, the contents of blood glucose (BG), alanine aminotransferase (ALT), aspartate aminotransferase (AST), total antioxidant capacity (T-AOC), hydrogen peroxide (H2O2), NO and insulin (Ins) were measured, liver p22 phox and p47 phox mRNA content was determined with real-time PCR and the expression of p22 phox and p47 phox protein was examined by Western blotting. In addition, pathological changes in liver were observed with HE staining. Sequoyitol could reduce the content of fasting blood glucose, ALT, AST, Ins and H2O2, restore insulin sensitive index (ISI) and weight, elevate liver tissue T-AOC and NO content, reduce the NADPH oxidase subunit liver tissue p22 phox and p47 phox mRNA and protein expression, as well as ameliorate liver pathologic lesions. The results showed that sequoyitol can ease the type 2 diabetic rat liver oxidative stress by lowering NADPH oxidase expression.

AIM: Sesamin (Ses) is one of the major lignans in sesame seeds with antihypertensive and antifibroticactivities, but its effect is weak. In this experiment, we combined vitamin E (VitE) and Ses to observe their effects on blood pressure and left ventricular fibrosis in spontaneously hypertensive rats (SHRs), and to explore its possible mechanism. METHODS: Male SHRs were randomly divided into SHRs+Ses+VitE group, SHRs+Ses group, SHRs+VitE group, and SHRs model group. Wistar-Kyoto (WKY) rats were setted as a normal control group. After 10 weeks of administration, systolic blood pressure (SBP) of rats was measured by tail cuff method, and the left ventricle was taken to calculate the organ index. Collagen deposition was observed by Masson staining, and cardiac collagen volume fraction (CVF) was analyzed. Expressions of collagen I, collagen III, transforming growth factor-β
