1.Effects of postconditioning with ischema and propofol on retinal injury induced by limb ischemia-reperfusion in rats

Objective To evaluate the effects of postconditioning with ischema and propofol on retinal injury induced by limb ischemia-reperfusion (LIR) in rats.Methods Twenty-four male Wistar rats were randomly divided into 3 groups (n =8 each) using a random number table:sham operation group (group S),group LIR,and propofol postconditioning + postconditioning + ischemic LIR group (group P + I + LIR).The model of LIR was established by separating and ligating the femoral artery and vein of bilateral hind limbs for 60 min followed by reperfusion.In group P + I + LIR,the animals were subjected to 4 episodes of 60 min ischemia-5 min reperfusion-5 min ischemia-5 min reperfusion and to 4 cycles of propofol 500 μg· kg-1 · min-1 infused intraperitoneally at the same time (during reperfusion)-no propofol infusion during second ischemia.At 120 min of reperfusion,blood samples were collected from the abdominal aorta for determination of serum malondialdehyde (MDA) concentrations.The eyeball was removed and the retina was cut into sections which were stained with H.E.and examined by microscopy.Results Compared with group S,the serum MDA concentration was significantly increased in group LIR,and no significant change was found in the serum MDA concentration in group P + I + LIR.The serum MDA concentration was significantly lower in group P + I + LIR than in group LIR.The structure of retina was normal in group S,oedema was found in all the layers of retina,vacuolar degeneration was observed in the most of ganglionic layers,and the structure disturbance was found in group LIR.The pathological changes of retina were significantly attenuated in group P + Ⅰ + LIR.Conclusion Postconditioning with ischema and propofol can attenuate retinal injury induced by LIR,and the mechanism is related to inhibition of lipid peroxidation in rats.
2.Efficacy of thrombolytic therapy combined with psychological intervention in treatment of patients with acute myocardial infarction

Objective To observe the clinical efficacy of thrombolytic therapy combined with psychological intervention in treatment of patients with acute myocardial infarction.Methods 80 cases were treated in department of emergency, Hangzhou First People’s hospital were randomly divided into observation group and control group from February 2014 to February 2015.40 cases in each group.Observation group was given psychological intervention conventional therapy plus thrombolytic therapy, control group were treated with thrombolytic therapy alone, recorded and analysed of therapeutic effect of two groups of patients.Results NOS (nitric oxide synthase), ET (endothelin) values of observation group post-treatment were (45.9 ±5.1)U/mL,(87.3 ±7.1) pg/mL,which were better than control group (39.1 ±3.6)U/mL,(96.5 ±9.3)pg/mL (P<0.05); observation group total effective rate was 75.0% better than control group ( 70.0%) , but the two groups had no significant difference in efficiency.Conclusion Thrombolytic therapy combined with psychological intervention in the treatment of acute myocardial infarction is effective , precise, and can be effectively improve the symptoms of acute myocardial infarction, it is worthy of further research and application.
3.Clinical internship and innovation of psychiatry from the perspective of medical undergraduates

Objective To evaluate the current methods for psychiatry clinical internship and needs for innovation from the perspectives of medical undergradutes.Methods Independent interview on patients and their relatives as well as social psychological interview were conducted for five-year psychiatry majors in clinical internship (n =72).Self-made questionnaire was used to investigate the teaching feedback concerning the practice effect and teaching method tendency.Data were analyzed by t test and P < 0.05 signifies statistically significant differences.Results Medical students considered the clinical internship as the main source of psychiatry knowledge (rising from 29.4 % (20/69) to 36.2% (25/69),t =4.67,P <0.01).Respective interviewing with patients and their families and then doing focus-group discussion was the most acceptable method for medical undergraduates:82.6%(57/69) of medical undergraduates thought this method helped expose defects of clinical skills and prejudice against psychiatry; 54.3% (37/69) of medical undergraduates thought this method was more beneficial to acquire psychiatry knowledge.The second acceptable method was the demonstration of a typical case by teacher:27.3% (19/69) thought this method helped expose defects of clinical skills and prejudice against psychiatry; 27.2% (19/69) thought this method was more beneficial to acquire psychiatry knowledge.62.0% (43/69) of medical undergraduates ranked the characteristics of the symptomatology and core symptoms differentiation as the first place.49.0% (34/69) considered the training of interviewing skills with patients as the second important in the clinical training.91.3% (63/69) expected to see more kinds of mental disorders.65.2 % (45/69) expected to learn more about the interviewing skills.Conclusions It is necessary to pay more attention to the method of doing respective interviewing with patients and their families and then making focus-group discussion.Meanwhile training of interviewing skills in the clinical internship of psychiatry should be enhanced.
4.INVESTIGATION OF MELATONIN ON SEX HORMONES AND OVULATION

To investigate the effect of melatonin(Mel) on follicle-stimulating hormone(FSH), luteinizing hormone(LH), estradiol(E 2) and ovulation, 100 healthy women were divided into 5 groups to take 0mg, 12mg, 30mg, 75mg, and 300mg Mel per day, respectively, for 3 months. The levels of FSH, LH, E 2 were determined by enzyme-linked immunosorbent assay(ELISA) before and after medication, the development of dominant follicle was detected by ultrasound. We have found that the levels of FSH, LH and E 2 were all lower and the dominant follicle was smaller than those of the control (0mg) group at 3 months. In this study we demonstrated the direct involvement of melatonin in modulation of the female reproductive system in a dose-dependent manner, and it may be put into effect through melatonin receptors in human hypothalamus-pitutary-ovary cells.

Physical training induces beneficial adaptations,but long excessive exercise may lead to severe damage to the skeletal muscles,liver,heart,kidneys and immune functions. Over the past few decades,health scholars have been searching for natural components that can prevent or improve the damage induced by hyperkinesis. The mechanisms of organ damage induced by long overtraining include immunosuppression,metabolism disorder,hormone disturbance,oxidative damage, etc. Natural poly-saccharides have interventional effects on these injuries,possibly by improving immunity,regulating metabolism and ameliorating free radical damage.
6.Physico-chemical properties and antimicrobial activity of silver ion dressings

Objective To detect the physico-chemical properties , cytotoxicity and antimicrobial activity of dressings containing Ag +.Methods The morphology of this dressing was shown by the scanning electron microscope ( SEM ) .The cytotoxicity was detected by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT) method.Using the shake flask test method , the antibacterial effect of dressings was studied .Result There was no significant difference in the water vapor transmission rate between the dressings (P>0.05).Compared with the alginate calcium dressing ,silver ion dressings had a stronger swelling rate .A higher ion concentration would lead to a much larger swelling rate and slower degradation . A lower cytotoxicity was exhibited among the dressings .The silver ion dressing had stronger bacteriostasis to Gram-positive ( G+) and Gram-negative ( G-) bacteria than the alginate calcium dressing .Conclusion The experiment has proved that the silver ion dressing has stronger antibacterial activity and lower cytotoxicity , and it is more effective for wound surface healing, with a shortened treatment course .
7.Fluorescence Assay for Phospholipase C Activity Using Liposome Probes

A simple assay for detection of phospholipase C (PLC) activity was developed based on a fluorescence liposome probe using the Liss Rhod PE-loaded phospholipid liposomes.The liposome probe was prepared by the coassembly of 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC) and fluorescent lipid (Liss Rhod PE).The probe showed very low background fluorescence due to fluorescence self-quenching effect of Liss Rhod PE.As the PLC enzyme selectively digested lipid, the Rhod fluorescence was recovered from its quenched state, leading to the sensitive detection of PLC.This assay provided a limit of detection (at a signal-to-noise ratio of 3) of 2 U/L for PLC.In the presence of PLC inhibitor, the fluorescent response of the sensor for PLC decreased, indicating that the assay could also be used for screening PLC inhibitors.
8.Effects of extractive pericarpium trichosanthes on apoptosis of HUVECs induced by high glucose

Aim To study the inhibitory effects of ex-tractive pericarpium trichosanthes ( EPT) on high glu-cose-induced apoptosis in human umbilical vein endo-thelial cells ( HUVECs ) and its underlining mecha-nisms. Methods HUVECs were cultured. Effects of EPT at different concentrations on the high-glucose-in-duced apoptosis in HUVECs were observed. The cell viability of HUVECs was determined by MTT colori-metric method. Cell apoptosis was identified by Ho-echst staining. The intracellular activity of Caspase-3 was detected with colorimetry. Protein expression and p65 nuclear translocation of NF-κB were detected by Western blot and immunofluorescence staining. Re-sults Treated with 30 mmol · L-1 glucose for 48 hours had a significantly decrease on cell viability com-pared with control, and the apoptotic rate and Caspase-3 activity were increased markedly, the protein expres-sion of NF-κB was upregulated and p65 nuclear trans-location in HUVECs increased; Pre-incubation with EPT(12. 5,25,50 mg·L-1 ) for 1 hour enhanced the cell viability, and decreased the apoptotic rate and Caspase-3 activity and downregulated the expression of NF-κB in a dose-dependent manner. Moreover, EPT could inhibit p65 translocation. Conclusion EPT can protect HUVECs against the apoptosis induced by high glucose in vitro,and its mechanism may be related with downregulation of NF-κB expression and inhibition of the intracellular activity of Caspase-3 .
9.Analysis on published scientific papers from 2001 to 2010 by professionals of provincial Center for Disease Control and Prevention in China

Objectives Through analyzing the published scientific papers from 2001 to 2010 by the professionals of the 31 provincial Center for Disease Control and Prevention in China,offer reference for making plan about scientific research,disciplinary areas,personnel training.Methods Literature quantitative analysis and health statistics methods were used to analyze these papers.Results The professionals in 31 provincial CDC published a total of 22079 papers,Zhejiang 1669(7.56%),Guangxi 1579 (7.15 %),Jiangsu 1410 (6.39 %) are the top 3 provinces.The ratio of published papers in Zhonghua medical journal among all the papers are 1366(6.19%),the first three provinces Tianjinlll (13.67%).Beijing160 (13.57%).Shaanxi34 (10.59%).Average papers published by the eastern,central and western regions are 1131,452,444,eastern above western regions (P =0.0065.P =0.0028).Conclusion In recent ten years,the quantity and quality of papers published by the professionals of provincial CDC in China were improved.The unbalanced development exist among eastern,central and western regions,But the majority CDC's papers should be strengthened further.
10.Human endostatin antiangiogenic gene therapy mediated by recombinant adeno-associated virus vector in nude mouse with endometriosis

Objective To study the therapeutic effect of recombinant adeno-associated virus carrying human endostatin gene therapy on endometriosis in mice model. Methods Recombinant adeno-associated virus vector carrying human endostatin gene and enhanced green fluorescent proteins gene (rAAV2-endostatin-EGFP) was constructed. Endometrium was from 12 patients with leiomyoma undergoing hysterectomy in Second Hospital, Tianjin Medical University between November and December 2008. Endometriosis models of nude mice were established by transplanting human endometrial fragments intooperitoneal surface. After 1 week, those 60 mice were divided into 3 groups: treatment group including 20 mice injected with rAAV2-endostatin-EGFP to ectopic lesion, control group including 20 mice injected with rAAV2-EGFP to ectopic lesion and blank control group including 20 mice injected with phosphate buffered saline (PBS) to the ectopic lesion. At 1, 2 and 3 weeks after treatment, those mice underwent laparotemy to observe the location and size of ectopic lesion in abdominal cavity. The expression of endostain protein, number of gland, microvessel density (MVD) and vascular endothelial growth factor (VEGF) were measured in ectopic lesions. The serum level of estradiol and progesterone were detected in nude mice among every groups. Results (1) All endometriosis of nude mice models were established successfully through peritoneum transplanting. After 1 week's treatment, flat lesion nodes, decreased gland number and narrow and atrophy glandular cavity were observed by light microscope. (2) The endostatin gene was transferred into nude mice successfully and expressed effectively. It was observed that endostatin protein expression was shown with enhanced green fluorescent proteins in ectopic lesion. (3) Glands number of ectopic lesion in rAAV2-endostatin-EGFP group(7.8±1.9,7.0±1.5 and 5.5±1.7) were significantly less than 10.1± 1.7, 10.2±2.0 and 9.8±2.4 in rAAV2-EGFP control group and 10.2±2.2,10.0±2.0 and 9.7±2.2 in PBS control group at 1,2 and 3 weeks after treatment(all P<0.05). Glands number of ectopic lesion in rAAV2-endostatin-EGFP group at 3 weeks was significantly less than those at 1 and 2 weeks after treatment (P<0.05). (4) MVD of ectopic lesion in rAAV2-endostatin-EGFP group (12.2±1.5,11.4±2.1 and 9.0±1.4) was significantly less than those at rAAV2-EGFP control group (16.5±1.7,16.5±1.9 and 16.9±1.9) and PBS control group (16.2±1.6,16.0±1.6 and 16.3±1.7) at 1,2 and 3 weeks after treatment (all P<0.05) . MVD of ectopic lesion in rAAV2-endostatin-EGFP group at 3 weeks was significantly less than those at 1 and 2 weeks after treatment(P<0.05). (5) The rate and density of VEGF expression at ectopic lesion in rAAV2-endostatin-EGFP group (35%, 30%, 25% and 1.60±0. 43,1.33± 0. 30,1.03±0.36) were significantly less than those at rAAV2-EGFP control group (80% ,75% ,85% and 2.43±0.53,2.43±0.29,2.66±0.45) and PBS control group (85% ,90% ,90% and 2.36±0.53,2.64± 0.57,2.53±0.52) at one 1, 2 and 3 ,weeks after treatment (all P<0.05). The expression of VEGF at ectopic lesion in rAAV2-endostatin-EGFP group at 3 weeks was significantly less than those at 1 and 2 weeks after treatment (P<0.05). (6) The level of estradial and progesterone in serum of nude mice of rAAV2-endostatin-EGFP group [ E_(2)> : (48±7 ) pmol/L, P: (61±8 ) nmol/L ] did not reach statistical difference when compared with those at rAAV2-EGFP control group [ E_(2): (50±9) pmol/L, P: (60±10) nmol/L] and PBS control group [E_(2):(48±7)pmol/L,P: (58±10)nmol/L,P>0.05]. Conclusions The recombinant adeno-asseciated virus carrying human endostatin gene therapy could inhibit angiogenesis at endometriotic lesions and not influence steroid level. The antiangiogenic gene therapy might become a novel option for endometriosis.
