1.Serum paraoxonase and its relationship to atherosclerosis

Human serum paraoxonase (PON1) is a high density lipoprotein (HDL)associated enzyme capable of hydrolyzing lipid peroxides in vitro .PON1 has recently attracted attetion as a key factor against oxidative modification of LDL by involving in degradation of oxidized phospholipid and may therefore play an important role on the prevention of the atherosclerotic process of HDL.The concentration and activity of PON1 have been shown to be associated with the risk of atherosclerosis.In addition,the genetic polymorphisms of PON1 have been proposed as a genetic marker of risk for coronary artery disease.The measurement of the genetic polymorphisms of PON1 may be helpful in the prevention of coronary artery disease.
2.Observation on serumγ-glutamyl transpeptidase levels of patients with sub-clinical schistosomiasis before and after pathogen treatment

Objective To observe the changes of serumγ?glutamyl transpeptidase(GGT)levels before and after the patho?gen treatment in patients with subclinical schistosomiasis,and explore its clinical value in the diagnosis and treatment of sub?clinical schistosomiasis. Methods Totally 109 patients with subclinical schistosomiasis,who were found in the endemic inves?tigation of schistosomiasis in Ezhou City,were selected as the investigation subjects,and then they were treated with praziquan?tel. The serum GGT levels of the subjects before and after the treatment were detected and compared. Results Before the treat?ment,the average value of the GGT levels of the 109 patients was(48.1 ± 45.9)IU/L,among which,the GGT levels of 69 cases (63.3%)were normal,and the levels of 40 cases(36.7%)were increased. After the treatment,the average GGT level of the pa?tients was(32.1 ± 23.4)IU/L,which decreased by 33.3%comparing with that before the treatment,and the difference had a statistical significance(U=2.17,P=0.01). The GGT levels of 65 patients decreased in different degrees. Among the 40 pa?tients whose GGT levels had increased before the treatment,the GGT levels of 31 ones returned to the normal. Conclusion The GGT level detection can accurately reflect the liver function in the patients with subclinical schistosomiasis ,and also it has certain clinical application value to judge the liver function damage and recovery of the patients before and after the pathogen treatment.
3.AEG-1 promotes metastasis of breast cancer MCF-7 cells

Objective The objective of this study was to observe the effect of AEG-1 gene on the metastasis in breast cancer MCF-7 cells.Methods AEG-1 siRNAs were transfected into MCF-7 cells to silence AEG-1 expression,and negative siRNA was used as a control.Transwell chamber was used to detect the ability of cell migration and invasion of MCF-7 cells.CCK8 assay was used to detect the cell proliferation of MCF-7 cells.At the same time,the effect of VEGF on the angiogenesis was investigated by detecting the changes of lumen formation in HUVEC cells.Results The migration,invasive and proliferative abilities were significantly inhibited in MCF-7 cells transfected with AEG-1 siRNA.Knockdown AEG-1 was significantly decreased the level of VEGF in the supernatant of MCF-7 cells.Knockdown AEG-1 was also significantly inhibited the angiogenesis activity in HUVEC cells.Conclusion Knockdown AEG-1 can significantly inhibit the migration of MCF-7 cells,including cell migration,invasion,proliferation and angiogenesis.These results suggest that AEG-1 plays an important role in the metastasis process of breast cancer and opens up new ideas for future treatment breast cancer.
4.The expression of Caspase-8 in gastric carcinoma tissues and its clinical significance

ObjectiveTo investigate the expression characteristics of Caspase-8 in gastric carcinoma tissues and its correlation with clinical pathological features.Methods The mRNA and protein of Caspase-8 in gastric carcinoma and its surrounding tissues were detected by reverse transcription polymerase chain reaction (RT-PCR) and immunohistochemistry,respectively.ResultsThe Caspase-8 mRNA in gastric carcinoma tissues was obviously lower than that in its surrounding ones (0.154±0.065 vs 0.394±0.107,t =14.04,P <0.01),which was correlated with lymph node metastasis,tumor stage,differentiation grade (P<0.01).The Caspase-8 protein in gastric carcinoma tissues was also significantly lower than that in its surrounding ones (31.48% vs 85.19%,x2 =32.04,P < 0.01),which was correlated with tumor stage,differentiation grade (P<0.05).ConclusionsCaspase-8 is low-expressed in gastric carcinoma,which may play an important role in the occurrence and progression of gastric carcinoma.
5.Percutaneous coronary intervention results in increases of lipoprotein(a) and oxidized lipoprotein(a) in patients with acute coronary syndrome

Objective To investigate possible changes of lipoprotein(a) [Lp(a)] and oxidized Lp (a) [ox-Lp(a) ] levels after PCI and it mechanisms. Methods Bloods were selected from 75 patients with ACS undergoing PCI, and at 24 hours, 2 and 3 days, and 6 months pre-and post-PCI treatment, and from 29 control patients pre-and post-coronary angiography without undergoing PCI. The levels of Lp(a) , ox-Lp(a) , Lp(a) immune complexes (IC) and its autoantibody were determined by ELISA. The extents of CAD were determined by coronary angiography. The differences of variants pre-and post-operations were analyzed by paired samples t test. The differences of levels of Lp(a) and ox-Lp(a) among time points after PCI were analyzed by ANOVA. Correlations between Lp(a) and ox-Lp(a) , and between angiographic variables and Lp(a), ox-Lp(a) levels were calculated. Results Compared to pre-PCI, Lp(a) [233.10 (152.86-328.79) mg/L vs 202.05 (106.15-271.42) mg/L, t=6. 81, P<0.01], ox-Lp(a) [19.05 (10.98-31.80) mg/L vs 10. 51 (4.98-17.97) μg/ml, t = 13. 22,P <0. 01] and Lp(a)-IC [2.72 (1.604.91) AU vs 2. 11 (1.04-3. 97) AU, t = 3. 34, P < 0. 01 ] levels significantly increased immediately in post-PCI, while its antoantibody levels significantly decreased (A = 0. 81 ± 0. 33 vs A = 0. 72 ± 0. 28, t = 5.58, P < 0. 01). Strong correlations were noted between levels of ox-Lp( a) and Lp( a) both in pre-PCI (r =0. 66, P <0.01) and post-PCI (r = 0. 62, P <0. 01). PCI resulted in rapidrise of Lp(a) and ox-Lp(a) levels and then decreased quickly in 24 hours, returned to baseline in 2-3 days. The changes of Lp(a) and ox-Lp(a) levels in pre-and post-PCI were positively related with severity of ACS. In contrast, in the angiography-only control group, no significant changes were noted in Lp(a) , ox-Lp(a) , Lp(a)-IC and Lp(a) autoantibodies levels between the pre-and post-angiography samples. Conclusion PCI results in acute plasma acute increases of levels of Lp(a) and ox-Lp(a) ,and the changes are related with lesion severity of the coronary artery.
6.The effect of scan duration on ALFF of resting-state fMRI

Objective To examine the reliability and similarity of amplitude of low-frequency fluctuation (ALFF)for different scan duration.Methods 10 min 10 s resting state fMRI data acquired from 13 healthy volunteers separately were divided into 10 pieces of the same length.The reliability and similarity of ALFF were assessed for each scan length (1-10 min).Results Spatial maps of ALFF steadily appeared in each time period (1-10 min);but during the time evolution,both spatial distribution and extent of ALFF increased gradually.At only one minute,there were detectable statistical differences within the group.When the time ran-ging from 5 to 10 min,relatively stable ALFF results in the group achieved.Conclusion ALFF is a relatively stable index,it can be easily affected by scan duration.In the case of 1-5 min,it should be carefully analyzed and interpretated particularly.
7.Determination of Ultratrace Copper in Barium Phosphate Laser Glass by Inductively Coupled Plasma Mass Spectrometry

With the use of high pure HF and HNO3 reagents, and autoclaves made of high purity Rerfluoroalkoxy ( PFA ) material, a solution sample digestion technique effective for phosphate samples, subjected to high temperature fusion, was established. The whole procedure was concise, fast and of low blank value. Key factors such as the amount and ratios of the reagents, the digestion temperature and time, were systematically optimized, it was found that within 0. 5 h at 150 ℃, only 1. 7 mL of total reagent consumption could lead to a complete sample decomposition. Most importantly, the samples were not required to be ground to fine powder, which greatly reduced the risk of contamination. In addition, an effective liquid_liquid extraction procedure based on 5_nonylsalicylaldehyde oxime as the extractant was established for matrix separation and analyte preconcentration. Under the optimal extraction conditions of 5 mL of 15% extractant, 0. 5% HNO3 of extraction acidity and 20% HNO3 of back_extraction acidity, a matrix separation efficiency of over 99. 999% could be realized and a preconctration factor of 10 could be obtained, which resulted in complete elimination of the matrix_induced interference and great enhancement of the analytical sensitivity. After optimization of the operation parameters of ICP_MS, high signal to background detection of Cu in 20%HNO3 at 840 W of plasma power and low sample uptake rate were realized. The detection limits of 2. 5 ng/g, RSD of 3. 3% for six detections of parallel samples, and the recovery of 94. 3% for spike test were obtained, respectively. The method was finally applied to three real samples analysis, and the results agreed well with the data from laser adsorption loss experiment.

9.Determination of Active Ingredient Oleanolic Acid in She Medicine Clematis Florida var. Plena by HPLC

Objective:To establish a method for the determination of oleanolic acid in She medicine Clematis florida var. plena. Methods:The HPLC method was carried on a Lanbo-Kromasil C18 chromatography column using acetonitrile-water ( 86∶ 14 ) as the mobile phase. The detection wavelength was 205 nm, the flow rate was 1. 0 ml·min-1 , and the column temperature was 25 ℃. Re-sults:The calibration curve of oleanolic acid was linear within the range of 0. 52-15. 60μg (r=0. 999 9). The average recovery of oleanolic acid was 92. 13%(RSD=3. 0%, n=9). Conclusion:The method is simple, rapid, accurate and reproducible, which can be used as an ideal method for the quality control of She medicine Clematis florida var. plena. .
10.Dynamic observation and assessment of iodine nutritional status of population in Meizhou City before and after salt iodine concentration reduction

Objective To master the status of iodine content in drinking water and iodine nutritional status of population before and after the salt iodine concentration reduction in 2012 in Meizhou City, and to provide a scientific basis for safety assessment of salt iodine content after adjustment of the policy.Methods Using stratified random sampling method, drinking water samples were collected, and iodide content was measured by cerous sulfate catalytic spectrophotometry.Urine samples of children aged 8-10 were collected, in which the iodide content was quantitatively tested by arsenic cerium catalytic spectrophotometry.Samples of household edible salt were collected to determine iodine content by direct titration in 2012 before (September 2011), and after the salt iodine concentration reduction (September 2012 and September 2013).Results A total of 422 water samples were measured, the median iodine content of water was 2.4 μg/L.The water iodine median was 2.1 μg/L in centralized water supply (n =163),and 2.9 μg/L in decentralized water supply (n =259), the difference was statistically significant (U =-2.526, P < 0.05).Totally 800 and 803 urine samples of children aged 8-10 were collected in 2011 and 2012, median urinary iodine was 216.5 and 207.5 μg/L, respectively, which were higher than that in 2013 (n =807, 190.0 μg/L, x2 =17.040,24.868, all P < 0.05).Urinary iodine ≥300 μg/L ratio was significantly decreased (2013 than 2011 and 2012,19.3% (156/807) vs.26.5% (212/800) and 24.5% (197/803), x2 =6.363, 11.695, all P < 0.05), and urinary iodine < 100 μg/L rose in the proportion (2013 than 2011, 18.0% vs.13.5%, x2 =6.045, P < 0.05).A total of 2 410 household salt samples were tested, and the coverage rate of iodized salt, iodized salt qualified rate, the consumption rate of qualified iodized salt were all higher than 98.8%.Totally 800, 803 and 807 salt samples were collected in 2011, 2012 and 2013, the salt iodine medians were 31.0, 27.7 and 25.4 mg/kg, respectively, the difference between the salt iodine medians was statistically significant (H =91.422, P < 0.05).Conclusions Iodine excess risk is significantly decreased, and the level of iodine nutritional status of the city's population is appropriate after the salt iodine concentration reduction in 2012.The salt iodine concentration adjusted is suitable and safe.
