1.Fast limit test of trace aconitine in Xiaohuoluo Pills

AIM:To establish a capillary electrophoresis with field-amplified sample stacking method for the separation and determination of aconitine alkaloid in Xiaohuoluo Pills. METHODS: An uncoated fused-silica capi-llary column(50 ?m i.d.?43 cm,effective length 35 cm) was used.The running buffer was 60 mmol/L phosphate electrolyte solution(pH=6.0)-acetonitrile(8(∶)2).The ruuning voltage was 10 kV and the capillary inlet was dipped in water for 5 s with 3.5 kPa prior to electrokinetic injection(12 kV,30 s).The detection wavelength was 235 nm. RESULTS: This method allowed 400 fold enrichment of aconitine alkaloid.A good linear relation was obtained in the range of 37.5~2.4?10~3 ng/mL (r=0.999 5),with the detection limit of 9.4 ng/mL.The average recovery was 97.6% with the RSD of 1.9%. CONCLUSION: The method is simple,rapid,specific and selective with high stacking efficiency;it provides a new reliable means for production and quality control of Xiaohuoluo Pills.
2.Comparison of clinical and MRI characteristics of pituitary macroadenoma between different genders

Objective To compare the clinical and MRI characteristics of pituitary macroadenoma between male and female patients. Methods Two hundred and thirty patients with pathologically diagnosed pituitary macroadenomas (104 mlaes and 126 females) were divided into two groups according to different gender. Patient age, the ratio of intratumoral hemorrhage and cavernous sinus invasion, and tumor volume were calculated in each group and compared between the two groups. The constitution of different pathological types was also compared. Results The age, ratio of intratumoral hemorrhage and cavernous sinus invasion, and tumor volume was (44.8±13.2) years old, 21.15% (22/104), 50.00% (52/104), and 6100.48 (mm3, median) in the male group and (44.8±11.9) years old, 28.57% (36/126), 48.41% (61/126) and 5037.05 (mm3, median) in the female group, respectiveluy. There was no statistical difference between the two groups regarding to patient age, the ratio of intratumoral hemorrhage and cavernous sinus invasion, tumor volume, and constitution of pathological types (P=0.972, P=0.197, P=0.811, P=0.189, P=0.093, respectively). Conclusion Pituitary macroadenomas show no gender preference in clinical and MRI characteristics.

4.The effects of 5-FU and L-Arg on the human liver carcinoma model in nude mice

Aim To observe the effect of 5-FU and L-Arg on the human liver carcinoma model in nude mice and study the mechanism.Methods We established the human liver carcinoma model in nude mice with BEL-7402 cell and gave normal saline,5-FU,5-FU+L-Arg by intraperitoneal injection. The tumor size was measured.The necrotic degree and range were observed by the microscope. Immunohistochemistry method and chemical colorimetry were performed to determine the expression and activity of iNOS. Nitrate reductase method was adopted to test the concentration of NO in the tumor tissue. The one-way ANOVA,Bonferroni and Kruskal-Wallis H tests were adopted for the statistical analysis.Results 5-FU combined with L-Arg promoted the tumor suppressive action apparently. The necrotic range enlarged. The expression and activity of iNOS increased. The production of NO also increased.Conclusions 5-FU combined with L-Arg has agonist effects on the inhibition of the growth of human liver carcinoma in nude mice.It may be related with inducing the synthesis of iNOS and the increase of NO.

6.Effect of health education improvement on promotion of timely vaccination among migrant children in community

Objective To explore the effect of health education improvement on promotion of timely vaccination among migrant children in community and to provide references for community health service center.Methods A total of 155 migrant children's parents were recruited from July 2015 to February 2016,and improved health education intervention was carried out on the basis of routine health education to promote parents' health belief on timely vaccination.The compliance of parents' participation in health education activities,the health belief of parents themselves on timely vaccination at different stages of intervention and parents' satisfaction,and the rate of timely vaccination of migrant children before and after intervention were evaluated.Results Regarding the compliance of health education activities for parents in 12 weeks,the first 3 weeks was 85% ~ 91%,the last 4 weeks were 95% and above.Six months after intervention,the score of parents' health belief on timely vaccination was higher than that after 3 months of intervention and before intervention,and the difference was statistically significant (P<0.05).The timely vaccination rates in the study group for migrant children was higher than that in the historical groups (P<0.05).After intervention,the satisfaction rate for parents was 95.484%.Conclusion Health education improvement could effectively promote parents' health belief on timely vaccination,satisfaction rate and rate of timely vaccination among migrant children.

Objective: To investigate the expression of histone deacetyase4 (HDAC4) in human liver carcinoma cell line Bel-7402 and to explore the regulatory effects of HDAC4 on the proliferation and differentiation of Bel-7402. Methods: Carcinoma cells Bel-7402 was treated with different concentrations of sodium phenylbutyrate (SPB), an inhibitor of HDAC4. Expression of HDAC4 mRNA in Bel-7402 cells was analyzed by RT-PCR before and after SPB treatment. Reverse microscope was used to observe the morphological changes of Bel-7402 cells. MTT assay and flow cytometry were adopted to describe the proliferation and cell cycle of Bel-7402 cells. Expression of P27 protein was determined by immunohistochemical method. 8.Expression of bone morphogenetic proteins and their receptors in the normal adult rat spinal cord

Objective To observe the expression distribution of bone morphogenetic proteins (BMP) in the spinal cord of normal adult rats. Methods Expression of BMP2, BMP4, and BMP7, and their receptors BMPR Ⅰa, BMPR Ⅰb, and BMP Ⅱ were detected by immunochemistry analysis in the spinal cord of normal adult rats. Results Expression of BMPR Ia or BMPR Ib was observed in the motor neurons of the anterior horn, sensory neurons of the dorsal horn, oligodentrocytes, some microglia, and some astrocytes. Expression of receptor BMPR Ⅱ was found in the oligodentrocytes and motor neurons in the gray matter of anterior horn. It was also expressed in some glial fibrillary acidic protein (GFAP)-positive astrocytes in the white matter but not in the gray matter. BMP2 and BMP4 were not expressed in the spinal cord of normal adult rats by immunohistochemistry. BMP7 was expressed in all the APC-positive oligodentrocytes, all the NeuN-positive motor neurons in the anterior horn, and some astrocytes in the normal spinal cord. Phosphated pSmad 1/5/8 protein was expressed in all the oligodentrocytes, all the neurons, and some astrocytes, especially in the GFAP-positive astrocytes which were RC2-positive radial glia in the subventricular zone.Conclusion BMP7, BMP receptors, and phosphated pSmad 1/5/8 are expressed in many types of cells whereas BMP2 and BMP4 are not expressed in the spinal cord of normal adult rats, which suggests an important function of BMP signal pathway in the neuron and glia of spinal cord.
9.An observation on clinical efficacy of Shenqi Fuzheng injection for treatment of patients with acute exacerbation of chronic obstructive pulmonary disease

Objective To investigate the clinical therapeutic effect of Shenqi Fuzheng injection for treatment of patients with acute exacerbation of chronic obstructive pulmonary disease (AECOPD).Methods A prospective clinical study was conducted. Fifty-eight consecutive patients with AECOPD were admitted in Departments of Respiratory Disease and Critical Care Medicine in Zhuozhou City Hospital of Hebei Province from January 2012 to December 2013. They were randomly divided into western medicine (WM) control group (28 cases, the routine treatment of WM) and integrated traditional Chinese medicine (TCM) with WM group (30 cases, on the basis of conventional therapy, Shenqi Fuzheng injection 250 mL intravenous drip was given once a day for a therapeutic course of 10 days). The duration of mechanical ventilation, the successful rate of weaning from ventilator, the rate of using ventilator again after weaning, the length of stay in intensive care unit (ICU), and mortality were recorded respectively in the two groups. Before and after treatment, the arterial blood gas analysis, acute physiology and chronic health evaluation Ⅱ (APACHE Ⅱ) score, clinical pulmonary infection score (CPIS), pulmonary function and dyspnea score were evaluated. Results Compared with the WM control group, the duration of mechanical ventilation (hours: 104±16 vs. 125±24) and the length of stay in ICU (days: 6.3±2.1 vs. 7.2±3.6) were significantly shorter, the rate of successful weaning from ventilator was obviously higher [73.3% (22/30) vs. 60.7% (17/28)], and the rate of using ventilator again after weaning was remarkably lower [13.3% (4/30) vs. 28.6% (8/28)] in the combined TCM and WM group, the differences between the two groups being statistically significant (allP < 0.05); the mortality was lower in the combined group [10.0% (3/30) vs. 10.7% (3/28)], but there was no statistically significant difference (P > 0.05). Compared with those before treatment, the pH value, arterial partial pressure of oxygen (PaO2), forced expiratory volume in 1 second (FEV1), forced vital capacity(FVC) and the ratio of FEV1/FVC were all significantly higher in the two groups after treatment, while the partial pressure of arterial carbon dioxide (PaCO2), APACHE Ⅱ score, CPIS score, residual volume/total lung capacity (RV/TLC), and the dyspnea score were all lower in the two groups after treatment, the more obvious changes in levels being after 10 days of treatment in combined TCM and WM group [pH: 7.44±0.04 vs. 7.40±0.08, PaCO2 (mmHg, 1 mmHg = 0.133 kPa): 59.1±11.9 vs. 68.1±12.4, PaO2 (mmHg): 70.5±6.9 vs. 65.1±7.4, APACHE Ⅱ score: 14.5±4.2 vs. 17.4±2.2, CPIS score: 5.3±2.4 vs. 7.6±1.4, FEV1 (L): 1.60±0.47 vs. 1.54±0.34, FEV1/FVC: (65.33±2.65)% vs. (62.00±3.25)%, FVC (L): 1.72±0.21 vs. 1.66±0.21, RV/TLC: (42.13±1.67)% vs. (43.12±0.95)%, dyspnea scores: 1.71±0.54 vs. 2.32±0.65, allP < 0.05].Conclusion Shenqi Fuzheng injection possesses certain clinical value in treatment of patients with AECOPD, as it can obviously improve the pulmonary function and the data of arterial blood gas analyses, and effectively relieve the clinical symptoms.
10.Role of the autophagy in the treatment of neuropathic pain with pulsed radiofrequency

Objective:To detect the effect of pulse radiofrequency (PRF) treatment on the neuropathic pain established by L5-spinal nerve ligation (SNL) on rats,and to investigate if PRF treatment would affect the expression of autophagy related protein LC3 and autophagy related receptor P62 at the dorsal horn.Methods:A total of 36 male Sprague-Dawley rats were randomly divided into 3 groups:a Sham group,a SNL group,and a SNL+PRF group.The 50% paw withdrawal mechanical threshold (PWMT) was detected at 1 day before and 1,3,7,14 and 28 days post-operation by using Von-Frey filaments.The autophagy related protein LC3 and autophagy related receptor P62 were investigated by Western blot.Results:Compared with the Sham group,the PWMT significantly decreased in the SNL group at each time points (P<0.05);in SNL+PRF group,PRF treatment could elevate the PWMT at the 1st day post-operation and lasted for 28 days (P<0.05).What's more,SNL could elevate the LC3-Ⅱ and P62 levels at the 7th day post-operation (P<0.05),which were decreased by the PRF treatment (P<0.05).Conclusion:PRF treatment could improve SNL-induced the neuropathic pain,which might be partly due to the regulatory effects on the autophagy levels at the spinal dorsal horn.
