1.Effect of Xiaoyao pill combined with low-dose corticosteroid on serum estrogens in menopause syndrome and its clinical efficacy

Objective To explore the effect of Xiaoyao pill combined with low-dose corticosteroid on serum estrogens levels in the treatment of patients with menopause syndrome and its clinical efficacy.Methods 96 patients with menopause syndrome from April 2013 to February 2015 in department of obstetrics and gynecology of Lishui second people’s hospital were selected and divided into 2 groups according to a random number table, the control group (48 cases) were treated with small dose of hormone therapy, the treatment group (48 cases) were treated with Xiaoyao pill combined with small dose of hormone therapy.The serum estrogens levels, Kupperman score, endometrial thickness and clinical efficacy were compared. Results The total effective rate of control group(79.17%) was lower than that of treatment group(93.75%)(P<0.05).Compared with control group post-treatment, the serum follicle stimulating hormone (FSH) and luteinizing hormone (LH) levels were lower, estradiol (E2) level was higher, the Kupperman score was lower, the endometrial thickness was higher than those of control group, there were significant differences ( P <0.05 ) . Conclusion Xiaoyao pill combined with small dose of hormone could improve serum E2, FSH and LH levels in the treatment of patients with menopause syndrome with high security.
2.The speciifc inhibition of siRNAs targeting 5' untranslated region of the enterovirus 71 genome

Objective To screen and identify effective small interfering RNA (siRNAs) targeting the conserved 5’ untrans-lated region (UTR) of the enterovirus 71 (EV71) genome. Methods Double-stranded siRNAs were designed to target the 5’UTR of the EV71 genome. The cytotoxicity effect of siRNAs on rhabdomyosarcoma (RD) cells was evaluated. The cytopathic effect on EV71-infected RD cells was observed under phase-contrast microscopy and the effective siRNAs were screened out by cell viability assay and real-time TaqMan RT-PCR assay. Results All the siRNAs did not exhibit any cytotoxic effects on the viability and growth of RD cells. Transfection of si-1 and si-2 targeting the 5’ UTR in EV71 genome into RD cells signiifcantly delayed and alleviated the cytopathic effects of EV71 infection, increased cell viability and reduced the EV71 RNA transcripts. And the inhibitory effect of si-1/si-2 on EV71 replication was sequence-speciifc. Conclusions The 2 siRNAs (si-1 and si-2) targeting the conserved 5’UTR of the EV71 genome may have broad spectrum antiviral effects on many epidemic EV71 strains.
3.Association of polymorphism in neurogenic differentiation factor 1 gene with type 2 diabetes.

OBJECTIVETo evaluate the role of polymorphism in the neurogenic differentiation factor 1(Neuro D) gene in Chinese patients with type 2 diabetes mellitus.
METHODSThe genotypes of codon 45 variant (GCC-->ACC) in the Neuro D gene were determined by mismatch PCR-restriction fragment length polymorphism assay in 448 Chinese, including 124 subjects with normal glucose tolerance and 324 patients with type 2 diabetes mellitus. The diabetic patients were divided into two groups cutting off with the age of 40 at onset.
RESULTSNo homozygote of the Ala45Thr variant was found in these subjects. The frequencies of AT heterozygous type were significantly higher in early-onset type 2 diabetic group than those in the control group and in the late-onset type 2 diabetic group (chi(2)=7.85, P=0.005; chi(2)=8.81, P=0.003). The frequencies of Thr45 allele in the early-onset type 2 diabetic group were significantly different from those of the control group (13.4% vs 5.2%, chi(2)=7.15, P=0.008) and the late-onset type 2 diabetic group (13.4% vs 5.8%, chi(2)=8.13, P=0.004). The presence of Thr45 allele was shown to have an association with early-onset type 2 diabetes (OR=2.52, 95% CI: 1.42-4.49). Furthermore, the subjects carrying the variant appeared to have lower serum concentration of C-peptide in diabetic group. However, the frequencies of polymorphism genotypes of Neuro D gene showed no difference between the late-onset type 2 diabetic group and the control group.
CONCLUSIONThe genetic polymorphism in the Neuro D is associated with the development of early-onset type 2 diabetes. The presence of Thr45 allele may represent a risk factor for early-onset type 2 diabetes among Chinese.

4.Effects of forced-air warming blanket on the quality of postoperative recovery and cognitive recovery in elderly patients undergoing laparoscopic radical resection of colorectal cancer

Objective:To investigate the effects of forced-air warming blankets combined with conventional warming strategies on the quality of postoperative recovery and cognitive recovery in elderly patients undergoing laparoscopic radical resection of colorectal cancer.Methods:This was a prospective, randomized, controlled trial.A total of 70 patients aged 65-75 years, who were classified as American Society of Anesthesiologists(ASA)grade-Ⅱ or Ⅲ, undergoing the laparoscopic radical resection of colorectal cancer under general anesthesia, were enrolled.Patients were randomly divided into two groups: the forced-air warming group(group FAW, n=35)and the conventional warming group(group CW, n=35). The inadvertent perioperative hypothermia(IPH), postoperative shivering, postoperative agitation, anesthesia recovery time and other postoperative complications were compared between the two groups.The post-operative quality of recovery scale(PQRS)was used to evaluate the quality of postoperative recovery and the recovery of cognitive function before and 1, 3, and 7 days after operation.Results:As compared with the group CW, the group FAW showed that the incidences of IPH, postoperative shivering and agitation were decreased(5.7% vs.22.8%, 2.8% vs.28.6%, 5.7% vs.31.4%, χ2=4.200, 10.057 and 7.652, P=0.042, 0.003 and 0.006), and the satisfaction degree of patients was increased at 48 hours after operation( P<0.01). The postoperative wake-up time was prolonged in the group CW as compared with the group FAW( P<0.01). Compared with the group CW, the proportion of patients with excellent overall recovery quality was increased in the group FAW 1 day after surgery( P<0.05). There was no significant difference in general recovery quality and cognitive recovery between the two groups at 3 and 7 days after operation. Conclusions:For elderly patients undergoing laparoscopic radical resection of colorectal cancer, the forced-air warming blanket combined with conventional warming strategies is more helpful to decrease the incidence of IPH, reduce postoperative shivering and improve the overall recovery quality at 1 day after operation, but no significant effect on postoperative recovery of cognitive function is found.
5.Role of long non-coding RNA ENST 933 in breast cancer and its underlying mechanism

Objective To investigate the regulatory mechanism of long non-coding RNA(lncRNA)ENST00000579933(hereinafter referred to as lncRNA ENST 933)on the development of breast cancer(BC).Methods The expression level of lncRNA ENST 933 in BC tissues as well as in BC cell lines(MCF-7,MDA-MB-231,MDA-MB-453)was detected by fluorescence-based real-time quantitative polymerase chain reaction RT-qPCR.The effects of lncRNA ENST 933 and miR-588 on the proliferation,migration,invasion and cell cycle in BC were determined using CCK-8 assay,clone formation assay,EdU,Transwell assay,flow cytometry,Western blotting and in vivo transplantation tumor assay.Finally,dual luciferase reporter gene,RNA immunoprecipitation assay,RT-qPCR,Western blotting and rescue assay were used to evaluate the interactions among lncRNA ENST 933,miR-588,and eukaryotic initiation factor 6(EIF6).Results The expression of lncRNA ENST 933 was up-regulated in both BC tissues and BC cell lines(P<0.05).Overexpression of lncRNA ENST 933 could enhance the proliferation,migration and invasion abilities of BC cells and promote the growth of transplanted tumors,while knockdown of lncRNA ENST 933 resulted in inhibited development of BC and arrested cell cycle(P<0.05).The expression of miR-588 was obviously down-regulated in BC tissues,and its overexpression inhibited cell proliferation,migration and invasion abilities in the BC cells(P<0.05).Moreover,the results of rescue assay indicated that lncRNA ENST 933 promoted the expression of target gene EIF6 by competitively binding with miR-588.Western blotting revealed that miR-588 decreased the phosphorylation of AKT and mTOR(P<0.05).Conclusion lncRNA ENST 933 promotes the progression of BC by modulating the miR-588/EIF6 axis.
6.Less common variants association study and statistical analysis

In the past decade,based on the “common disease-common variant” hypothesis,genomewide association studies (GWAS) have been extensively used to dissect the genetic components of complex diseases and quantitative traits.However,the identified disease-associated common variants by GWASs can only explain small part of the corresponding disease heritability.“Common disease-rare variant” hypothesis has been proposed to explore the missed heritability.With the development of the next generation sequencing technology,various association studies of less common variants are ongoing.This paper provides an overview of the study designs and statistical tests of these variants.
7.One case of repeated episodes of shaking head after general anesthesia of plastic surgery

A 49-year-old patient with breast atrophy and abdominal fat accumulation was admitted in the Plastic Surgery Hospital of the Chinese Academy of Medical Sciences on January 29, 2018. The patient turned her entire head quickly to the left and to the right each 90 degree without any obvious predisposing factor after general anesthesia. Each episode lasted 3 to 10 seconds and wasn’t associated with limbs and facial tissue twitch. She was consciousness clear after the onset without other obvious discomfort. Intravenous administration of sedative and analgesic drugs could not improve patient’s situation significantly, followed by psychological supporting treatment. The patient returned to normal about 3 hours after onset without giving any other special therapy. Except postoperative agitation, psychiatric disorders and the related histories of medication, the possible inducing factors include but not limited to epilepsy and hysteria. The etiology, pathogenesis, diagnosis and treatment of this case need further discussion. However, it is important that it gives clinical medical staff good suggestion which they should conduct a comprehensive analysis on the treatment of clinical problems, not only attach importance to organic lesion, but also pay attention to the psychological and mental factors of the patients. It will provide guidance for taking appropriate drugs and psychotherapy in order to achieve the best therapeutic effects.
