1.A Comparison of Three Diagnostic Criterions for Metabolic Syndrome Applied in Population of Xilinhaote City

Objective To compare the differences of three diagnostic criterions for metabolic syndrome (MS) proposed by the International Diabetes Federation (IDF) in 2005, the Third Report of National Cholesterol Education Program-Adult Treatment Panel Ⅲ (NCEP-ATPⅢ) in 2005 and Chinese Diabetes Society(CDS)in 2004 in population of Xilinguole city in inner mongolia. Methods 1 060 employees aged more than 20 yrs in enterprises taken health examination in Xilinguole hospital were investigated by epidemiological survey and the prevalence rates of MS and its components were calculated according to these three definitions respectively, and compared the agreement rates and difference between these definitions. Results The age-adjusted MS prevalence rates were 25.0%, 27.9% and 22.5% according to IDF (2005), ATPⅢ (2005) and CDS (2004) respectively. The agreement rate of diagnosis between IDF and ATPⅢ was the highest, and the Kappa value was 0.929. The kappa values between IDF and CDS,ATPⅢ and CDS were 0.726 and 0.763, respectively. There were 4.3% and 10.8% subjects respectively without MS diagnosed by IDF and CDS criterion presented at least 3 risk factors respectively. For Mongolia nationality people, when the waist circumference was 90cm in male and BMI was 25kg/m2 in female, the sensitivity and specificity were relative better, and the distance of ROC curve was the shortest, but for Han nationality, BMI≥25 kg / m2 in male and waist circumference ≥80 cm was better. Conclusion ATPⅢ definition can screen out the highest prevalence of MS and risk factors aggregation among three definitions, and it's best for screening for the high risk population of cardio-cerebrovascular diseases. The cutoff value of waist circumference (WC) in IDF and ATP Ⅲ seems better for Mongolia nationality men; while the cutoff value in CDS was better for Han nationality men. We should combine waist circumference and body mass index to judge the condition of obesity.
2.Establishment of Bacterial Endotoxins Test for Brozopine

Objective:To establish bacterial endotoxin test for brozopine. Methods: Interference pre-test and interfering factors test were conduced on 3 batches of samples from 2 manufacturers to confirm the applicability of bacterial endotoxin test and the non-in-terfering concentration. The bacterial endotoxin test was carried out based on the method described in the second part of Chinese Phar-macopeia (2010 edition) and relevant standards and guidelines. Results: The three batches of brozopine showed no interference in bacterial endotoxin test at the concentration less than or equal to 2. 5 mg·ml-1 . The bacterial endotoxin test of the three bathes of samples all met the requirements. Conclusion:Bacterial endotoxin test can be used for the quality control of brozopine.
3.RNF213 gene polymorphisms and susceptibility of intracranial vascular stenosis disease: a meta-analysis

Objective To investigate the correlation between RNF213 gene polymorphisms (rs112735431,rs138130613,and rs148731719) and the susceptibility of intracranial vascular stenosis disease.Methods The literature of studies on the correlation between RNF213 gene polymorphisms and intracranial vascular stenosis disease was collected according to the related databases.Using the Stata 12.0 software and selecting suitable genetic model,the heterogeneity was analyzed and the pooled odds ratio (OR) and its 95% confidence interval (CI) were calculated.Results A total of 12 articles were included after screening.The result of meta-analysis showed that the rs112735431 polymorphism had a significant correlation with the susceptibility of moyamoya disease (MMD) in all genetic models,especially the most significant dominant model (AA + GA genotype vs.GG genotype:OR 101.46,95% CI 59.41-173.27;P <0.001),at the same time,the polymorphism of this site also had significant correlation with the nonMMD intracranial large artery stenosis/occlusion (AA + GA genotype vs.GG genotype:OR 13.82,95% CI 4.48-42.61;P< 0.001);the rs138130613 polymorphism had significant correlation with the susceptibility of MMD in Chinese population (OR 5.01,95% CI 1.57-15.98;P=0.006);and no correlation between the rs148731719 polymorphism and the susceptibility of MMD was observed.Conclusions The RNF213 gene rs112735431 polymorphism is a susceptible factor of MMD,at the same time,the polymorphism of this site is also associated with the formation of non-MMD intracranial large artery stenosis.Systematic study on the molecular function of RNF213 may have important significance for diagnosis and treatment of such vascular stenosis diseases.
4.Effects of stem cell factor in combination with erythropoietin on erythropoiesis in patients with anemia associated with non-Hodgkin's lymphoma

Purpose:To explore the effects of stem cell factor(SCF) in combination with erythropoietin (EPO) on erythropoiesis in patients with anemia associated with non-Hodgkin's lymphoma (NHL). Methods:Bone marrow (BM) mononuclear cells (MNC) from NHL patients and normal simples were cultured for 7 consecutives days in the presence or absence of SCF, EPO, and SCF+EPO in liquid medium. 5.ASPS induces G_2/M arrest of H446 cells by activation of ERK signal pathway

Objective To investigate ASPS induced G_2/M arrest in lung cancer cell line H446 and its effect on ERK MAP kinase signal transduction pathways. Methods Cell cycle phases were inspected by flow cytometery (FCM) ; Western blot analysis was used to inspect the proteins of ERK, p-ERK. Results Compared with control group, G_2/M phase cells increased with concentration significantly, G_0/G_1 phase cells were not different, G_2/M phase cells and G_0/G_1 phase cells were not different when pre-incubated with PD98059 prior to exposure to ASPS of different concentrations, protein of p-ERK was significantly increased, expression of ERK was no different. Conclusion ASPS may induce G_2/M arrest of H446 cells possibly by activation ERK MAP kinase pathways.
6.Sampling investigation into the problems related to mental health in 1 498 adolescents in some cities and rural areas in Hubei province

BACKGROUND: The incidence of mental problems in adolescents is increasingly higher and has gradually become an important factor affecting their health and growth.OBJECTIVE: To probe into the mental health status and its influencing factors in adolescents in some cities and rural areas in Hubei Province.DESIGN: Clustering stratified random sampling, cross-sectional survey.SETTING: Department of Maternal, Child and Adolescent Health, Tongji Medical College, Huazhong University of Science and Technology.PARTICIPANTS: According to the selected investigation spots of National Surveillance of Constitution and Health of Students, we selected representative urban areas (Wuhan City and Huangshi City) and rural areas (Yun County, Xiantao City, and Xiaogan City) in Hubei Province as sample spots. Totally 1 498 adolescents, aged 16-18 years, were surveyed by clustering stratified random sampling. There were 605 adolescents in urban group and 893 in rural group.METHODS: The self-made survey scale was used to investigate general conditions, such as age, parents' occupation and educational level, family type, social atmosphere, school type and atmosphere, and interpersonal relationship. Symptom Checklist-90 (SCL-90) was used to determine the adolescents' mentalhealth status; 9 subscales (somatization, obsessivecompulsive, interpersonal sensitivity, depression, anxiety, hostility, phobic anxiety, paranoid ideation, and psychosis) were set as the observation indexes. SCL-90 is an inventory which is rated on a 5-point scale. Factor score of SCL-90 ≥ 3 or total score ≥ 160 was considered mental disorders. The family educational pattern was investigated through Egma Minnen av Bardndosnaupporstran(EMBU) by questionnaires. It contained positive rearing pattern and negative rearing pattern. Positive rearing pattern (namely, affectional warmth and understanding) contributes to mental health, while negative rearing pattern (e.g. punishment, firm control, excessive interference, partialism or rejection, denial and excessive protection) is adverse to mental health. The family environment was investigated with Family Environment Scale(FES). Four subscales (intimacy, conflict, intellectual-cultural orientation, and positive-recreational orientation) with high reliability and validity were set as indexes. All participants were given unified instructions and required to fill in anonymous questionnaires. The questionnaires were then collected by the teacher in charge of the class.Some indexes of adolescents in the urban and rural areas were observed including the score of SCL-90 and its relation with general conditions,parental rearing pattern and family environment quality. The total scores of symptom checklist 90 were set as dependent variables for stepwise regression analysis of related factors.MAIN OUTCOME MEASURES:①The incidence of mental health ,gender and regional difference of adolescents in some cities and rural areas in Hubei province; ②The score of SCL-90 and its comparison with national norm;③Correlation analysis of the total scores of SCL-90 with general condition and parents' educational type.RESULTS: ① The general incidence of mental disorders was 18.13 % in the 1 498 adolescents in Hubei province. It was 18.20% in boys and 18.04% in girls with no significant difference (P > 0.05). Regional differences: The incidence in rural areas [23.74% (212/893)] was significantly higher than that in urban areas [16.69% (101/605), P < 0.05]. Among all kinds of mental disorders, compulsion, interpersonal sensitivity, hostility,paranoid ideation, and depression were more common. ② Comparison with the national norm: The factor scores of SCL-90 and family environmentalquality were significantly lower that those of the national norm.③ Single factor analysis: The total score of SCL-90 was obviously correlated with age, gender, parents' educational level and expectation, and school atmosphere (P > 0.01). It was negatively correlated with positive rearing pattern (affectional warm, understanding) (P > 0.01), but positively correlated with negative rearing pattern (punishment, firm control, excessive interference,partialism or rejection, and denial). It had significant positive correlation with conflict and significant negative correlation with intimacy, intellectualcultural orientation and positive-recreational orientation(P > 0.01).④Multiple factor analysis: The mental health status of the students was closely related to age, gender, parents' educational level and expectation, interpersonal sensitivity as well as school atmosphere, parental rearing pattern,and quality of family environment.CONCLUSION: The incidence of mental problems in adolescents in some urban and rural areas in Hubei Province is relatively high. Score factors of SCL-90 are all higher than those of the national norm and the mental health level is poor. This has obvious correlation with family factors including age, gender, parents' educational level and expectation, and school atmosphere. Moreover, the quality of family environment and parental rearing pattern are the major factors affecting their mental health.
7.The effects of proinflammatory cytokines on expression of serotonin transporters in human enterocyte-like cell line

Objective To determine the effect of proinflammatory cytokines including interleukin-lbeta (IL-1β) and tumor necrosis factor-Alpha (TNF-α) on human serotonin transporter (SERT) in human enterocyte-like cell line Caco-2.Methods Caco2 cells were cultured for 5 days,then were divided into control group.IL-1β treated group (50 ng/ml) and TNF-α treated group (50ng/ml) for 2.24,48 and 72 hours.The expression of SERT mRNA was detected by RT-PCR at 2,24 and 48 hours and its protein expression was measured by Western blotting at 24,48 and 72 hours.Results The expression of SERT mRNA at 2,24 and 48 hours was lower both in IL-1β treated group (1.393±1.184,1.064±0.625 and 1.013±0.415,respectively) and TNF-α treated group (1.000±0.000,0.829±0.162and 0.945±0.147,respectively) in comparison with control group (2.282±1.367,1.586±0.421 and 1.86 ±O.496,respectively).There was significant difference among three groups (P<0.01).The expression of SERT protein at 24,48 and 72 hours was lower in IL-1β treated group and TNF-α treated group than in control group.Conclusions The IL-1β and TNF-α can inhibit the expressions of SERT mRNA and protein in Caco-2 cells,which indicates that IL-1β and TNF-α may change visceral sensitivity by influencing peripheral 5-hydroxytryptamine levels.
8.Regulation of histone acetylation and apoptosis by trichostatin in HL-60 cells.

In order to examine the strong anticancer action and low toxicity of Trichostatin A (TSA), the effect of TSA was examined on the growth inhibition, acetylation of histone H3 and apoptosis in HL-60 cells by employing MTT, immunocytochemical techniques, and Annexin-V-FITC/ PI assay. Our results showed that TSA could inhibit proliferation of HL- 60 cells in a time- and dose-dependent manner, and the IC50 at the 36th h was 100 ng/ml. The apoptosis-inducing effect of TSA on HL-60 cells was also time- and dose-dependent. But it didn't demonstrate apparent apoptosis induction in NPBMNCs within specific dose and time range. Both of the acetylation of histone H3 in HL-60 cells and NPBMNCs increased significantly (P<0.05) after treated with 100 ng/ml TSA for 4 h. However, there was no significant differences between the two groups (P>0.05). It is concluded that TSA can inhibit growth and induce apoptosis of HL-60 cells in a time- and dose-dependent manner, and is able to selectively induce apoptosis in HL-60 cells but does not respond in NPBMNCs under the same conditions. The difference of TSA between HL-60 cells and NPBMNCs can't be explained by the regulation of histone acetylation.

9.Expression and clinical significance of Survivin and PTEN in cervical carcinoma

Objective To study the expression and clinical significance of Survivin and PTEN in cervical carcinoma. Methods The expression of Survivin and PTEN in 60 cases of cervical carcinoma, 15 cases of CIN Ⅰ , 15 cases of CIN Ⅱ, 15 cases of CIN Ⅲ and 15 cases of normal cervical tissues were detected by immunohistochemical staining. Results The expression positive rates of Survivin were 86.67 %, 80.00 %,33.33 %, 20.00 % and 0, respectively, and those of PTEN were 21.67 %, 40.00 %, 80.00 %, 86.67 % and 100.00 % in cervical carcinoma, CIN Ⅲ, CIN Ⅱ, CIN Ⅰ and normal cervical tissues, respectively. The expression positive rates of Survivin were increased along with normal cervical epithelium, CIN Ⅰ, CIN Ⅱ,CIN Ⅲ and invasive carcinoma of cervix;while those of PTEN were inverse order.Compared with the expression positive rates of Survivin and PTEN protein in the CIN Ⅱ group, those in the CIN Ⅲ group had significant differences (P <0.01 and P <0.05, respectively). Expressions of Survivin and PTEN were correlated to the size of focus(P <0.05), the depth of tumor and pelvic lymphnode metastasis (P <0.05), but not to clinical staging, pathological types, pathological grading and age(P >0.05). There was a negative correlation between the expression of PTEN and Survivin in cervical carcinoma. Conclusion The expressions of Survivin and PTEN are correlated with invasion and metastasis of cervical carcinoma, and be related to clinical pathotology.Survivin and PTEN may play a important role in the formation, proliferation, differentuation and metastasis of cervical carcinoma. They may be used as markers of early diagnose, efficacy and prognosis evaluation.
10.Relationship of intestinal flora imbalance and immunoglobulin in patients with ulcerative colitis

Objective To investigate the relationship among the activity of diseases , the alteration of intestinal flora and the serum immuoglobulin (Ig) in patients with ulcerative colitis (UC) and to further evaluate the clinical value of fecal cue ratio in assessing humoral immune abnormalities. Methods Clinical data of 401 UC patients admitted in Peking University First Hospital from January 2013 to May 2015 were analyzed. According to Mayo index , patients were divided into inactive and active groups , and the active group was further divided into the mild, moderate and severe group. Serum immunoglobulin (IgG, IgM, IgA, C3 and C4) and fresh fecal bacteria cue proportion were measured. Statistical analysis was made to find the relevance among the above indexes. Results With the increase of disease activity , the intestinal flora imbalance aggravated (Spearmann correlation coefficient equals 0.295, P < 0.05) and serum IgM decreased significantly. However, serum C3 increased obviously. Serum C4 increased in mild to moderate group but decreased significantly in severe group. With the aggravation of intestinal flora imbalance , serum IgM and IgA decreased but C3 increased (P < 0.05). Conclusions The measurement of intestinal flora cue proportion, Ig, C3 and C4 may contribute to better determining the severity of diseases and provide beneficial information for therapy.
