AIM: Tissue engineered periosteum fabricated by porcine small intestinal submucosa (SIS) combined with rabbit bone marrow-derived mesenchymal stem cells (BMSCs), are similar to the natural periosteum in the morphology, structure and function. The study was designed to explore the cell attachment in process of constructing tissue-engineered periosteum by different wetting methods to determine the most effective method of fabricating tissue-engineered periosteum. METHODS: The experiment was carried out in the Institute of Orthopedics, the Second Affiliated Hospital of Lanzhou University from September 2005 to March 2007.①Two New Zealand purebred rabbits, 20 days old, were used in this study.②Bone marrow was obtained from healthy rabbits, and BMSCs were isolated by using density gradient centrifugation for culture in vitro. After induction, BMSCs were seeded to SIS wetted with fetal bovine serum (experimental group) and pretreated with phosphate buffered saline (control group), to construct the tissue-engineered periosteum that was cultured for two weeks in vitro. ③The adhesion rates were calculated, and alkaline phosphatase (ALP) activity was detected after seeding. The growth, dissociation and proliferation of the cells were observed by inverted microscope and scanning electron microscope. RESULTS: ①At 5 hours and 12 hours after seeding, the adhesion rates were higher in the experimental group than in the control group [(57.34?2.23)%, (39.16?5.22)%, P

The effects of amino acid electuary on experimental liver fibrosis in rats with chronic liver injury induced by CCI4 were studied. The results showed that the electuary could prevent the decrease in body weight and food intake, reduce the levels of serum glutamic-pyruvic transaminase activities, elevate contents of total serum protein and serum albumin, normalize the serum ratio of BCAA to AAA and disordered amino acid pattern and suppress degeneration, necrosis and fibrosis of liver tissues in rats with chromc liver injury. These results indicate that the amino acid electuary has beneficial effects in prevention and treatment of liver fibrosis.

The effects of amino acid electuary (Ganan Elemental Granules) on experimental acute hepatic failure induced by D-galactosamine in rats were studied. The results showed that the electuary could significantly prevent the decrease in body weight and food intake, the prolongation in time of blood coagulation time and increase in the levels of serum glutamic-pyruvic transaminase activity, elevate contents of total serum protein and serum albumin, normalize the serum ratio of BCAA to AAA and disordered amino acid pattern, increase survival of animals and improve encephalopathy in rats with acute hepatic failure. These results suggest that the amino acid electuary has a better protective effects on acute hepatic failure.

AIM: To establish a new human endometial carcinoma cell line for basic and clinical study. METHODS: The minced tumor tissues were inoculated subcutaneously into the back of nude mice. After forming solid tumor, the mice were killed and the transplant was taken out for primery culture. RESULTS: The established cell line was maintained by serial passages, named EAC. EAC cell line grew rapidly, steadily, and it had similar microscopic morphology to adenocarcinoma cell. Chromosome analysis reveal EAC chromosome counts ranging in hyperdiploid. The expression of ER, PR, P53, MDM2 were negative by SP immunohistochemistry. Tumorigenicity of EAC in nude mice was 100%. CONCLUSION: A new human endometrial carcinoma, EAC, is established successfully, which is helpful for the basic and clinical study of endometrial carcinoma.
