Objective To investigate the expression of tissue transglutaminase(tTG) and vascular endothelial growth factor(VEGF) in epithelial ovarian carcinoma and the relationship of invasion and metastance in ovarian carcinoma.Methods The expression of tTG and VEGF was detected in 55 malignant and 21 benign ovarian neoplasms by immunohistochemical SP method.The clinical significance of tTG and VEGF and their relationship were analyzed.Results The expression rates of tTG and VEGF in malignant epithelial ovarian tumors were significantly higher than in benign epithelial ovarian tumors(tTG:92.7％ vs.38.1％,x2 =32.620,P ＜0.01 ;VEGF:90.9％ vs.47.6％,x2 =19.570,P ＜0.01).In ovarian cancer,the expression of tTG in tumor cells and stromal cells was significantly related to clinical phases and the presence of ascites.The expression of VEGF in tumor cells had a significant revelance to clinical phases,pathological grades and ascites.The expression of tTG in ovarian cancer cells was positively correlated with VEGF(r =0.8856,x2 =43.14,P ＜0.01).Conclusion Tissue transglutaminase and VEGF were detected not only in tumor cells,but also instromal cells.High expression of tTG and VEGF may play an importaant role in the pathogenesis of ovarian cancer and their synergistic effect may act as an important factor in promoting the invasion and metastasis of this cancer.

ObjectiveTo observe the effects of free fatty aids (FFAs) on the apoptosis and migration of human breast cancer cells ( MDA-MB-231 ).MethodsBreast cancer cells were exposed to oleic acid (OA) and palmitic acid (PA) with different concentrations (50,200,and 500 umol/L).Quantity RT-PCR was conducted to detect mRNA expression of PTEN,and Western blot to detect protein expression of PTEN and Bcl-2.Cell apoptosis was measured with TUNEL method,and migration measured with transwell method.ResultsThe mRNA and protein expressions of PTEN in MDA-MB-231 cells were down-regulated after OA stimulation,but up-regulated after PA stimulation ( all P ＜ 0.05 ).The protein expression of Bcl-2 increased after OA stimulation and decreased after PA stimulation ( both P ＜ 0.05 ).PA increased the apoptosis,and both OA and PA enhanced the invasion of MDA-MB-231 cells.ConclusionsPA may promote apoptosis of MDA-MB-231 cells through changing the expression of the tumor suppressor PTEN,but also increases the invasion of the cancer cells.The invasion of cancer cells could be enhanced by OA stimulation as well.

ObjectiveTo investigate the expression of EMMPRIN and MMP-9 in epithelial ovarian carcinoma and the relationship of invasion and metastasis in ovarian carcinoma.MethodsThe expression of EMMPRIN and MMP-9 was detected in 55 malignant and 21 benign ovarian neoplasms by immunohistochemical Sp method.ResultsThe expression rate of EMMPRIN and MMP-9 in malignant, benign epithelial ovarian tumors was 89.1％, 14.3％ and 96.4％, 23.8％, respectively.There were significance different between malignant tumors and benign tumors.In ovarian cancer, the expression of EMMPRIN in tumor cells had a significant relevance to clinical phases and pathology types, while the expression of EMMPRIN in stromal ceils was associated with clinical phases, pathology types and pathological grades.The expression of MMP-9 in tumor cells had a significant relevance to clinical phases and pathology types while the expression of MMP-9 in stromal cells was associated with clinical phases, pathology types and ascites.ConclusionsEMMPRIN and MMP-9 were detected not only in tumor cells, but also in stoma cells.High expression of EMMPRIN and MMP-9 may play an important role in the pathogenesis of ovarian cancer, and their synergistic effect may act as an important factor in promoting the invasion and metastasis of ovarian cancer.

In the opinion of dialectical materialism, concept is developing with the deepening and broadening of human being's mind. The developing process of syndrome concept in traditional Chinese medicine also takes the same way. In the Sui and Tang Dynasties, the connotation of syndrome concept was defined and the embryonic form of syndrome differentiation of zang-fu viscera was formed. In the Song, Jin and Yuan Dynasties, the development of syndrome concept embodied in optimizing the theoretical research based on clinical practice and experiences according to many medical masters in different denominations. The syndrome differentiation of eight principles was established to standardize and perfect the principles of syndrome differentiation in the Qing Dynasty. Modern research of syndrome concept is manifested in objectifying the process of research of syndrome and diseases in different levels and aspects by use of advanced scientific technology.

A low calcium medium developed for epidermal keratinocytes were prepared according to the MCDB 153 modified formula and used in human epidermal keratinocyte culture compared with DMEM culture system. The observation by contrast microscopy and electron microscopy showed that in the low calcium medium keratinocytes grew as a monolayer of high proliferation and had many characteristics of basal cells, with a more rounded shape and large intercellular spaces. Increasing the calcium ion concentration in the medium or changing the other culture conditions the cells in these cultures could be induced stratification and terminal differentiation. The results suggest that the growth, proliferation and differentiation of cultured human epidermal keratinocytes can be controlled and regulated someway.

Objective To observe the clinical effectiveness and adverse side effects of percutaneous embedding of ~(125)I particles in combination with chemotherapy for advanced non-small cell lung cancer(NSCLC).Methods Twenty-one patients with advanced NSCLC(IIIa-IIIb phase) received percutaneous embedding of ~(125)I particles in combination with concurrent chemotherapy.The chemotherapy was conducted in accordance with TP regime(paclitaxel at 135 mg/m~2 and cisplatin at 30 mg/m~2 were administered on day 1 and on day 2 to 4,respectively with 28 days as a cycle).12～75 ~(125)I particles were embedded with CT-guided percutaneous puncture one week after chemotherapy.The radioactivity quantum was 22～33 MBq per particle.The overall radioactivity quantum of embedded particles was 264-1650 MBq with one or more puncture points.Chemotherapy was then continued for two cycles.Results All the 21 patients completed the therapy,among whom,23.80% got complete response(CR),66.67% got partial response(PR) and 9.53% had stable or progressive disease(NC+PD) with an overall response(CR+PR) of 90.48%.The complications included hemopneumothorax(33.34%),hemorrhage(4.77%),particle migration(4.77%).The incidence rates of radiation esophagitis and radiation pneumonia with grade 1～2 were 4.77%(1/21) and 4.77%(1/21) respectively.No radiation esophagitis or radiation pneumonia with grade 3～4 was found.The incidence rate of grade 3～4 acute myelosuppression comprises 52.38% leucocytopenia,9.53% thrombocytopenia,28.58% decrease of hemoglobin,grade III-IV nausea and 42.86%vomiting,38.10% grade III-IV alopecia.The 1-year survival rate was 80.96%.The 12-month local control rate was 85.72%. Conclusion The results have shown that CT-guided percutaneous embedding of ~(125)I particles in combination with chemotherapy was effective and well-tolerable with few complications and good compliance.

Objective:To explore the influence of different blood glucose level at hospitalization on no-reflow and prognosis in patients with acute myocardial infarction (AMI),but without diabetes after emergency percutaneous coronary intervention (PCI).Methods:A total of 678 non-diabetic AMI patients undergoing PCI were selected.Ac-cording to blood glucose level at hospitalization,patients were divided into normal group (n=312),blood glucose level higher group (higher group ,n=201)and blood glucose level extremely high group (extremely high group,n=165).Clinical characteristics,no-reflow after PCI and incidence of cardiovascular events within 30d after PCI were compared among three groups.Results:Compared with normal group and higher group,there were significant rise in age,percentages of >1 location of myocardial infarction and Killip's ≥ class II,white blood cell count instant af-ter hospitalization,peak value of creatine kinase (CK)and CK-isoenzyme (CK-MB)in extremely high group,P <0.01 all;Compared with normal group,the age,peak value of CK and CK-MB significantly rose in higher group (P<0.01 all).Along with blood glucose rose,incidence rate of no-reflow significantly increased (17.0% vs.38.8%vs.49.7%),left ventricular ejection fraction (LVEF)on 30d after PCI gradually reduced [(68.6± 5.7)% vs. (62.2±6.4)% vs.(55.6±6.9)%]in above-mentioned 3 groups in turn,there was significant difference between any two groups,P <0.05 or <0.01.Conclusion:Stress hyperglycemia possesses great influence on no-reflow and prognosis in non-diabetic AMI patients after PCI.

OBJECTIVE: To investigate the in vivo possibility of osteogenesis and angiogenesis of tissue-engineered periosteum in rabbits.METHODS: The marrow mesenchymal stem cells (MSCs) derived from New Zealand rabbits were adhered to small intestinal submucosa (SIS) to fabricate the tissue-engineered periosteum. Totally 12 New Zealand rabbits were received critical bone defect in bilateral radii to prepare models. The tissue-engineered periosteum was randomly implanted in one side of bone defect,and the other side was treated by SIS. At 4 weeks after operation, the angiogenesis of tissue engineered bone was detected by Tetracycline fluorescence microscopy and formaldehyde-ink perfusion method; simultaneously, the new bone formation was firmed by haematoxylin-eosin staining.RESULTS: Animals showed normal daily behaviors and non-infection wounds healing. The gross observation showed that bone defects in the experimental side were bridged with newly formed bone; while the defects of the control side were remained empty.Tetracycline fluorescence microscopy and hisotological examination could confirm the new bone tissue formation in the experimental side. The ink staining in new bone specimens suggested that there were abundant of neovasculization in tissue-engineered bone.CONCLUSION: Tissue-engineered periosteum can form new bone in allogenic rabbits and can be vascularized by some inherent mechanism for new bone tissue survivor.
