Clinical Medicine of China 2016;32(9):830-833
Objective To examine the expressions of hypoxia inducible factor?1α( HIF?1α) and trans?forming growth factor?β1 ( TGFβ1) in patients with endometrial carcinoma,to investigate their clinical signifi?cances. Methods Fifty?four patients with endometrial carcinoma and 20 patients with normal endometrium were selected in Yijishan Hospital of Wannan Medical College from2012 to 2015. Expression of HIF?1αand TGF?β1 were detected by immunohistochemical method respectively, the correlations between HIF?1α,TGF?β1 and clinic pathologic characteristics and prognostic significance were analyzed. Results The positive expression rates of HIF?1αand TGF?β1 were 59. 26%( 32/54) and 64. 81%( 35/54) in endometrial carcinoma respectively, signifi?cantly higher than of expression of patients with normal endometrium ( 5. 00%( 1/20 ) , 0 ( 0 );χ2 =17. 390, 24. 596;P<0. 01). The expressions of HIF?1α were correlated with FIGO stage(χ2=6. 712,P=0. 010),tumor thrombus in pulse tube(χ2=4. 804,P=0. 036),deep myometrial invasion(χ2=6. 484,P=0. 011),and lymph node metastasis(χ2 = 5. 732, P= 0. 017 ) . The expression of TGF?β1 were correlated with FIGO stage (χ2=4. 878,P=0. 027) ,deep myometrial invasion(χ2=4. 060,P=0. 044) and lymph node metastasis(χ2=5. 211, P=0. 022). Expression of HIF?1αwas positively correlated with expression of TGF?β1(r=0. 705,P=0. 000) . Conclusion Increased expressions of HIF?1αand TGF?β1 are all involved in carcinogenesis, progression and metastasis of endometrial carcinoma, which may be considered as a target in endometrial carcinoma therapy.

Objective To investigate the value of neutrophil-to-lymphocyte ratio in evaluation of prognosis in patients with early cervical cancer before radical resection. Methods A restropective study was performed in 76 patients who underwent radical resection for early cervical cancer in our hospital between 2007 and 2009. All patients were diagnosed by pathology. All patients did not accept neoadjuvant therapy , the median value of neutrophit-to-lymphocyte ratio (NLR) was 1.94 (0.73-9.31 in range). Based on this value of 1.94 as threshold, all patients were divided into 2 groups: a low NLR(≤1.94, n=38) group and a high NLR ( >1.94, n=38) group. Univariate and multivariate analyses were performed to assess the effectiveness of preoperative NLR to the prognosis in patients who underwent radical hysterectomy combined with pelvic lymph node dissection. Results The preoperative NLR was different significantly in the depth of stromal infiltration, lymphatic metastas, FIGO stage and pathological type between the low NLR group and the high NLR group. The high NLR group, lymphatic metastas, postoperative radiotherapy and FIGO stageⅡ were all risk factors for prognosis and disease-free survival in univariate analysis.Multivariate analysis revealed that NLR, lymphatic metastas and FIGO stageⅡwere independent risk factors for disease-free survival. FIGO stageⅡand lymphatic metastasis were independent risk factors for overall survivaI. Conclusion Preoperative NLR was found to correlate to unfavorable histopathologic features of cervical cancer. The preoperative NLR may be used as a useful and easy biomarker for disease-free survival in patients with cervical cancer, but not an independent risk factor for poor prognosis.

AIM:To investigate the effects of down-regulation of protein kinase C (PKC) on the activity of storeoperated Ca2 + channels (SOC) and the proliferation of airway smooth muscle cells (ASMCs). METHODS:Rat bronchial smooth muscle cells were isolated and cultured. Fluo-3 /AM fluorescence was measured by laser confocal microscope to assessing intracellular Ca2 +. Downregulation of PKC activity was achieved by incubation of ASMCs with PKC activator phorbol-12-myristate-13-acetate (PMA,10 ?mol/L) or phorbol 12,13 -dibutyrate (PDBu,1 ?mol/L) for 24 h. The proliferation of ASMCs was assayed by calculating the reduction rates of Alamar blue. RESULTS:Down-regulation of PKC activity by longterm exposure of PMA or PDBu inhibited the proliferation of ASMCs,the similar results were obtained by using PKC inhibitor chelerythrine. Both downregulation of PKC activity and inhibition of PKC activity by chelerythrine reduced Ca2 + entry through SOC channels. Low concentration of PMA (0. 1 ?mol/L) promoted the proliferation of ASMCs,and this effect was inhibited by SOC blocker SKF-96365. CONCLUSION:Inhibition or down -regu-lation of PKC activity results in the inhibition of SOC channels, suggesting that PKC is involved in the activation of these channels. Ca2 + entry through SOC channels might contribute to PKC-promoted proliferation of ASMCs.

Objective To investigate the clinical efficacy of Wumei Wan (Fructus Mume Pills) for diabetic gastroparesis with the syndrome of cold-heat mixture. Methods Sixty-eight diabetic gastroparesis patients with the syndrome of cold-heat mixture were evenly randomized into treatment group and control group. On the basis of treatment for controlling the blood glucose level, the treatment group was given oral use of Wumei Wan and the control group was given oral use of Cisapride. After treatment for one month, the therapeutic effect of both groups was evaluated, and the changes of gastrointestinal symptoms, gastric emptying, 2-hour postprandial glucose(2hPG), glycosylated hemoglobin(HbA1c), and fasting plasma glucose(FPG) in both groups were also observed. Results (1)The total effective rate of the treatment group was 88.24%, and that of the control group was 67.65%, the difference being significant(P<0.05). (2 ) After treatment , the scores of gastrointestinal symptoms of nausea and vomiting, gastric fullness, anorexia, belching and acid regurgitation, abdominal and gastric pain, diarrhea, and constipation in the treatment group were markedly improved(P<0.05 or P<0 . 01 compared with those before treatment ). In the control group, symptom relief was shown in nausea and vomiting, anorexia, and diarrhea(P<0.05). The treatment group had better effect on relieving nausea and vomiting, abdominal and gastric pain, diarrhea, and constipation than the control group(P<0.05 or P<0.01). (3)After treatment, 2hPG, FPG and HbA1c levels were obviously decreased in both groups(P<0.05 compared with those before treatment) , and the decrease in the treatment group was superior to that in the control group (P<0.05). (4)After treatment, half gastric emptying time, full gastric emptying time, and the contraction of gastric antrum were improved in both groups(P<0.05), and the improvement of gastric emptying in the treatment group was superior to that in the cont rol group(P<0.05). Conclusion Wumei Wan has better effect for the treatment of diabetic gastroparesis than western medicine Cisapride.

Objective To compare the impact on the results tested with arterial blood gas analysis and peripheral blood with glucose meter for critical patients in ICU in different temperature states. Methods The samples of venous biochemical test, blood arterial blood gas analysis and peripheral blood of 196 cases of critical patients in ICU were collected synchronously, and measure the D-value, correlation and bias adjustment factor of glucose blood tested with a synchronous fasting blood glucose test and venous/biochemical analyzer in different temperature states and different blood glucose groups, and the results of blood glucose test were analyzed. Results In normal temperature state, hypoglycemia simultaneous rapid intravenous glucose monitoring blood glucose level results compared with the lowest positive rate 3.31%(5/151), while the pairwise comparison showed there was both statistical signifcance between hypoglycemia group and target group (χ2=38.469), hyperglycemia group and target group (χ2=15.504) when choosing a synchronous fasting blood glucose test and intravenous blood glucose test (P<0.01). In high temperatures state, hypoglycemia simultaneous rapid intravenous glucose monitoring blood glucose level results compared with the lowest positive rate 0. There was both statistical significance between hypoglycemia group and target group (χ2=18.187), hypoglycemia group and hyperglycemia group (χ2=12.857) when choose a synchronous fasting blood glucose test and intravenous blood glucose test (P<0.01). Conclusions In high temperatures state, a synchronous fasting blood glucose test can not reflect the true value of blood glucose for critical patients.

Acta Nutrimenta Sinica 1956;0(04):-
Objective : To study the protective effects of two antioxidants on the influence of passive smoking on learning and memory ability of mouse offspring. Method: Passive smoking model of pregnant mice was established. Learning and memory ability was evaluated by water maze and long term potentiation (LTP). Nitric oxide (NO) content and nitric oxide synthase (NOS) activity of brain, vitamin E(VE) concentration and reactive oxygen species (ROS) of serum were determined. Results: Latency (the swimming time from beginning to endpoint) and errors (the number of entering blind end) in control and two antioxidant groups were shorter as compared with tobacco smoking (TS) group after 6 d in water maze test, and still shorter after 10 d in control and TS+VE groups. LTP was inhibited in TS group but increased significantly in two antioxidant groups. NOS activitiy was significantly higher in TS group in comparisonwith the control. NO content of TS+VE group was significantly lower than TS and TS+Q groups. Serum VE concentration and ROS level were correlated with the results of latency in water maze and LTP. Conclusion: Passive smoking of the pregnant mice may restrain LTP formation through disturbance of hippocampus function, and reduce the learning and memory ability of the offspring and VE may protect such effects.
