1.Expression of Caspase-3 mRNA in frontal cortex and hippocampus of chronic stress-induced depression rats treated by electro-acupuncture

ObjectiveTo observe the expression of Caspase-3 mRNA in prefrontal cortex and hippocampus of chronic stress-induced depression rats,and to detect the machnisms of antidepression by electro-acupuncture.MethodsSprague-Dawley rats were randomly divided into four groups:control group,model group,model +electro-acupuncture group and model + paroxetine group,12 rats in each group.Open-field test was used to observe the changes of movements,and real-time fluorescence quantitative PCR (RT-PCR) method was used to detect Caspase-3 mRNA levels in prefrontal cortex and hippocampus.Results ①Open-field test:after stress,compared with control group rats,the model group rats' crossing numbers (29 ± 7),rearing times (6 ± 2) were apparently less than those of control group( (66 ± 13),( 10 ±2) ; P<0.05,P>0.05).In comparison with model group,the crossing times and rearing times being increasing in degree in electro-acupuncture group( (61 ±9),( 13 ±1 ) ) and paroxetine group( (39 ± 10),(8 ± 1 ),P<0.01,P>0.05).② Compared with the control group,Caspase-3 mRNA in prefrontal cortex and hippocampus significantly increased in model group(P <0.05 ) ;and compared with model group,Caspase-3 mRNA in prefrontal cortex in electro-acupuncture group and paroxetine group significantly decreased(P < 0.05 ),and both expression of Caspase-3 mRNA in hippocampus in electro-acupuncture group and expression of Caspase-3 mRNA in hippocampus in paroxetine group decreased (P > 0.05 ).ConclusionChronic stress can increase the expression of Caspase-3 mRNA in prefrontal cortex and hippocampus of chronic stress-induced depression rats,while electro-acupuncture can decrease the expression of caspase-3 mRNA,which may be an important way to anti-depression by electro-acupuncture.
2.Construction of cDNA Expression Library of Unfed Female Haemaphysalis longicornis and Immuno-Screening

Objective To construct a cDNA expression library from unfed female tick Haemaphysalis longicornis for screening and cloning potential antigenic genes.Methods Total RNA was isolated from unfed female ticks,mRNA was purified and a library of oligo(dT)-primed cDNA with added directional EcoR Ⅰ/Hind Ⅲ linkers was constructed from the purified mRNA.The constructed cDNA was ligated to the EcoRⅠ/HindⅢ arms of the ?SCREEN vector.Pure phage stocks were harvested by plaque purification and converted to plasmid subclones by plating phage on host strain BM25.8.Recombinant plasmids that were subcloned to E.coli BM25.8 were isolated and transformed into E.coli JM109.Recombinant plasmids abstracted from JM109 were identified by PCR and sequencing.Rusults The recombinant phage DNA was packaged by using phage-marker packaging extracts,resulting in a primary cDNA library with a size of 1.8?106 pfu.Data showed 100% of the library were recombinant and the titer of the amplified library was 2.4?109 pfu/ml.Forty-two clones of encoding immunodominant antigens were obtained from the cDNA library.Sequence analysis revealed 12 unique cDNA sequences and the encoded putative proteins showed similarities to H.longicornis tropomyosin mRNA,Rhipicephalus annulatus unknown larval protein mRNA,chromosome 2R of Drosophila melanogaster,mitochondrial DNA of H.flava,clones HqL09 unkown mRNA and Hq05 mRNA of H.qinghaiensis,and myosin alkali light chain protein mRNA.Conclusion The cDNA expression library from unfed female H.longicornis was successfully constructed and screening of protective genes may provide candidate antigens of the tick.
3.A Clinical Observation of Xiaoji Decoction for Intermediate and Late Stages of Lung Cancer

ive] To observe the therapeutic effects of Xiaoji Decoction (XD) in treating intermed iate and late stages of lung cancer. [ Methods] One hundred and twelve patients with lung cancer in stage ? -? were randomly allocated to Group A, Group B and Group C. Group A ( n = 49) was treated with XD alone, Group B ( n = 33 ) was treated with various chemotherapeutic regimens according to the histo-logical types of lung cancer: CAP regimen for lung squamous carcinoma, EP regimen for lung adenocarci-noma and CE regimen for small cell carcinoma of lung, In Group C, 30 patients were treated with chemo therapy combined with XD. Symptomatic relief, life quality, tumor size, distant metastasis, sub-types of T lymphocytes, survival period and side effects in the three groups were evaluated after one course of treatment. [Results] The effects of XD on tumor size were evaluated by the effective rate and stabilized rate. 4.Low anterior laparoscopic resection of rectal cancer with specimen removal through a preventive ostomy incision

Objective:To evaluate the safety and feasibility of cancer tissue specimen delivery through a preventive ostomy incision during laparoscopic radical resection of rectal cancer .Methods:A total of 155 patients undergoing laparoscopic radical rectal cancer combined with prophylactic ileostomy at Peking University Third Hospital from Oct 2016 to Sep 2021 were retrospectively divided into two groups according to where the specimens were delivered through prophylactic colostomy incision (46 cases) or through newly made suprapubic incision (109 cases).Results:The by prophylactic colostomy incision delivery group had shorter surgery time [(243±66) min vs. (281±73) min, t=3.003, P<0.01] and shorter postoperative hospital stay [(7.5±2.2) d vs. (8.8±4.3)d, t=2.516, P<0.05], while there were no significant differences in intraoperative blood loss, intraoperative blood transfusion, postoperative first time of flatus, surgery-related complications and ostomy-related complications between the two groups (all P>0.05). Though the ostomy size in the prevention colostomy group was larger ( P<0.01), but there were no significant differences in the ostomy related complications between the two groups ( P>0.05). Conclusions:Laparoscopic radical resection of rectal cancer with specimen delivery through a preventive ostomy incision is of more aesthetic advantages without causing higher postoperative complications.
