1.Research progress of glioma stem cells and the related signal transduction pathways

Gliomas harbor a small population of cells termed glioma stem-like cells (GSC),which have the ability to undergo self-renewal and initiate tumorigenesis.GSCs are resistant to a wide variety of chemotherapeutic agents and possess a remarkable ability to recover from cytotoxic therapy.Furthermore,GSCs play a crucial role in RT failure,as tumors surviving RT are enriched in GSCs.Therefore,an alternative strategy involving selective targeting of this functionally distinct chemo-and radiation resistant small group of GSCs rather than the bulk of the tumor may be more successful in treating this deadly disease.GSC exhibit various alterations to signaling pathway activity,which are associated with self-renewal and neoplastic proliferation.These altered pathways may represent possible targets for GSC.
2.Therapeutic evaluation of the correction of the severe bi-maxillary protrusion cases by Tweed-Merrifield technique

Objective:To evaluate the influence of Tweed-Merrifield technique in correction of severe bimaxillary protrusion adult patients on the measurement of the dental and skeletal changes after orthodon-tic treatment by Johnston analysis and the regular cephalomatric analysis.Methods:Twelve adolescent patients with severe bimaxillary protrusion were included in this self-control retrospective study.Lateral cephalometric radiographs were taken before and after treatments.All the radiographs were traced and analyzed by the method of Johnston analysis.Other measurements were evaluated using a series of 1 3 li-near and angular measurements including SNA,SNB,ANB,U1 -SN,U1 -NA,U1 /NA,L1 -NB, U1 /NB,L1 /MP,U1 -L1 ,(U1 +L1 )/2-AB,MP/SN and MP/FH from regular cephalomatric analysis. These measurements were also applied to compare the differences between pre-and post-treatments,which clarify the dental and skeletal changes by Johnston analysis.The effect of orthodontic correction was de-termined using the non-parameters test.Results:The maxillary moved backforward by 1 .3 mm according to the stable skull base,while the mandible moved forward by 2.1 2 mm.The relative position between the maxillary and mandible (ABCH)changed 3.42 mm.The upper and lower incisors retracted signifi-cantly.The upper and lower molars moved slightly forward and the relative positions of upper and lower molars and anterior teeth after treatment were 3.44 mm and 4.23 mm respectively.After treatment,the parameters of ANB、U1 -NA、U1 /NA、U1 -SN、L1 -NB、L1 /NB and L1 -M were reduced by -(1 .98 ± 1 .55)°(P =0.01 2),-(5.08 ±4.6)mm (P =0.002),-(1 1 .79 ±1 .21 )°(P =0.004),-(1 3.55 ±6.32)°(P =0.047), -(3.1 7 ±3.07)mm (P =0.01 0), -(6.84 ±2.55)°(P =0.038)and-(4.1 3 ±2.24)°(P =0.048)on average,whose changes had the statistically significant effects.Con-clusion:Tweed-Merrifield technique (directional force technique)can stabilize anchorage molar,retract anterior teeth and significantly improve the hard and soft tissue profile for patients with bimaxillary protru-sion,and make a good vertical control which means this technique is applicable to the patients who need strong anchorage.Even for the severe bimaxillary protrusion adult patients,the Tweed-Merrifield tech-nique can control the anchoragewell and make the profiles improved greatly.
3.Diagnostic value of 18F-FDG PET/CT in the preoperative advanced gastric carcinoma comparing with multi-slice spiral computed tomography

Objective Evaluate the diagnostic value of PET/CT in the preoperative advanced gastric carcinoma comparing with mult;-slice spiral computed tomography. Methods Thirty-nine advanced gastric carcinoma patients taking PET/CT and abstract 40 advanced gastric carcinoma patients performing MSCT before operation,were done the TNM staging, and the Results were compared with the operation histopathology. Results For diagnosing primary lesions, regional lymph nodes, N3 lymph nodes ,PET/CT was accurate in 92.3% , 66.7% and 100.0%.Instead, MSCT was accurate in 82.0% , 50.0% and 62.5%. Conclusions (1) PET/CT and MSCT have high accuracy in diagnosing primary lesions and regional lymph nodes. But for detecting the N3 lymph nodes and distant metastasis, PET/CT has higher accuracy than MSCT. PET/CT combining abdominoscopy may decrease or avoid exploratory laparotomy.
4.Medial gastrocnemius muscle transferring reconstruction the patella tendon with long following-up

Objective To evaluate the long following-up outcome of the medial gastrocnemius muscle transferring reconstruction the patella tendon after the wide resection of aggressive bone tumors in the proximal tibia. Methods With the 69 patients of the osteogenetic sarcoma in the proximal tibia were treated with the wide resection and reconstruction the patella tendon. After the long following up the knee extensor,function and complications were evaluated. Results With the 69 patients, the 45 survival patients were followed up for the average 68.6 (24-128) months. The local recurrence rate was 8.7%(6/69). The strength of knee extending was in the average of grade 4.2(3.6-5.0), the degree of knee flexion was in the average of 95°(75°-135°), the degree of knee extension was in the average of-2°(0°-12°), the knees of five patients cannot fully extension. The MSTS functional score was in the average of 77% (23.1/30). Conclusion During the limb salvage of the proximal tibial aggressive bone tumors, the medial gastrocnemius muscle transferring reconstruction the patella tendon could offer the knee extension strength; improve the soft tissue coverage and functional results.
5.MELD and CTP scores in predicting short-term prognosis in patients with severe chronic hepatitis B

Objective To apply the model for end-stage liver disease(MELD)and the ChildTurcotte-Pugh(CTP)in predicting short-term prognosis of patients with severe chronic hepatitis B,and to evaluate their clinical value.Methods Data of 115 patients with severe chronic hepatitis B were retrospectively analyzed and all patients were divided into survival gnmp and fatal group by survival status at 3rd month of the treatment.The scores of MELD and CTP were obtained.The accuracies of MELD and CTP were evaluated by the receiver operating characteristic(ROC)curve.The difference between two systems was analyzed by Kaplan-Meier survival curve.Results MELD and CTP scores in fatal group were significantly higher than those in the survival group(t=4.891 and 3.949,P<0.05),and two systems were significantly correlated(γ=0.500,P=0.000).MELD and CTP scores were good for predicting the fatality rate of patients with severe chronic hepatitis B within 3 months(C-statistic:0.765 and 0.834,respectively),and there was no significant difference between the two systems(Z=1.516,P>0.05).Patients with CTP<10.5 or MELD<27.5 had longer survival time(Z:17.88 and 25.28,P=0.000)and higher survival rates(χ2=16.88 and 31.59.P=0.000)than those with CTP>10.5 or MELD>27.5.Conclusion MELD and CTP scores have similar predictive valHe for short-term prognosis of patients with severe chronic hepatitis B.and clinical data should be contained to better predict the short-term prognosis.
6.Biological behaviors of two novel syngeneic human osteosarcoma cell lines

Objective To characterize and compare the different biological behaviors of two novel human osteosarcoma cell lines, Zos and Zos-M, established respectively from the primary site and the skip metastasis of an osteosarcoma patient. Methods Two novel human osteosarcoma cell lines, Zos and Zos-M,were developed using tissue plant culture method. The vitro examinations included observations of morphology, analysis of karyotype and cell cycle, calculation of doubling time and growth curve, detection of osteoblastic markers and matrigel invasion assay. Subcutaneous, intratibial and intravenous inoculations into nude mice were performed to study the in vivo tumorigenicity and metastatic potentials of both cell lines.MTT were used to detect sensitivity of the cell lines to chemotherapeutic drugs. RT-PCR was performed to assess the expression of and some metastasis-related genes. Results Both cell lines proliferated actively and remained stable for more than 100 passages in vitro without interruption. The morphology and expression of osteoblastic markers of Zos and Zos-M were conformed to the characteristic of osteosarcoma. The karyotype analysis displayed aneuploidy and various structural abnormalities. The population doubling time of Zos and Zos-M were 33.65 h and 31.58 h respectively. Both cell lines were less sensitive to the current chemotherapy protocols compared to U-2OS. Zos and Zos-M were 100% tumorigenic by subcutaneous and othotopic injection. 37.5% of nude mice injected Zos-M and none of nude mice injected Zos developed lung metastasis after intravenous injection. The comparison of the expression patterns of some metastasis-related genes revealed that the decreased expression of cadherin-11 in Zos-M may correlate with a high potential of metastases. Conclusion The two novel established human osteosarcoma cell lines, Zos and Zos-M and related animal models could serve as models for the study of drug resistance and screening of new therapeutics for osteosarcoma. In addition, the study also provide tools for the study of metastasis because the same genetic background and different potential of metastasis of Zos and Zos-M.
7.Th1/Th2 cytokine balance in patients with severe chronic hepatitis B and its relationship with prognosis

Objective To investigate the balance of Th1/Th2 cytokines and its relationship with prognosis of severe chronic hepatitis B ( CHB ). Methods Peripheral blood samples were collected from 112 severe CHB patients, 30 CHB patients and 30 healthy controls. IL-4, IFN-γ levels and HBV DNA loads were measured by ELISA and fluorescent PCR, respectively. The levels of cytokines in different stages, and their correlations with HBV DNA loads and short-term prognosis were analyzed. Results Higher levels of IL-4, IFN-γ and Th1/Th2 ratios in peripheral blood were detected in patients with severe CHB than those with CHB and the healthy controls (Z = 8.968, 10. 004 and 26. 067, P =0. 009, 0. 007 and 0. 000). IL4 levels in patients with end-stage server CHB were markedly higher than those in other stages ( Z = 3. 672 and 3. 158, P= 0.000 and 0.002), while their Thl/Th2 ratios were lower (Z=3. 161 and 2. 166, P=0.002 and 0. 030). No significant differences on levels of IL-4, IFN-γ and Th1/Th2 ratios were observed in severe CHB patients with different HBV DNA levels (Z =4.431, 2.626 and 0. 140, P =0.219, 0.403 and 0. 987). Elevated IL-4 was closely correlated with the high case-fatality rate within 12 weeks. Conclusions The balance between Th1 and Th2 cytokines is- disturbed in patients with severe CHB. Thl/Th2 ratio decreases with the aggravation of diseases, which may indicate unfavorable short-term prognosis.
8.Recent results of concurrent chemoradiotherapy with lobaplatin and paclitaxel in advanced esophageal carcinoma

ObjectiveTo investigate the recent treatment result and the toxic responses of chemoradiotherapy with lobaplatin and paclitaxel in advanced esophageal carcinoma. MethodsFrom April 2010 to May 2011, 26 initial treatment patients withadvanced esophageal carcinoma were enrolled. The chemotherapy consisted of paclitaxel (135 mg/m2) on day 1 and lobaplatin (35 mg/m2) on day 2,and one cycle continued 21 days. Radiotherapy started on the second day after the fist cycle of chemotherapy. The median total dose was 60 Gy (2 Gy/Fraction,30 fractions,completed in 6 weeks).ResultsThere were 52 cycles in all and mean 2 cycles per patient.CR occurred in 7 patients (26.9 %),PR in 17 patients (65.4 %) and SD in 2 patients (7.7 %).The total effective rate was 92.3 % (CR+PR).The toxic responses mainly represented as following,myelotoxicity principally showed leucopenia at grade Ⅰ - Ⅱ occurred 46.1% (12/26) and at grade Ⅲ-Ⅳ 46.1% (12/26),thrombopenia at grade Ⅰ - Ⅱ occurred 30.8 % (8/26) and at grade Ⅲ-Ⅳ 7.7 % (2/26),hypohemoglobinemia at grade Ⅰ - Ⅱ occurred 42.3 % (11/26) and at grade Ⅲ 15.4 % (4/26).Baldness occurred 84.6 %(22/26). Pain of muscles or arthralgia occurred 69.2 %(18/26). Responses of gastrointestinal tract at grade Ⅰ - Ⅱ occurred 11.5 % (3/26).Radiation induced esophagitis occurred 88.5 % (23/26) and tracheitis 19.2 % (5/26).ConclusionThe toxic responses of concurrent chemoradiaotherapy with lobaplatin and paclitaxel in advanced esophageal carcinoma are tolerant, and the recent treatment result is good.Further study is needed.
9.Cinobufagin-induced apoptosis in osteosarcoma cells in vitro

Objective To study the growth inhibition,apoptosis induction effects of cinobufagin(CB)on human osteosarcoma(OS) cell line U2OS,MG63 and SaO2 in vitro and the underlying mechanism of action of cinobufagin in OS cells.Methods Cell viability was assessed by MTT assay.Cell-cycle status,apoptosis-inducing effects were evaluated by flow cytometry,fluorescent staining and DNA fragmentation assays.Inhibitors of apoptosis proteins (IAPs) and Bcl-2 family proteins including Bax,cleaved-PARP,xIAP,cIAP-1,survivin and p65 were tested by Western blot.Results MTT assay showed that CB could inhibited the growth of U2OS,MG63 and SaO2 cells in a dose-and time-dependent manner.The 48 h IC50 of CB on U2OS,MG63 and SaO2 cells were (104.83± 16.96) nmol/L,(47.07±7.5) nmol/L,and (136.72±10.08) nmol/L respectively.The induction of G2/M cell-cycle arrest was seen in the cells treated with CB.After cells were cultured for 12 h in the presence of 100 nmol/L CB,the percentages of cells in the G0/G1 phase were decreased,while G2/M phase were increased in U2OS,MG63 and SaOS2 cells,respectively.The results showed CB inhibited the proliferation of osteosarcoma cells through blocking the cell cycle in G2/M phase.Induction of apoptosis was confirmed by Hoechst 33258 and Annexin V/PI staining.After treating with 100 nmol/L CB for 48 h,the extents of apoptosis were 33.6%±6.4%,36.4%±7.8% and 29.3%±5.1%,respectively.These results indicate that the anti-tumor activity of cinobufagin in osteosarcoma cells was due to a G2/M cell cycle arrest and apoptosis inducing effect.Western blot showed that CB could induce the apoptosis related family proteins Bax,cleaved-PARP up-regulation,xIAP,cIAP-1,survivin and p65 downregulation in OS cells.Conclusion CB can inhibit the cell viability and induce G2/M cell cycle arrest and apoptosis in U2OS,MG63 and SaO2 cells.The apoptosis-inducing effect of CB is confirmed by the regulation of apoptosis related proteins IAPs and Bcl-2 in vitro.
10.Preliminary study on validity and reliability of the chronic HBV-infections related stigma scale

ObjectiveTo evaluate the reliahility and validity of the chronic HBV-infections related stigma scale.MethodsThe initial items and construct of the scale were developed according to theoretical analysis and interviews of experts and patients.A total of 151 patients with chronic HBV-infection were administered by convenient sampling method in this pilot study. The reliability and the validity of the scale were then evaluated.ResultsThe response rate of the scale was 94.5%.The Cronbach α coefficients of all dimeusions ranged from 0.75-0.87.The results of correlation analysis showed that there were higher correlation coefficients ( r ranged from 0.62-0.86) between items and their hypothesized subscales than those with other subscales ( r ranged from 0.14-0.55).The scale distinguished between patients with low subscale scores ( the subscale scores were ( 1.89 ±0.30 ),( 1.86 ± 0.29 ),( 1.96 ± 0.23 ),( 2.29 ± 0.45 ),( 1.59 ± 0.42 ) independently) and those with high subscale scores(the subscalc scores were (3.62 ±0.44),(3.99 ±0.41 ),(3.79 ±0.37),(4.13 ±0.34),(3.10 ±0.53 ) independently) (P < 0.01 ).Confirmatory factor analysis showed that the main indices of goodness of fit CFI was 0.94,NNFI 0.92,RMSEA 0.087.ConclusionThe chronic HBV-infections related stigma has good psychometric properties regarding to reliability and validity.
