1.Research on the effects of piperine on proliferation and invasion of human bladder cancer T24 ;Cells

Objective To investigate the effects of differentpiperine concentrationson the prolif-eration and invasion of bladder cancer T24 cells.Methods Bladder cancer T24 cells were treated with different concentrations of piperine(5,1 0,20,40,80,and 1 60μmol/L).Then MTT assay,Transwell exper-iments,and western blot were used to detect the expression of bax and bcl-2,and the proliferation and ap-optosis of T24 cells.Results After a 24-hour treatment of piperine,IC50 value was 38.73μmol/L.With the increaseof piperine concentrations,the activity of T24 cell was significantly inhibited in a dose-depend-ent manner.Westernblot showed that the expression of anti-apoptotic protein bcl-2 reduced,and the pro-apoptotic protein bax increased.Conclusion Piperine could inhibit the proliferation of bladder cancer T24 cells and promote the apoptosis of T24 cells.
2.Immune Regulation and Therapeutic Observation of Anti-Cough in Children Treated with Pediatric Long-term Cough Decoction

This study was aimed to observe the effect of Pediatric Long-term Cough Decoction in the treatment of long-term cough among children. A total of 96 cases were diagnosed in traditional Chinese medicine (TCM) as qi-yin deficiency, and lung-heat accumulation with constipation. The western medicine diagnosis was mycoplasma pneumoniae pneumonia (MPP) infection in the recovery period after chronic cough. All cases were randomly divided into the TCM group and control group, with 48 cases in each group. In TCM group, the Pediatric Long-term Cough Decoction was used to supplement qi and nourish yin, clear away the lung-heat and excrete feces. In the control group, montelukast was used. For both groups, clinical effect of score and CD4+ T lymphocytes, CD8+ T lymphocytes and CD4+/CD8+ ratio were observed one day before treatment and thirty days after treatment. The results showed that after thirty-day treatment, the clinical effect of score in both groups was obviously improved (P < 0.01). The TCM group was better than the control group (P< 0.01). Compared with pretreatment, the posttreatment CD4+ T lympho-cytes, CD8+ T lymphocytes and CD4+/CD8+ ratio in TCM group were obviously improved (P < 0.05). The CD4+ T lymphocytes and CD4+/CD8+ ratio in the control group were obviously improved (P < 0.05). However, there was no change on the CD8+ T lymphocytes. The regulation effect on CD4+ T lymphocytes and CD8+ T lymphocytes of TCM group was better than the control group (P< 0.01). It was concluded that the Pediatric Long-term Cough Decoction can effectively improve clinical symptoms of chronic cough in children and regulate immune function.

RNA dot blot hybridization was employed to investigate the alterations in C-myc, C-H-ras transcript levels in bone marrow and peripheral blood cells from 24 patients with acute nonlymphocytic leukemia. 10 cases of normal bone marrow cells were used as control. We found that the positive rates and intensities of expression of C-myc and C-H-ras in acute nonlymphocytic leukemia were hipher than those of normal control. The C-myc and C-H-ras expressed simultaneously in 10 patients. After having been cultured 5 days with RA and low dose Ara-C combined with low dose Harr, the C-myc transcipt level of C-myc in acute nonlymphocytic leukemia was decreased obviously.
6.Molecular Cloning and Biochemical Characterization of Protocatechuate 3, 4-dioxygenase in Burkholderia sp.NCIMB 10467

Strain NCIMB 10467, a lignin degrader, was reclassified as genus Burkholderia according to its 16S rDNA sequence. It seems that the metabolism of protocatechuate by this strain is diverse under the induction of various substrates. A 9505-bp DNA fragment extending from α conserved region of the gene, which encodes β subunit of orthocleavage protocatechuate 3, 4-dioxygenase(P34D; EC 1.13.11.3),was obtained by genome walking. Sequence analysis revealed two deduced open reading frames, pcaG and pcaH, encoding the α and β subunits of P34D respectively in this fragment. The P34D activity could be detected when pcaGH were expressed in E. Coli and the disruption of pcaH in strain NCIMB 10467 has lead to loss of its ability to catabolize protocatechuate. It was proved that the cloned pcaGH were encoding a functional protocatechuate 3, 4-dioxygenase which was necessary for the protocatechuate metabolism in this strain.
7.Sclerotherapic effects of different concentrations of lauromacrogol on endometrial cyst models in rats in two retention ways

Objective To compare the sclerotherapic effects of different concentrations of lauromacrogol on endometrial cyst models in rats in two ways :10 minutes retention with extraction and retention without extraction .Methods Ninety SD rats after modeling were randomly divided into three groups :group A ,10 minutes retention with extraction;group B ,retention without extraction;group C , normal saline .While both A and B group were randomly divided into four subgroups with different concentrations :1% ,0 5.% ,0 2.5% ,and 0 1.25% . Seven days after treatment the cure rates and histopathological changes were observed .Results Cure rates in group A were 86 7.% ,71 4.% ,50% and 20% ;in group B were 100% ,100% ,87 5.% and 37 5.% .In both A and B groups ,the cure rates were not significantly different between 1% ,0 5.% and 0 2.5% concentration subgroups ( P >0 0.5) ,the cure rates of these subgroups in group B had statistical significance with 0 1.25% concentration subgroups and the saline control group( P <0 0.5) ,but there were no significant difference between 0 2.5% and 0 1.25% concentration subgroups in group A .Compared the differences of the cure rates among the same concentration subgroups , it had no statistical significance in groups A and B of 1% and 0 1.25% concentration subgroups( P >0 0.5) , but between 0 5.% and 0 2.5% concentration subgroups ,it had statistical significance( P <0 0.5) .Conclusions It will be conductive to enhancing the sclerotherapic effect if the lauromacrogol concentration remains above 0 2.5% and does not be extracted from the cyst models .
8.Effects of Jiaweitaohechengqitang on expression of?-SMA protein in hepatic fibrosis rats induced by CCl_4

Objective:To study the effects of Jiaweitaohechengtang on expression of?-SMA protein in hepatic fibrosis rats induced by CCl4.Methods:Hepatic fibrosis models were induced by CCl 4 .All the experimental rats were divided into six groups:normal control group,model control group,colchicine positive treatment group,jiaweitaohechenqitang groups with high- dosage,medium-dosage and low-dosage.Drugs were given to rats for 8 weeks by intragastric administration.Scores of hepatic fibrosis were assessed with semiquantitive scoring system(SSS)through tianlanghong staining.Protein expression levels of ?-smooth muscle actin(?-SMA)were determined with immunohistochemical steining method.Results:SSS in model control group were higher than those in normal control group,but lower than those in three treatment groups with jiaweitaohechengqitang. Compared with nornal control group,the protein expression levels of?-SMA in liver tissues in model control group increased markedly,but decreased when it was compared to jiaweitaohechengqitang groups.There was a significant difference between model control group and three treatment groups.Conclusion:Jiaweitaohechengqitang has therapeutic effects on hepatic fibrosis.The mechanism may be ralated to the inhabitition of the activation of HSC.
9.Evaluation of clinical effects of AO clavicula hook plate in treatment of complete acromioclavicular dislocation

Objective To evaluate the clinical effects of AO clavicula hook plate in treatment of acromioclavicular dislocation. Methods According to the specific anatomic, physiological and biomechanical conditions of the patients, 43 cases of complete acromioclavicular dislocation were treated either by AO clavicula hook plate (23 cases),or by Kirschner pin and steer wire tension band (20 cases). Results The results were evaluated by radiographic representations and joint function recovery according to Karlsson's criteria. The excellent results of the 2 methods were 95.6% and 65% respectively. There was significant difference. Conclusions The AO clavicula hook plate can meet the clinic requirements and be used with satisfactory results in the treatment of acromioclavicular dislocation.
10.Effects of Acupuncture from Tiaokou to Chenshan and Exercise on Shoulder-hand Syndrome at Stage I after Stroke

Objective To observe the effect of acupuncture penetrated from Tiaokou (ST38) to Chenshan (BL57) combined with exer-cise on affected limbs of shoulder-hand syndrome (SHS) at stage I after stroke. Methods 40 inpatients with SHS at stage I after stroke from January, 2013 to December, 2014 were randomly divided into treatment group (n=20) and control group (n=20). The treatment group accept-ed acupuncture from Tiaokou to Chenshan on unaffected sides and exercise of affected upper limbs, and the control group accetpted routine acupuncture on Triple Shoulder Acupoints on affected sides. They were assessed with Pain Rating Index (PRI), Fugl-Meyer Assessment (FMA) and modified Barthel Index (MBI) before and 4 weeks after intervention. Results The incidence of improvement was 90% in the treatment group, and 80%in the control group (χ2=0.784, P=0.376). The scores of PRI decreased (t>5.37, P<0.001), and the score of FMA and MBI increased (t>4.66, P<0.001) significantly after intervention. The score of FMA and MBI improved more in the treatment group than in the control group (t>2.25, P<0.05). Conclusion Acupuncture from Tiaokou to Chenshan combined with exercise is more effective on SHS at stage I after stroke than conventional Triple Shoulder Acupoints.
