Objective To investigate the level and clinical significance of nerve growth factor ( NGF) , transforming growth factor ( TGF )-β1 , estradiol ( E2 ) and testosterone ( T ) in serum and ex-pressed prostatic secretion (EPS) of patients with category Ⅲprostatitis. Methods From August 2011 to January 2012, 64 patients with (chronic prostatitis/chronic pelvic pain syndrome , CP/CPPS) and 20 health people were enrolled in this study.In CP/CPPS group, the age of patients ranged from 18 to 56 years, mean (36.6±9.3) years.The history of CP/CPPS ranged from 3 months to 6 years, mean 2 years.All patients were asked to complete NIH-CPSI questionnaires with CP/CPPS, including group ⅢA 35 cases and groupⅢB 29 cases.The age of healthy controls ranged from 25 to 41 years.The average healthy control age was (33.1±3.9) years.EPS and serum samples from CP/CPPS and control group were collected and frozen . NGF, TGF-β1 , E2 and T level in EPS and serum were measured by ELISA and radioimmunoassay and com -pared in each group. Results The mean E2, E2/T, TGF-β1 level in serum of patients with CP/CPPS were (175.7±82.4) pmol/L, (7.9±6.7), (2 216.2±581.6) ng/L, which were higher than that in healthy controls, (131.7±49.4) pmol/L, (4.6±2.4), (1 599.8±469.5) ng/L.The mean T level in CP/CPPS pa-tients′serum was (24.7±8.9) nmol/L, which was lower than that in controls (29.2±7.0) nmol/L.The E2/T (34.5±29.8), TGF-β1(6 859.3±5 229.4 ng/L), NGF (467.0±164.3 ng/L) levels in EPS of CP/CPPS patients were higher than that in controls (16.5±13.8), (1 774.1±1 304.3) ng/L, (310.8±106.6) ng/L. The TGF-β1 level in EPS of CP/CPPS patients showed the positive correlation ship with urination symptom score (6.1±2.4) (r=0.641, P<0.05).The NGF level in EPS of CP/CPPS patients also showed the positive correlation ship with pain score (7.6±2.6) (r=0.497, P<0.05).E2/T,TGF-β1 levels in serum and E2/T, TGF-β1,NGF levels in EPS of group ⅢA were (7.1±4.6), (2131.5±412.0)ng/L and (31.5±22.3), (7 667.1±5 652.4)ng/L, (440.6±134.3)ng/L, which were significantly higher than those in healthy con-trol (P<0.05).E2/T, TGF-β1 levels in serum and E2/T, TGF-β1, NGF levels in EPS of group ⅢB were (8.9±8.5), (2 340.5±728.2) ng/L and (38.2±37.1), (5 884.4±4 574.3) ng/L, (498.9±192.1) ng/L, which were also higher than those in healthy control ( P<0.05) . Conclusions Hormonal imbalance in es-tradiol and testosterone with TGF-β1 , NGF higher levels in EPS is closely related with pathogenesis and clin-ical symptom of category III chronic nonbacterial prostatitis .
4.Prevention of thoracolumbar osteoporotic compression fracture with transpedicular intracorporeal bone graft with allogenic bone

Objective To evaluate the clinical efficacy of transpedicular intracorporeal bone graft with allogenic bone in treatment of thoracolumbar osteoporotic compression fracture. Methods A total of 45 patients with thoracolumbar osteoporotic compression fractures were treated with posterior short segment pedicle screw fixation and transpedicular intracorporeal bone graft with allogenic bone. Anteroposte-rior and lateral X-ray photographs were taken before and after operation and at follow up period to determine the ratio of anterior and middle compressed body height to the normal height, the vertebral angle and the superior-inferior endplate angle. The extent of local pain was measured by VAS score. The implant failure was also recorded during follow-up. Results The operative reduction and interbody bone grafting exerted a satisfactory effect on the ratio of anterior and middle body height to the normal height, the vertebral angle and the superior-inferior endplate angle. Local back pain disappeared immediately after surgery in 34 patients out of 38 patients followed up for more than two years. No implant failure was found during follow-up. Conclusions Early treatment with posterior short segment pedicle screw fixation and transpedicular intracorporeal bone graft with allogenic bone can effectively correct local deformity, prevent late vertebral collapse and implant failure and is an ideal treatment method for thoracolumbar osteoporotic compression fracture.
5.Applied Value of MR Dynamic Contrast-Enhancement at 3.0T MR Magneton in Diagnosing Infiltrating Ductal Carcinoma of Breast

Objective To explore the value of MR dynamic contrast-enhancement in diagnosis and differential diagnosis of infiltrating ductal carcinoma of breast at 3.0T MR magneton.Methods 17 cases of breast infiltrating ductal carcinoma underwent plain MRI and MR dynamic contrast-enhanced scan using 3.0T MR scanners with dedicated breast coil.All cases were confirmed by surgery and pathology.MRI signal intensity,morphology and hemodynamic characteristics of lesions were analysed.Results 17 breast infiltrating ductal carcinomas in 17 cases all appeared as masses,low(8/17)or equal(9/17)signal intensity on T_1WI,high(14/17)or equal(3/17)signal intensity on T_2WI.On morphology,the lesions were irregular and lobulated in shape(82.4%,14/17);undefinite margins(12/17)or non-smooth margins(15/17),glitch sign or astral sign(7/17);the lesions showed non-uniform marked enhancement(10/17)or ring enhancement(5/17)after intravenous administration of contrast agent.82.4%(14/17)of the lesions,the blood vessels could be seen gathering around the lesions on the maximum intensity projection(MIP)image.On hemodynamics,the early enhanced ratio for all cases was over 90%;88.2%(15/17)of the lesions,peak enhancement was less than three minutes;the time-signal intensity curve of the lesions appeared as type Ⅱ was 35.3%(6/17)and type Ⅲ was 58.8%(10/17)mostly.Conclusion Dynamic contrast-enhanced MRI manifestations of breast infiltrating ductal carcinoma are of certain characteristics,which may contribute to the diagnosis and differential diagnosis.
6.Effect of apolipoprotein E deficiency on astrocyte destruction

Objective To explore the effect of apolipoprotein E deficiency on astrocyte destruction in the ex vivo spinal cord slice model. Methods Vibratome-cut transverse spinal cord slices from C57BL/6 postnatal day 7 ApoE -/-mice and wild-type mice were cultured on transwell porous supports. After 7 days in the culture , spinal cord slices were exposed to LPS for 5 days. The expression of AQP4, GFAP and Iba1 was detected by immunofluorescence. The concentration of TNF-α and NO were detected by ELISA and method of nitrate reductase, respectively. Results Marked loss of AQP4 and GFAP staining were shown in LPS-affected groups, not in the control group, and the lesion score was higher in ApoE-/-group than WT-group (P<0.05). In addition, the expression of Iba1 and concentration of TNF-α, NO in the LPS-affected groups were more than that of the control group (P < 0.05), and they were higher in the ApoE-/-LPS group than the WT-LPS group (P < 0.05). Conclusions ApoE deficiency exacerbates astrocyte injury, likely through promoting the release of pro-inflammatory mediators from microglia.
7.The cloning and sequencing of H-2Kk gene cDNA of 615 mice.

OBJECTIVEThe purposes of this study were to clone and sequence the major histocompatibility complex type I (MHC I) molecular antigen recognizing gene (H-2Kk) of 615 mice, and to provide the functional gene for transgenic therapy.
METHODSThe 1.4 kb full-length fragment of H-2Kk gene complementary DNA (cDNA) was amplified from the total RNA of 615 mouse liver by using reverse transcription polymerase chain reaction (RT-PCR). The cDNA was inserted into PGEM3Zf(+) vector directionally, and the competent E. coli JM109 was transformed with the ligated product. The recombinant PGEM3Zf(+)-H-2Kk cDNA plasmid was obtained using restricted enzyme analysis of the transfectants. The complete sequence of 615 mouse H-2Kk cDNA was determined by using Sanger's method.
RESULTSThe sequences of 615 mouse H-2Kk cDNA were 99% similar with those of H-2Kk cDNA which were reported by other researchers, and the sequences encoding antigen recognizing regions (ARS) were identical with each other.
CONCLUSIONThe authors cloned the MHC I molecular antigen recognizing gene (H-2Kk) of 615 mice successfully and got the functional gene of MHC I.

8.Correlation between sex hormone level in serum and expressed prostatic secretion with erectile function in patients with type Ⅲ prostatitis

Objective To explore the correlation between the levels of estradiol E2 and testosterone T in serum and expressed prostatic secretion(EPS) with the erectile function in the patients with type Ⅲ prostatitis(CP/CPPS) .Methods The E2 and T lev‐els in serum and EPS from 64 cases of CP/CPPS ,including 35 cases of type Ⅲ A and 29 cases of Ⅲ B ,and 20 individuals of physical examination were detected by using the radioimmunoassay .All cases were evaluated by the scores of NIH‐CPSI and the Internation‐al Index of Erectile Function 5(IIEF‐5) .64 patients were grouped according to the IIEF‐5 scores ,the erectile dysfunction(ED) group(32 cases) and the non‐ED group(32 cases) .Results The mean E2/T levels in serum and EPS of the Ⅲ A group and the Ⅲ B group were higher than those in the control group ,the difference had statistical significance(P<0 .05) .20 cases(57 .14% ) of ED were found in the Ⅲ A group ,which were more than 12 cases(41 .38% ) of ED in the Ⅲ B group ,but there was no statistically signifi‐cant difference (>0 .05 .There was a positive correlation between the IIEF‐5 score and the T level in serum and EPS in the CP/CPPS group(r=0 .218 ,r=0 .231 ,P<0 .05) .There was a negative correlation between the IIEF‐5 score and the E2/T level in ser‐um and EPS(r= -0 .189 ,r= -0 .652 ,P<0 .05) ,which had no correlation with the NIH‐CPSI score(P>0 .05) .The serum T level in the ED group was (6 .32 ± 1 .86)ng/mL ,which was lower than(7 .89 ± 2 .92)ng/mL in the non‐ED group and (8 .41 ± 2 .02)ng/mL in the control group ;the .E2/T level in EPS in the ED group was (55 .02 ± 29 .26) ,which was higher than (14 .06 ± 9 .36) in the non‐ED group and (16 .45 ± 13 .76) in the control group ,the differences among them were statistically significant (P<0 .05) .Con‐clusion The imbalance degree of hormone estradiol and testosterone in serum and EPS is related with erectile function in the pa‐tients with CP/CPPS .
9.Mechanism of Saffron in Treatment of Parkinson's Disease Based on Network Pharmacology and Molecular Docking

[Objective]To investigate the therapeutic effect and molecular mechanism of Chinese medicine Saffron on Parkinson's disease(PD)based on network pharmacology and molecular docking.[Methods]The pharmacodynamic components of Saffron included in the analysis were determined through the pharmacological database and literature research of the Chinese medicine system.With the help of compound target database and disease target database,the corresponding targets of saffron components and PD targets were predicted,respectively.Enrichment analysis of compound-disease intersection targets was made by using R procedures and referencing correlated R-packages.Construction of protein-protein interaction(PPI)networks and analysis of node-related topological parameters.Simulate molecular docking to verify binding properties of representative compounds and key targets.The pharmacodynamic properties of Saffron acid on key targets were preliminarily evaluated by experiments.[Results]The 8 components of saffron with druggable potential corresponded to 279 targets,PD had 2 183 disease targets,and a total of 98 intersection targets of compounds and diseases were obtained.Enrichment analysis showed that the compound-disease intersection target was involved in biological processes such as reactive oxygen radical synthesis and metabolism,neuroinflammation and mitogen-activated protein kinase(MAPK)signaling pathway.The key targets identified by PPI network analysis were tumor necrosis factor(TNF)and signal transducer and activator of transcription 3(STAT3).Molecular docking showed that saffron glycosides and saffron acids had stronger binding activity with TNF.Experiments had shown that saffron acid can reduce the transcription levels of lipopolysaccharide-induced cytoinflammatory factor TNF,interleukin-1β(IL-1β)and inducible nitric oxide oxide synthase(iNOS).[Conclusion]Saffron can be used as a candidate for PD treatment,and its neuroprotective effect may involve the interaction and synergy of many targets and signaling pathways,which deserves special attention to the activity of saffron acid against central nervous system inflammation.
10.Hemodynamics of deep vein in lower limb during perioperative period of total hip arthroplasty

Objective To investigate hemodynamic changes of deep vein in lower limb during the perioperative period of total hip arthroplasty (THA) and early diagnosis of deep vein thrombosis (DVT). Methods Doppler ultrasound, hemorheology detection and plasma D-dimer testing were done on 62 patients treated with THA. Statistical analysis was carried out on the data of patients with or without DVT to study the early diagnosis of DVT. Results The results of Doppler ultrasound showed DVT in 8 patients. Compared with postoperative concentration of plasma D-dimer, the preoperative con-centration of plasma D-dimer was significandy higher in patients with or without DVT (P < 0.05). The levels of hemorheological indices were significantly increased at postoperative day 7 (P < 0.05). Con-clusions Doppler ultrasound combined with plasma D-dimer testing and hemorheology detection are helpful in early diagnosis of DVT.
