Objective To investigate the protective effect of penehyclidine hydrochloride pretreatment on focal cerebral ischemia/reperfusion injury and expression of caspase-3 in rats.Methods Sixty male Sprague-Dawley rats weighing 250-300 g were randomly divided into 3 groups(n=20): control (C) group in which sham operation was performed; ischemia-reperfusion (I-R)group, in which saline (2.0 mg?kg-1)was administered for intraperitoneal injection 30 min before anesthesia,then the ischemia model was made by occlusion of the left middle cerebral artery for 120 min; penehyclidine hydrochloride pretreatment (P) group, in which penehyclidine hydrochloride(2.0 mg?kg-1)was administered for intraperitoneal injection 30 min before anesthesia,then the same 120 min focal cerebral ischemia was performed as described in I-R group.At the end of 24 h reperfusion the animals were decapitated and the brains were removed.The volume of cerebral infarction was detected with TTC staining;Coronal sections including hippocampal tissue were obtained for HE staining.The levels of caspase-3 mRNA expression in hippocampus were detected by using semi-quantitative RT-PCR technique.Results Compared with group I-R,the volume of cerebral infarction in group P were reduced significantly(P
2.Analysis of preoperative risk factors of postoperative complications in patients undergoing endovascular repair surgery

Objective To summarize the perioperative outcomes of patients undergoing endovascular repair,and to screen out related preoperative risk factors in order to provide reference for the promotion of postoperative rehabilitation of patients.Methods The clinical data of 2 896 patients undergoing elective endovascular repair in the Department of Vascular Surgery,Zhongshan Hospital,Fudan University from Jan.2013 to Dec.2014 were analyzed to screen out related preoperative risk factors of postoperative complications retrospectively.Results A total of 148 patients had postoperative complications,including 2 cases of death during hospitalization.The incidence of complications in the patients older than 75 years old,with hypertension,diabetic mellitus,renal dysfunction,chronic obstructive pulmonary disease (COPD) or ASA physical status more than grade Ⅱ is 9.7%,8.1 %,14.3 %,8.0 %,29.7 %,6.3 %,respectively.The multivariate Logistic regression analysis identified that patients older than 75 years old (P =0.000,OR =43.29,95%CI:22.51-83.28),with hypertension (P=0.000,OR=3.822,95 %CI:2.37-6.16),diabetic mellitus (P=0.025,OR=1.714,95%CI:1.07 -2.75),renal dysfunction (P =0.017,OR =2.692,95 % CI:1.19-6.07) or COPD (P =0.000,OR =7.158,95%CI:3.83-13.37) and ASA physical status more than grade Ⅱ (P =0.000,OR =27.77,95%CI:13.79-55.93) were the independent risk factors with postoperative complications of endovascular repair surgery.Conclusions The patients older than 75 years old,with hypertension,diabetic mellitus,renal dysfunction or COPD and ASA physical status more than grade Ⅱ were the independent risk factors for endovascular repair surgery.

Objective To investigate the effects of obesity on functional residual capacity ( FRC ) , ventilatory function and gas exchange during and after cholecystectomy. Methods Twenty ASA Ⅰ or Ⅱ patients of both sex aged 26-58yr undergoing elective cholecystectomy were divided into 2 groups according to the body mass index (BMI): control group BMI 30 (n = 10) . The patients were premedicated with intramuscular luminal 1.0 g and atropine 0.5 mg. Anesthesia was induced with midazolam 2-3 mg, fentanyl 5?g?*kg-1, propofol 1.5-2.0 mg?kg and vecuronium 0.1 mg?kg-1 and maintained with infusion of propofol (at 8-10 mg?kg-1?h-1) and fentanyl (at 1-2 ?g?kg-1?h-1) and intermittent i.v. boluses of vecuronium after tracheal intubation. The patients were mechanically ventilated (VT 10ml?kg-1,I:E=1:2, FiO2 100%) and PETCO2 was maintained at 35-40 mm Hg by adjusting the respiratory rate. Pethidine 1 mg?kg-1 was given i.m. every 8 h after operation for analgesia. BP, HR, ECG, SpO2, PET CO2 , FiO2, VT , RR were monitored during anesthesia. Arterial blood samples were taken before anesthesia (T0, baseline), 15 min after induction of anesthesia (T1), 30 min after skin incision (T2) and 6, 24 h after operation (T3,4) for blood gas analysis. FRC (closed-circuit helium dilution method), forced vital capacity (FVC), forced expiratory volume at 1 second (FEV10) and maximal mid-expiratory flow volume (MMF) were measured before and 6 h and 24 h after operation.Results FRC was significantly decreased after induction of anesthesia, during and after operation compared with the baseline value (T0) in both groups and the decrease in FRC was significantly larger in obesity group than in control group (P

Aim To investigate the role of peripheral benzodiazepine receptor in rat cardiac mitochondrial permeability transition.Methods The isolated rat cardiac mitochondria were incubated with different doses(50,100,200 ?mol?L-1) of PBR antagonist 1-(2-chlorophenyl-N-methyl-1-methylpropyl)-3-isoquinolinecarboxamide (PK 11195). In additional group(CsA group), 5 ?mol?L-1 cyclosporine A (CsA), an inhibitor of MPT was added 5 minutes before the addition of 100 ?mol?L-1 PK 11195. Negative control group(Con group) was given none treatment. Positive control group(Ca2+ group) was given 150 ?mol?L-1 CaCl2. 6.Comparisons of the life quality and its influential factors among the senior citizens in communities, nursing homes and elderly wards in Shanghai Minhang district

Objective To understand the life quality of elderly person in communities, nursing homes and elderly wards, and to find out the related influencing factors for providing references for policy-makers to improve the life quality of the elderly. Methods The senior citizens in 9 communities, 3 villages, 12 nursing homes and 5 elderly wards in Shanghai Minhang District were enrolled by stratified cluster sampling. And the life quality and its influential factors among the inhabitants in communities, nursing homes and elderly wards were compared by adopting Short-Form 36 (SF-36) questionnaire. The major influential factors on the life quality were gender, education, marriage, profession, age, disease, residence, entertainment, physical exercise for community; Age, disease, residence, entertainment, physical exercise for nursing home; Age, etc for elderly ward. Conclusions The influences of demographic factors on life quality of the senior citizens in communities are more significant than in nursing home and elderly wards. Variations on the degree of satisfaction (in residence, entertainment, health care and transportation) and the situation of physical exercise have made vast difference in the life quality of the senior citizens both in communities and nursing homes.
7.Research in simulation training model for cooperation between doctors and nurses

Objective To discuss the construction of training model for cooperation between doctors and nurses and evaluate its teaching effect.Methods 101 nursing students and 80 clinical medicine students were involved in an overall training for the purpose of cooperation between doctors and nurses by using of Sim-man and then test and questionnaire were conducted to evaluate the effect of the training.Results Statistics showed that training for cooperation between doctors and nurses had improved in such factors as attitude to the cooperation,communication skills,team work,and adaptability (operant skill was note included).In addition,88% of the subjects supported the views that this kind of training should be offered before their internship.Conclusions Training for cooperation between doctors and nurses by using Sim-man can cultivate awareness of cooperation between doctors and nurses and improve team work,and set a better foundation for good collaboration between doctors and nurses.
8.Determination of osthole and imperatorin in Compound Shajiziyou Suppository by RP-HPLC

AIM:To develop a HPLC method of determining osthole and imperatorin in Compound Shajiziyou Suppository(oil of Semen Hippophae,Fructus Cnidii,Radix Sophorae flavescentis,etc.). METHODS: Phenomnex(luna) C_(18) column was used at(25 ?C).The mobile phase consisted of methanol-water(65(∶)35).The flow rate was 1.0 mL/min.The detection wavelength was set at 310 nm. RESULTS: The linear ranges of osthole and imperatorin were 0.051-0.816 ?g(r=0.999 6) and 0.026-0.416 ?g(r=0.999 8),respectively,The average recoveries were 98,4% with RSD of 1.4% and 97.6% with RSD of 2.0% respectively. CONCLUSION: This method is simple,accurate,reproducible and can be used for the determination of osthole and imperatorin in Compound Shajiziyou Suppository.
9.Identification of serum biomarkers for diagnosing stage I lung adenocarcinoma by MALDI-TOF mass spectrometry

Objectire To identify specific biomarkers that could improve early diagnsis of lung adenuearcinoma using matrix-assisted laser desorptian/ionization (MALDI) technology. Methods Serum samples were isolated from 17 patients with stage I lung adenuearcinoma and 17 age- and sex-matched healthy controls, and the serum proteomic profiles were obtained by matrix-assistcd laser desorption ionization time of flight (MALDI-TOF) mass spectrometry. Results Compared with healthy control group, two highly expressed potential biomarkers were identified with the relative molecular weights of 6 631.64 Da and 4 964. 21 Da. The two best novel protein peaks were automatically chosen for the system training and the development of the constructed model. The constructed model was then used to test an independent set of masked serum samples from 15 lung adenocarcinoma patients and 22 healthy individuals. The analysis yielded a sensitivity of 93.3 %, and a specificity of 95.5 %. Conclusion These results suggest that MALDI-TOF-MS ProteinChip technology is a quick, convenient, and high-output analyzing method that is capable of selecting several relatively potential biomarkers from the serum of lung adenocarcinoma patients and may have a clinical value in the future, and will provide clues to identifying new serologic btomarkers of lung adenocarcinoma.
10.Mechanism of Inhibition of propofol on intracellular calcium increase in aortic smooth muscle cells

Objective:To test the effects of propofol on intracellular calcium free concentration ([Ca~(2+)]i) and inositol 1,4,5-triphosphate (IP_3) biological synthesis induced by norepinephrine (NE) and 5-hydroxytryptamine (5-HT) in aortic smooth muscle cells (ASMC)of rats for the mechanism of relaxtion of propofol on vascular smooth muscle.Method: Using the flurospectrophotometry and Fura-2/AM loading method,the changes of [Ca~(2+)]i levels in primary culture ASMC were measured,and using the specific, IP_3 assay system and isotope radioactive protein binding experiment IP_3 production levels in aortic smooth muscle were measured. Result:The baselines of [Ca~(2+)]i was decreased when primary culture ASMC was pretreated with propofol in 72 hours. Propofol inhibited [Ca~(2+)]i increase induced by NE and 5-HT in dose-dependent way. With extracellular calcium free or calcium channel blocker(Verapamil),inhibition of propofol on NE and 5-HT increasing [Ca~(2+)]i levels were decreased,but could not be cancelled. Propofol depressed IP_3 biological synthesis induced by NE and 5-HT in dose-dependent way. Conclusion:Relaxation of propofol on aortic smooth muscle is closely related to inhibiting IP_3- induced calcium release to decrease intracellular calcium concentration.
