1.Lung resistance protein and multidrug resistance of gastric cancer

Lung resistance protein (LRP) is a gene which is found early and is closely related to multidrug resistance. LRP is expressed in most normal tissues with secretory function and is involved in secretion and cell detoxification function. LRP can also be seen expressed in malignant tumors including gastric cancer. There is a certain correlation between LRP expression and the prognosis of malignant tumor patients. LRP plays the role of multi-drug resistance mainly by preventing chemotherapy drugs into the nucleus and pumping chemother apy drugs out of the cells.
2.HPLC Determination of Bufogenin in Liuying Pills

Reversed high-pressure liquid chromatography was applied in content determination of bufogenin in Liuying Pills. The sample was extracted with chloroform. Using the calibration curve method of external peak area for quantitation, ODS as chromatographic column, 60% methanol as mobile phase, and detection wavelength at 298nm, the recovery reached 97.7%(CV=0.5%).
3.Optimization and identification of in vitro isolation and culture condition of human umbilical cord blood mesenchymal stem cells

BACKGROUND: Compared with the bone marrow mesenchymal stem cells, umbilical cord blood mesenchymal stem cells (UCB-MSCs) is an ideal source of tissue engineered seed cells, but the culture success rate is low.OBJECTIVE: To explore establish a stable reliable method to isolate and culture UCB-MSCs by optimizing medium choice,centrifugation speed and time, incubation density, choice of growth factor, first time of medium change.DESIGN, TIME AND SETTING: The cytological in vitro study was performed at the Department of Blood Transfusion, Bethune International Peace Hospital of Chinese PLA from January to October 2008.MATERIALS: A total of 20 samples of neonatal UCB by full-term uterine-incision delivery were supplied by Stem Cell Center,Bethune International Peace Hospital of Chinese PLA. Parturient and their family member signed the informed consent.METHODS: Under sterile conditions, UCB-MSCs were isolated by combination of density gradient centrifugation (1 500 r/min, totally 15 minutes) and different adherent time method. UCB-MSCs were incubated in DMEM/F12 medium, supplemented with 10% human UCB serum, 5 μg/L granulocyte-macrophage colony-stimulating factor (GM-CSF), 15 pg/L interleukin-3. MSCs at 1 ×1010/L were incubated in plastic flask coated with human UCB serum at 37℃ and 5% CO2 saturated humidity. The medium was changed following 3 days of culture. Non-adhered cells were removed. Subsequently, the medium was changed once every other 24 hours. When 80% confluence, UCB-MSCs were digested by the mixture of pancreatin-athylenediamine tetraacatic acid.MAIN OUTCOME MEASURES: Morphological changes of UCB-MSCs were observed by inverted phase contrast microscopy. Cell immunophenotypes were determined by flow cytometry.RESULTS: A small quantity of adherent round cells were determined after 24 hours, and adherent cells became more at 48 hours,with a few monopole spindle cells. Cell colonies were detected at day 7. Fibroblast-like cells arranged parallelly, presented whirlpool-shape and unclear boundary, with 80% 80% confluence 2-3 weeks following culture. At the second passage, these calls adhered at hour 12, and reached 80% 90% confluence at day 10. Flow cytometry showed that these calls were positive for CD29 and CD44, but negative for hematopoietic lineage marker CD34.CONCLUSION: MSCs can be successfully isolated from human UCB by using this modified method in vitro, with short culture cycle and high cell purification. Adherent cells have the same immunophenotype as bone marrow mesenchymal stem cells.
4.Effect of TRAIL on Expression of Multidrug Resistance Gene GST-π in Drug-resistant Human Gastric Cancer Cell Line SGC7901/VCR

Background:Multidrug resistance of tumor cells is one of the important factors that cause failure of chemotherapy in advanced gastric cancer. Tumor necrosis factor-related apoptosis-inducing ligand(TRAIL)may enhance the killing effect of chemotherapeutics on tumor cells,and reverse drug-resistant cell lines to sensitive cell lines,but its mechanism is not yet clear. Aims:To study the effect of TRAIL on expression of multidrug resistance gene glutathione S-transferase-π(GST-π)in drug-resistant human gastric cancer cell line SGC-7901 / VCR and the potential mechanism of TRAIL in reversing multidrug resistance of gastric cancer cells. Methods:SGC-7901 / VCR cells were treated with TRAIL in different doses (50,100,200 and 400 μg/ L)for 48 hours. After treatment,expression of GST-π mRNA in SGC-7901 / VCR cells and concentration of GST-π in culture supernatant were detected by RT-PCR and ELISA,respectively. Results:TRAIL could inhibit mRNA expression and protein secretion of GST-π in SGC-7901 / VCR cells in a dose-dependent manner within a certain range(≤200 μg/ L). The relative expression levels of GST-π mRNA in 50,100,200 and 400 μg/ L TRAIL groups were 0. 89 ± 0. 04,0. 77 ± 0. 08,0. 65 ± 0. 06 and 0. 61 ± 0. 03,respectively,and the concentrations of GST-π in culture supernatant in these groups were(57. 56 ± 1. 19)ng/ mL,(56. 30 ± 0. 80)ng/ mL,(31. 41 ± 1. 65)ng/ mL and (30. 80 ± 1. 34)ng/ mL,respectively,all were significantly lower than those in control group[1. 01 ± 0. 13 and(58. 62 ± 1. 38)ng/ mL,P all < 0. 05]. Conclusions:TRAIL may play a potential role in reversing multidrug resistance of gastric cancer cells through down-regulating GST-π expression.
5.Relationship between Functional Gastrointestinal Disorders and Psychological Factors and Related Treatment

Background:Functional gastrointestinal disorders(FGIDs)is a common disease of digestive system which severely affects the quality of life. Recently,more and more attentions have been paid to the relationship between FGIDs and psychological factors. Aims:To investigate the relationship between FGIDs and psychological factors and related treatment. Methods:A total of 270 patients with FGIDs admitted from Sept. 2013 to Sept. 2014 at Anhui No. 2 Province People’s Hospital were enrolled. Hamilton Anxiety Scale(HAMA)and Hamilton Depression Scale(HAMD)were used to evaluate the mental state of patients,and symptom severity of digestive system was scored. Patients with psychological abnormalities received conventional therapy to relieve digestive symptoms,and those who failed from conventional therapy were assigned into refractory group and others were assigned into response group. Patients in refractory group were further divided into conventional therapy group and combined therapy group for receiving paroxetine in addition to conventional therapy. Results:The rates of anxiety and depression were 61. 5%(166 / 270)and 57. 0%(154 / 270),respectively. The scores of HAMA and HAMD were positively correlated with the score of digestive symptoms(r = 0. 585,P < 0. 01;r = 0. 584,P <0. 01)in patients with anxiety and depression in response group. The scores of HAMA,HAMD and digestive symptoms in response group after conventional therapy were significantly decreased(P < 0. 05),while the scores of HAMA and HAMD in refractory group after therapy were significantly higher than those in response group before therapy(P < 0. 05). The scores of HAMA,HAMD and digestive symptoms in combined therapy group were significantly lower than those in conventional therapy group(P < 0. 05). Conclusions:Relationship is found between FGIDs and psychological factors,and antidepressants might contribute to the amelioration of symptoms of FGIDs.

7.Significance of microvessel density in bone marrow of patients with aplastic anemia

Objective: To investigate the changes and significance of microvessel density(MVD) in bone marrow of patients with aplastic anemia. Methods: 30 aplastic anemia cases,20 hyperplastic anemia cases,and 20 normal individuals′ paraffin-embedded bone marrow biopsys were stained with HE and CD34 by immunohistochemistry method respectively;and the vol% of hematopoietic tissue area in bone marrows were detected accordingly. Results:The difference between the MVD of HE and CD34 was not significant in aplastic anemia cases ,and the difference between the MVD of HE and CD34 was not significant in normal individuals either. While the MVD of HE was significantly lower than that of CD34 in hyperplastic anemia cases. The difference of the MVD between light and heavy pathogenetic condition in aplastic anemia cases was significant.The MVD of CD34 were significantly different among the cases of aplastic anemia, hyperplastic anemia,and normal individuals.The MVD showed significant linear correlation with the vol% of hematopoietic tissue area (r=0.74,P
9.STUDIES ON THE RELATION BETWEEN THE NUTRITIONAL STATUS OF LACTATING MOTHERS AND MILK COMPOSITIONS AS WELL AS THE MILK INTAKE AND GROWTH OF THEIR INFANTS TN BEIJING (III) THE LIPID COMPOSITION OF BREAST MILK

A nutritional survey of lactating mothers in different lactating periods was carried out in urban, suburban and rural districts of beijing in 1983-1984. Milk were collected, and its lipid compositions, including specific gravity, fat, cholesterol and fatty acids were analyzed. The results were as follows:1.The average of specific gravity of 207 milk samples were from 1.018 to 1.022. Though the value of the rural district was higher (1.022). The difference was rather small.2.216 milk samples were determined for the fat content (g%). The average of urban district was 3.78, of suburban 3.31 and of rural 3.08. Differ-eces between urban and other two districts were significant.3 .Milk cholesterol content was determined in 194 samples. The content in colostrum was the highest, 23.4mg/100g, but decreased gradually and steadied at about 10mg/100g in three months. No differece was observed among three districts.4.221 milk samples were analyzed for its fatty acid composition. The main components were oleic acid (29-37%), palmitic acid (17-25%) and lino-leic acid (12-25%), with 1-2% of erucic acid. The linoleic acid content was the highest in rural district.5.Statistical analysis showed that the correlations between milk fat and cholesterol content were significant in all the three districts, but the regression coefficient of urban district was significantly lower than that of rural district.6.There was significant correlation between fat intake of mothers and their milk fat content in total 188 samples, but no correlation was found when three districts were examined separately. All the results showed that the dietary fat intake of mothers might influence their milk fat content. The characteristics of fatty acids pattern of Chinese breast milk was the higher linoleic acid content as compared with other countries. The essential fatty acids in breast milk could meet the infant's need. The cholesterol allowance from breast milk was about 70-85mg per day.
