Influences of the concentrations of luminol, zymosan & cells and opsonizing time of zymosan on luminol-dependent chemiluminescence( LDCL ) of peritoneal macrophages ( PM?) in rats were investigated by the method of orthogonal analysis. The result showed that the best levels were 100?mol/L of luminol, 10 mg/ml of zymosan, 5? 10~6/ml of cells and 45 min for opsonizing time. The LDCL of PM? was enhanced by incubation with TGP for 12 h in a concentration-dependent manner in the range of 0.09-2.25 ?g/ml of TGP, but LDCL was diminished with the concentration of 11.25 ?g/ml of TGP . The concentration-effect curve seems to be bell-shaped. The data suggested that TGP had two-sided regulatory effects on the LDCL of PM? in rats, depending on the concentration used.

Method for measuring interleukin 1 ( IL -1 ) and effects of total glucosides of paeony(TGP) on IL -1 production were studied. The results showed that the best concentration for LPS -induced IL-1 production of M s in rats was 8mg/L, the best dilution for IL-1 of LPS-induced M s production was 1 : 90. TGP ( 0 .5 - 12 .5mg/L ) enhanced LPS-induced IL -1 production in a concentration dependent manner, but diminished in the higher concentration of TGP ( 62 .5~l25mg/L ) , the concentration-effect curve seems to be bell-shaped. The data suggested that TGP had two-sided regulatory effects on LPS -induced IL production of M s in rats.

Effects of melatonin ( MT ) and supernatants from cultured pineal glands stimulated by 10 ?mol/L isoproterenol (ISO) for 0, 30, 60, 120 min on interleukin I ( IL-1 ) production by peritoneal macrophage ( PM? ) in rats were studied. Theses results showed that LPS-induced IL-1 production by PM? was enhanced by in vitro treatment with MT (0.1~1 ?mol/L ) , but IL-1 production was not influenced by MT without LPS. LPS-induced IL-1 production by PM? was enhanced in various degree by the supernatants, action of supernatants for 60min was best. The data is suggested that LPS-activated M?s may be one of target cells for MT.

Effects of supernatants from cultured rat pineal glands on ConA and LPS induced proliferative responses of splenocytes in mice, ConA induced interleukin 2 (IL-2) production of splenocytes in rats were studied. These results showed that ConA and LPS induced proliferative responses of splenocytes were enhanced by supernatants from cultured rat pineal glands stimulated with 10 ?mol/L isoproterenol (ISO) for 60 min (Dilution. 1 : 45 ) , ConA induced IL-2 production of splenocytes in rats was also enhanced by the supernatants (Dilution: 1 : 135 ) .

Effects of melatonin ( MT) on Con A or LPS-induced prolifera-tive responses of splenocytes in mice and Con A induced interleukin 2 ( IL-2 ) production of splenocytes in rats were studied. The results showed that Con A and LPS induced proliferative responses of splenocytes were enhanced by in vitro treatment with MT (0.01~1?mol/L and 0.01~10?mol/L, reinduced-pectively ) . ConA IL-2 production of splenocytes were also enhanced by in vitro treatment with MT (1~10?mol/L), but MT has no effects oa above functions of splenocytes without activating by ConA or LPS. These data suggested that mitogen activated lymphocytes might be one of the target cells for MT.

Objective To investigate the chemical constituents in the vines of Piper hancei for obtaining a more comprehensive understanding on its effective components. Methods Compounds were separated by column chromatography with silica gel and polyamide, and their structures were elucidated by spectral analysis and chemical evidence (IR, UV, MS, (~1H-NMR), (~(13)C-NMR)). Results Nine compounds were isolated from the chloroform extract fraction. Their structures were identified as: futoamide (Ⅰ), trichostachine (Ⅱ), retrofractamide A (Ⅲ), pipercide (Ⅳ), guineensine (Ⅴ), piperine (Ⅵ), piperettine (Ⅶ), piperovatine (Ⅷ), and (2E, 4E)-N-isobutyl-7-(3, 4-methylenedioxyphenyl)-hepta-2, 4-dienamide (Ⅸ). (Conclusion )Compounds Ⅱ-Ⅳ and Ⅶ-Ⅸ are isolated from the plant for the first time.

Effects of total glucosides of paeony ( TGP) on interleukin2 ( IL-2 ) production were studied with lL-2-dependent marine splenic lymohocytes. The results showed that IL-2 production by splenocytes was enhanced with low concentration of TGP (0.5-12.5 mg/L) , and diminished over the rang of 12.5-62.5mg/L. The concentration-effect curve seems to be bell-shaped. The data suggested that TGP hap two-sided regulatory effects on IL 2 production by splenocytes.

Objective:To evaluate the efficacy and safety of selective intra-arterial thrombolysis with urokirmse for acute cerebral infarction.Methods:Thirty-eight patients with acute cerebral infarction were treated by selective intra-arterial thrombolysis with urokinase within 6 hours of onset.Head CT seans were performed immediately and 24 hours after the procedures to find out whether intracerebral hemorrhage(ICH)had occurred.The activities of daily living and functional outcome of the patients were evaluated by the Barthel Index(BI)and the National Institutes of Health Stroke Scale(NmSS).Results:Of the 38 patients,21 achieved complete recanalization,8 achieved partial recanalization,3 occurred symptomatic intracranial hemorrhage,and l died.At day 90,22 patients had a favorable outcome(BI≥90),11(50≤BI＜90)had a better outcome,and 4 had a worse one(BI＜50);at 6 months,24 patients had a favorable outcome,9 had a better outcome.and 4 had a worse one.The NIHSS scores at day 14 after the procedures were significantly superior to the levels betore the procedures(12.68±7.43 versus 15.38±4.32,P＜0.011.Conclusions: Intra-arterial thrombolysis with urokinase can recanalize the occluded vessels and improve the acute clinical symptoms and long-term prognosis of patients.

Objective To investigate the effect of candesartan cilexetil on rotenone-induced Parkinson's disease (PD) in rats.Methods Forty 10-week-old male Lewis rats were chosen in our study and equally randomized into control group,rotenone group,rotenone+candesartan cilexetil group and candesartan cilexetil group (n=10); rotenone (2.5-3.0 mg/[kg· d]) was given for 4 weeks to the rats of rotenone group and rotenone+candesartan cilexetil group by subcutaneous osmotic minipumps implantation under the back; rats in the rotenone+candesartan cilexetil group and candesartan cilexetil group were orally administered candesartan cilexetil.Neurological behavioral measurements were performed to evaluate the motor features; tyrosine hydroxylase (TH) and α-synuclein immunoreactivities in the substantia nigra pars compacta (SNc) were observed.Protein level of α-synuclein was determined by Westem blotting.Results The weight of rats in the rotenone group reduced to (297.3±12.2) g,with significant difference as compared with that of the other three groups (P＜0.05); decreased TH immunoreactivity (377.0±41.6) cells/mm2) and increased α-synuclein immunoreactivity (0.75±0.02) in the SNc of rats in the rotenone group was noted,enjoying significant differences as compared with the other three groups (P＜0.05); these values in the rotenone+candesartan cilexetil group were (337.2±26.3) g,(639.7±46.0) cells/mm2 and 0.57±0.01,respectively (P＜0.05).Western blotting confirmed that rotenone up-regulated the expression ofα-synuclein in the SNc,and candesartan ceilexetil markedly attenuated the increase (P＜0.05).Conclusion Candesartan cilexetil can protect rotenone-induced PD in rats through decreasing TH-positive cell apoptosis and α-synuclein deposition.
