1.The effect of an inhibitor of inducible nitric oxide synthase on the embryo development of mice

Objective: To determine whether aminoguaidine(AG),a selective inhibitor of inducible nitric oxide synthase (NOS), affect the embroy development of mice, and to study the mechanisms of iNOS affecting on pregnancy. Methods: AG(10 mg*kg-1*d-1 or 20 mg*kg-1*d-1) were injected subcutaneously to mice from day 3,7,14. kill mice at day19. The weight of fetus and placenta, the number of fetus and fetal resorptions were measured. Histological changes of placenta and umbilical cord were observed. Immunohistochemistry and NADPH histochemistry were adopted to study NOS activity of placental and umbilical cord respectively. Results: The conceptus in the uterus were resorbed in the early pregnancy. In the early and middle pregnancy the weight of pregnant mouse and the number of fetus of AG groups was decreased. The number of fetal resorptions was increased. But the weight of fetus and placenta were not affected. The expression of iNOS in the placenta were no difference significantly between the two groups with image analysis. Conclusion: AG inhibits the implantation and growth of embryo in the early and middle pregnancy, buy doesn't affect the formed fetus and placenta significantly.

Objective To evaluate which parameters of transvaginal color Doppler ultrasonography(TVCDU) can effectively reflect blood flow in ovarian tumors. Methods TVCDU was preoperatively used to determine all the parameters of blood flow in 60 women with epithelial ovarian cancer(EOC) and 36 with benign ovarian cystadenomas (control group). Microvessel density(MVD) was tested in the tissue samples by immunohistochemical method(SP method). The parameters of TVCDU in EOC were compared with those in control group.The correlation between MVD and the parameters in EOC group and control group was analyzed. Objective To observe the inhibitory effect of xiaoaiping injection and octreotide on H22 tumor-bearing mice and find the best drug concentration,then to explore its mechanism.Methods Establish a mouse H22 subcutaneous tumor model.After tumor the experiment animals were divided into normal control group,model group,Xiaoaiping low,medium and high dose group,octreotide group,and the group of XAP low,medium and high dose groups were combined with OCT.Calculate the tumor's volume and draw the tumor growth curve.Intraperitoneal injection for 14 days,Inhibitory rate was calculated; To observe its pathological changes by light microscope; The ratio.of CD3 + NK1.1-T cells,CD3-NK1.1 + NKcells,CD3 + NK1.1 + NK-Tcells in peripheral blood were measured by flow cytometry.Results Compared with the control group,H22 liver cancer in different treatment group had a certain inhibition effect on growth,The inhibitory effect of the combination group was better than single-agent group,High-dose Xiaoaiping + octreotide was best,Tumor model group compared with normal control group,The ratio of T cells,NK cells and NKT cells was significantly lower(P <0.05) ; T cells,NK cells and NKT cells after treatment in each group had some enhancement,High-dose Xiaoaiping + octreotide was the most obvious,the ratio of T cells,NK cells and NKT cells of the combination group was significantly more than the single-drug group and the same concentration of octreotide monotherapy Xiaoaiping group(P < 0.05).Conclusion High-dose Xiaoaiping + octreotide is the best drug for the inhibitory drug concentration.The inhibition of tumor growth may pass to improve the tumorbearing mice with immune status and enhance the body's anti-tumor capacity.

5.Expression of Insulin-like Growth Factor Binding Protein 2 in Glioma: Related to Prognoses

Objective To determine the levels of insulin-like growth factor binding protein-2 (IGFBP-2) in glioma tumor tissue and their clinical significance. Methods The levels of IGFBP-2 in glioma samples from 45 patients were detected with immunohistochemistry. Their correlation with tumor grade, p53 and Ki67 expression levels and the outcome of patients were tested. Results IGFBP-2 increased in glioma tissue in a grade-dependent manner, and significantly correlated with p53 and Ki67 expression. High IGFBP-2 level is significantly associated with earlier tumor recurrence and shorter overall survival. Conclusion Tumor IGFBP-2 levels can supply more prognostic information to the tumor grading system.
6.Purpura, abdominal pain and massive ascites

Objective To explore the clinical characteristics of Henoch-Sch(o)nlein purpura complicated with acute necrotizing acute pancreatitis,in order to provide information for the diagnosis and differentiation of Henoch-Scht(o)nlein purpura complicated with acute abdominal disease.Methods There was a case present with purpura,abdominal pain,massive ascites and poor spirit in Department of Pediatrics,the First Affiliated Hospital of Zhengzhou University.The clinical manifestations and physical examination results were summarized and discussed,some pediatric specialists from nephrology department,surgery department,digestive system department were invited to discuss the case.The treatment was adjusted according to result of discussion,the final diagnosis was tracked.Results The case was firstly diagnosed with Henoch-Sch(o)nlein purpura,acute diffuse peritonitis with cause in dispute.After the discussion,the intestinal necrosis,perforation could not be excluded,acute pancreatitis required further identification.Exploratory laparotomy was received because of the changing condition after discussion.The intraoperative diagnosis was acute necrotizing pancreatitis.The patient recovered well with medical treatment.Conclusion When children with Henoch-Sch(o)nlein purpura had an acute abdominal disease,acute necrotizing pancreatitis should be considered in addition to intestinal necrosis,perforation.
7.Curative effect observation of plasma eardrum perforation homochronous with palatoplasty in the treatment of chronic secretory otitis media.

OBJECTIVE:
To investigate the effect of plasma eardrum perforation homochronous with palatoplasty on hearing and tympanic cavity pressure in cleft palate children with secretory otitis media.
METHOD:
Fifty-six cases of cleft palate children with secretory otitis media were randomly divided into observation group and control group. For the control group, palatoplasty was carried out alone. For the observation group, homochronous with palatoplasty, plasma eardrum perforation was performed with the diameter of the aperture 2-3 mm, which was decided by observing viscosity of tympanum fluid under microscope or otoscope.
RESULT:
Compared with the simple cleft palate repairing palatoplasty, the perforation in the observation group could keep for 6-8 weeks after the surgery. Followed up 6 months later, the children in observation group improved better in terms of hearing restoration and tympanometry.
CONCLUSION
By means of plasma eardrum perforation homochronous with palatoplasty, the children of cleft palate with secretory otitis media could improve hearing and tympanometry in a short period, recovering the function of middle ear.

8.EXPRESSION AND BIOLOGICAL ACTIVITY ANALYSIS OF RECOMBINANT ADENOVIRUS EXPRESSION VECTOR FOR RECEPTOR TrkC OF NEUROTROPHIN-3

Objective To detect the expression and the biological activity of a recombinant adenovirus expression vector carrying human neurotrophin-3 (NT-3) receptor TrkC gene (Adeno-TrkC) in neural stem cells. Methods The expression of TrkC mRNA in 293 cells infected with Adeno-TrkC was detected by RT-PCR, and the expression of TrkC protein in neural stem cells infected by Adeno-TrkC was detected with immunocytochemistry and Western blotting. The effects of human neurotrophin_3 (NT-3) on the neural stem cells infected by Adeno-TrkC differentiating into neuron-like cells and astrocyte-like cells in vitro were observed. Results The transcription of TrkC mRNA and the expression of TrkC protein was detected in 293 cells and neural stem cells infected by Adeno-TrkC. This kind of TrkC was able to make more neural stem cells differenting into neuron-like cells in vitro with its ligand NT-3 and the percentage of neuron-like cell’s differentiation was 55^2% which was higher than that of other control groups.Conclusion TrkC protein is expressed in neural stem cells transfected with Adeno-TrkC and is able to make neural stem cells differenting into neuron-like cells. The present study may provide the basis on gene therapy of central nervous injury using further NT-3 and TrkC.
9.Early warning signs of severe preeclampsia

Objective To identify the early warning signs of severe preeclampsia (SPE). Methods A case-control (1: 2) observational study was conducted. Forty-seven pregnant women with SPE, who attended the prenatal clinics of Peking University Third Hospital regularly from Jan. 2002 to Dec. 2007, were selected as the study group, including 12 early onset and 35 late onset ones. The control group consisted of 94 healthy singleton pregnant women at the same period. Clinical data were collected and analyzed. Results (1) The basal body mass index (BMI) showed no difference between the study and control group [(23.27±4.31)kg/m2 vs (21.52±3.09)kg/m2, P>0.05]. (2) The net increase of BMI in the study group before the onset of SPE was higher than that in the control [(5.60±2.17)kg/m2 vs (4.85±1.52)kg/m2, P<0.05] and the increase of BMI per week was also higher [(0.74±0.41)kg/(m2*w)-1 vs (0.23±0.18)kg/(m2*w)-1, P<0.01]. The sensitivity and specificity of BMI increase per week in predicting SPE was 84% and 81% at a cut-off value of 0.39 kg/(m2*w)-1, respectively, and 79% and 91% at 0.41 kg/(m2*w)-1 correspondingly. (3) During the third trimester and before the onset of SPE, the weight gain per week in the study group was higher than that of the control [(0.93±0.70)kg vs (0.63±0.20)kg, P<0.01]. Significant difference was also found in the net weight gain between the two groups (P<0.01), but not in the percentage of women with excessive weight gain (>0.50 kg/w) [60%(25/42) in the study group vs 63%(53/84) in the control group, P>0.05]. (4) Higher percentage of women experienced pre-hypertension in the study group than in the controls [17%(8/47) vs 5%(5/94), P<0.01]. (5) In the study group, 53%(25/47) of the women had edema before SPE onset, but the figure dropped to 18% (17/94) in the controls(P<0.01). (6) Eight women in the study group and one in the control group suffered from hypoproteinemia before SPE onset with the average level of plasma albumin of (32.6±1.6)g/L and(38.4±2.1)g/L(P<0.01), respectively. (7) Proteinuria was reported in 10 cases (21%)in the study group and 4(4%) in the controls (P<0.01). (8) Logistic regression analysis showed that the risk factors for SPE included edema (OR=6.16,95%CI:2.29-16.57),pre-hypertension (OR=6.21,95%CI:1.56-24.77),proteinuria (OR=9.68,95%CI:1.86-50.30), and weight gain >0.85 kg/w during the third trimester (OR=11.60,95%CI:3.54-37.97). Conclusions Edema, excessive weight gain,pre-hypertension and hypoproteinemia are early warning signs of SPE. Pregnant women with the above signs required close monitoring during prenatal care.
10.Research progress in feeder layers of human embryonic stem cells

Human embryonic stem cell (hES cells) lines can be derived from the inner cell mass (ICM) of preimplantation blastocysts.hES cells are commonly defined as undifferentiated pluripotent cells that can proliferate and have the capacity of both self-renewal and differentiation into one or more types of specialized cells.hES cells remain undifferentiated when culture on feeder layers,such as murine embryonic fibroblasts (MEFs).It is believed that various factors secreted from feeder layers are necessary to prevent hES cell from differentiation.In this review,we will summarize the advantages and disadvantages of various types of feeder cells by which a reference for future research will be provided.
